1Fecal microbiota transplantation from young donor mice improves ovarian function in aged mice显示文摘Advanced maternal age is characterized by declines in the quantity and quality of oocytes in the ovaries,and the aging process is accompanied by changes in gut microbiota composition.However,little is known about the relationship between gut microbiota and ovarian aging.By using fecal microbiota transplantation(FMT)to transplant material from young(5-week-old)into aged(42-week-old)mice,we find that the composition of gut microbiota in FMT-treated mice presents a“younger-like phenotype”and an increase of commensal bacteria,such as Bifidobacterium and Ruminococcaceae.Moreover,the FMT-treated mice show increased anti-inflammatory cytokine IL-4 and decreased pro-inflammatory cytokine IFN-γ.Fertility tests for assessing ovarian function reveal that the first litter size of female FMT-treated mice is significantly higher than that of the non-FMT group.Morphology analysis demonstrates a dramatic decrease in follicle atresia and apoptosis as well as an increase in cellular proliferation in the ovaries of the FMT-treated mice.Our results also show that FMT improves the immune microenvironment in aged ovaries,with decreased macrophages and macrophage-derived multinucleated giant cells(MNGCs).These results suggest that FMT from young donors could be a good choice for delaying ovarian aging.Li Xu Qiankun Zhang Xiaowei Dou Yipeng Wang Jianwei Wang Yong Zhou Xingyin Liu Jing Li 2022Journal of Genetics and Genomics2022,49,11:2
2Noise level estimation method with application to EMD-based signal denoising显示文摘This paper proposes a new signal noise level estimation approach by local regions. The estimated noise variance is applied as the threshold for an improved empirical mode decomposition(EMD) based signal denoising method. The proposed estimation method can effectively extract the candidate regions for the noise level estimation by measuring the correlation coefficient between noisy signal and a Gaussian filtered signal. For the improved EMD based method, the situation of decomposed intrinsic mode function(IMFs) which contains noise and signal simultaneously are taken into account. 3Evaluation of menopausal status among breast cancer patients with chemotherapy-induced amenorrhea显示文摘Objective: In patients with chemotherapy-induced amenorrhea(CIA), the menopausal status is ambiguous and difficult to evaluate. This study aimed to establish a discriminative model to predict and classify the menopausal status of breast cancer patients with CIA.Methods: This is a single center hospital-based study from 2013 to 2016. The menopausal age distribution and accumulated incidence rate of CIA are described. Multivariate models were adjusted for established and potential confounding factors including age, serum concentration of estradiol(E2) and follicle-stimulating hormone(FSH),feeding, pregnancy, parity, abortions, and body mass index(BMI). The odds ratio(OR) and 95% confidence interval(95% CI) of different risk factors were estimated.Results: A total of 1,796 breast cancer patients were included in this study, among whom, 1,175(65.42%) were premenopausal patients and 621(34.58%) were post-menopause patients. Five hundred and fifty patients were included in CIA analysis, and a cumulative CIA rate of 81.64% was found in them. Age(OR: 1.856, 95% CI:1.732-1.990), serum concentration of E2(OR: 0.976, 95% CI: 0.972-0.980) and FSH(OR: 1.060, 95% CI:1.053-1.066), and menarche age(OR: 1.074, 95% CI: 1.009-1.144) were found to be associated with the patients' menopausal status. According to multivariate analysis, the discriminative model to predict the menopausal status is Logit(P)=-28.396+0.536 Age-0.014 E2+0.031 FSH. The sensitivities for this model were higher than 85%, and its specificities were higher than 89%.Conclusions: The discriminative model obtained from this study for predicting menstrual state is important for premenopausal patients with CIA. 9A novel mutation in ROR2 led to the loss of function of ROR2 and inhibited the osteogenic differentiation capability of bone marrow mesenchymal stem cells(BMSCs)显示文摘Receptor tyrosine kinase-like orphan receptor 2(ROR2)has a vital role in osteogenesis.However,the mechanism underlying the regulation of ROR2 in osteogenic differentiation is still poorly comprehended.A previous study by our research group showed that a novel compound heterozygous ROR2 variation accounted for the autosomal recessive Robinow syndrome(ARRS).This study attempted to explore the impact of the ROR2:c.904C>T variant specifically on the osteogenic differentiation of BMSCs.Methods:Coimmunoprecipitation(CoIP)-western blotting was carried out to identify the interaction between ROR2 and Wnt5a.Double-immunofluorescence staining was used for determining the expressions and co-localization of ROR2 and Wnt5a in bone marrow mesenchymal stem cells(BMSCs).Western blot(WB)analysis and quantitative reverse transcription polymerase chain reaction(RT-qPCR)were conducted to identify the expression levels of ROR2 in the BMSCs transfected with LV-shROR2 or LV-ROR2-c.904C>T.The alkaline phosphatase(ALP)activity was