| 3 | Determination of copy number for 5S rDNA and centromeric sequence RCS2 in rice by Fiber-FISH显示文摘The copy number of 5S rDNA and centromerie sequence RCS2 was determined by extended DNA fiber based fluorescence in situ hybridization (Fiber-FISH) in rice (Oryza sativa ssp. indica cv. Guangluai No. 4) genome. In order to determine the copy number, it is necessary to know the basepair number that a given length DNA fiber contains under a microscope. Therefore, the length of two DNA frag-ments, in which the basepair number had been already known, was measured. The insert sequence of the tested BAC 38D17 was 136 kb and its extended DNA was 56.4 μm long, 2.41 kb/μm on average, while that of the tested BAC 44B4 was 144.5 kb in total and 55.7 μm long, 2.60 kb/μm on average under the microscope. They were very close to the theoretical value of B-DNA in the Watson-Crick DNA model, which is 2.97 kb/μm. | 5 | FISH analysis of the integration patterns in transgenic rice co-transformed by micro-projectile bombardment显示文摘Using multi-color fluorescence in situ hybridization (FISH), we localized transferred barnase-psl and pHctinG DNA sequences onto chromosomes of two trans-genie rice plants, named Q12 and Q13, both of which were produced by micro-projectile bombardment. In both Q12 and Q13, each detected cell showed 2-3 signal spots on their chromosomes respectively. The signals of both barnase-psl and pHctinG were mostly detected in the adjacent chromosomal sites in which their signals were overlapped and could be recognized by the signal color on the metaphase chromosomes. Fiber FISH further demonstrated that the multiple copies in each of the two DNA sequences distributed adjacently on the DNA fiber in Q13. | 6 | Adaptive watermark generation mechanism based on time series prediction for stream processing显示文摘The data stream processing framework processes the stream data based on event-time to ensure that the request can be responded to in real-time.In reality,streaming data usually arrives out-of-order due to factors such as network delay.The data stream processing framework commonly adopts the watermark mechanism to address the data disorderedness.Watermark is a special kind of data inserted into the data stream with a timestamp,which helps the framework to decide whether the data received is late and thus be discarded.Traditional watermark generation strategies are periodic;they cannot dynamically adjust the watermark distribution to balance the responsiveness and accuracy.This paper proposes an adaptive watermark generation mechanism based on the time series prediction model to address the above limitation.This mechanism dynamically adjusts the frequency and timing of watermark distribution using the disordered data ratio and other lateness properties of the data stream to improve the system responsiveness while ensuring acceptable result accuracy.We implement the proposed mechanism on top of Flink and evaluate it with realworld datasets.The experiment results show that our mechanism is superior to the existing watermark distribution strategies in terms of both system responsiveness and result accuracy. | Yang SONG Yunchun LI Hailong YANG Jun XU Zerong LUAN Wei LI | 2021 | Frontiers of Computer Science2021,15,6: | 0 |
| 7 | Complete Freund’s adjuvant-induced decrement of pruriceptor-mediated suppression of itch显示文摘Peripheral inflammation is always accompanied by a noxious sensation,either pain or itch,providing a protective warning for the occurrence of pathological changes;however,the mechanisms determining whether pain,itch,or both will be elicited under certain inflammatory statuses are still far from clear.Complete Freund’s adjuvant(CFA)contains heat killed and dried Mycobacterium tuberculosis widely used to induce inflammatory pain models,but how CFA treatment affects itch sensation and the possible mechanisms are still unclear.In this study,using itch behavior testing and calcium imaging,we showed that both the behaviors and calcium responses associated with Transient Receptor Potential Vanilloid 1(TRPV1)-mediated histamine-dependent itch and Transient Receptor Potential Ankyrin 1(TRPA1)-mediated histamine-independent itch were significantly suppressed by CFA treatment.Furthermore,to explore the possible cellular mechanisms,high-throughput single-cell RNA sequencing and real-time PCR were used to detect CFA-induced changes of itch-related genes in dorsal root ganglion(DRG)neurons.Our results revealed that although both nociceptive Trpv1^(+)and Trpa1^(+)DRG neurons were increased after CFA treatment,most known pruriceptors,including Hrh1^(+),Mrgpra3^(+),Mrgprd^(+),Htr3a^(+),Htr1f^(+),IL31ra^(+),Osmr^(+),and Lpar3^(+)DRG neurons,were significantly decreased,which may explain that CFA treatment caused itch suppression.This study indicated that itch sensation was affected after CFA treatment,although negatively,and comprehensive but not specific suppression of different pruriceptors was observed after CFA treatment,suggesting that a unified adaptive change of increased pain and decreased itch will occur simultaneously under CFA-induced inflammatory conditions. | Wen Yin Li Liu Yuxi Zhou Yunchun Zhang Dexu Kong Saihong Xu Dan Tang Dan Huang Daxiang Wen Yingfu Jiao Yinghui Fan Po Gao Weifeng Yu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,5: | 0 |

| 9 | Low concentration isopropanol gas sensing properties of Ag nanoparticles decorated In_(2)O_(3) hollow spheres显示文摘In order to detect low concentrations of volatile organic compounds (VOCs) for the early diagnosis of lung cancer, sensors based on hollow spheres of In_(2)O_(3) were prepared through the soft template method. Ag nanoparticle decorated In_(2)O_(3) composites were synthesized via dipping and annealing. The microstructure, phase composition, element distribution, and state of Ag were analyzed by the scanning electron microscopy (SEM), X-ray diffraction (XRD), high-resolution transmission electron microscopy (HRTEM), and X-ray photoelectron spectroscopy (XPS). The gas sensing tests showed that Ag-In_(2)O_(3) sensors had the highest response to isopropanol at 300 ℃. The best response of Ag-In_(2)O_(3) composite sensor was 5.2, which had a significant improvement compared with only In_(2)O_(3). Moreover, the response and recovery time of Ag-In_(2)O_(3) composite sensor was significantly shortened.
