Understanding primary and secondary sources of ambient oxygenated volatile organic compounds in Shenzhen utilizing photochemical age-based parameterization method显示文摘Oxygenated volatile organic compounds(OVOCs) are key intermediates in the atmospheric photooxidation process. To further study the primary and secondary sources of OVOCs,their ambient levels were monitored using a proton-transfer reaction mass spectrometer(PTR-MS) at an urban site in the Pearl River Delta of China. Continuous monitoring campaigns were conducted in the spring, summer, fall, and winter of 2016. Among the six types of OVOC species, the mean concentrations of methanol were the highest in each season(up to 13–20 ppbv), followed by those of acetone, acetaldehyde and acetic acid(approximately 2–4 ppbv), while those of formic acid and methyl ethyl ketone(MEK) were the lowest(approximately 1–2 ppbv). As observed from a diurnal variation chart, the OVOCs observed in Shenzhen may have been affected by numerous factors such as their primary and secondary sources and photochemical consumption. The photochemical age-based parameterization method was used to apportion the sources of ambient OVOCs. Methanol had significant anthropogenic primary sources but negligible anthropogenic secondary sources during all of the seasons. Acetone, MEK and acetic acid were mostly attributed to anthropogenic primary sources during each season with smaller contributions from anthropogenic secondary sources. Acetaldehyde had similar contributions from both anthropogenic secondary and anthropogenic primary sources throughout the year.Meanwhile, anthropogenic primary sources contributed the most to formic acid. | Bo Zhu Yu Han Chuan Wang Xiaofeng Huang Shiyong Xia Yingbo Niu Zixuan Yin Lingyan He | 2019 | Journal of Environmental Sciences2019,31,1: | 13
Experimental investigation and prediction model for UCS loss of unsaturated sandstones under freeze-thaw action显示文摘Sandstone is widely distributed in cold regions and the freeze-thaw deterioration of them has caused many geological engineering disasters. As an important and direct index of frost resistance, the strength loss of sandstones under freeze-thaw actions should be investigated to provide a guidance for the stability assessment of geological engineering. In this research, the UCS(Uniaxial compressive strength)loss of six typical sandstones with different water contents after 0,20,40 and 60 freeze-thaw cycles was measured in the laboratory. The experimental results indicated that the freeze-thaw damage was more serious in sandstones containing high water contents, and the critical saturations for causing a significant loss of UCS under freeze-thaw were 60%-80%for these sandstones.Below this critical saturation, the UCS loss of the sandstones was mainly caused by water weakening rather than freeze-thaw damage.Besides,a developed strength prediction model was proposed by combining the exponential decay function and multiple linear regression method. The initial porosity, elastic modulus and tensile strength of fresh sandstones were a good parameter combination to accurately determine the decay constant in this developed model. The main novelty of this model is that it can accurately and easily estimate the UCS loss of sandstones after any freeze-thaw cycle only using the initial parameters of fresh sandstones, but it does not need to perform freeze-thaw and mechanical strength experiments. This study not only provides an accurate prediction model of UCS under freeze-thaw,but also makes a contribution to better understanding the frost resistance mechanism of sandstones. | Shibing Huang Yingbo He Shilin Yu Chen Cai | 2022 | International Journal of Mining Science and Technology2022,32,1: | 2

Aryl hydrocarbon receptor activation drives polymorphonuclear myeloid-derived suppressor cell response and efficiently attenuates experimental Sjögren’s syndrome显示文摘Myeloid-derived suppressor cells(MDSCs)comprise heterogeneous myeloid cell populations with immunosuppressive capacity that contribute to immune regulation and tolerance induction. We previously reported impaired MDSC function in patients with primary Sjögren’s syndrome(pSS)and mice with experimental SS(ESS).However, the molecular mechanisms underlying MDSC dysfunction remain largely unclear. In this study,we first found that aryl hydrocarbon receptor(AhR)was highly expressed by human and murine polymorphonuclear MDSCs(PMN-MDSCs).Indole-3-propionic acid(IPA),a natural AhR ligand produced from dietary tryptophan, significantly promoted PMN-MDSC differentiation and suppressive function on CD4^(+)T cells. In contrast, feeding a tryptophan-free diet resulted in a decreased PMN-MDSC response,a phenotype that could be reversed by IPA supplementation. The functional importance of PMN-MDSCs was demonstrated in ESS mice by using a cell-depletion approach.Notably,AhR expression was reduced in PMN-MDSCs during ESS development, while AhR antagonism resulted in exacerbated ESS pathology and dysregulated T effector cells, which could be phenocopied by a tryptophan-free diet.Interferon regulatory factor 4(IRF4),a repressive transcription factor, was upregulated in PMN-MDSCs during ESS progression.Chromatin immunoprecipitation analysis revealed that IRF4 could bind to the promoter region of AhR,while IRF4 deficiency markedly enhanced AhR-mediated PMN-MDSC responses.Furthermore, dietary supplementation with IPA markedly ameliorated salivary glandular pathology in ESS mice with restored MDSC immunosuppressive function.Together, our results identify a novel function of AhR in modulating the PMN-MDSC response and demonstrate the therapeutic potential of targeting AhR for the treatment of pSS. | Yanxia Wei Na Peng Chong Deng Futao Zhao Jie Tian Yuan Tang Sulan Yu Yacun Chen Yu Xue Fan Xiao Yingbo Zhou Xiaomei Li Hejian Zou Ke Rui Xiang Lin Liwei Lu | 2022 | Cellular & Molecular Immunology2022,19,12: | 1

