1Effect of yttrium on the microstructure of a semi-solid A356 Al alloy显示文摘The semi-solid slurry of an A356 Al alloy, which was grain-fined by yttrium, was manufactured by low temperature pouring. The effects of grain-refining on the morphology and the grain size of the primary α phase in the semi-solid A356 Al alloy were researched. The results indicate that the semi-solid A356 Al alloy with particle-like and rosette-like primary α-Al can be prepared by low temperature pouring from a liquid grain-refined A356 alloy. The grain size and particle morphology of primary α-Al in the A356 Al alloy are markedly improved by the addition of 0.5 wt.% Y. 2Cyanobacteria-/cyanotoxin-contaminations and eutrophication status before Wuxi Drinking Water Crisis in Lake Taihu, China显示文摘After the appalling 'Wuxi Drinking Water Crisis', increasing investigations concerning the contaminations of cyanobacterial blooms and their toxins in Lake Taihu have been performed and reported in the last two years. However, information regarding these issues before the crisis in 2007 remained insufficient. To provide some background data for further comparisons, the present study reported our investigations conducted in 2004, associated with the cyanotoxin contaminations as well as the eutrophication status in Lake Taihu. Results from the one-year-study near a drinking water resource for Wuxi City indicated that, unlike the status in recent two years, cyanobacteria and chlorophyta are the co-dominance species throughout the year. The highest toxin concentration (34.2 ng/mL) in water columns occurred in August. In bloom biomass, the peak value of intracellular toxin (0.59 μg/mg DW) was determined in October, which was lag behind that in water column. In addition, MC-RR was the major toxin variant throughout the year. During the study period, nutrients levels of total nitrogen and phosphorus were also recorded monthly. 3Cotton GhPOX1 encoding plant class Ⅲ peroxidase may be responsible for the high level of reactive oxygen species production that is related to cotton fiber elongation显示文摘The accumulation of reactive oxygen species(ROS) is involved in plant cell development.In plant,class Ⅲ peroxidases are heme-containing enzymes encoded by a large multi-gene family participated in the release or consumption of ROS.The specific function of each member of the family is still elusive.Here,we showed that ROS was significantly generated during cotton fiber initiation and elongation,whereas,application of NADPH oxidase inhibitor diphenyleneiodonium(DPI) and peroxidase inhibitor salicylhydroxamic acid(SHAM) to the wild-type cotton ovule culture significantly suppressed fiber growth,respectively.Their inhibitory effects were caused by the reduction of superoxide radical(O2-).Ten GhPOX genes(cDNAs) encoding cotton class Ⅲ peroxidases were isolated,among them eight GhPOX genes were reported for the first time.Microarray analyses indicated that GhPOX1 was the mostly predomi-nantly expressed in fast-elongating cotton fiber cells.Real-time quantitative PCR analysis revealed the transcript level of GhPOX1 was over 400-fold higher in growing fiber cells than in ovules,flowers,roots,stems and leaves.To reveal the role of GhPOX1 in plant devel-opment,its Arabidopsis orthologue atpox13 mutant was demonstrated to be defective in branch root development.Taken together,the data suggest that GhPOX1 plays an important role during fiber cell elongation possibly by mediating production of reactive oxygen spe-cies.Wenqian Mei Yongmei Qin Wenqiang Song Jun Li Yuxian Zhu 2009Journal of Genetics and Genomics2009,36,3:19
4miR-503-3p promotes epithelialemesenchymal transition in breast cancer by directly targeting SMAD2 and E-cadherin显示文摘Although progress in clinical and basic research has significantly increased our understanding of breast cancer, little is known about the molecular mechanism underlying breast cancer metastasis. Identification of effective therapeutic targets to prevent breast cancer metastasis is urgently needed. The function of mi R-503-3p has been investigated in other cancers, but its role in breast cancer remains undefined.Here, we found that mi R-503-3p was overexpressed in breast cancer tissue and plasma compared with adjacent normal breast tissue and with plasma from healthy individuals. Moreover, we identified mi R-503-3p to be an oncogene of breast cancer cell proliferation, migration and invasion. Upregulation of mi R-503-3p in breast cancer cells inhibited expression of epithelialemesenchymal transition(EMT)-related protein SMAD2 and the epithelial marker protein E-cadherin by directly binding to their m RNA30 untranslated region, whereas increased expression of mesenchymal marker proteins, including vimentin and N-cadherin. 