| 2 | Propofol differentially induces unconsciousness and respiratory depression through distinct interactions between GABAa receptor and GABAergic neuron in corresponding nuclei显示文摘Propofol is the most commonly used intravenous anesthetic worldwide.It can induce loss of consciousness prior to the occurrence of severe respiratory suppression,which is also a pharmacodynamic feature of all general anesthetics.However,the neural mechanisms underlying this natural phenomenon are controversial and highly related to patient safety.In the present study,we demonstrated that the pharmacodynamic effects of propofol(50 and 100 μM)on suppression of consciousness-related excitatory postsynaptic currents in the medial prefrontal cortex(mPFC)and centromedian nucleus of the thalamus(CMT)were lower than those in the kernel respiratory rhythmogenesis nucleus pre-Botzinger complex(PrBo).Furthermore,we unexpectedly found that the GABAa receptor β3 subunit is the key target for propofol's action and that it is mutually and exclusively expressed in GABAergic neurons.It is also more abundant in the mPFC and CMT,but mainly co-localized with GABAergic neurons in the PrBo.As a result,the differentiated expression pattern should mediate more neuron suppression through the activation of GABAergic neurons in the mPFC and CMT at low doses of propofol(50 μM).However,PrBo GABAergic neurons were only activated by propofol at a high dose(100 μM).These results highlight the detailed pharmacodynamic effects of propofol on consciousness-related and respiration-related nuclei and provide the distinct interaction mechanism between the β3 subunit and GABAergic neurons in mediating the suppression of consciousness compared to the inhibition of respiration. | Junli Jiang Yingfu Jiao Po Gao Wen Yin Wei Zhou Yunchun Zhang Yanjun Liu Daxiang Wen Yuan Wang Liang Zhou Tian Yu Weifeng Yu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,8: | 11 |

| 4 | Physical location of rice Gm-6,Pi-5(t) genes in O.officinalis with BAC-FISH显示文摘A fluorescence in situ hybridization (FISH) procedure was adopted to physically map two rice BAC clones 24E21 and 4F22 linked to Gm-6 and Pi-5(t) in O. offi-cinalis. FISH results showed that the two BAC clones were located at 4L. The percentage distance from the centromere to the hybridization sites was 72 ± 2.62 for 24E21 and 54± 5.43 for 4F22, the detection rates were 52.70% and 61.2%. The results obtained from the BAC and plasmid clones, RG214 and RZ565 of cultivated rice and O. officinalis were the same. This suggested that the markers, RG214 and RZ565 of cultivated rice and O. officinalis were in the same BAC clones. The homologous sequences of Gm-6 and Pi-5(t) in O. officinalis were positions that signals existed on the 4L. Many signals were observed when no Cot-1 DNA blocked. This also showed that repetitive sequences were some ho-molgous between cultivated rice and O. officinalis. The identification of chromosome 4 of O. officinalis is based on Jena et al. (1994). | 5 | Comparative genome research between maize and rice using genomic in situ hybridization显示文摘Using the genomic DNAs of maize and rice as probes respectively, the homology of maize and rice genomes was assessed by genomic in situ hybridization. When rice genomic DNAs were hybridized to maize, all chromosomes displayed many multiple discrete regions, while each rice chromosome delineated a single consecutive chromosomal region after they were hybridized with maize genomic DNAs. | 7 | Determination of copy number for 5S rDNA and centromeric sequence RCS2 in rice by Fiber-FISH显示文摘The copy number of 5S rDNA and centromerie sequence RCS2 was determined by extended DNA fiber based fluorescence in situ hybridization (Fiber-FISH) in rice (Oryza sativa ssp. indica cv. Guangluai No. 4) genome. In order to determine the copy number, it is necessary to know the basepair number that a given length DNA fiber contains under a microscope. Therefore, the length of two DNA frag-ments, in which the basepair number had been already known, was measured. The insert sequence of the tested BAC 38D17 was 136 kb and its extended DNA was 56.4 μm long, 2.41 kb/μm on average, while that of the tested BAC 44B4 was 144.5 kb in total and 55.7 μm long, 2.60 kb/μm on average under the microscope. They were very close to the theoretical value of B-DNA in the Watson-Crick DNA model, which is 2.97 kb/μm. | 9 | The mechanics and deformation of high temperature steel frame rapidly cooled by spray water in fire fighting显示文摘A finite element is established for analyzing the dynamical mechanics and deformation of steel frame at high temperature when it is rapidly cooled down by spray water in