1Study on Extraction Technology of Theabrownine from Liubao Tea and Its Antioxidant Activity显示文摘[Objective]This study aimed to provide theoretical reference for the extraction of theabrownine from Liubao tea,determination of the biological activity,and development and utilization of theabrownine. [Method]Orthogonal experimental design was adopted to investigate the effects of solid-liquid ratio,extraction temperature,extraction duration,and extraction frequency on the yield of theabrownine extracted from Liubao tea. The extraction technology was optimized,and the antioxidant activity of theabrownine was analyzed. [Result] The results showed that extraction frequency and extraction temperature had significant effects on the yield of theabrownine extracted from Liubao tea,while solid-liquid ratio and extraction duration had no significant effect. The optimal conditions for extracting theabrownine from Liubao tea were determined as follows: solid-liquid ratio was 1∶30,extraction temperature was 100 ℃,extraction duration was 80 min,and extraction frequency was 4 times. Under these conditions,the yield of theabrownine reached 22.92%. Results of antioxidant activity determination showed that hydroxyl radical-scavenging activity of theabrownine was as high as 74%,and the scavenging effects was significantly higher than that of Vitamin C under low concentrations; nitroso-scavenging activity of theabrownine reached 73%,with no significant difference from that of Vitamin C; superoxide anion free radical-scavenging activity of theabrownine could be above 60%,which was lower than that of Vitamin C. 2An algorithm and observability research of autonomous navigation and joint attitude determination in asteroid explorationdescent stage显示文摘In this paper, an autonomous relative navigation and joint attitude determination algorithm in asteroid exploration descent stage is researched based on feature point information of perpendicular asteroid surface image observed by optical navigation camera, distance vectors from spacecraft to asteroid measured by three angled installed lidars and relative velocity increment measured by accelerometer when the relative distance vector to the centroid of asteroid can not be obtained. The inertial attitude of spacecraft is determined by sun vector, star vectors and inertial angular velocity respectively measured by sun sensor, star trackers and inertial reference unit. 3Effect of Kushen(Radix Sophorae flavescentis) extract on laryngeal neoplasm Hep2 cells显示文摘OBJECTIVE:To evaluate the effect of fermented extract of Kushen(Radix Sophorae Flavescentis) or non-fermented ESF on laryngeal neoplasms Hep2 cells.METHODS:Use 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay to explore the effect of fermented ESF and non-fermented ESF on Hep2 cells,and detect the mRNA and protein expression level of Bcl-2,Bax and Caspase-3 with reverse transcription polymerase chain reaction(RT-PCR) andWestern blot.RESULTS:Both fermented ESF and non-fermented ESF could inhibit laryngeal neoplasm's Hep2 cells,but and the cells did not response to the dilution 1:320 of fermented ESF,nor to the 1:1280 dilution of non-fermented ESF.As time progressed,the dilution 1:80 of fermented ESF and 1:320 dilution of non-fermented ESF could significantly reduce Bcl-2 mRNA and protein expression and down-regulate Caspase-3 mRNA and protein expression.Bax mRNA and protein were not expressed in Hep2 cells.CONCLUSIONS:Both fermented ESF and non-fermented ESF could inhibit the proliferation of Hep2 cells,and the effect of non-fermented ESF was significantly better than that of the fermented.Yingzi Wang Chunchao Han Xinsheng Fang Xinyuan Shi Ailing Feng Kuibang He Shenghai Zhang Xiuyu Sun 2013Journal of Traditional Chinese Medicine2013,33,2:1
4Inhibiting DNA methylation alleviates cisplatininduced hearing loss by decreasing oxidative stress-induced mitochondria-dependent apoptosis via the LRP1-PI3K/AKT pathway显示文摘Cisplatin-related ototoxicity is a critical side effect of chemotherapy and can lead to irreversible hearing loss.This study aimed to assess the potential effect of the DNA methyltransferase(DNMT)inhibitor RG108 on cisplatin-induced ototoxicity.Immunohistochemistry,apoptosis assay,and auditory brainstem response(ABR)were employed to determine the impacts of RG108 on cisplatin-induced injury in murine hair cells(HCs)and spiral ganglion neurons(SGNs).Rhodamine 123 and TMRM were utilized for mitochondrial membrane potential(MMP)assessment.Reactive oxygen species(ROS)amounts were evaluated by Cellrox green and Mitosox-red probes.Mitochondrial respiratory function evaluation was performed by determining oxygen consumption rates(OCRs).The results showed that RG108 can markedly reduce cisplatin induced damage in HCs and SGNs,and alleviate apoptotic rate by protecting mitochondrial function through preventing ROS accumulation.Furthermore,RG108 upregulated BCL-2 and downregulated APAF1,BAX,and BAD in HEI-OC1 cells,and triggered the PI3K/AKT pathway.Decreased expression of low-density lipoprotein receptor-related protein 1(LRP1)and high methylation of the LRP1 promoter were observed after cisplatin treatment.RG108 treatment can increase LRP1expression and decrease LRP1 promoter methylation.In conclusion,RG108 might represent a new potential agent for preventing hearing loss induced by cisplatin via activating the LRP1-PI3K/AKT pathway.Yingzi He Zhiwei Zheng Chang Liu Wen Li Liping Zhao Guohui Nie Huawei Li 2022Acta Pharmaceutica Sinica B2022,12,3:1
