1Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20
2Data processing and initial results of Chang'e-3 lunar penetrating radar显示文摘To improve our understanding of the formation and evolution of the Moon,one of the payloads onboard the Chang'e-3(CE-3) rover is Lunar Penetrating Radar(LPR). This investigation is the first attempt to explore the lunar subsurface structure by using ground penetrating radar with high resolution. We have probed the subsurface to a depth of several hundred meters using LPR. In-orbit testing, data processing and the preliminary results are presented. 3Bio-Mobilization of Potassium from Clay Minerals: I. By Ectomycorrhizas显示文摘A pot experiment was carried out to investigate effect of ectomycorrhizal fungi on eucalyptus growth and K bio-mobilization from soils and clay minerals. In the experiment, sands mixed with soil, KCI-saturated vermiculate and mica, respectively, were used to nurse eucalyptus seedlings which were nonectomycorrhized or ectomycorrhized by an ectomycorrhizal fungus Pisolithus tinctorius strain XCI (Pt XC1) isolated from a forest soil from Xichang, Sichuan Province, China, and a worldwide well-known ectomycorrhizal fungus Pisolithus tinctorius strain 2144 (Pt 2144) obtained in Australia. More depletion of HCl-soluble K by mycorrhizas from the soil and minerals than nonmycorrhizas suggested that mycorrhizas had a great ability to mobilize K present in the interlayer and feldspar. Mycorrhizal seedlings depressed greatly K digested with HF-HCIO4 from substrates after consecutive extractions of soils and minerals by water, ammonium cetate and boiling HCl, while nonmycorrhizal seedlings reduced it little if any, showing that the mycorrhizal seedlings could mobilize and then utilize the structural K in mineral lattice. Ectomycorrhizal fungi played a very important role not only in promoting the growth of eucalyptus seedlings but also in mobilizing K in soils and minerals. The infection of Pt XC1 led to a better growth of eucalyptus seedlings and more K accumulation in the seedlings than that of Pt 2144. 4Fast-track surgery in elderly patients undergoing colorectal cancer radical resection显示文摘Purpose:To investigate the efficacy of applying fast-track surgery(FTS)to elderly patients undergoing radical resection of colorectal cancer.Methods:Elderly patients undergoing radical resection of colorectal cancer received FTS(n=31)or routine(n=31)nursing care.The time to first anal exhaust,oral feeding and leaving the bed,duration of postoperative hospital stay and the incidence of complications were compared between the two groups.Results:Patients receiving FTS nursing demonstrated significantly shorter times to exhaust,oral feeding and leaving the bed compared with those receiving routine nursing(all p<0.01).Furthermore,there were significantly fewer incidences of postoperative pulmonary and urinary tract infections and intestinal adhesion in patients receiving FTS nursing(all p<0.05).Conclusion:Application of FTS in elderly patients undergoing radical resection of colorectal cancer facilitates an early rehabilitation after surgery,but places higher demands on nursing care.Hong-Yan Kong Ai-Ling Yang Cai-Ya Ying Zhen-Fang Kong Ling-Ling Yuan Shan-Shan Hu Shun Zhang 2014International Journal of Nursing Sciences2014,1,4:6
5Optimal Power Allocation for Multiuser OFDM-Based Cognitive Heterogeneous Networks显示文摘Heterogeneous network(HetNet) as a promising technology to improve spectrum efficiency and system capacity has been concerned by many scholars, which brings huge challenges for power allocation and interference management in multicell network structures. Although some works have been done for power allocation in heterogeneous femtocell networks, most of them focus centralized schemes for single-cell network under interference constraint of macrocell user. In this paper, a sum-rate maximization based power allocation algorithm is proposed for a downlink cognitive Het Net with one macrocell network and multiple microcell networks. The original power allocation optimization problem with the consideration of cross-tier interference constraint, maximum transmit power constraint of microcell base station and inter-cell interference of microcell networks is converted into a geometric programming problem which can be solved by Lagrange dual method in a distributed way. 6Tuning Superhydrophobic Materials with Negative Surface Energy Domains显示文摘Hydrophobic/superhydrophobic materials with intrinsic water repellence are highly desirable in engineering fields including