| 1 | Upregulated expression of Ezrin and invasive phenotype in malignantly transformed esophageal epithelial cells显示文摘AIM: To investigate the correlation between ezrin expression and invasive phenotype formation in malignantly transformed esophageal epithelial cells.
METHODS: The experimental cell line employed in the present study was originated form the progressive induction of a human embryonic esophageal epithelial cell line (SHEE)by the E6E7 genes of human papillomavirus (HPV) type 18.The cells at the 35th passage after induction called SHEEIMM were in a state of immortalized phase and used as the control,while that of the 85th passage denominated as SHEEMT represented the status of cells that were malignantly transformed. The expression changes of ezrin and its mRNA in both cell passages were respectively analyzed by RT-PCR and Western blot. Invasive phenotype was assessed in vivo by inoculating these cells into the severe combined immunodeficient (SCID) mice via subcutaneous and intraperitoneal injection, and in vitro by inoculating them on the surface of the amnion membranes, which then was determined by light microscopy and scanning electron microscopy.
RESULTS: Upregulated expression of ezrin protein and its mRNA was observed in SHEEMT compared with that in SHEEIMM cells. The SHEEMT cells inoculated in SCID mice were observed forming tumor masses in both visceral organs and soft tissues in a period of 40 days with a special propensity to invading mesentery and pancreas, but did not exhibit hepatic metastases. Pathologically, these tumor cells harboring larger nucleus, nucleolus and less cytoplasm could infiltrate and destroy adjacent tissues. In the in vitro study,the inoculated SHEEMT cells could grow in cluster on the amniotic epithelial surface and intrude into the amniotic stroma. In contrast, unrestricted growth and invasiveness were not found in SHEEIMM cells in both in vivo and in vitroexperiment.

| 3 | Application of nonlinear chemical fingerprinting to identification,evaluation and clinical use of Glycyrrhiza显示文摘Nonlinear chemical fingerprinting is a new technique for analyzing traditional Chinese medicines (TCMs). We developed such a method and applied it to Glycyrrhiza, a significant TCM. Nonlinear chemical fingerprints were obtained for Glycyrrhiza from four different production areas, and for 14 other TCMs with similar induction time. The calculation of their similarities was based on similarity system theory. Using intuitive visual comparison of the data and the similarity calculation results, Glycyrrhiza was discriminated from the 14 other TCMs. There was a linear relationship between the content of active TCM components and the induction time. This was used to compare the content of active components in Glycyrrhiza from the four different production areas, and establish an equivalent-dose relationship for these four samples. | 6 | Principle of nonlinear chemical fingerprint by using dissipative components in samples as well as calculation and evaluation of similarity显示文摘Under the conditions of constant temperature and pressure,different influences of samples with different chemical components on the mechanism of nonlinear chemical reaction will cause different changes of the potential-time relationship curve of the nonlinear chemical reaction system.Using it as the character,and using the B-Z nonlinear chemical system to use acetone and substrates in samples as main dissipative substances qua an example,the principle of nonlinear chemical fingerprint has been researched and discussed in detail.At the same time,the general method for calculating the system similarity about nonlinear chemical fingerprint was also put forward,and similarities of nonlinear chemistry fingerprints of different batches of Guhan Yangshengjing and 18 sorts of other samples were calculated by Euclidean distance,correlation coefficient,included angle cosine and system similarity,at the same time,the various similarities were analyzed.The results showed that,both of correlation coefficient and included angle cosine are unable to be used as the criterion for quantitatively evaluating the similarity of nonlinear chemistry fingerprint;as non-parametric similarity,Euclidean distance can accurately reflect the feature differences in the fingerprints,but as parametric similarity,sometimes,Euclidean