1Upregulation of CD81 in trophoblasts induces an imbalance of Treg/Th17 cells by promoting IL-6 expression in preeclampsia显示文摘The disturbance of maternal immune tolerance to a semiallogeneic fetus is recognized as one of the key pathologies of preeclampsia(PE),in which an imbalance between the inflammation-limiting regulatory T cells(Tregs)and the inflammationmediating Th17 cells plays an essential role.Previously,we reported that the abnormal upregulation of tetraspannin CD81 in trophoblast cells(fetal component)participated in the pathogenesis of PE.However,as one of the potential immune regulatory molecules,whether CD81 induces PE by interfering with the balance of the maternal immune system has not yet been clarified.Thus,we investigated the relationship between the upregulation of CD81 in trophoblast cells and the imbalance of Treg and Th17 cells in mothers.Here,we demonstrated that upregulation of CD81 in trophoblast cells was accompanied by a decrease in Treg cells and an increase in Th17 cells in both the basal plate(placental maternal side)and peripheral blood of patients with PE.In vitro culture of naïve T cells with medium from the CD81-overexpressing trophoblast cell line HTR-8 resulted in enhanced differentiation of T cells into Th17 cells and decreased the formation of Tregs,which was dependent on the paracrine signaling of IL-6 in trophocytes,induced by CD81.In a CD81-induced PE rat model,we found a significant shift of T cell differentiation towards Th17 cells,and administration of IL-6 antibody mitigated the PE phenotype and the imbalance of the Treg/Th17 cells.These results define a vital regulatory cascade involving trophocyte-derived CD81,IL-6,and maternal Treg/Th17 cells in the pathogenesis of PE and suggests new therapeutic approaches based on CD81 and IL-6 downregulation to prevent human PE.Hailin Ding Yimin Dai Yi Lei Zhiyin Wang Dan Liu Ruotian Li Li Shen Ning Gu Mingming Zheng Xiangyu Zhu Guangfeng Zhao Yali Hu 2019Cellular & Molecular Immunology2019,16,3:15

3Targeting integrin pathways:mechanisms and advances in therapy显示文摘Integrins are considered the main cell-adhesion transmembrane receptors that play multifaceted roles as extracellular matrix(ECM)-cytoskeletal linkers and transducers in biochemical and mechanical signals between cells and their environment in a wide range of states in health and diseases.Integrin functions are dependable on a delicate balance between active and inactive status via multiple mechanisms,including protein-protein interactions,conformational changes,and trafficking.Due to their exposure on the cell surface and sensitivity to the molecular blockade,integrins have been investigated as pharmacological targets for nearly 40 years,but given the complexity of integrins and sometimes opposite characteristics,targeting integrin therapeutics has been a challenge.To date,only seven drugs targeting integrins have been successfully marketed,including abciximab,eptifibatide,tirofiban,natalizumab,vedolizumab,lifitegrast,and carotegrast.Currently,there are approximately 90 kinds of integrin-based therapeutic drugs or imaging agents in clinical studies,including small molecules,antibodies,synthetic mimic peptides,antibody-drug conjugates(ADCs),chimeric antigen receptor(CAR)T-cell therapy,imaging agents,etc.A serious lesson from past integrin drug discovery and research efforts is that successes rely on both a deep understanding of integrin-regulatory mechanisms and unmet clinical needs.Herein,we provide a systematic and complete review of all integrin family members and integrin-mediated downstream signal transduction to highlight ongoing efforts to develop new therapies/diagnoses from bench to clinic.In addition,we further discuss the trend of drug development,how to improve the success rate of clinical trials targeting integrin therapies,and the key points for clinical research,basic research,and translational research.Xiaocong Pang Xu He Zhiwei Qiu Hanxu Zhang Ran Xie Zhiyan Liu Yanlun Gu Nan Zhao Qian Xiang Yimin Cui 2023Signal Transduction and Targeted Therapy2023,8,2:3
4AN INTEGRO-DIFFERENTIAL PARABOLIC VARIATIONAL INEQUALITY ARISING FROM THE VALUATION OF DOUBLE BARRIER AMERICAN OPTION显示文摘This paper studies the nonlinear variational inequality with integro-differential term arising from valuation of American style double barrier option. First, the authors use the penalty method to transform the variational inequality into a nonlinear parabolic initial boundary problem(i.e., penalty problem). Second, the existence and uniqueness of solution to the penalty problem are proved by using the Scheafer fixed point theory. Third, the authors prove the existence of variational inequality' solution by showing the fact that the penalized PDE converges to the variational inequality. 5Standard pancreatoduodenectomy versus extended pancreatoduodenectomy with modified retroperitoneal nerve resection in patients with pancreatic head cancer:a multicenter randomized controlled trial显示文摘Background:The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial,and more high-level clinical evidence is needed.This study aimed to evaluate the outcome of extended pancreatoduodenectomy(EPD)with retroperitoneal nerve resection in pancreatic head cancer.Methods:This multicenter randomized trial was performed at 6 Chinese highvolume hospitals that enrolled patients between October 3,2012,and September 21,2017.Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments(preoperative chemotherapy or chemoradiation)within three months were randomly assigned to undergo standard pancreatoduodenectomy(SPD)or EPD,with the latter followed by dissection of additional lymph nodes(LNs),nerves and soft tissues 270◦on the right side surrounding the superior mesenteric artery and celiac axis.The primary endpoint was overall survival(OS)by intention-to-treat(ITT).The secondary endpoints were disease-free survival(DFS),mortality,morbidity,and postoperative pain intensity.Results:TheR1 ratewas slightly lower with EPD(8.46%)thanwith SPD(12.56%).The morbidity and mortality rates were similar between the two groups.The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort(23.0 vs.20.2 months,P=0.100),while the median DFS was superior in the EPD group(16.1 vs.13.2 months,P=0.031).Patients with preoperative CA19–9<200.0 U/mL had significantly improved OS and DFS with EPD(EPD vs.SPD,30.8 vs.20.9 months,P=0.009;23.4 vs.13.5 months,P<0.001).The EPD group exhibited significantly lower locoregional(16.48%vs.35.20%,P<0.001)andmesenteric LNrecurrence rates(3.98%vs.10.06%,P=0.022).The EPD group exhibited less back pain 6 months postoperation than the SPD group.Conclusions:EPD for pancreatic head cancer did not significantly improve OS,but patients with EPD treatment had significantly improved DFS.In the subgroup analysis,improvements in bothOS and DFS in the EPD armwere observed in patients with preoperative CA19–9<200.0 U/mL.EPD could be used as an effective surgical procedure for patients with pancreatic head cancer,especially those with preoperative CA19–9<200.0 U/mL.Qing Lin Shangyou Zheng Xianjun Yu Meifu Chen Yu Zhou Quanbo Zhou Chonghui Hu Jing Gu Zhongdong Xu LinWang Yimin Liu Qingyu Liu MinWang Guolin Li He Cheng Dongkai Zhou Guodong Liu Zhiqiang Fu Yu Long Yixiong Li Weilin Wang Renyi Qin Zhihua Li Rufu Chen 2023Cancer Communications2023,43,2:3
6Silver nanoparticles-decorated and mesoporous silica coated single-walled carbon nanotubes with an enhanced antibacterial activity for killing drug-resistant bacteria显示文摘The mounting threat of antibiotic-resistant bacterial infections has made it imperative to develop innovative antibacterial strategies.Here we propose a novel antibacterial nanoplatform of silver nanoparticles-decorated and mesoporous silica coated single-walled carbon nanotubes constructed via a N-[3-(trimethoxysiltyl)propyl]ethylene diamine(TSD)-mediated method(SWCNTs@mSiO2-TSD@Ag).In this system,the outer mesoporous silica shells are able to improve the dispersibility of SWCNTs,which will increase their contact area with bacteria cell walls.Meanwhile,the large number of mesopores in silica layers act as microreactors for in situ synthesis of Ag NPs with controlled small size and uniform distribution,which induces an enhanced antibacterial activity.Compared with TSD modified mesoporous silica coated single-walled carbon nanotubes(SWCNTs@mSiO2-TSD)and commercialAg NPs,this combination nanosystem of SWCNTs@mSiO2-TSD@Ag exhibits much stronger antibacterial performance against multi-drug-resistant bacteria Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus)in vitro through damaging the bacterial cell membranes and a fast release of silver ions.Furthermore,the in vivo rat skin infection model verifies that SWCNTS@mSiO2-TSD@Ag have remarkably improved abilities of bacterial clearance,wound healing promoting as well as outstanding biocompatibility.Therefore,this novel nanoplatform indicates promising potentials as a safe and powerful tool for the treatment of clinical drug-resistant infections.Yu Zhu Jia Xu Yanmao Wang Cang Chen Hongchen Gu Yimin Chai Yao Wang 2020Nano Research2020,13,2:2
7Use of angiotensin-converting enzyme inhibitors and angiotensin Ⅱ receptor blockers in context of COVID-19 outbreak:a retrospective analysis显示文摘The possible effects of angiotensin-converting enzyme inhibitors(ACEIs)or angiotensin Ⅱ receptor blockers(ARBs)on COVID-19 disease severity have generated considerable debate.We performed a single-center,retrospective analysis of hospitalized adult COVID-19 patients in Wuhan,China,who had definite clinical outcome(dead or discharged)by February 15,2020.Patients on anti-hypertensive treatment with or without ACEI/ARB were compared on their clinical characteristics and outcomes.The medical records from 702 patients were screened.Among the 101 patients with a history of hypertension and taking at least one anti-hypertensive medication,40 patients were receiving ACEI/ARB as part of their regimen,and 61 patients were on antihypertensive medication other than ACEI/ARB.We observed no statistically significant differences in percentages of in-hospital mortality(28%vs.34%,P=0.46),ICU admission(20%vs.28%,P=0.37)or invasive mechanical ventilation(18%vs.26%,P=0.31)between patients with or without ACEI/ARB treatment.Further multivariable adjustment of age and gender did not provide evidence for a significant association between ACEI/ARB treatment and severe COVID-19 outcomes.Our findings confirm the lack of an association between chronic receipt of reninangiotensin system antagonists and severe outcomes of COVID-19.Patients should continue previous antihypertensive therapy until further evidence is available.Jiuyang Xu Chaolin Huang Guohui Fan Zhibo Liu Lianhan Shang Fei Zhou Yeming Wang Jiapei Yu Luning Yang Ke Xie Zhisheng Huang Lixue Huang Xiaoying Gu Hui Li Yi Zhang Yimin Wang Frederick G.Hayden Peter W.Horby Bin Cao Chen Wang 2020Frontiers of Medicine2020,14,5:2