7The effects of Xingnao Jieyu capsules on post-stroke depression are similar to those of fluoxetine显示文摘The Xingnao Jieyu capsule has been shown to effectively relieve neurologic impairments and lessen depression. It remains poorly understood whether this capsule can be used to treat post-stroke depression. Thus, in the present study, we established a rat model of post-stroke depression using left middle cerebral artery occlusions in combination of chronic unpredictable stress and solitary housing during development. Experimental rats received intragastric perfusion with 0.82, 0.41, and 0.20 g/kg Xingnao Jieyu capsules separately dissolved in 2 mL distilled water. Fluoxetine served as a positive control. The treatment was conducted over 28 days. Sugar water consumption test, open-field test, real-time fluorescent quantitative PCR and immunohistochemical staining results demonstrated that intragastric perfusion with various doses of Xingnao Jieyu capsules increased sugar water consumption, voluntary behaviors and synaptotagmin mRNA and protein expression in rats with post-stroke depression. These therapeutic effects were similar to those of fluoxetine. 8Tomato SlAN11 regulates flavonoid biosynthesis and seed dormancy by interaction with bHLH proteins but not with MYB proteins显示文摘The flavonoid compounds are important secondary metabolites with versatile human nutritive benefits and fulfill a multitude of functions during plant growth and development.The abundance of different flavonoid compounds are finely tuned with species-specific pattern by a ternary MBW complex,which consists of a MYB,a bHLH,and a WD40 protein,but the essential role of SlAN11,which is a WD40 protein,is not fully understood in tomato until now.In this study,a tomato WD40 protein named as SlAN11 was characterized as an effective transcription regulator to promote plant anthocyanin and seed proanthocyanidin(PA)contents,with late flavonoid biosynthetic genes activated in 35S::SlAN11 transgenic lines,while the dihydroflavonol flow to the accumulation of flavonols or their glycosylated derivatives was reduced by repressing the expression of SlFLS in this SlAN11-overexpressed lines.The above changes were reversed in 35S::SlAN11-RNAi transgenic lines except remained levels of flavonol compounds and SlFLS expression.Interestingly,our data revealed that SlAN11 gene could affect seed dormancy by regulating the expressions of abscisic acid(ABA)signaling-related genes SlABI3 and SlABI5,and the sensitivity to ABA treatment in seed germination is conversely changed by SlAN11-overexpressed or-downregulated lines.Yeast two-hybrid assays demonstrated that SlAN11 interacted with bHLH but not with MYB proteins in the ternary MBW complex,whereas bHLH interacted with MYB in tomato.Our results indicated that low level of anthocyanins in tomato fruits,with low expression of bHLH(SlTT8)and MYB(SlANT1 and SlAN2)genes,remain unchanged upon modification of SlAN11 gene alone in the transgenic lines.These results suggest that the tomato WD40 protein SlAN11,coordinating with bHLH and MYB proteins,plays a crucial role in the fine adjustment of the flavonoid biosynthesis and seed dormancy in tomato.Yongfeng Gao Jikai Liu Yongfu Chen Hai Tang Yang Wang Yongmei He Yongbin Ou Xiaochun Sun Songhu Wang Yinan Yao 2018Horticulture Research2018,5,1:13
9Analysis for retrieval and validation results of FY-3 Total Ozone Unit (TOU)显示文摘Retrieval experiment was made for global total column ozone using the first year measurements of Total Ozone Unit (TOU) on board the second generation polar orbiting meteorological satellite of China, FY-3/A. The retrieval results were analyzed and validated by comparison with AURA/OMI, Meteop/GOME-2 global ozone products and ground-based ozone measurement data. The qualititative comparisons over the globe especially over Antarctica and the Tibetan Plateau show that the spatial and temporal distribution characteristics are consistent with OMI and GOME-2 products. The quantitative comparisons with ground-based measurements and AURA/OMI ozone product were made over 74 stations, the TOU total ozone retrieval has a 3% rms relative error compared with AURA/OMI ozone product and 4.2% rms relative error with ground-based measurements. The maximum difference between satellite retrieval and ground-based measurements was found in the Antarctica ozone hole. 10Pediatric reference intervals in China(PRINCE):design and rationale for a large,multicenter collaborative cross-sectional study显示文摘There is a lack of accurate pediatric reference intervals(RIs) in China, with most commonly used RIs established without consideration of the effect of age and gender. The Pediatric Reference Intervals in China(PRINCE) project aims to establish and verify pediatric RIs for 31 common laboratory measurands.The project will be a large, multicenter