distance can not accurately reflect the relative extent of characteristic difference in the nonlinear chemical fingerprints;system similarity can most truthfully reflect the characteristic difference in the nonlinear chemical fingerprints,and is the best evaluating method among the four ones.Therefore,system similarity can be used to quantitatively calculate the similar extent between the nonlinear chemical fingerprints.An economical,simple and convenient,easy pushing and effective method for identifying and evaluating complicated samples has successfully been put forward. | ZHANG TaiMing ZHAO Zhe FANG XuanQi QIAO JunXi XIANG FengQin ZHU Rong LIANG YiZeng DING Feng | 2012 | Science China Chemistry2012,55,2: | 13 |
| 7 | Separation and identification of differentially expressed nuclear matrix proteins between human esophageal immortalized and carcinomatous cell lines显示文摘AIM: To separate and identify differentially expressed nuclear matrix proteins (NMPs) between the immortalized human esophageal epithelial cell line (SHEE) and the malignantly transformed esophageal carcinoma cell line (SHEEC), and to provide new ways for finding specific markers and the pathogenesis of esophageal carcinoma.METHODS: SHEE and SHEEC cell lines were used to extract NMPs. The quality of NMPs was monitored by Western blot analysis including DNA topoisomerase Ⅱα, proliferation cell nuclear antigen (PCNA) and histone. NMPs of SHEE and SHEEC were analyzed by two-dimensional electrophoresis (2-DE), silver staining and PDQuest6.2 image analysis software. Three spots in which the differentially expressed NMlPs were more obvious, were selected and analyzed with matrix-assisted laser desorption/ionization time of flying mass spectrometry (MALDI- TOF-MS) and database search.RESULTS: Western blot analysis revealed that DNA topoisomerase Ⅱα and PCNA were detected, and the majority of histones were deleted in NMPs of SHEE and SHEEC. After 2-DE image analysis by PDQuest6.2 software, the 2-DE maps were detected with an average of 106±7.1 spots in SHEE and 132±5.0 spots in SHEEC. Most of them were matched one another (r=0.72), only 16 protein spots were found differing in intensity. Three NMPs including cytoskeletal tropomyosin,FK506bindingprotein6,similartoretinoblastoma binding protein 8 were preliminarily identified by MALDI- TOF-MS.CONCLUSION: These differentially expressed NMPs may play an important role during malignant transformation from SHEE to SHEEC. | 8 | High-performance Li-ion batteries based on graphene quantum dot wrapped carbon nanotube hybrid anodes显示文摘Since Akira Yoshino first proposed the usage of the carbonaceous materials as an anode of lithium ion batteries(LIBs)in 1985,carbonaceous materials such as graphite and graphene have been widely considered as LIB anodes.Here,we explored the application of novel carbonaceous UB anodes incorporating graphene quantum dots(GQDs).We fabricated a freestanding all-carbon electrode based on a porous carbon nanotube(CNT)sponge via a facile in-situ hydrothermal deposition technique,creating coaxial structure of GQD-coated CNTs(GQD@CNTs)through electrostatic interaction and n-n stacking with tunable loading and functionalization.This hybrid structure combined conductive CNTs with highly active GQDs,in which GQDs with predesigned functional groups provided massive storage sites for Li ions and the 3D CNT frameworks avoided the agglomeration of GQDs,together contributing to a high specific capacity(700 mAh·g^-1 at 100 mA·g^-1 after 100 cycles)and rate performance.Even at a high current density of 1,000 mA·g^-1,the reversible specific capacity remained at 483 mAh g-1 after 350 cycles.In particular,the mechanism study demonstrated the important role of oxygen functional groups of GQDs in promoting the performance of the LIB anodes by controlled grafting of GQDs onto various porous-carbon and metal-foam based structures. | Xuewei Zhao Yizeng Wu Yunsong Wang Huaisheng Wu Yawei Yang Zhipeng Wang Linxiu Dai Yuanyuan Shang Anyuan Cao | 2020 | Nano Research2020,13,4: | 7 |
