| 1 | Gd-EOB-DTPA-enhanced MRI radiomic features for predicting histological grade of hepatocellular carcinoma显示文摘Background:Prediction models for the histological grade of hepatocellular carcinoma(HCC)remain unsatisfactory.The purpose of this study is to develop preoperative models to predict histological grade of HCC based on gadolinium-ethoxybenzyl diethylenetriamine pentaacetic acid(Gd-EOB-DTPA)-enhanced magnetic resonance imaging(MRI)radiomics.And to compare the performance between artificial neural network(ANN)and logistic regression model.Methods:A total of 122 HCCs were randomly assigned to the training set(n=85)and the test set(n=37).There were 242 radiomic features extracted from volumetric of interest(VOI)of arterial and hepatobiliary phases images.The radiomic features and clinical parameters[gender,age,alpha-fetoprotein(AFP),carcinoembryonic antigen(CEA),carbohydrate antigen 19-9(CA19-9),alanine aminotransferase(ALT),aspartate transaminase(AST)]were selected by permutation test and decision tree.ANN of arterial phase(ANN-AP),logistic regression model of arterial phase(LR-AP),ANN of hepatobiliary phase(ANN-HBP),logistic regression mode of hepatobiliary phase(LR-HBP),ANN of combined arterial and hepatobiliary phases(ANN-AP+HBP),and logistic regression model of combined arterial and hepatobiliary phases(LR-AP+HBP)were built to predict HCC histological grade.Those prediction models were assessed and compared.Results:ANN-AP and LR-AP were composed by AST and radiomic features based on arterial phase.ANN-HBP and LR-HBP were composed by AFP and radiomic features based on hepatobiliary phase.ANN-AP+HBP and LR-AP+HBP were composed by AST and radiomic features based on arterial and hepatobiliary phases.The prediction models could distinguish between high-grade tumors[Edmondson-Steiner(E-S)grade III and IV]and low-grade tumors(E-S grade I and II)in both training set and test set.In the test set,the AUCs of ANN-AP,LR-AP,ANN-HBP,LR-HBP,ANN-AP+HBP and LR-AP+HBP were 0.889,0.777,0.941,0.819,0.944 and 0.792 respectively.The ANN-HBP was significantly superior to LR-HBP(P=0.001).And the ANN-AP+HBP was significantly superior to LR-AP+HBP(P=0.007).Conclusions:Prediction models consisting of clinical parameters and Gd-EOB-DTPA-enhanced MRI radiomic features(based on arterial phase,hepatobiliary phase,and combined arterial and hepatobiliary phases)could distinguish between high-grade HCCs and low-grade HCCs.And the ANN was superior to logistic regression model in predicting histological grade of HCC. | Yingfan Mao Jincheng Wang Yong Zhu Jun Chen Liang Mao Weiwei Kong Yudong Qiu Xiaoyan Wu Yue Guan Jian He | 2022 | Hepatobiliary Surgery and Nutrition2022,11,1: | 6 |

| 11 | KRAS mutation-induced EndMT of brain arteriovenous malformation is mediated through the TGF-β/BMP-SMAD4 pathway显示文摘Objective Somatic KRAS mutations have been identified in the majority of brain arteriovenous malformations(bAVMs),and subsequent in vivo experiments have confirmed that KRAS mutation in endothelial cells(ECs)causes AVMs in mouse and zebrafish models.Our previous study demonstrated that the KRASG12D mutant independently induced the endothelial-mesenchymal transition(EndMT),which was reversed by treatment with the lipid-lowering drug lovastatin.However,the underlying mechanisms of action were unclear.Methods We used human umbilical vein ECs(HUVECs)overexpressing the KRASG12D mutant for Western blotting,quantitative real-time PCR,and immunofluorescence and wound healing assays to evaluate the EndMT and determine the activation of downstream pathways.Knockdown of SMAD4 by RNA interference was performed to explore the role of SMAD4 in regulating the EndMT.BAVM ECs expressing the KRASG12D mutant were obtained to verify the SMAD4 function.Finally,we performed a coimmunoprecipitation assay to probe