1Improvement of oxytetracycline production mediated via cooperation of resistance genes in Streptomyces rimosus显示文摘Increasing the self-resistance levels of Streptomyces is an effective strategy to improve the production of antibiotics.To increase the oxytetracycline(OTC) production in Streptomyces rimosus,we investigated the cooperative effect of three co-overexpressing OTC resistance genes:one gene encodes a ribosomal protection protein(otrA) and the other two express efflux proteins(otrB and otrC).Results indicated that combinational overexpression of otrA,otrB,and otrC(MKABC) exerted a synergetic effect.OTC production increased by 179%in the recombinant strain compared with that of the wild-type strain M4018.The resistance level to OTC was increased by approximately two-fold relative to the parental strain,thereby indicating that applying the cooperative effect of self-resistance genes is useful to improve OTC production.Furthermore,the previously identified cluster-situated activator OtcR was overexpressed in MKABC in constructing the recombinant strain MKRABC;such strain can produce OTC of approximately7.49 g L^((-1)),which represents an increase of 19%in comparison with that of the OtcR-overexpressing strain alone.Our work showed that the cooperative overexpression of self-resistance genes is a promising strategy to enhance the antibiotics production in Streptomyces.Shouliang Yin Xuefeng Wang Mingxin Shi Fang Yuan Huizhuan Wang Xiaole Jia Fang Yuan Jinliang Sun Tiejun Liu Keqian Yang Yuxiu Zhang Keqiang Fan Zilong Li 2017Science China(Life Sciences)2017,60,9:10

3The localization of type 2 diabetes susceptibility gene loci in northern Chinese Han families显示文摘We conducted a genome-wide scan, in which 358 well distributed fluorescent dye-labeled microsatellite marker sets were applied in 32 Chinese Han type 2 diabetes families from Northern China to search for the susceptibility gene loci. The data collected from screening all the chromosomes of genome were genotyped by using genescan and genotyping software, then, parametric and non-parametric multipoint test, and affected sib-pair analysis as well, were used to analyze the data. We identified some susceptibility gene loci residing in chromosomes 1,12,18,20, respectively, or precisely, located around D1S214, D1S207, D1S218, D1S235, D12S336, D18S61 and D20S118. 4Development of a new method for analyzing free aluminum ions(Al^(3+)) in seafood using HPLC-ICP-MS显示文摘This paper presents a novel method for simultaneous online examination of free aluminum ions(Al3+)in seafood,using solidphase extraction and high-performance liquid chromatography online with inductively coupled plasma mass spectrometry(SPEHPLC-ICP-MS),without post-column reaction.The optimum conditions for chromatographic separation of Al3+were achieved using an IonPac CS5A analytical column with an IonPac CG5A guard column.The mobile phase consisted of 0.040 mol/L LiOH,0.0060 mol/L 2,6-pyridinedicarboxylic acid,and 0.090 mol/L CH3COOH(pH 4.7).The free Al3+ions in seafood were extracted by shaking with the mobile phase at 70°C for 2 h.SPE was conducted using an Oasis MCX,3cc/60 mg,30μm column,which was activated and equilibrated with 2 mL of methanol and 4 mL of deionized water before use.HCl(0.075 mol/L,2 mL)was used to wash inorganic Al from the SPE column.The standard recoveries of Al3+were all above 89%and the relative standard deviations were all below 5%.The proposed method was successfully used for the examination of Al3+in seafood samples,and the results were similar to those obtained using the static equilibrium method.SHANG DeRong ZHAO YanFang ZHAI YuXiu NING JinSong DING HaiYan SHENG XiaoFeng 2013Chinese Science Bulletin2013,58,35:8