detected,and Alizarin Red S staining was done for evaluating the osteogenic differentiation of BMSCs.RT-qPCR was employed to identify the expression of the sphingomyelin synthase 1(SMS1)mRNA in the BMSCs transfected with LV-shROR2 or LV-ROR2-c.904C>T and the mRNA expression levels of Runt-related transcription factor 2(RUNX2),osteocalcin(OCN),and osteopontin(OPN).WB was performed to confirm the protein expressions of extracellular regulated protein kinases1(ERK),P-ERK,Smad family member1/5/8(Smad1/5/8),P-Smad1/5/8,P-P38,P38,RUNX2,OCN,and OPN in the BMSCs transfected with LV-shROR2/LV-ROR2-c.904C>T and sphingomyelin(SM).Results:The ROR2:c.904C>T mutant altered the subcellular localization of the ROR2 protein,which caused an impaired interaction between ROR2 and Wnt5a.The depletion of ROR2 restricted the osteogenic differentiation capability of BMSCs and downregulated the expression of SMS1.SM treatment could reverse the inhibition of osteoblastic differentiation in ROR2-depleted BMSCs.Conclusion:The findings of this work revealed that the ROR2:c.904C>T variant led to the loss of function of ROR2,which impaired the interaction between ROR2 and Wnt5a and also controlled the osteogenic differentiation capability of BMSCs.Furthermore,SM was revealed to be engaged in the osteoblastic differentiation of BMSCs regulated by ROR2,which renders SM a potential target in the therapy for ARRS.WENQI CHEN XIAOYANG CHU YANG ZENG YOUSHENG YAN YIPENG WANG DONGLAN SUN DONGLIANG ZHANG JING ZHANG KAI YANG 2023BIOCELL2023,47,7:0

11A telomere-to-telomere reference genome provides genetic insight into the pentacyclic triterpenoid biosynthesis in Chaenomeles speciosa显示文摘Chaenomeles speciosa(2n=34),a medicinal and edible plant in the Rosaceae,is commonly used in traditional Chinese medicine.To date,the lack of genomic sequence and genetic studies has impeded efforts to improve its medicinal value.Herein,we report the use of an integrative approach involving PacBio HiFi(third-generation)sequencing and Hi-C scaffolding to assemble a high-quality telomere-to-telomere genome of C.speciosa.The genome comprised 650.4 Mb with a contig N50 of 35.5 Mb.Of these,632.3 Mb were anchored to 17 pseudo-chromosomes,in which 12,4,and 1 pseudo-chromosomes were represented by a single contig,two contigs,and four contigs,respectively.Eleven pseudo-chromosomes had telomere repeats at both ends,and four had telomere repeats at a single end.Repetitive sequences accounted for 49.5%of the genome,while a total of 45515 protein-coding genes have been annotated.The genome size of C.speciosa was relatively similar to that of Malus domestica.Expanded or contracted gene families were identified and investigated for their association with different plant metabolisms or biological processes.In particular,functional annotation characterized gene families that were associated with the biosynthetic pathway of oleanolic and ursolic acids,two abundant pentacyclic triterpenoids in the fruits of C.speciosa.Taken together,this telomere-to-telomere and chromosome-level genome of C.speciosa not only provides a valuable resource to enhance understanding of the biosynthesis of medicinal compounds in tissues,but also promotes understanding of the evolution of the Rosaceae.Shaofang He Duanyang Weng Yipeng Zhang Qiusheng Kong Keyue Wang Naliang Jing Fengfeng Li Yuebin Ge Hui Xiong Lei Wu De-Yu Xie Shengqiu Feng Xiaqing Yu Xuekui Wang Shaohua Shu Zhinan Mei 2023Horticulture Research2023,10,10:0

14CosmicGrowth Simulations——Cosmological simulations for structure growth studies显示文摘I present a large set of high resolution simulations, called CosmicGrowth Simulations, which were generated with either 8.6 billion or 29 billion particles. As for the nominal cosmological model that can match nearly all observations on cosmological scales, I have adopted a flat Cold Dark Matter(CDM) model with a cosmological constant Λ(ΛCDM). The model parameters have been taken either from the latest result of the WMAP satellite(WMAP ΛCDM) or from the first year's result of the Planck satellite(Planck ΛCDM). Six simulations are produced in the ΛCDM models with two in the Planck model and the others in the WMAP model. In order for studying the non-linear evolution of the clustering, four simulations were also produced with 8.6 billion particles for the scale-free models of an initial power spectrum P(k) ∝ k^n with n = 0,-1,-1.5 or-2.0. Furthermore, two radical CDM models(XCDM) are simulated with 8.6 billion particles each. Since the XCDM have some of the model parameters distinct from those of the ΛCDM models, they must be unable to match the observations, but are very useful for studying how the clustering properties depend on the model parameters. The Friends-of-Friends(FoF) halos were identified for each snapshot and subhalos were produced by the Hierarchical Branch Tracing(HBT) algorithm. 