| 9 | Epstein-Barr virus in hepatocellular carcinogenesis显示文摘AIM: In recent years, studies have suggested that EpsteinBarr virus (EBV) is associated with HCC. The present study was to determine the prevalence of EBV in HCC patients,and whether EBV acted synergistically with hepatitis viruses in HCC carcinogenesis.METHODS: Liver tissue 115 HCC patients and 26 noncarcinoma patients were studied, Polymerase chain reaction (PCR) was performed to detect EBV BarnHI W DNA, EBV LMP1 DNA, HI3V X DNA, and HBV S DNA, Reverse transcription PCR (RT-PCR) was performed to detect HCV RNA and HDV RNA, Immunohistochemistry was performed to detect LMP1,HBsAg, HBcAg and HCV, The positive ratios were compared between HCC group and control group by X^2 test,RESULTS: Totally, 78 HCC samples whose 13-globulin DNA was positively detected by amplified PCR were selected,PCR was performed in all cases for EI3V DNA and HBV DNA,RT-PCR was performed in 18 cases for HCV RNA and HDV RNA. EBV BarnHI W and EBV LMP1 were positive in 18 and 6 cases, respectively. HBV X gene and HBV S gene were positive in 42 and 27 cases respectively. HCV was positive in one of the 18 cases, and none was positive for HDV. The positive rates were 28.2% (22 of 78) for EBV DNA (BarnHI W and/or LMP1) and 56.4% (44 of 78) for HBV DNA (X gene and/or S gene) respectively. In addition, 12 cases were positive for both EBV DNA and HBV DNA. Among the 26 cases in the control group, 2 cases were positive for EBV BarnHI W, 4 positive for HBV X gene and 3 positive for HBV S gene. The positive rates were 8.0% (2 of 26) and 23.1% (6 of 26),respectively, for EBV DNA and HBV DNA. The result of DNA sequencing of BamHI W was 100% homologous with the corresponding sequence of B95-8. There was significant difference in EBV infection rate between HCC patients and controls (X^2 = 4.622, P<0.05). The difference in HBV infection rate was also significant (X^2 = 8.681, P<0.05). However, there was no obvious correlation between HBV and EBV in HCC patients (X^2 = 0.835,/)>0.05). LMP1, HBV (HBsAg, HI3cAg) and HCV were detected positively in 25, 45 and 6 of 78 cases of HCC tissues respectively. In the 26 control cases,the corresponding positive cases were 2, 4 and O. The difference in EBV infection rate between HCC patients and control cases was statistically significant (X^2= 6.02,P<0.05). The difference in HBV infection rate was also statistically significant (X^2 = 10.03, P<0.05). In the 25 cases with positive LMP1 expression, 6 were in the nuclei of tumor cells, 9 in the cytoplasm of tumor cells and 10 in mesenchymal lymphocyte cytoplasm.CONCLUSION: The existence of EBV infection in HCC tissues suggests that EBV may be involved in the hepatocellular carcinogenesis in China. HBV infection may be a major cause of HCC. There is no correlation between EBV and HBV in the development of HCC. | 10 | Improving CNT-Si solar cells by metal chloride-to-oxide transformation显示文摘Transitional metal oxides(TMOs)are important functional materials in silicon-based and thin-film optoelectronics.Here,TMOs areapplied in carbon nanotube(CNT)-Si solar cells by spin-coating solutions of metal chlorides that undergo favorable transformation in ambient conditions.An unconventional change in solar cell behavior is observed after coating two particular chlorides(MoCl,and WCls,respectively),characterized by an initial severe degradation followed by gradual recovery and then well surpassing the original performance.Detailed analysis reveals that the formation of correspondina oxides(MoOa and WO.)enables two primary functions on both CNTs(p-type doping)and Si(inducing inversion layer),leading to significant improvement in open-circuit voltage and fill factor,with power conversion efficiencies up to 13.0%(MoOg)and 13.4%(WOg).Further combining with other chlorides to increase the short-circuit current,ultimate cells efficiencies achieve>16%with over 90%retention after 24 h,which are among the highes stable efficiencies reported for CNT-Si solar cells.The transformation of functional layers as demonstrated here has profoundinfluence on the device characteristics,and represents a potential strategy in low-cost manufacturing of next-generation high efficiency photovoltaics. | Huaisheng Wu Xuewei Zhao Yizeng Wu Qinghuan Ji Linxiu Dai Yuanyuan Shang Anyuan Cao | 2020 | Nano Research2020,13,2: | 3 |

The technique based on chromatographic fingerprinting is essentially a kind of high-through put and integral tools to explore the complexity of herbal medicines. In order to further control the comprehensive quality of TCMs,confirmation and identification of their important chemical components are necessary.