cross-sectional study:14,490 healthy children and adolescents aged up to 19 years will be surveyed by 10 children's hospitals and one pediatric department of a university hospital. To evaluate the feasibility and efficiency of the study methods, 602 children were surveyed in the pilot phase of the PRINCE study in April 2017: it found that some measurands were distinctly age dependent and that there were differences between values for males and females. The results of the pilot study affirmed the necessity of the PRINCE project for Chinese pediatrics. The pilot also indicated potential difficulties in the full survey, e.g., difficulties in recruiting children aged under 3 years and insufficient collection of blood samples from infants. 11Therapeutic efficacy of anti-CD19 CAR-T cells in a mouse model of systemic lupus erythematosus显示文摘Dysregulated B-cell activation plays pivotal roles in systemic lupus erythematosus(SLE),which makes B-cell depletion a potential strategy for SLE treatment.The clinical success of anti-CD19 CAR-T cells in treating B-cell malignancies has attracted the attention of researchers.In this study,we aimed to investigate the feasibility of applying anti-CD19 CAR-T cell therapy to SLE treatment in a mouse disease model.We constructed murine anti-CD19 CARs with either CD28 or 4-1BB as the intracellular costimulatory motif and evaluated the therapeutic function of the corresponding CAR-T cells by infusing them into MRL-lpr mice.Furthermore,anti-CD19 CAR-T cells were transferred to MRL-lpr mice before the onset of disease to determine their role in SLE prevention.According to our observations,compared with antibody treatment,the adoptive transfer of our anti-CD19 CAR-T cells showed a more sustained B-cell-depletion effect in MRL-lpr mice.The transfer of syngeneic anti-CD19 CAR-T cells not only prevented disease pathogenesis before the onset of disease symptoms but also displayed therapeutic benefits at a later stage after disease progression.We also tried to optimize the treatment strategy and found that compared with CAR-T cells with the CD28 costimulatory motif,CAR-T cells with the 4-1BB costimulatory motif showed better therapeutic efficiency without cell enrichment.Taken together,these results show that anti-CD19 CAR-T cell therapy was effective in the prevention and treatment of a murine model of SLE,indicating its potential for clinical use in patients.Xuexiao Jin Qin Xu Chengfei Pu Kaixiang Zhu Cheng Lu Yu Jiang Lei Xiao Yongmei Han Linrong Lu 2021Cellular & Molecular Immunology2021,18,8:12
12Distribution and seasonal variation of estrogenic endocrine disrupting compounds,N-nitrosodimethylamine, and N-nitrosodimethylamine formation potential in the Huangpu River, China显示文摘Detection of estrogenic endocrine disrupting compounds(EDCs) and N-nitrosodimethylamine(NDMA) in drinking water has led to rising concerns. There are, however, a paucity of studies on the distribution and seasonal variation of NDMA and NDMA formation potential(NDMA-FP) in natural waters, especially in China. For EDCs, limited studies have investigated the distribution and seasonal variation of estrone(E1), 17β-estradiol(E2), estriol(E3), 17α-ethinylestradiol(EE2), technical-nonylphenols(t-NP), and bisphenol A(BPA) in Shanghai. In this study, water samples were collected from 11 sampling sites along the Huangpu River in 2012. The distribution and seasonal variation of EDCs, NDMA, and NDMA-FP were investigated. The results showed that all of the 11 sampling sites were contaminated by the target compounds. Compared with E2 and EE2, higher E1 and E3 concentrations were usually detected in the Huangpu River. The values of 17β-estradiol equivalents(EEQ) suggest a high possibility of endocrine effects on exposed organisms in the Huangpu River. NDMA-FP and t-NP were the dominant contaminants among the eight target compounds. The detection rates of the target compounds and their concentrations were both higher in dry seasons than in wet seasons. 13The in vitro effect of lipopolysaccharide on proliferation, inflammatory factors and antioxidant enzyme activity in bovine mammary epithelial cells显示文摘Lipopolysaccharide(LPS) was selected as a stimulus to investigate its effect on cell viability and oxidative stress in bovine mammary epithelial cells(BMEC) by detecting the cell relative growth rate(RGR),antioxidant indicators and inflammatory factors. This information was used to provide the theoretical basis for the establishment of a LPS-induced oxidative damage model. The experiment was divided into two parts. The first part used a two-factor experimental design to determine the appropriate incubation time of LPS by detecting the RGR. The third-passage BMEC were divided into 24 groups with six replicates in each group. The first factor was LPS concentration, which