| 5 | Effects of Rhodiola rosea and its major compounds on insulin resistance in Caenorhabditis elegans显示文摘By establishing insulin resistance model of Caenorhabditis elegans,the effects of Rhodiola rosea extract and its compounds on glucose level,insulin signal intensity,antioxidant enzymes and cell apoptosis of C.elegans were investigated in present study.The mRNA expression of DAF-2,DAF-16,AkT-1,SKN-1,AAK-2 and SOD-3 were detected by qRT-PCR.The results revealed that R.rosea extract contain salidroside,kaempferol,chlorogenic acid,kaemphenol-7-O-glucoside,caffeic acid,and quercetin,etc.At the concentration of 200μg/mL,R.rosea extract treated C.elegans showed increased insulin signal intensity by 52.46%compared with the normal group,while,glucose content decreased by 72.13%and reactive oxygen species(ROS)level decreased by 36.84%.Compared with high glucose model group,the activities of superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px)increased by 10.9,14.2 and 27.9,respectively.After R.rosea extract intervention,the expression levels of DAF-2,AkT-1,AAK-2,SKN-1 and other genes significantly increased.These results also indicated that R.rosea extract could activate insulin receptor substrate(IRS)and adenosine 5′-monophosphate(AMP)-activated protein kinase(AMPK)pathways to enhance insulin signaling,promote nutrient metabolism,and achieve the effect for improving insulin resistance of C.elegans. | Hui Teng Hongting Deng Yanzi Wu Chang Zhang Chao Ai Hui Cao Jianbo Xiao Lei Chen | 2022 | Journal of Future Foods2022,2,4: | 0 |
| 6 | The protein corona modulates the inflammation inhibition by cationic nanoparticles via cell-free DNA scavenging显示文摘A central paradigm in nanomedicine is that when synthetic nanoparticles(NPs)enter the body,they are immediately cloaked by a corona of macromolecules(mostly proteins)that mediates the role of the physico-chemical properties in the NP biological functions(the“coronation paradigm”).In this work,we focused on the assessment of the“coronation paradigm”for cationic NPs(cNPs)used as rheumatoid arthritis(RA)drugs due to their ability to scavenge cell-free DNA(cfDNA).We fabricated series of cNPs uniformly coated with single or di-hydroxyl groups and different types of amino groups and showed that hydroxylated nanoparticles displayed a prolonged retention in inflamed joints and greater anti-inflammatory effect in collagen-induced arthritis(CIA)rats than the non-hydroxylated analogues.Especially,the cNPs with secondary amines and a di-hydroxyl shell showed the best performance among the tested cNPs.Proteomic analysis showed that the cNPs with a di-hydroxyl shell adsorbed less opsonin proteins than the cNPs carrying mono hydroxyl groups and non-hydroxylated ones,which may provide a mechanistic explanation for the different biodistribution profiles of cNPs.Thus,this study suggests that the protein corona mediates the effects of the surface chemistry on the fate and functions of cNPs as anti-RA drugs. | Xingliang Liu Huiyi Liang Yanzi Yan Jingjiao Wu Massimo Bottini Lixin Liu Yongming Chen | 2022 | Bioactive Materials2022,7,7: | 0 |