6Assessment and Comparison of in vitro Immunoregulatory Activity of Three Astaxanthin Stereoisomers显示文摘In recent years,the immune-modulatory role of all-trans astaxanthin from different pigment sources has been studied.It was reported that all-trans astaxanthin might exist as three stereoisomers,and the composition of all-trans stereoisomers in natural materials differs from that of synthetic products.However,the different biological effects of various all-trans stereoisomers still remain unclear.In the present study,we evaluated the bioactivity of three astaxanthin stereoisomers,(3S,3'S)-trans-,(3R,3'R)-transand meso-trans-astaxanthin,in regulating cell-mediated immune response using mice lymphocytes and peritoneal exudates cells(PECs) systems.After the treatment with three astaxanthin stereoisomers(20 μmol L-1),the lymphocyte proliferation capacity,neutral red phagocytosis of PECs and natural killer(NK) cell cytotoxic activity were comparatively assessed.The results showed that all three astaxanthin stereoisomers significantly promoted lymphocyte proliferation,phagocytic capacity of PECs,and cytotoxic activity of NK cells.Moreover,the(3S,3'S)-trans-astaxanthin exhibited a much higher response than others.SUN Weihong XING Lihong LIN Hong LENG Kailiang ZHAI Yuxiu LIU Xiaofang 2016Journal of Ocean University of China2016,15,2:7
7An experimental and numerical study of the freak wave speed显示文摘The propagation speed is one of the most important characteristics for describing freak waves. The research of freak wave speed is not only helpful for understanding the generation mechanism and evolution process of freak waves, but also applicable to the prediction. A stable and accurate method is proposed for the calculation of the freak wave speed, in which physical model tests are carried out to measure the motion of the largest wave crest along the wave tank. The linear regression relationship between the spatial position of the largest wave crest and instantaneous moment is established to calculate the speed of totally 248 cases of experimental freak waves and 312 supplementary cases of numerical freak waves. 8Effects of Sini San used alone and in combination with fluoxetine on central and peripheral 5-HT levels in a rat model of depression显示文摘OBJECTIVE: To investigate the effects of Sini San and fluoxetine on the levels of central and peripheral 5-HT in a rat model of depression, and provide new insight into the treatment of depression with integrated Chinese-Western Medicine.METHODS: A rat model of depression was established by chronic mild stress(CMS). Model rats received either Sini San, fluoxetine, a combination of the two drugs, or no drug treatment. Healthy naive rats were used as controls. Open field and sucrose preference tests were used to assess depression-like behavior. ELISA and immunohistochemistry were used to determine central and peripheral levels of 5-HT.RESULTS: In the group with no drug treatment,central 5-HT expression decreased while peripheral5-HT concentrations increased as CMS continued.Four weeks after CMS, Sini San alone was less effective in reducing depression-like behavior than fluoxetine alone or in combination with Sini San,but combined use was more effective than fluoxetine alone. Eight weeks after CMS, Sini San alone or in combination with fluoxetine was more effective in reducing depression-like behavior than fluoxetine alone. Furthermore Sini San and fluoxetine used alone or in combination notably increased central 5-HT expression and decreased peripheral 5-HT levels in the rat model.CONCLUSION:The results of the present study indicate that there is a synergistic action between the two medicines in the treatment of depression. Sini San exhibited a relatively long lag before its effects were observed; however, by eight weeks the Traditional Chinese Medicine appeared at least as effective as fluoxetine. 9Detection of phthalates migration from disposable tablewares to drinking water using hexafluoroisopropanol-induced catanionic surfactant coacervate extraction显示文摘Hexafluoroisopropanol(HFIP)-induced sodium dodecyl sulfate/dodecyltrimethylammonium bromide(SDS/DTAB) catanionic surfactant coacervate extraction method coupled with high performance liquid chromatography(HPLC) was used to detect the migration of phthalates from disposable tablewares to drinking water. The concentration factors are larger than 82 and extraction recoveries over 53% for water samples spiked with 100 or 200 ng/m L phthalates. Limit of detection is in the range of 1.0–2.6 ng/m L.Good linearity with correlation coefficients larger than 0.9985 is obtained in the concentration of20–1500 or 40–3000 ng/m L. Relative recoveries are from 82.4% to 123.6% for water samples spiked with30/60, 250/500, and 1500/3000 ng/m L phthalates, respectively. Relative standard deviations(RSDs) are0.4%–7.4% for intraday precision(n = 5) and 0.6%–7.8% for interday precision(n = 3). 