was 0(control), 0.1,1.0 and 10.0 μg/mL;the second factor was LPS incubation time(2,4, 6, 8,12 and 24 h). The optimum LPS incubation time was6 h according to the results of the first part of the experiment. The second part of the experiment was conducted using a single-factor experimental design, and the third-passage cells were divided into four groups with six replicates in each group. The cells were incubated with culture medium containing different concentrations of LPS(0 [control], 0.1, 1.0 and 10.0 μg/mL) for 6 h to select the appropriate concentration of LPS to measure the antioxidant indicators and inflammatory factors. The results showed the RGR was significantly reduced as the concentration of LPS and the incubation time increased;the interaction between concentration and incubation time was also significant. The cells treated with0.1 μg/mL of LPS for 6 h had no significant difference in the activities of glutathione peroxidase(GPx) and superoxide dismutase(SOD)(P > 0.05) compared with the cells in the control group. On the contrary,catalase(CAT) activity and malondialdehyde(MDA) content were markedly lower and higher, respectively, in the 0.1 μg/mL LPS-treated group for 6 h compared with the control group(P < 0.05). The activities of GPx, CAT and SOD in the BMEC treated with 1.0 or 10.0 μg/mL of LPS were significantly lower compared with the cells treated with 0.1 μg/mL of LPS and cells in the control group after 6 h of incubation; however, the opposite trend was detected in MDA content. There was no significant(P > 0.05)difference between the 10.0 and 1.0 μg/mL LPS-treated groups. Compared with the control group,interleukin-1, interleukin-6 and nitric oxide concentrations and the activity of inducible nitric oxide synthase in the 0.1 μg/mL LPS-treated group significantly increased(P < 0.0001), but the levels of tumour necrosis factor did not significantly change(P > 0.05). All of observed indicators were higher in the 1.0 and 10.0 μg/mL LPS-treated groups(P < 0.0001) compared with the other groups, but there was no significant(P> 0.05) difference between the 1.0 and 10.0 μg/mL LPS-treated groups. 14Antidepressant effect of Xingnao Jieyu decoction mediated by alleviating neuroinflammation in a rat model of post-stroke depression显示文摘OBJECTIVE: To identify the antidepressant effect of Xingnao Jieyu (XNJY) decoction on a post-stroke depression (PSD) rat model and the underlying molecular mechanism. METHODS: We established a rat PSD model by middle cerebral artery occlusion (MCAO) combined with chronic unpredictable mild stress (CUMS). Healthy SD rats were randomly divided into six groups: sham, PSD, fluoxetine (Flu), and XNJY groups at low, middle, and high doses. The sham group underwent sham operation, while the other groups underwent MCAO+CUMS. The Flu and XNJY decoction groups were intragastrically administered with Flu or different doses of XNJY for 21 consecutive days. Histopathological changes in the cortex and hippocampus were observed by staining with hematoxylin and eosin and Terminal-deoxynucleoitidyl Transferase Mediated Nick End Labeling. Iba1 positive cells were evaluated by immunofluorescence assay. The expressions of tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6), interleukin-1β(IL-1β), 5-hydroxytryptamine (5-HT), and norepinephrine (NE) in the cortex and hippocampus were measured by enzyme linked immunosorbent assay. RESULTS: The PSD group rats had a significant decrease in body weight, consumption of sucrose water, and locomotor activity but an increase in immobility time during a forced swimming test (P < 0.01) compared with sham group. Flu and different doses of XNJY significantly recovered these indices (P < 0.01). XNJY also inhibited neuronal damage and apoptosis in the cortex induced by PSD (P < 0.01). Furthermore, XNJY reduced the number of Iba1 positive cells and the expressions of TNF-α, IL-6, and IL-1β, in addition to recovered the levels of 5-HT and NE in the cortex and hippocampus (P < 0.01). CONCLUSION: The alleviation of neuroinflammation might be an important mechanism of the XNJY decoction against PSD. 15Operating condition influences on PCDD/Fs emissions from sinter pot tests with hot flue gas recycling显示文摘This study was designed to clarify the influence of operating conditions on the formation and emissions of polychlorinated-p-dibenzodioxins and dibenzofurans (PCDD/Fs) from a sintering process with hot flue gas recycling. A pilot scale sinter pot with simulated flue gas recycling was developed, and four key operational parameters, including temperature, oxygen content of the simulated waste flue gas, the coke rate of the sintering mixture, and the quicklime quality, were selected for exploring PCDD/Fs formation. The results showed that the temperature of the recycled