antiicing in aerocrafts,antidrag and anticorrosion in ships,and antifog and self-cleaning in optical lenses,screen,mirrors,and windows.However,superhydrophobic material should have small surface energy(SE)and a micro/nanosurface structure which can reduce solid-liquid contact significantly.The low SE is generally found in organic materials with inferior mechanical properties that is undesirable in engineering.Intriguingly,previous theoretical calculations have predicted a negative SE forθ-alumina(θ-Al_(2)O_(3)),which inspires us to use it as a superhydrophobic material.Here,we report the experimental evidence of the small/negative SE ofθ-Al_(2)O_(3) and aθ-Al_(2)O_(3)-based superhydrophobic coating prepared by one-step scalable plasma arcing oxidation.The superhydrophobic coating has complete ceramic and desired micro/nanostructure and therefore exhibits excellent aging resistance,wear resistance,corrosion resistance,high-temperature tolerance,and burning resistance.Owing to the rarity of the small/negative SE in inorganic materials,the concept to reduce SE byθ-Al_(2)O_(3) may foster a blowout to develop robust superhydrophobicity by complete inorganic materials.Zhongzhen Wu Liangliang Liu Shunning Li Shunping Ji Pinghu Chen Suihan Cui Zhengyong Ma Yuchang Weng Qian Huang Zhongcan Wu Hao Wu Yuan Lin Ricky KYFu Hai Lin Xiubo Tian Paul KChu Feng Pan 2019Research2019,,1:6
7The penetrating depth analysis of Lunar Penetrating Radar onboard Chang'e-3 rover显示文摘Lunar Penetrating Radar(LPR) has successfully been used to acquire a large amount of scientific data during its in-situ detection. The analysis of penetrating depth can help to determine whether the target is within the effective detection range and contribute to distinguishing useful echoes from noise.First, this study introduces two traditional methods, both based on a radar transmission equation, to calculate the penetrating depth. The only difference between the two methods is that the first method adopts system calibration parameters given in the calibration report and the second one uses high-voltage-off radar data. However, some prior knowledge and assumptions are needed in the radar equation and the accuracy of assumptions will directly influence the final results. Therefore, a new method termed the Correlation Coefficient Method(CCM) is provided in this study, which is only based on radar data without any a priori assumptions. The CCM can obtain the penetrating depth according to the different correlation between reflected echoes and noise. To be exact, there is a strong correlation in the useful reflected echoes and a random correlation in the noise between adjacent data traces. In addition, this method can acquire a variable penetrating depth along the profile of the rover, but only one single depth value can be obtained from traditional methods. Through a simulation, the CCM has been verified as an effective method to obtain penetration depth. The comparisons and analysis of the calculation results of these three methods are also implemented in this study. 9Bio-Mobilization of Potassium from Clay Minerals: Ⅱ. By Ectomycorrhizal Fungi显示文摘Ectomycorrhizal fungi, including Cenococcum geophilum SIV (Cg SIV), and Pisolithus tinctorius 2144 (Pt 2144), 441 (Pt 441) and XC1 (Pt XC1), were cultured in Pachlewski liquid medium with H2KPO4,KCl- saturated vermiculite and mica as K sources, respectively, to investigate the mechanism of K absorption and mobilization by the fungi. Fungal growth rate, K absorption and mobilization varied significantly among the fungal species. Faster growth and greater K accumulation in Pt XC1 than Pt 2 144, Pt 441 and Cg siv were observed. Ectomycorrhizal fungi depressed HCl-soluble K in minerals after successive extractions by water and NH4OAc. Ratio of the total amount of K, including water-, NH4OAc- and HCI-soluble K, lost from substrates to the K accumulated in fungal colonies was less than 60%. These reveal that the ectomycorrhizal fungi could utilize K in interlayer and structural pools, which are usually unavailable for plants in short period. Large differences in the depletion of K in interlayer and structural pools by fungi were observed at fungal harvest. Taking into account the nutrient absorption by ectomycorrhizal fungi in symbionts and the direct contact between hyphae and soils, the fungi species colonized on the root surfaces seemed to be related to the effectiveness of mycorrhizas to utilize K in soils. Ectomycorrhizal fungi differed in the efflux of protons and oxalate. Pt XC1 was observed to have greatest ability to effuse protons and oxalate among the fungi adopted in the experiment. Furthermore, the higher the concentrations of protons and oxalate in the liquid culture solutions, the larger the depletion of K in interlayer and structural pools in minerals by fungi. Protons could replace interlayer K and chelation of oxalate with Fe and Al in crystal lattice could cause weathering of clay minerals. 