1Transferring deep neural networks for the differentiation of mammographic breast lesions显示文摘Machine learning can help differentiating benign and malignant lesions seen on mammographic images. Conventional models require handcrafting features for lesion representation. Due to insufficient medical instances, the performance of convolutional neural networks(CNNs) can be further increased. This study makes use of transfer learning for mammographic breast lesion diagnosis and deep neural network(DNN) models pre-trained with large-scale natural images are employed. The diagnosis performance is evaluated with the prediction accuracy(ACC) and the area under the curve(AUC) on average. A histologically verified database is analyzed which contains 406 lesions(230 benign and 176 malignant). Involved models include transferred DNNs(GoogLeNet and AlexNet), shallow CNNs(CNN2 and CNN3) that are fully trained with medical instances and boosted by support vector machine(SVM), and two conventional methods which combine handcrafted features and SVM for lesion diagnosis. Experimental results indicate that GoogLeNet achieves the best performance(ACC=0.81, AUC=0.88), followed by AlexNet(ACC=0.79, AUC=0.83) and CNN3(ACC=0.73, AUC=0.82). 2Mendelian randomization studies on atherosclerotic cardiovascular disease: evidence and limitations显示文摘Epidemiological research has revealed a galaxy of biomarkers, such as genes, molecules or traits, which are associated with increased risk of atherosclerotic cardiovascular diseases(ASCVD). However, the etiological basis remains poorly characterized.Mendelian randomization(MR) involves the use of observational genetic data to ascertain the roles of disease-associated risk factors and, in particular, differentiate those reflecting the presence or severity of a disease from those contributing causally to a disease. Over the past decade, MR has evolved into a fruitful approach to clarifying the causal relation of a biomarker with ASCVD and to verifying potential therapeutic targets for ASCVD. In this review, we selected high-quality MR studies on ASCVD, examined the causal relationship of a series of biomarkers with ASCVD, and elucidated the role of MR in validating biomarkers as a therapeutic target by comparing the results from MR studies and randomized clinical trials(RCTs) for the treatment of ASCVD. The good agreement between the results derived by MR and RCTs suggests that MR could be performed as a screening process before novel drug development. However, when designing and interpreting a MR study, the assumptions and limitations inherent in this approach should be taken into account. 3Female CFOs and loan contracting: Financial conservatism or gender discrimination? – An empirical test based on collateral clauses显示文摘Based on signaling and gender discrimination theory, we examine whether chief financial officer(CFO) gender matters to bank–firm relationships and the designing of collateral clauses in bank loan contracting, and explore the potential path of influence. Data taken from Chinese listed companies between2009 and 2012 indicate that(1) female-CFO-led firms are less likely to obtain credit loans than male-CFO-led firms;(2) female-CFO-led borrowers are more likely to be required to provide collateral for loans than male-CFO-led borrowers; and(3) banks are more inclined to claim mortgaging collateral when lending to female-CFO-led firms and prefer to guarantee collateral when lending to male-CFO-led firms. Female-CFO-led borrowers seem to be granted more unfavorable loan terms than male-CFO-led borrowers, supporting the hypothesis that female CFOs experience credit discrimination. Further analysis reveals that regional financial development helps to alleviate lending discrimination against female CFOs. 4Microstructure,Mechanical,and Thermal Properties of B4C-TiB2-SiC Composites Prepared by Reactive Hot-pressing显示文摘B4C-TiB2-SiC composites with excellent properties were prepared by reactive hot-pressing using B4C,TiC,and Si powders as the raw materials.The phase transition process was investigated by heating the powder mixture to different temperatures and combined with XRD tests.TiB2 and SiC phases were synthesized through an in situ reaction,and the mechanical and thermal properties were improved simultaneously.Microstructure and mechanical properties were also studied,and the 60wt% B4C-21.6wt% TiB2-18.4wt% SiC composite showed a relative density of 99.1%,Vickers hardness of 34.6 GPa,flexural strength of 582 MPa,and fracture toughness of 5.08 MPa·m1/2.In addition,the values of thermal conductivity and thermal expansion coefficient were investigated,respectively.CAO Yaoqin 何强龙 王为民 2020Journal of Wuhan University of Technology(Materials Science)2020,35,6:2