Melatonin improves synapse development by PI3K/Akt signaling in a mouse model of autism spectrum disorder显示文摘Autism spectrum disorders are a group of neurodevelopmental disorders involving more than 1100 genes, including Ctnnd2 as a candidate gene.Ctnnd2knockout mice, serving as an animal model of autis m,have been demonstrated to exhibit decreased density of dendritic spines. The role of melatonin,as a neuro hormone capable of effectively alleviating social interaction deficits and regulating the development of dendritic spines,in Ctnnd2 deletion-induced nerve injury remains unclea r.In the present study,we discove red that the deletion of exon 2 of the Ctnnd2 gene was linked to social interaction deficits, spine loss, impaired inhibitory neurons, and suppressed phosphatidylinositol-3-kinase(PI3K)/protein kinase B(Akt) signal pathway in the prefrontal cortex. Our findings demonstrated that the long-term oral administration of melatonin for 28 days effectively alleviated the aforementioned abnormalities in Ctnnd2 gene-knockout mice.Furthermore, the administration of melatonin in the prefro ntal cortex was found to improve synaptic function and activate the PI3K/Akt signal pathway in this region. The pharmacological blockade of the PI3K/Akt signal pathway with a PI3K/Akt inhibitor,wo rtmannin, and melatonin receptor antagonists, luzindole and 4-phenyl-2-propionamidotetralin,prevented the melatonin-induced enhancement of GABAergic synaptic function. These findings suggest that melatonin treatment can ameliorate GABAe rgic synaptic function by activating the PI3K/Akt signal pathway, which may contribute to the improvement of dendritic spine abnormalities in autism spectrum disorders. | Luyi Wang Man Xu Yan Wang Feifei Wang Jing Deng Xiaoya Wang Yu Zhao Ailing Liao Feng Yang Shali Wang Yingbo Li | 2024 | Neural Regeneration Research2024,19,7: | 0

Cultivation of Hippeastrum vittatum Embryogenic Calli and Their Sensitivity to Antibiotics显示文摘In this study,an embryogenic callus induction and proliferation system for Hippeastrum vittatum was established, with the tender bulbs as explants. And then the sensitivity of the explants and calli to kanamycin and hygromycin was evaluated. The results suggested that the embryogenic calli were induced from tender bulbs and cultured in Murashige and Skoog( MS) medium supplemented with 0. 5 mg/L N-phenyl-N'-1,2,3-thiadiazol-5-ylurea( TDZ),1. 0 mg/L 2,4-dichlorophenoxyacetic acid( 2,4-D),and 30 g/L sucrose( pH5. 8) in the dark at 25 ± 1 ℃. Further study of the influence of kanamycin and hygromycin on callus induction and multiplication showed that, the lethal doses of kanamycin and hygromycin to bulb explants were 100 and 30 mg/L,respectively. All explants of H. vittatum died on the medium supplemented with 100 mg/L kanamycin or 30 mg/L hygromycin at the induction stage, and callus proliferation was completely inhibited by 100 mg/L kanamycin or 25 mg/L hygromycin, and all the calli died at last. These results will provide important reference for further studies of transgenic H. vittatum. | Bo YU Min LIU Yu ZHU Aidong ZHONG Lili HUANG Dongxiang ZHU Chunhui LI Genfa ZHU Yingbo SUN | 2018 | Agricultural Biotechnology2018,7,1: | 0

Responses of soil microbial communities to manure and biochar in wheat cultivation of a rice-wheat rotation agroecosystem in East China显示文摘Soil contamination in agroecosystems remains a global environmental problem. Biochar has been suggested as an organic amendment to alleviate soil pollution, sequester carbon(C), and improve soil fertility. However, information on how bacterial and fungal communities in acidic bulk and rhizosphere soils respond to swine manure and its biochar is still lacking. In this study, biochar and swine manure were applied at two rates of 1.5 and 3 t ha-1in a rice-wheat rotation field to assess how soil characteristics, especially p H and chemical element availability, correlate to compositional variations of bacteria and fungi in bulk and rhizosphere soils. Our results showed that high rates of biochar and manure promoted the bacterial richness in bulk and rhizosphere soils by increasing soil pH and reducing soil arsenic(As) and copper(Cu) availability. Compared with soil As and Cu availability, soil p H had opposite effects on beta diversity of both the bacterial and fungal communities. Specifically, biochar and swine manure applications stimulated the bacterial classes Gemmatimonadetes, Deltaproteobacteria, and Gammaproteobacteria by increasing soil pH and decreasing soil available chemical elements. Opposite trends were observed in fungal communities responding to biochar and manure. For example, biochar restrained the fungal class Eurotiomycetes by decreasing soil As and Cu availability, but manure inhibited Leotiomycetes mainly because of an increase in soil pH and a decrease in soil dissolved organic C. These suggest that both bacterial and fungal communities respond significantly to biochar and manure amendments in both bulk and rhizosphere soils, possibly because of their sensitive adaptation to variations in soil environmental factors, such as pH level and chemical element availability. | Jun MENG Wenjin LI Yingbo QIU Zhangtao LI Linze LI Yu LUO Haipeng GUO Yijun YU Shengdao SHAN Huaihai CHEN | 2023 | Pedosphere2023,33,6: | 0