5Reducing the Internal Difference Error Rate in Hospital Pharmacies:From the Angle of Quality Control Circle显示文摘Objective To improve the management level of pharmacy dispensing center,reduce dispensing errors and promote the safety of drug use.Methods Hospital pharmacies could be managed according to the theory of quality control circle(QCC).Based on the ten steps of QCC,the internal difference error rate in pharmacies could be reduced.Results and Conclusion The error rate of pharmacies was reduced from 2.74‰to 0.57‰,and the goal achievement rate was 108.466.Besides,the progress rate reached 84.82%.The abilities of circle members were improved,and the operation of pharmacy was more standardized.The activity of QCC is helpful to reduce the internal difference error rate,improve the operation level of pharmacy and ensure the safety of drug use.Wang Shuling Lei Yingzi Liu Linchuan Wang Ruili He Yalan 2020Asian Journal of Social Pharmacy2020,15,2:1

7Transcription factor CsTT8 promotes fruit coloration by positively regulating the methylerythritol 4-phosphate pathway and carotenoid biosynthesis pathway in citrus (Citrus spp.)显示文摘Carotenoids directly influence citrus fruit color and nutritional value,which is critical to consumer acceptance.Elucidating the potential molecular mechanism underlying carotenoid metabolism is of great importance for improving fruit quality.Despite the well-established carotenoid biosynthetic pathways,the molecular regulatory mechanism underlying carotenoid metabolism remains poorly understood.Our previous studies have reported that the Myc-type basic helix-loop-helix(bHLH)transcription factor(TF)regulates citrus proanthocyanidin biosynthesis.Transgenic analyses further showed that overexpression of CsTT8 could significantly promote carotenoid accumulation in transgenic citrus calli,but its regulatory mechanism is still unclear.In the present study,we found that overexpression of CsTT8 enhances carotenoid content in citrus fruit and calli by increasing the expression of CsDXR,CsHDS,CsHDR,CsPDS,CsLCYE,CsZEP,and CsNCED2,which was accompanied by changes in the contents of abscisic acid and gibberellin.The in vitro and in vivo assays indicated that CsTT8 directly bound to the promoters of CsDXR,CsHDS,and CsHDR,the keymetabolic enzymes of the methylerythritol 4-phosphate(MEP)pathway,thus providing precursors for carotenoid biosynthesis and transcriptionally activating the expression of these three genes.In addition,CsTT8 activated the promoters of four key carotenoid biosynthesis pathway genes,CsPDS,CsLCYE,CsZEP,and CsNCED2,directly promoting carotenoid biosynthesis.This study reveals a novel network of carotenoid metabolism regulated by CsTT8.Our findings will contribute to manipulating carotenoid metabolic engineering to improve the quality of citrus fruit and other crops.Quan Sun Zhengchen He Ranran Wei Yingzi Yin Junli Ye Lijun Chai Zongzhou Xie Wenwu Guo Juan Xu Yunjiang Cheng Qiang Xu Xiuxin Deng 2023Horticulture Research2023,10,11:0
8Inhibition of histone methyltransferase PRMT5 attenuates cisplatininduced hearing loss through the PI3K/Akt-mediated mitochondrial apoptotic pathway显示文摘This study aimed to evaluate the therapeutic potential of inhibiting protein arginine methyltransferase 5(PRMT5)in cisplatin-induced hearing loss.The effects of PRMT5 inhibition on cisplatin-induced auditory injury were determined using immunohistochemistry,apoptosis assays,and auditory brainstem response.The mechanism of PRMT5 inhibition on hair cell survival was assessed using RNA-seq and Cleavage Under Targets and Tagment-quantitative polymerase chain reaction(CUT&Tag-qPCR)analyses in the HEI-OC1 cell line.Pharmacological inhibition of PRMT5 significantly alleviated cisplatin-induced damage to hair cells and spiral ganglion neurons in the cochlea and decreased apoptosis by protecting mitochondrial function and preventing the accumulation of reactive oxygen species.CUT&Tag-qPCR analysis demonstrated that inhibition of PRMT5 in HEI-OC1 cells reduced the accumulation of H4R3me2s/H3R8me2s marks at the promoter region of the Pik3ca gene,thus activating the expression of Pik3ca.These findings suggest that PRMT5 inhibitors have strong potential as agents against cisplatininduced ototoxicity and can lay the foundation for further research on treatment strategies of hearing loss.Zhiwei Zheng Benyu Nan Chang Liu Dongmei Tang Wen Li Liping Zhao Guohui Nie Yingzi He 2023Journal of Pharmaceutical Analysis2023,13,6:0