5RECQL4 regulates DNA damage response and redox homeostasis in esophageal cancer显示文摘Objective:RECQL4(a member of the RECQ helicase family)upregulation has been reported to be associated with tumor progression in several malignancies.However,whether RECQL4 sustains esophageal squamous cell carcinoma(ESCC)has not been elucidated.In this study,we determined the functional role for RECQL4 in ESCC progression.Methods:RECQL4 expression in clinical samples of ESCC was examined by immunohistochemistry.Cell proliferation,cellular senescence,the epithelial-mesenchymal transition(EMT),DNA damage,and reactive oxygen species in ESCC cell lines with RECQL4 depletion or overexpression were analyzed.The levels of proteins involved in the DNA damage response(DDR),cell cycle progression,survival,and the EMT were determined by Western blot analyses.Results:RECQL4 was highly expressed in tumor tissues when compared to adjacent non-tumor tissues in ESCC(P<0.001)and positively correlated with poor differentiation(P=0.011),enhanced invasion(P=0.033),and metastasis(P=0.048).RECQL4 was positively associated with proliferation and migration in ESCC cells.Depletion of RECQL4 also inhibited growth of tumor xenografts in vivo.RECQL4 depletion induced G0/G1 phase arrest and cellular senescence.Importantly,the levels of DNA damage and reactive oxygen species were increased when RECQL4 was depleted.DDR,as measured by the activation of ATM,ATR,CHK1,and CHK2,was impaired.RECQL4 was also shown to promote the activation of AKT,ERK,and NF-k B in ESCC cells.Conclusions:The results indicated that RECQL4 was highly expressed in ESCC and played critical roles in the regulation of DDR,redox homeostasis,and cell survival.Guosheng Lyu Peng Su Xiaohe Hao Shiming Chen Shuai Ren Zixiao Zhao Yaoqin Gong Qiao Liu Changshun Shao 2021Cancer Biology & Medicine2021,18,1:2
6CUL4B facilitates HBV replication by promoting HBx stabilization显示文摘Objective:Hepatitis B virus(HBV)infection is a major public health problem worldwide.However,the regulatory mechanisms underlying HBV replication remain unclear.Cullin 4 B-RING ubiquitin E3 ligase(CRL4 B)is involved in regulating diverse physiological and pathophysiological processes.In our study,we aimed to explain the role of CUL4 B in HBV infection.Methods:Cul4 b transgenic mice or conditional knockout mice,as well as liver cell lines with CUL4 B overexpression or knockdown,were used to assess the role of CUL4 B in HBV replication.Immunoprecipitation assays and immunofluorescence staining were performed to study the interaction between CUL4 B and HBx.Cycloheximide chase assays and in vivo ubiquitination assays were performed to evaluate the half-life and the ubiquitination status of HBx.Results:The hydrodynamics-based hepatitis B model in Cul4 b transgenic or conditional knockout mice indicated that CUL4 B promoted HBV replication(P<0.05).Moreover,the overexpression or knockdown system in human liver cell lines validated that CUL4 B increased HBV replication in an HBx-dependent manner.Importantly,immunoprecipitation assays and immunofluorescence staining showed an interaction between CUL4 B and HBx.Furthermore,CUL4 B upregulated HBx protein levels by inhibiting HBx ubiquitination and proteasomal degradation(P<0.05).Finally,a positive correlation between CUL4 B expression and HBV pg RNA level was observed in liver tissues from HBV-positive patients and HBV transgenic mice.Conclusions:CUL4 B enhances HBV replication by interacting with HBx and disrupting its ubiquitin-dependent proteasomal degradation.CUL4 B may therefore be a potential target for anti-HBV therapy.Haixia Shan Bo Wang Xiaodong Zhang Hui Song Xi Li Yongxin Zou Baichun Jiang Huili Hu Hao Dou Changshun Shao Lifen Gao Chunhong Ma Xiaoyun Yang Xiaohong Liang Yaoqin Gong 2022Cancer Biology & Medicine2022,19,1:2
7CUL4B negatively regulates Toll-like receptor-triggered proinflammatory responses by repressing Pten transcription显示文摘Toll-like receptors (TLRs) play critical roles in innate immunity and inflammation. The molecular mechanisms by which TLR signaling is fine-tuned remain to be completely elucidated. Cullin 4B (CUL4B), which assembles the CUL4B-RING E3 ligase complex (CRL4B), has been shown to regulate diverse developmental and physiological processes by catalyzing monoubiquitination for histone modification or polyubiquitination for proteasomal degradation. Here, we identified the role of CUL4B as an intrinsic negative regulator of the TLR-triggered inflammatory response. Deletion of CUL4B in macrophages increased the production of proinflammatory cytokines and decreased anti-inflammatory cytokine IL-10 production in response to pathogens that activate TLR3, TLR4, or TLR2. Myeloid cell-specific Cul4b