flue gas had a major affect on PCDD/Fs formation, and a high temperature could significantly increase their formation during sintering. A clear linear correlation between the temperature of recycling flue gas and PCDD/Fs emission (r = 0.93) was found. PCDD/Fs could be reduced to a certain extent by decreasing the level of oxygen in the recycled flue gas, while sintering quality was unchanged. The coke rate had no significant influence on the formation of PCDD/Fs, but the quality of quicklime used in the sintering mixture could affect not only the amount of PCDD/Fs emissions but also the sintering productivity. Compared with a benchmark sinter pot test, PCDD/Fs emissions markedly decreased with improvements to quicklime quality. However, the reduction in PCDD/Fs emissions realized by using high-quality quicklime was limited by the temperature of the inlet gas. 16Enhanced Expression of miR-425 Promotes Esophageal Squamous Cell Carcinoma Tumorigenesis by Targeting SMAD2显示文摘Esophageal squamous cell carcinoma(ESCC) is one of the most common and deadly cancers in the world. Currently, clinical therapy of ESCC remains limited and the five-year survival rate is poor. The function of miR-425 has been reported in multiple human cancers.However, the tumorigenic role and clinical significance of miR-425 in ESCC remains unclear. We found that enhanced expression of miR-425 in ESCC cell lines not only promoted cell proliferation and colony formation, but also increased cellular metastasis. Furthermore, we revealed the mechanism that miR-425 inhibited the expression of SMAD2 by targeting the second binding site in the 30-untranslated region(30-UTR) in ESCC. This mode of action influenced not only SMAD2 mRNA expression but also protein expression. In addition, we detected the expression of miR-425 in ESCC tissues and plasma. Moreover, we analyzed the relationship between miR-425 expression and SMAD2 m RNA expression. We found that miR-425 was overexpressed in ESCC tissues and the plasma relative to adjacent normal tissues and plasma of healthy individuals. Furthermore, there was a negative correlation between miR-425 expression and SMAD2. 19FY-3 satellite Ultraviolet Total Ozone Unit显示文摘FY-3 satellites are Chinese second-generation polar orbit meteorological satellite series. Ultraviolet Total Ozone Unit (TOU) is one of the main payloads on FY-3 satellite and the first instrument for daily global coverage of total ozone monitoring in China. The main purpose of TOU is to measure the Earth backscatter ultraviolet radiation for retrieving daily global map of atmospheric ozone. TOU will provide the important parameters for environmental monitoring, climate forecasting and global climate changing research. At present, the in-orbit testing of TOU has accomplished and won a consummation, and it will be delivered to National Satellite Meteorological Center for the operational phase of the project. In this paper we introduce the instrument of TOU and its measuring principle, in general. We also analyze the recent working status of the instrument, including the sensitivity, measuring precision of solar irradiance, diffuser degradation and wavelength drift. The inversion results show that TOU can provide good global ozone maps, and a comparison with the OMI total ozone product shows that their RMS deviation is about 5%. 20TRAP1 Shows Clinical Significance and Promotes Cellular Migration and Invasion through STAT3/MMP2 Pathway in Human Esophageal Squamous Cell Cancer显示文摘Tumor necrosis factor receptor-associated protein 1(TRAP1), an important member of mitochondrial heat shock protein 90 family, is involved in multiple biological processes in several types of tumors. However, its pathological role in esophageal squamous cell cancer(ESCC) remains unknown. Herein, we demonstrated the clinical value of TRAP1, and its role in apoptosis and motility in ESCC. The clinical potential of TRAP1 was investigated through immunohistochemical analysis in 328 ESCC samples, which revealed that strong TRAP1 expression was associated with increased risk of lymph node metastasis, while high TRAP1 expression correlated with poor prognosis. Expression of TRAP1 was found to be an independent prognostic factor for patients with ESCC. Additionally, the upregulation of TRAP1 antagonized cisplatin-induced apoptosis while its downregulation sensitized cells to cisplatin-induced apoptosis. As revealed by the transwell assay, TRAP1 overexpression promoted cellular migration and invasion as compared to the control groups. In contrast,silencing of endogenous TRAP1 expression attenuated the ability of migration and invasion. Finally, the molecular mechanism investigated in the present study demonstrated that TRAP1-mediated migration and invasion occurred through STAT3/MMP2 signaling pathway.