10Human liver chimeric mouse model based on diphtheria toxin-induced liver injury显示文摘AIM To establish an inducible liver injury mouse model and transplant human hepatocytes to obtain liverhumanized mice.METHODS We crossed three mouse strains,including albumin(Alb)-cre transgenic mice,inducible diphtheria toxin receptor(DTR) transgenic mice and severe combined immune deficient(SCID)-beige mice,to create Alb-cre/DTR/SCID-beige(ADSB) mice,which coincidentally harbor Alb-cre and DTR transgenes and are immunodeficient. As the Cre expression is driven by the liver-specific promoter Alb(encoding ALB),the DTR stop signal flanked by two lox P sites can be deleted in the ADSB mice,resulting in DTR expression in the liver. ADSB mice aged 8-10 wk were injected intraperitoneally(i.p.) with diphtheria toxin(DT) and liver damage was assessed by serum alanine aminotransferase(ALT) level. Two days later,mouse livers were sampled for histological analysis,and human hepatocytes were transplanted into the livers on the same day. A human ALB enzyme-linked immunosorbent assay was performed 7,14,21 and 28 d after transplantation. Human CD68 immunohistochemistry was performed 30 and 90 d after transplantation.RESULTS We crossed Alb-cre with DTR and SCID-beige mice to obtain ADSB mice. These mice were found to have liver damage 4 d after i.p. injection of 2.5 ng/g bodyweight DT. Bodyweight began to decrease on day 2,increased on day 7,and was lowest on day 4(range,10.5%-13.4%). Serum ALT activity began to increase on day 2 and reached a peak value of 289.7 ± 16.2 IU/m L on day 4,then returned to background values on day 7. After transplantation of human liver cells,peripheral blood human ALB level was 1580 ± 454.8 ng/m L(range,750.2-3064.9 ng/m L) after 28 d and Kupffer cells were present in the liver at 30 d in ADSB mice.CONCLUSION Human hepatocytes were successfully repopulated in the livers of ADSB mice. 14Association of sexually transmitted infection with semen quality in men from couples with primary and secondary infertility显示文摘This study aims to compare the prevalence of sexually transmitted infections(STIs)with semen quality in men from couples with primary and secondary infertility.Semen samples were collected from 133 men who requested fertility evaluation.Seminal tract infection with Ureaplasma spp.(UU),Mycoplasma hominis(MH),Mycoplasma genitalium(MG),Chlamydia trachomatis(CT),Neisseria gonorrhoeae(NG),and herpes simplex virus-2(HSV-2)was assessed by PCR-based diagnostic assays.Among all patients,the prevalence of STIs was higher in men from couples with primary infertility than that in men from couples with secondary infertility(39.7%vs 21.7%,P=0.03).The prevalence of UU was 28.8%and 13.3%in men from couples with primary and secondary infertility,respectively.Men from couples with primary infertility were more likely to be positive for UU than men from couples with secondary infertility(P=0.04).Regarding the UU subtype,the prevalence of Ureaplasma urealyticum(Uuu)and Ureaplasma parvum(Uup;including Uup1,Uup3,Uup6,and Uup14)did not differ between the two groups.No associations between the prevalence rates of MH,MG,and CT were found in men from either infertility group.A lower sperm concentration was associated with STI pathogen positivity in men with primary infertility according to the crude model(P=0.04).The crude and adjusted models showed that semen volume(both P=0.03)and semen leukocyte count(both P=0.02)were independently associated with secondary infertility.These findings suggest the importance of classifying the type of infertility during routine diagnosis of seminal tract infections.Shun Bai Yuan Li Mei-Hong Hu Li Wu Li-Jun Shui Xiao-Han Wang Yi-Xun Liu Qiu-Ling Yue Li-Na Yu Kai-Qiang Fu Xian-Hong Tong Xue-Chun Hu Bo Xu 2022Asian Journal of Andrology2022,24,3:2