11Expression of hippocampal corticosteroid receptors,as well as corticotrophin-releasing hormone and vasopressin in the hypothalamic paraventricular nucleus,in fornix transected rats显示文摘BACKGROUND:The hippocampus regulates the hypothalamic-pituitary-adrenal axis through negative feedback.The hypothalamic paraventricular nucleus receives neuronal input from the hippocampus via the fornix. OBJECTIVE:To explore whether the negative feedback effect of the hippocampus on the hypothalamic-pituitary-adrenal axis is contributed to the inhibitory effect of mineralocorticoid receptor (MR) and glucocorticoid receptor(GR) in the hippocampus on the paraventricular nucleus via the fornix. DESIGN,TIME AND SETTING:Randomized,controlled,animal experiment.The study was performed at the Department of Histology and Embryology,China Medical University between September 2006 and September 2008. MATERIALS:Rabbit anti-rat anti-MR and rabbit anti-rat anti-GR antibodies were purchased from Santa Cruz Biotechnology,USA.Rabbit anti-rat anti-corticotrophin releasing hormone(CRH) and rabbit anti-rat anti-arginine vasopressin antibodies were purchased from Wuhan Boster. METHODS:A total of 90 male,Wistar rats were randomly divided into model and sham-surgery groups(n=45).Fornix transection was performed in the model group,while the sham-surgery group underwent surgery,but no fornix transection. MAIN OUTCOME MEASURES:Immunohistochemistry was used to examine MR and GR expression in the hippocampus,as well as CRH and anti-arginine vasopressin in the paraventricular nucleus.Western blot was used to measure alterations in MR,GR,and CRH protein expression following fomix transection. RESULTS:Compared with the sham-surgery group,there were no obvious changes in MR and GR expression in the hippocampus,or CRH and anti-arginine vasopressin expression in the paraventricular nucleus within 4 days of fornix transection.However,after 7-10 days,significantly decreased MR and GR expression in the hippocampus,and increased CRH and anti-arginine vasopressin expression in the paraventricular nucleus were observed(P<0.05-0.01). 12Three-dimensional numerical simulation of obfique wave action on vertical walls显示文摘A three-dimensional numerical model based on the potential theory was developed to study the oblique wave action on vertical walls. A source term inside the domain was used to generate incident waves and outgoing waves were dissipated by sponge layers and trans- mitted by radiation boundaries. The finite difference method was used to solve the governing equations and boundary conditions in the regular transformed domain in σ-coordinate. Satisfactory agreements between the numerical predictions and experimental results of wave force were obtained. 14A nanocleaner specifically penetrates the blood-brain barrier at lesions to clean toxic proteins and regulate inflammation in Alzheimer's disease显示文摘Insurmountable blood-brain barrier(BBB) and complex pathological features are the key factors affecting the treatment of Alzheimer's disease(AD).Poor accumulation of drugs in lesion sites and undesired effectiveness of simply reducing Aβ deposition or TAU protein need to be resolved urgently.Herein,a nanocleaner is designed with a rapamycin-loaded ROS-responsive PLGA core and surface modification with KLVFF peptide and acid-cleavable DAG peptide [R@(ox-PLGA)-KcD].DAG can enhance the targeting and internalization effect of nanocleaner towards neurovascular unit endothelial cells in AD lesions,and subsequently detach from nanocleaner in response to acidic microenvironment of endosomes to promote the transcytosis of nanocleaner from endothelial cells into brain parenchyma.Then exposed KLVFF can capture and carry Aβ to microglia,attenuating Aβ-induced neurotoxicity.Strikingly,rapamycin,an autophagy promoter,is rapidly liberated from nanocleaner in the high ROS level of lesions to improve Aβ degradation and normalize inflammatory condition.This design altogether accelerates Aβ degradation and alleviates oxidative stress and excessive inflammatory response.Collectively,our finding offers a strategy to target the AD lesions precisely and multi-pronged therapies for clearing the toxic proteins and modulating lesion microenvironment,to achieve efficient AD therapy.Ting Lei Zhihang Yang Xue Xia Yuxiu Chen Xiaotong Yang Rou Xie Fan Tong Xiaolin Wang Huile Gao 2021Acta Pharmaceutica Sinica B2021,11,12:4
