1The mechanosensitive lncRNA Neat1 promotes osteoblast function through paraspeckle-dependent Smurf1 mRNA retention显示文摘Mechanical stimulation plays an important role in bone remodeling. Exercise-induced mechanical loading enhances bone strength,whereas mechanical unloading leads to bone loss. Increasing evidence has demonstrated that long noncoding RNAs(lnc RNAs) play key roles in diverse biological, physiological and pathological contexts. However, the roles of lnc RNAs in mechanotransduction and their relationships with bone formation remain unknown. In this study, we screened mechanosensing lnc RNAs in osteoblasts and identified Neat1, the most clearly decreased lnc RNA under simulated microgravity. Of note, not only Neat1 expression but also the specific paraspeckle structure formed by Neat1 was sensitive to different mechanical stimulations, which were closely associated with osteoblast function. Paraspeckles exhibited small punctate aggregates under simulated microgravity and elongated prolate or larger irregular structures under mechanical loading. Neat1 knockout mice displayed disrupted bone formation, impaired bone structure and strength, and reduced bone mass. Neat1 deficiency in osteoblasts reduced the response of osteoblasts to mechanical stimulation. In vivo, Neat1 knockout in mice weakened the bone phenotypes in response to mechanical loading and hindlimb unloading stimulation. Mechanistically, paraspeckles promoted nuclear retention of E3 ubiquitin ligase Smurf1 m RNA and downregulation of their translation, thus inhibiting ubiquitination-mediated degradation of the osteoblast master transcription factor Runx2, a Smurf1 target. 2MicroRNA-activated hydrogel scaffold generated by 3D printing accelerates bone regeneration显示文摘Bone defects remain a major threat to human health and bone tissue regeneration has become a prominent clinical demand worldwide.The combination of microRNA(miRNA)therapy with 3D printed scaffolds has always posed a challenge.It can mimic physiological bone healing processes,in which a biodegradable scaffold is gradually replaced by neo-tissue,and the sustained release of miRNA plays a vital role in creating an optimal osteogenic microenvironment,thus achieving promising bone repair outcomes.However,the balance between two key factors-scaffold degradation behavior and miRNA release profile-on osteogenesis and bone formation is still poorly understood.Herein,we construct a series of miRNA-activated hydrogel scaffolds(MAHSs)generated by 3D printing with different crosslinking degree to screened the interplay between scaffold degradation and miRNA release in the osteoinduction activity both in vitro and in vivo.Although MAHSs with a lower crosslinking degree(MAHS-0 and MAHS-0.25)released a higher amount of miR-29b in a sustained release profile,they degraded too fast to provide prolonged support for cell and tissue ingrowth.On the contrary,although the slow degradation of MAHSs with a higher crosslinking degree(MAHS-1 and MAHS-2.5)led to insufficient release of miR-29b,their adaptable degradation rate endowed them with more efficient osteoinductive behavior over the long term.MAHS-1 gave the most well-matched degradation rate and miR-29b release characteristics and was identified as the preferred MAHSs for accelerated bone regeneration.This study suggests that the bio-adaptable balance between scaffold degradation behavior and bioactive factors release profile plays a critical role in bone regeneration.These findings will provide a valuable reference about designing miRNAs as well as other bioactive molecules activated scaffold for tissue regeneration.Ting Pan Wenjing Song Hongbao Xin Haiyue Yu He Wang Dandan Ma Xiaodong Cao Yingjun Wang 2022Bioactive Materials2022,7,4:2

9USP31 acetylation at Lys1264 is essential for its activity and cervical cancer cell growth显示文摘Ubiquitin-specific protease 31(USP31)is a member of deubiquitinase family that is involved in nuclear factor-KB activation and sarcomagenesis.However,little is known about posttranslational modification in the regulation of its activity and cervical cancer cell growth.In our study,we found that the Lys1264 residue of USP31 can be modified with an acetyl group by high-resolution mass spectrometry in HeLa cell line,and site-specific mutagenesis can significantly increase USP31 ubiquitin hydrolase activity and decrease the expression of p65.When being transfected with a plasmid expressing mutated USP31,the number of cancer cells was significantly decreased.We also observed that mutated USP31 could promote apoptosis but not cell cycle by flow cytometer analysis.Overexpression of mutated USP31 could reverse the effect in USP31 knockdown cell line.To further investigate its activity in tumorigenesis,deacetylase sirtuin 1(Sirtl)was shown to interact with USP31 by co-immunoprecipitation and blocking the function of Sirtl by knockdown or the inhibitor nicotinamide could increase the acetylation of USP31.When Lys1264 of USP31 mutated,Sirtl could not remove its acetylation and alter the expression level of p65.Finally,inhibition or knockdown of Sirtl suppressed USP31 activity in HeLa cell line,leading to cisplatin-induced apoptosis resistance.Therefore,acetylation at Lys1264 suppresses USP31 activity and plays a protective role in cancer cell growth.Our study contributes to understanding the mechanism of USP31 activity regulation and its role in tumorigenesis.Yaoyao Hou Yingjun Fan Xian Xia Jicheng Pan Jianjun Hou Xixia Liu Xinpeng Chen 2021Acta Biochimica et Biophysica Sinica2021,53,8:1

15Optimization of working parameters for 3MGY-200 axial air-assisted sprayer in kiwifruit orchards显示文摘Axial air-assisted sprayers can distribute pesticides efficiently in kiwifruit orchards.Because of improper parameter settings,most sprayers deliver either too much or too little pesticide.To identify appropriate sprayer parameters for kiwifruit trees,the vertical distribution profiles of the applied liquid spray were examined in this study.The effects of spray fan speed(SFS),spray pressure(SP)and spray distance(SD)on the distributions of the sprayed liquid in the vertical profiles were studied.Combined actions of the above parameters were systematically analysed using the quadratic general rotary design test method.Regression equations for the spray liquid distributions and working factors are presented.Field confirmation experiments were carried out to optimize the parameters.Data analysis showed that the optional sprayer working parameters are those of Group 3,with an SFS equal to 1900 r/min and SP equal to 3.25 MPa.The results of this study provide a reference for future applications of this type of axial air-assisted sprayer in kiwifruit orchards.Chenchen Gu Zhijie Liu Guanting Pan Yingjun Pu Fuzeng Yang 2020International Journal of Agricultural and Biological Engineering2020,13,2:1
