| 4 | Aquaporin 9 is involved in CRC metastasis through DVL2-dependent Wnt/β-catenin signaling activation显示文摘Background:Aquaporin 9(AQP9)is permeable to water or other small molecules,and plays an important role in various cancers.We previously found that AQP9 was related to the efficacy of chemotherapy in patients with colorectal cancer(CRC).This study aimed to identify the role and regulatory mechanism of AQP9 in CRC metastasis.Methods:The clinical significance of AQP9 was analysed by using bioinformatics and tissue microarray.Transcriptome sequencing,Dual-Luciferase Reporter Assay,Biacore,and co-immunoprecipitation were employed to demonstrate the regulatory mechanism of AQP9 in CRC.The relationship between AQP9 and CRC metastasis was verified in vitro and in vivo by using real-time cell analysis assay,high content screening,and liver metastasis models of nude mice.Results:We found that AQP9 was highly expressed in metastatic CRC.AQP9 overexpression reduced cell roundness and enhanced cell motility in CRC.We further showed that AQP9 interacted with Dishevelled 2(DVL2)via the C-terminal SVIM motif,resulting in DVL2 stabilization and the Wnt/b-catenin pathway activation.Additionally,we identified the E3 ligase neural precursor cell expressed developmentally downregulated 4-like(NEDD4L)as a modulator regulating the ubiquitination and degradation of AQP9.Conclusions:Collectively,our study revealed the important role of AQP9 in regulating DVL2 stabilization and Wnt/β-catenin signaling to promote CRC metastasis.Targeting the NEDD4L–AQP9–DVL2 axis might have therapeutic usefulness in metastatic CRC treatment. | Yiting Liu Qianling Gao Xingzhi Feng Guanxing Chen Xuefei Jiang Daici Chen Zihuan Yang | 2023 | Gastroenterology Report2023,11,1: | 0 |

| 10 | Targeting macrophagic 17β-HSD7 by fenretinide for the treatment of nonalcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD)is the most common chronic liver disease worldwide and macrophage polarization plays an important role in its pathogenesis.However,which molecule regulates macrophage polarization in NAFLD remains unclear.Herein,we showed NAFLD mice exhibited increased 17β-hydroxysteroid dehydrogenase type 7(17β-HSD7)expression in hepatic macrophages concomitantly with elevated M1 polarization.Single-cell RNA sequencing on hepatic non-parenchymal cells isolated from wild-type littermates and macrophage-17β-HSD7 knockout mice fed with high fat diet(HFD)for 6 weeks revealed that lipid metabolism pathways were notably changed.Furthermore,17β-HSD7 deficiency in macrophages attenuated HFD-induced hepatic steatosis,insulin resistance and liver injury.Mechanistically,17β-HSD7 triggered NLRP3 inflammasome activation by increasing free cholesterol content,thereby promoting M1 polarization of macrophages and the secretion of pro-inflammatory cytokines.In addition,to help demonstrate that 17β-HSD7 is a potential drug target for NAFLD,fenretinide was screened out from an FDA-approved drug library based on its 17β-HSD7 dehydrogenase inhibitory activity.Fenretinide dose-dependently abrogated macrophage polarization and pro-inflammatory cytokines production,and subsequently inhibited fat deposition in hepatocytes co-cultured with macrophages.In conclusion,our findings suggest that blockade of 17β-HSD7 signaling by fenretinide would be a drug repurposing strategy for NAFLD treatment. | Xiaoyu Dong Yiting Feng Dongqin Xu Mengya Zhang Xiao Wen Wenhao Zhao Qintong Hu Qinyong Zhang Hui Fu Jie Ping | 2023 | Acta Pharmaceutica Sinica B2023,13,1: | 0 |
| 11 | Mo5+ doping induced interface polarization for improving performance of planar perovskite solar cells显示文摘In this paper,we investigate how interface-induced polarization affects the photovoltaic performance of hybrid perovskite solar cell(PSC)devices.The polarization of the hole transport layer(HTL)is regulated through incorporating metalliclike Mo Ox into PEDOT:PSS.The common Mo O3 doped into PEDOT:PSS is used as a reference,and the device that used PEDOT:PSS-Mo Ox as the HTL shows an enhanced Jsc and FF compared to the reference device.The open-circuit photovoltage decay and impedance spectroscopy measurements indicate that trap-assisted recombination is effectively suppressed at the interface between the hybrid perovskite and the PEDOT:PSS-Mo Ox HTL,while severe trap assisted recombination takes place at the perovskite/PEDOT:PSS and perovskite/PEDOT:PSS-Mo O3 interface.We attribute these experimental findings to the fact that the incorporation of metallic-like Mo5+into PEDOT:PSS enhances the conductivity of HTL and the interface polarization between PEDTOT:PSS layer and perovskite,which helps to induce an interface polarization electric field to facilitate separation of charges and screen the recombination between the traps and free charges. | Yurong Jiang Yue Yang Yiting Liu Shan Yan Yanxing Feng Congxin Xia | 2020 | Journal of Semiconductors2020,41,5: | 0 |