15Groupiness of Sea Waves and Their Characteristic Parameters显示文摘Three methods for studying wave groups and their main parameters for describing wave groupiness are reviewed in this paper. Then they are analyzed and compared combined with field data from both aspects of group height and group length. A method and two parameters that can describe wave groupiness are suggested. The groupiness parameters of sea waves at three field stations are given. The effects of groupiness on both distributions of the wave height and the phase of component waves are investigated. 17Design and evaluation of a novel transdermal patch containing diclofenac and teriflunomide for rheumatoid arthritis therapy显示文摘The aim of this study was to design a compound transdermal patch containing diclofenac(DA)and teriflunomide(TEF)for the treatment of rheumatoid arthritis(RA).The various organic amines salts of DA were prepared and their forming was confirmed using DSC and FTIR.The percutaneous permeation of organic amines salt of DA was investigated in vitro using a two-chamber diffusion cell with excised rabbit skin as transdermal barrier.The formulation of the patch was optimized in terms of the concentration of percutaneous permeation enhancer and the loading dose of drugs.The pharmacokinetic behavior of the optimal formulation was studies in rabbits and the anti-inflammatory and analgesic effects of the optimal patch were evaluated with the adjuvant arthritis model in rats and the pain model in mice,respectively.The result showed that skin penetration of diclofenactriethylamine(DA-TEtA)salt was better than other organic amine salts.Based on previous study of our laboratory,teriflunomide-triethylamine(TEF-TEtA)significantly enhanced the skin permeation of TEF.10%of azone(AZ)was the best enhancer for the two drugs.The optimal patch formulation was composed of 2%of TEF-TEtA,6%of DA-TEtA and 10%of AZ.The cumulative permeated amount of DA-TEtA in vitro was comparable with that of the commercial diclofenac-diethylamine(DA-DEtA)patch.The absolute bioavailability of TEFTEtA was 42%,which could achieve the therapeutic drug levels.In animal study,the optimized compound patch containing DA-TEtA and TEF-TEtA displayed significant antiinflammatory and analgesic effect,which indicated the potential of the compound patch.Yuxiu Zhang Dongmei Cun Xin Kong Liang Fang 2014Asian Journal of Pharmaceutical Sciences2014,9,5:3

19Ultra-low dark count InGaAs/InP single photon avalanche diode显示文摘A low noise InGaAs/InP single photon avalanche diode(SPAD) is demonstrated. The device is based on planar type separate absorption, grading, charge and multiplication structure. Relying on reasonably designed device structure and low-damage Zn diffusion technology, excellent low-noise performance is achieved. Due to its importance, the physical mechanism of dark count is analyzed through performance characterization at different temperatures. 20An efficient focusing model for generation of freak waves显示文摘Based on the Longuet-Higgins wave model theory, the previews studies have shown that freak waves can be generated in finite space and time successfully. However, as to generating high nonlinear freak waves, the simulation results will be unrealistic. Therefore, a modified phase modulation method for simulating high nonlinear freak waves was developed. The surface elevations of some wave components at certain time and place are positive by modulating the corresponding random initial phases, then the total surface elevation at the focused point is enhanced and furthermore a freak wave event is generated. The new method can not only make the freak wave occur at certain time and place, but also make the simulated wave surface time series satisfy statistical properties of the realistic sea state and keep identical with the target wave spectrum.