15Performance evaluation of lunar penetrating radar onboard the rover of CE-3 probe based on results from ground experiments显示文摘Lunar Penetrating Radar(LPR) onboard the rover that is part of the Chang'e-3(CE-3) mission was firstly utilized to obtain in situ measurements about geological structure on the lunar surface and the thickness of the lunar regolith, which are key elements for studying the evolutional history of lunar crust. Because penetration depth and resolution of LPR are related to the scientific objectives of this mission,a series of ground-based experiments using LPR was carried out, and results of the experimental data were obtained in a glacial area located in the northwest region of China. 16Rare earth texture analysis of rectangular extruded Mg alloys and a comparison of different alloying adding ways显示文摘The microstructures and textures of four rectangular extruded Mg alloys, Mg–3Gd, Mg–3Nd, Mg–1Gd–1Nd and Mg–1.5Gd–1.5Nd(all in wt%) were investigated by using optical microscopy(OM), X-ray diffraction(XRD) and electron backscatter diffraction(EBSD). The results demonstrate that the extruded alloys all reveal fully recrystallized microstructure and weak non-basal texture.For textures of the four extruded alloys, it is shown that the basal planes of majority grains tilt away from the sheet plane normal toward the extrusion direction. Compared with two binary alloys, the ternary alloys possess weaker texture strengths. Through orientation distribution function analysis, two types of rare earth textures, {0113} <0223>and {1215} <1213>, were found in the extruded alloys.Texture component {1215} <1213> is relatively weak, but it plays a critical role in the low texture intensities of two ternary alloys. A pairing effect of Gd and Nd elements has an influence on the texture modification. 17Utilization of electroless plating to prepare Cu-coated cotton cloth electrode for flexible Li-ion batteries显示文摘Flexible,lightweight and high conductivity substrates are required for the development of next-generation flexible Li-ion batteries(LIBs).In addition,the interfacial strength between the active material and flexible substrate should be optimized for high-performance LIBs.Herein,cotton cloth(CC)is employed as a flexible substrate,and electroless plating is utilized to deposit a layer of Cu nanoparticles,which enhances the conductivity of CC and acts as a precursor for the active material,i.e.,CuO.The results reveal that the in situ etching and subsequent heat treatment converted Cu film into CuO nanowires on CC substrate.Moreover,carbon nano tubes(CNTs)are introduced to enhance the connectivity of CuO nano wires.Consequently,the CuO/CNT/CC electrode rendered a high areal capacity of>700μAh-cm^(-2)after100 charge/discharge cycles as well as excellent rate capability.The current work presents a novel route to develop desirable substrates for next-generation flexible Li-ion batteries.Mai Shi Shun Wu Zhen-Dong Han Shuang Li Yu-Ting Pan Shuang Yuan Qiang Wang 2021Rare Metals2021,40,2:2
18Epidemiological Survey of Hemoglobinopathies Based on Next-Generation Sequencing Platform in Hunan Province,China显示文摘Objective This study was aimed at investigating the carrier rate of,and molecular variation in,α-andβ-globin gene mutations in Hunan Province.Methods We recruited 25,946 individuals attending premarital screening from 42 districts and counties in all 14 cities of Hunan Province.Hematological screening was performed,and molecular parameters were assessed.Results The overall carrier rate of thalassemia was 7.1%,including 4.83%forα-thalassemia,2.15%forβ-thalassemia,and 0.12%for bothα-andβ-thalassemia.The highest carrier rate of thalassemia was in Yongzhou(14.57%).The most abundant genotype ofα-thalassemia andβ-thalassemia was-α^(3.7)/αα(50.23%)andβ^(IVS-Ⅱ-654)/β^(N)(28.23%),respectively.Fourα-globin mutations[CD108(ACC>AAC),CAP+29(G>C),Hb Agrinio and Hb Cervantes]and sixβ-globin mutations[CAP+8(C>T),IVS-Ⅱ-848(C>T),-56(G>C),beta nt-77(G>C),codon 20/21(-TGGA)and Hb Knossos]had not previously been identified in China.Furthermore,this study provides the first report of the carrier rates of abnormal hemoglobin variants andα-globin triplication in Hunan Province,which were 0.49%and 1.99%,respectively.Conclusion Our study demonstrates the high complexity and diversity of thalassemia gene mutations in the Hunan population.The results should facilitate genetic counselling and the prevention of severe thalassemia in this region.XI Hui LIU Qin XIE Dong Hua ZHOU Xu TANG Wang Lan TANG De Guo ZENG Chun Yan WANG Qiong NIE Xing Hui PENG Jin Ping GAO Xiao Ya WU Hong Liang ZHANG Hao Qing QIU Li FENG Zong Hui WANG Shu Yuan ZHOU Shu Xiang HE Jun ZHOU Shi Hao ZHOU Fa Qun ZHENG Jun Qing WANG Shun Yao CHEN Shi Ping ZHENG Zhi Fen MA Xiao Yuan FANG Jun Qun LIANG Chang Biao WANG Hua 2023Biomedical and Environmental Sciences2023,36,2:1