15Elucidating galaxy assembly bias in SDSS显示文摘We investigate the level of galaxy assembly bias in the Sloan Digital Sky Survey (SDSS) main galaxy sample using ELUCID,a state-of-the-art constrained simulation that accurately reconstructed the initial density perturbations within the SDSS volume.On top of the ELUCID haloes,we develop an extended HOD model that includes the assembly bias of central and satellite galaxies,parameterized as Q_(cen)and Q_(sat),respectively,to predict a suite of one-and two-point observables.In particular,our fiducial constraint employs the probability distribution of the galaxy number counts measured on 8 h^(-1)Mpc scales N_8^(g) and the projected cross-correlation functions of quintiles of galaxies selected by N_(8)^(g) with our entire galaxy sample.We perform extensive tests of the efficacy of our method by fitting the same observables to mock data using both constrained and non-constrained simulations.We discover that in many cases the level of cosmic variance between the two simulations can produce biased constraints that lead to an erroneous detection of galaxy assembly bias if the non-constrained simulation is used.When applying our method to the SDSS data,the ELUCID reconstruction effectively removes an otherwise strong degeneracy between cosmic variance and galaxy assembly bias in SDSS,enabling us to derive an accurate and stringent constraint on the latter.Our fiducial ELUCID constraint,for galaxies above a stellar mass threshold M_(*)=10^(10.2)h^(-2)M_(⊙),is Q_(cen)=-0.09±0.05 and Q_(sat)=0.09±0.10,indicating no evidence for a significant (>2σ) galaxy assembly bias in the local Universe probed by SDSS.Finally,our method provides a promising path to the robust modelling of the galaxy-halo connection within future surveys like DESI and PFS.Andres N.Salcedo Ying Zu Youcai Zhang Huiyuan Wang Xiaohu Yang Yiheng Wu Yipeng Jing Houjun Mo David HWeinberg 2022Science China(Physics,Mechanics & Astronomy)2022,65,10:0

19A giant central red disk galaxy at redshift z=0.76:Challenge to theories of galaxy formation显示文摘We report a giant red central disk galaxy in the XMM-LSS north region. The region is covered with a rich variety of multi-band photometric and spectroscopic observations. Using the photometric data of the Canada-France-Hawaii Telescope Legacy Survey(CFHTLS) and spectroscopic observation of the baryon oscillation spectroscopic survey(BOSS), we find that the galaxy has a stellar mass of ~1011.6 M⊙. The galaxy has a red color and has an old stellar population, and thus its star formation has stopped.With the photometric image data of the Hyper Suprime-Cam(HSC) Subaru Strategic Program, we demonstrate that its luminosity profile is perfectly described by a Sérsic form with n=1.22 consistent with that of a disk. We also analyze its environment based on the VIMOS Public Extragalactic Redshift Survey(VIPERS) photometric catalog, and find that its close neighbors are all less massive, indicating that our observed galaxy is sitting at the center of its dark matter host halo. Existence of such a giant red central disk galaxy seriously challenges the current standard paradigm of galaxy formation, as there is no known physical mechanism to explain the quenching of its star formation. 20Simultaneous integrated dose reduction intensity-modulated radiotherapy effectively reduces cardiac toxicity in limited-stage small cell lung cancer显示文摘Objective:To assess the clinical outcomes and toxicities of once daily(QD)simultaneous dose reduction intensity-modulated radiotherapy(SDR-IMRT-QD;SDR-QD)versus conventional QD IMRT(C-QD)and twice daily(BID)IMRT in patients with limited-stage small cell lung cancer(LS-SCLC).Methods:After propensity score matching(PSM),a retrospective analysis involving 300 patients with LS-SCLC treated using SDR-QD,C-QD,or BID was performed from January 1,2014 to December 31,2019.The prescribed irradiation dose in the SDR-QD cohort was 60 Gy/PGTV and 54 Gy/PTV QD.The radiation dose was 60 Gy for both PGTV and PTV QD in the C-QD cohort.The radiation dose was 45 Gy for both PGTV and PTV in the BID cohort.Toxicities,short-term effects,and survival outcomes were recorded.A meta-analysis on the protective effects of pharmaceuticals for cardiac toxicities induced by anti-tumor therapy was performed.Results:The median overall survival time(MST)in the 3 cohorts were 32.7 months(SDR-QD),26.3 months(C-QD),and 33.6 months(BID);the differences between groups were statistically significant.Lower toxicities and doses to organs-at-risk(OARs)occurred in the SDR-QD and BID cohorts.Further,the cardiac dose dosimetric parameter Vheart40 was negatively associated with survival(r=-0.35,P=0.007).A Vheart40 value of 16.5%was recommended as a cut-off point,which yielded 54.7%sensitivity and 85.7%specificity for predicting negative survival outcomes.The meta-analysis indicated that pharmaceuticals significantly reduced the cardiac toxicities induced by chemotherapy,but not radiotherapy.Conclusions:SDR-QD was shown to have similar toxicities and survival compared with BID,but fewer toxicities and better survival than C-QD.In addition,cardiac dose exposure was negatively associated with survival.Thus,16.5%of the cardiac dosimetric parameter Vheart40 is recommended as the cut-off point,and a Vheart40>16.5%predicts poor survival.Jing Luo Jiawei Song Li Xiao Jiajia Zhang Yipeng Cao Jun Wang Ping Wang Lujun Zhao Ningbo Liu 2023Cancer Biology & Medicine2023,20,6:0