the mechanism by which lovastatin affects SMAD4.Results HUVECs infected with KRASG12D adenovirus underwent the EndMT.Transforming growth factor beta(TGF-β)and bone morphogenetic protein(BMP)signalling pathways were activated in the KRASG12D-mutant HUVECs and ECs in bAVM tissue.Knocking down SMAD4 expression in both KRASG12D-mutant HUVECs and ECs in bAVM tissues inhibited the EndMT.Lovastatin attenuated the EndMT by downregulating p-SMAD2/3,p-SMAD1/5 and acetylated SMAD4 expression in KRASG12D-mutant HUVECs.Conclusions Our findings suggest that the KRASG12D mutant induces the EndMT by activating the ERK-TGF-β/BMP-SMAD4 signalling pathway and that lovastatin inhibits the EndMT by suppressing TGF-β/BMP pathway activation and SMAD4 acetylation. | Hongyuan Xu Ran Huo Hao Li Yuming Jiao Jiancong Weng Jie Wang Zihan Yan Junze Zhang Shaozhi Zhao Qiheng He Yingfan Sun Shuo Wang Yong Cao | 2023 | Stroke & Vascular Neurology2023,8,3: | 1 |

| 17 | AuPt bimetallic nanoalloys supported on SBA-15:A superior catalyst for quinoline selective hydrogenation in water显示文摘The structure and size of bimetallic catalysts play a crucial role in many important chemical transformations.Controlled synthesis of bimetallic nanoparticles avoiding overgrowth and aggregation can be achieved by surfactants,which are always detrimental to catalytic performances and understanding of structure-property relationship.Preparation of surface-clean bimetallic catalysts with uniform size and well-defined structure is still challenging.Herein the Au-Pt bimetallic nanoparticles immobilized on SBA-15 were prepared by facile adsorption-reduction method.Characterizations showed that Au-Pt bimetallic nanoparticles were evenly confined within the mesopores of SBA-15,possessing the uniform size of 6.0 nm and existing in the form of alloy structure.For the first time the Au-Pt bimetallic nanoalloys with Au-to-Pt molar ratio of 5:1(Au5Pt1@SBA-15)exhibited a lot higher activity than monometallic Au@SBA-15 and Pt@SBA-15 catalysts with excellent selectivity towards 1,2,3,4-tetrahydroquinoline for chemoselective hydrogenation of quinoline in water under very mild conditions.It was superior to other heterogeneous catalysts reported to date.The observed properties were related to the synergic effect between Au and Pt.The Pt sites of high electron density originated from the electron transfer between Au and Pt enhanced the ability of H_(2) dissociation and then provided a new and main approach to H_(2) splitting,while the Au sites accounted for the adsorption and activation of quinoline.This precise synthetic strategy will be very helpful to explore surface-clean gold-based bimetallic catalysts with high performance for selective hydrogenations. | Jianbo Zhao Haifeng Yuan Guang Yang Yingfan Liu Xiaomei Qin Zheng Chen Cheong Weng-Chon Liming Zhou Shaoming Fang | 2022 | Nano Research2022,15,3: | 0 |
| 18 | Cotton GhBRC1 regulates branching,flowering,and growth by integrating multiple hormone pathways显示文摘Cotton architecture is partly determined by shoot branching and flowering patterns.Gh BRC1 was previously identified by RNA-seq analysis of nulliplex-branching and normal-branching cotton.However,the roles of Gh BRC1 in cotton remain unclear.In the present study,investigations of nuclear localization and transcriptional activity indicated that Gh BRC1 has characteristics typical of transcription factors.Gene expression analysis showed that Gh BRC1 was highly expressed in axillary buds but displayed different expression patterns between the two branching types.Overexpression of Gh BRC1 in Arabidopsis significantly inhibited the number of branches and promoted flowering.In contrast,silencing Gh BRC1 in cotton significantly promoted seedling growth.Gh BRC1 was induced by multiple hormones,including strigolactones,which promoted