knockout mice were more susceptible to septic shock when challenged with lipopolysaccharide, polyinosinic-polycytidylic acid or Salmonella typhimurium infection. We further demonstrated that enhanced TLR-induced inflammatory responses in the absence of CUL4B were mediated by increased GSK3β activity. Suppression of GSK3β activity efficiently blocked the TLR-triggered increase in proinflammatory cytokine production and attenuated TLR-triggered death in Cul4b mutant mice. Mechanistically, CUL4B was found to negatively regulate TLR-triggered signaling by epigenetically repressing the transcription of Pten, thus maintaining the anti-inflammatory PI3K-AKT-GSK3β pathway. The upregulation of PTEN caused by CUL4B deletion led to uncontrolled GSK3β activity and excessive inflammatory immune responses. 10Study of risk factors for intraoperative hypothermia during pediatric burn surgery显示文摘Background Intraoperative hypothermia(core temperature<36.0°C)is common during the perioperative period and can result in adverse consequences,especially in children.We aimed to determine the incidence of intraoperative hypothermia and its risk factors in pediatric patients during burn surgery.Methods In the present study we enrolled 197 pediatric patients with burn injury undergoing surgical debridement and skin grafting.Factors,such as total burn surface area(TBSA),were collected and analyzed to identify the potential risk factors for intraoperative hypothermia.Results The incidence of intraoperative hypothermia among all patients was 17.8%.Compared with patients with normothermia,children with hypothermia were associated with larger TBSA(25%vs 15%,p<0.001)and with less intraoperative active warming(34.28%vs 54.93%,p<0.05).In addition,compared with patients with moderate-degree burn,patients with severe and extremely severe burn were associated with much higher risk of intraoperative hypothermia[severe:odds ratio(OR)=3.805,95%confidence interval(CI)=1.396-10.368,p=0.009;extremely severe:OR=6.933,95%CI=2.604-18.462,p<0.001].TBSA was the only independent risk factor that emerged as being strongly associated with intraoperative hypothermia(OR=1.068,p=0.001)and could be used to predict the occurrence of hypothermia when combined with other factors.TBSA for predicting intraoperative hypothermia by receiver operating characteristic(ROC)curve analysis showed a good predictive ability with an area under the ROC curve of 0.758.Conclusion TBSA is an important risk factor for intraoperative hypothermia in pediatric patients with burn.Yaoqin Hu Yangfan Tian Manqing Zhang Jialian Zhao Qiang Shu 2021World Journal of Pediatric Surgery2021,4,1:1

16Local corruption and corporate cash holdings:Sheltering assets or agency conflict?显示文摘This study investigates the impact of corruption on corporate cash holdings in China. The political extraction argument predicts that firms might shelter liquid assets to avoid extraction by corrupt officials. Using data on A-shared listed firms between 2007 and 2012, we find that firms located in more corrupt regions hold less cash, supporting this hypothesis. Political resources help to diminish the risk of exploitation, reducing the extent to which liquid assets are sheltered. We find that the negative association between corruption and cash holding is more significant for non-state-owned enterprises(Non-SOEs)than for state-owned enterprises(SOEs). Moreover, the cash holdings of Non-SOEs without political connections are more sensitive to corruption than those of Non-SOEs with political connections. 18Compact low-birefringence polarization beam splitter using vertical-dual-slot waveguides in silicon carbide integrated platforms显示文摘The polarization beam splitter is a key component for polarization manipulation in photonic integrated circuits,but it is challenging to design for low-refractive-index optical materials,due to the low birefringence of the waveguides.We propose what we believe is a novel compact vertical-dual-slot waveguide-based coupling scheme for silicon carbide,enabling efficient low-birefringence polarization splitting by extensively modulating the transverse-magnetic mode distribution.We numerically and experimentally demonstrate the device in the 4H-siliconcarbide-on-insulator integrated platform,with a small footprint of 2.2 μm × 15 μm.The device,easy to fabricate via a single lithography process as other components on the chip,exhibits low insertion loss of <0.71 dB and <0.51 dB for the transverse-electric and transverse-magnetic polarized light,respectively,and polarization extinction ratio of >13 dB,over 80 nm wavelength range.XIAODONG SHI JINGJING ZHANG WEICHEN FAN YAOQIN LU NIANHUA PENG KARSTEN ROTTWITT AND HAIYAN OU 2022Photonics Research2022,10,1:1