10Isolation and Identification of Squash Leaf Curl China Virus in Zucchini显示文摘[Objectives]In 2019,virus diseases occurred widely on zucchini planted in Shandong Province.The disease symptom was different from previous reports.This study aimed to identify the pathogen causing the zucchini virus disease.[Methods]Ten diseased zucchini leaves were collected in the field and used as materials for PCR and sequencing.[Results]PCR detection and sequencing showed that the nucleotide sequence of the amplified fragment had the highest identity with the squash isolate of squash leaf curl China virus(SLCCNV)(MW389919.1)in Guangdong Province.Primers were further designed for amplifying the full-length SLCCNV.The full-length DNA-A was 2730 bp(OM692270.1),and the full-length DNA-B was 2711 bp(OM692269.1).Through sequence alignment,it was found that the DNA-A sequence shared identity of 89.65%-99.42%with registered SLCCNV,and the identity with the SLCCNV-GDHY pumpkin isolate(MW389919.1)in Guangdong was the highest,at 99.42%.The DNA-B sequence was identical with registered SLCCNV in the range of 81.82%-97.29%,and the identity with the SLCCNV-GDHY pumpkin isolate(MW389918.1)in Guangdong,was the highest,at 97.29%.Therefore,it was speculated that SLCCNV is the pathogen of zucchini virus disease.Since the virus was first found on zucchini in Shandong,it was named SLCCNV-SD.[Conclusions]This study provides materials for the research on the spread of SLCCNV in China and the analysis of population genetic characteristics,as well as a reference for the prevention and control of the virus in zucchini.Huixian SHI Yimin GU Xingsheng LI Xiping SUN Yangdong GUO Xiaowei YUAN 2022Agricultural Biotechnology2022,11,5:1
11An Improved Dynamic Modelling for Exploring Ball Bearing Vibrations from Time-Varying Oil Film显示文摘Bearings are key components in rotating machinery,which is widely used in many fields,such as CNC machines,wind turbines and induction machines.The increasingly harsh operation environment can lead to wear and tear on raceways and reduce the precision and reliability of bearing or even machinery.Lubrication could relieve the wear to some degree,which is benefit to prolong the bearing’s life.Thus,investigation on the vibration responses under the influence of oil film is of great significance.However,for mechanism analysis,how to include the oil film into the bearing dynamic model affects the result and efficiency of solution.To address this problem,this study proposed a fast algorithm through load distribution and interpolation when calculating oil film stiffness and thickness during the solution of bearing vibration model.Analysis of oil film on vibration is carried out and a bearing test rig is designed to verify the proposed model.Numerical simulation result shows that rotational speed and load have vital effect on oil film and vibration.The experimental result is consistent with the simulation,which shows that the proposed model has a better performance on modeling bearing vibration and the method of considering oil film is reasonable.Minmin Xu Zhenzhen Song Xiaoxi Ding Guoxing Li Yimin Shao James Xi Gu 2022Journal of Dynamics, Monitoring and Diagnostics2022,1,2:1
12Association between sleep duration and incidence of type 2 diabetes in China: the REACTION study显示文摘Backgrounds: Inadequate sleep duration is associated with a higher risk of type 2 diabetes and the relationship is nonlinear. We aim to assess the curve relationship between night sleep duration and the incidence of type 2 diabetes in China.Methods: A cohort of 11,539 participants from the REACTION study without diabetes at baseline (2011) were followed until 2014 for the development of type 2 diabetes. The average number of hours of sleep per night was grouped. Incidence rates and odds ratios (ORs) were calculated for the development of diabetes in each sleep duration category.Results: Compared to people who sleep for 7 to 8 h/night, people with longer sleep duration (≥9 h/night) had a greater risk of type 2 diabetes (OR: 1.27;95% CI: 1.01-1.61), while shorter sleep (<6 h/night) had no significant difference in risk of type 2 diabetes. When the dataset was stratified based on selected covariates, the association between type 2 diabetes and long sleep duration became more evident among individuals <65 years of age, male, body mass index <24 kg/m2 or with hypertension or hyperlipidemia, no interaction effects were observed. Furthermore, compared to people persistently sleeping 7 to 9 h/night, those who persistently slept ≥9 h/night had a higher risk of type 2 diabetes. The optimal sleep duration was 6.3 to 7.5 h/night.Conclusions: Short or long sleep duration was associated with a higher risk of type 2 diabetes.