seedling growth and seed germination of Arabidopsis plants overexpressing Gh BRC1.Consistent with these findings,RNA-seq analysis of virus-induced gene silencing treated cotton revealed that a large number of genes were differentially expressed between Gh BRC1-silenced and control plants,and these genes were significantly enriched in plant hormone signalling pathways.Together,our data indicates that Gh BRC1 regulates plant branching and flowering through multiple regulatory pathways,especially those regulating plant hormones,with functions partly differing from those of Arabidopsis BRC1.These results provide insights into the molecular mechanisms controlling plant architecture,which is important for breeding cotton with ideal plant architecture and high yield. | Quan Sun Yuanhui Xie Huimin Li Jinlei Liu Rui Geng Ping Wang Zongyan Chu Ying Chang Guanjun Li Xiao Zhang Youlu Yuan Yingfan Cai | 2022 | The Crop Journal2022,10,1: | 0 |
| 19 | Effects of Al_(2)O_(3)-SiO_(2) Raw Material Types on Properties of Anorthite Based Insulation Refractories显示文摘To optimize their Al_(2)O_(3)-SiO_(2) raw materials,anorthite based insulation refractories were prepared by the in-situ sintering process combined with the foaming method after sintering at 1350℃for 3 h,using green and pollution-free kaolin,kyanite,andalusite and sillimanite as Al_(2)O_(3)-SiO_(2) raw materials,respectively,and industrial CaCO_(3) as the CaO source.Effects of Al_(2)O_(3)-SiO_(2) raw material types on the physical properties,phase composition and microstructure were investigated.The results are as follows.All samples prepared by different Al_(2)O_(3)-SiO_(2) raw materials have hexagonal flake anorthite and a small amount of mullite and corundum.Their bulk density and thermal conductivity decrease in the order of using kaolin,andalusite,kyanite and sillimanite as the Al_(2)O_(3)-SiO_(2) raw material,but their apparent porosity increases.Moreover,in the sample with kaolin,the bonding between anorthite crystals on the pore walls is closer than that of the other samples,which is conducive to increasing the cold crushing strength.The bonding between anorthite crystals on pore walls gradually decreases in the order of using kyanite,andalusite and sillimanite as the Al_(2)O_(3)-SiO_(2) raw material,thus their cold crushing strength decreases accordingly.In comprehensive consideration,the properties of the sample from kyanite are the optimal.Its apparent porosity,thermal conductivity and cold crushing strength are 84.6%,0.141 W·m^(-1)·K^(-1) and 1.89 MPa,respectively. | DU Juan GUO Huishi YANG Jialin LI Wenfeng GUI Yanghai ZHAO Zhiqiang LIU Yingfan | 2024 | China's Refractories2024,33,1: | 0 |
| 20 | Design, synthesis, molecular modeling, and biological evaluation of acrylamide derivatives as potent inhibitors of human dihydroorotate dehydrogenase for the treatment of rheumatoid arthritis显示文摘Human dihydroorotate dehydrogenase(DHODH) is a viable target for the development of therapeutics to treat cancer and immunological diseases, such as rheumatoid arthritis(RA), psoriasis and multiple sclerosis(MS). Herein, a series of acrylamide-based novel DHODH inhibitors as potential RA treatment agents were designed and synthesized. 2-Acrylamidobenzoic acid analog 11 was identified as the lead compound for structureeactivity relationship(SAR) studies. The replacement of the phenyl group with naphthyl moieties improved inhibitory activity significantly to double-digit nanomolar range.Further structure optimization revealed that an acrylamide with small hydrophobic groups(Me, Cl or Br)at the 2-position was preferred. Moreover, adding a fluoro atom at the 5-position of the benzoic acid enhanced the potency. The optimization efforts led to potent compounds 42 and 53-55 with IC50 values of 41, 44, 32, and 42 nmol/L, respectively.
