| 1 | PrP mutants with different numbers of octarepeat sequences are more susceptible to the oxidative stress显示文摘One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important role.However,the detailed mechanism is less clear.In this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12(PrP-PG12)octarepeats were generated and PrP proteins were expressed both in E.coli and in mammalian cells.Protein aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from E.coli after treatment of H2O2.MTT and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type PrP.Statistically significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted octarepeats.Remarkably more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to H2O2.Furthermore,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2O2 at certain extent than the mutated PrPs and mock. | 2 | Survey of spinal cord injury-induced neurogenic bladder studies using the Web of Science显示文摘OBJECTIVE:To identify global trends in research on spinal cord injury-induced neurogenic bladder, through a bibliometric analysis using the Web of Science. DATA RETRIEVAL:We performed a bibliometric analysis of studies on spinal cord injury-induced neurogenic bladder using the Web of Science.Data retrieval was performed using key words'spinal cord injury','spinal injury','neurogenic bladder','neuropathic bladder','neurogenic lower urinary tract dysfunction','neurogenic voiding dysfunction','neurogenic urination disorder'and 'neurogenic vesicourethral dysfunction'. SELECTION CRITERIA:Inclusion criteria:(a)published peer-reviewed articles on spinal cord injury-induced neurogenic bladder indexed in the Web of Science;(b)type of articles:original research articles and reviews;(c)year of publication:no limitation.Exclusion criteria:(a)articles that required manual searching or telephone access;(b)Corrected papers and book chapters. MAIN OUTCOME MEASURES:(1)Annual publication output;(2)distribution according to journals; (3)distribution according to subject areas;(4)distribution according to country;(5)distribution according to institution;and(6)top cited publications. RESULTS:There were 646 research articles addressing spinal cord injury-induced neurogenic bladder in the Web of Science.Research on spinal cord injury-induced neurogenic bladder was found in the Science Citation Index-Expanded as of 1946.The United States,Ireland and Switzerland were the three major countries contributing to studies in spinal cord injury-induced neurogenic bladder in the 1970s.However,in the 1990s,the United States,the United Kingdom,the Netherlands,Germany and Japan published more papers on spinal cord injury-induced neurogenic bladder than Switzerland,and Ireland fell off the top ten countries list.In this century,the United States ranks first in spinal cord injury-induced neurogenic bladder studies,followed by France,the United Kingdom,Germany,Switzerland and Japan.Subject categories including urology, nephrology and clinical neurology,as well as rehabilitation,are represented in spinal cord injury-induced neurogenic bladder studies. | 4 | Magnetic reconnection driven by intense lasers显示文摘Laser-driven magnetic reconnection(LDMR) occurring with self-generated B fields has been experimentally and theoretically studied extensively, where strong B fields of more than megagauss are spontaneously generated in highpower laser–plasma interactions, which are located on the target surface and produced by non-parallel temperature and density gradients of expanding plasmas. For properties of the short-lived and strong B fields in laser plasmas, LDMR opened up a new territory in a parameter regime that has never been exploited before. Here we review the recent results of LDMR taking place in both high and low plasma beta environments. We aim to understand the basic physics processes of magnetic reconnection, such as particle accelerations, scale of the diffusion region, and guide field effects. | 5 | Electron acceleration in a coil target-driven low-βmagnetic reconnection simulation显示文摘Magnetic reconnection driven by a capacitor coil target is an innovative way to investigate low-βmagnetic reconnection in the laboratory,whereβis the ratio of particle thermal pressure to magnetic pressure.Low-βmagnetic reconnection frequently occurs in the Earth’s magnetosphere,where the plasma is characterized byβ≲0.01.In this paper,we analyze electron acceleration during magnetic reconnection and its effects on the electron energy spectrum via particle-in-cell simulations informed by parameters obtained from experiments.We note that magnetic reconnection starts when the current sheet is down to about three electron inertial lengths.From a quantitative comparison of the different mechanisms underlying the electron acceleration in low-βreconnection driven by coil targets,we find that the electron acceleration is dominated by the betatron mechanism,whereas the parallel electric field plays a cooling role and Fermi acceleration is negligible.The accelerated electrons produce a hardened power-law spectrum with a high-energy bump.We find that injecting electrons into the current sheet is likely to be essential for further acceleration.In addition,we perform simulations for both a double-coil co-directional magnetic field and a single-coil one to eliminate the possibility of direct acceleration of electrons beyond thermal energies by the coil current.The squeeze between the two coil currents can only accelerate electrons inefficiently before reconnection.The simulation results provide insights to guide future experimental improvements in low-βmagnetic reconnection driven by capacitor coil targets. | Jiacheng Yu Jiayong Zhong Yongli Ping Weiming An | 2023 | Matter and Radiation at Extremes2023,8,6: | 0 |
| 6 | Identification of potential pathogenic mutations in Chinese children with first branchial cleft anomalies detected by whole-exome sequencing显示文摘Importance:First branchial cleft anomalies(FBCAs)are rare congenital malformations,accounting for<8%of all branchial cleft anomalies.However,little is currently known about the cause of FBCAs at the molecular level.Objective:To identify genomic alterations related to the genetic etiology of FBCAs in Chinese children.Methods:We performed whole-exome sequencing of samples from 10 pediatric patients with FBCAs.Data analysis was carried out using the Burrow-Wheeler Alignment software package,and the dbSNP database for comparisons.Rare variants were further validated by Sanger sequencing.Insertion/deletions(indels)were examined using the Genome Analysis Toolkit.Results:We identified 14 non-synonymous mutations in seven potential FBCA-susceptibility genes(TRAPPC12,NRP2,NPNT,SH3RF2,RHPN1,TENM4,and ARMCX4).We also detected 133 shared small indels in 125 genes.Gene Ontology analysis indicated that most of the identified genes played critical roles in development and differentiation pathways involved in regulating organ development.Interpretation:We characterized the mutational landscape in pathways involved in development and differentiation in Chinese children with FBCA.The results identified potential pathogenic genes and mutations related to FBCA,and provide molecular-level support for the branchial theory of FBCA pathogenesis. | Yeran Yang Wei Liu Yaqiong Jin Min Chen Jie Lu Yongbo Yu Huimin Ren Shujing Han Ping Chu Yongli Guo Jie Zhang Xin Ni | 2021 | Pediatric Investigation2021,5,3: | 0 |