fire fighting. The simulation result shows that remarkable mechanical coupling effects are produced in the process, and the sectional stress in rapid cooling down is found considerably larger than that in heating-up. Meanwhile, the stress and deformation of a beam mainly related to cooling rate and location are much larger than those of a column in rapid cooling. In fire fighting, the structure on the first or second floor was more dangerous than those on other floors in rapid cooling. | 10 | Mouse Karyotype Obtained by Combining DAPI Staining with Image Analysis显示文摘In this study, mitotic metaphase chromosomes in mouse were identified by a new chromosome fluorescence banding technique combining DAPI staining with image analysis. Clear 4’, 6-diamidino-2-phenylindole (DAPI) multiple bands like G-bands could be produced in mouse. The MetaMorph software was then used to generate linescans of pixel intensity for the banded chromosomes from short arm to long arm. These linescans were sufficient not only to identify each individual chromosome but also analyze the physical sites of bands in chromosome. Based on the results, the clear and accurate karyotype of mouse metaphase chromosomes was established. | 11 | Polymorphisms of potential drug resistant molecular markers in Plasmodium vivax from China-Myanmar border during 2008-2017显示文摘Background:Plasmodium vivax remains the predominant species at the China–Myanmar border,imposing a major challenge to the recent gains in regional malaria elimination.To closely supervise the emerging of drug resistance in this area,we surveyed the variations in genes potentially correlated with drug resistance in P.vivax parasite and the possible drug selection with time.Methods:A total of 235 P.vivax samples were collected from patients sufering uncomplicated malaria at Yingjiang,Tengchong,and Longling counties,and Nabang port in China,Yunnan province,and Laiza sub-township in Myanmar,from 2008 to 2017.Five potential drug resistance genes were amplifed utilizing nested-PCR and analyzed,including pvdhfr,pvdhps,pvmdr1,pvcrt-o,and pvk12.The Pearson’s Chi-squared test or Fisher’s exact test were applied to determine the statistical frequency diferences of mutations between categorical data.Results:The pvdhfr F57I/L,S58R,T61M and S117T/N presented in 40.6%,56.7%,40.1%,and 56.0% of the sequenced P.vivax isolates,and these mutations signifcantly decreased with years.The haplotype formed by these quadruple mutations predominated in Yingjiang,Tengchong,Longling and Nabang.While a mutation H99S/R(56.6%)dominated in Laiza and increased with time.In pvdhps,the A383G prevailed in 69.2% of the samples,which remained the most prevalent haplotype.However,a signifcant decrease of its occurrence was also noticed over the time.The S382A/C and A553G existed in 8.4% and 30.8% of the isolates,respectively.In pvmdr1,the mutation Y976F occurred at a low frequency in 5/232(2.2%),while T958M was fxed and F1076L was approaching fxed(72.4%).The K10 insertion was detected at an occurrence of 33.2% in pvcrt-o,whereas there was no signifcant diference among the sites or over the time.No mutation was identifed in pvk12.Conclusions:Mutations related with resistance to antifolate drugs are prevalent in this area,while their frequencies decrease signifcantly with time,suggestive of increased susceptibility of P.vivax parasite to antifolate drugs.Resistance to chloroquine(CQ)is possibly emerging.However,since the molecular mechanisms underneath CQ resistance is yet to be better understood,close supervision of clinical drug efciency and continuous function investigation is urgently needed to alarm drug resistance. | Zhensheng Wang Chunyan Wei Yunchun Pan Zhihua Wang Xin Ji Qianqian Chen Lianhui Zhang Zenglei Wang Heng Wang | 2022 | Infectious Diseases of Poverty2022,11,2: | 2 |
| 12 | Dexmedetomidine maintains blood-brain barrier integrity by inhibiting Drp1-related endothelial mitochondrial dysfunction in ischemic stroke显示文摘Stroke is the second leading cause of death and long-term disability worldwide,which lacks effective treatment.Perioperative stroke is associated with much higher rates of mortality and disability.The neuroprotective role of dexmedetomidine(Dex),a highly selective agonist of alpha2-adrenergic receptor,has been reported in a stroke rat model,and it was found that pretreatment of Dex before stroke could alleviate blood–brain barrier(BBB)breakdown.However,the underlying mechanisms are still unknown.As the brain endothelial cells are the main constituents of BBB and in high demand of energy,mitochondrial function of endothelial cells plays an important role in the maintenance of BBB.Given that dynamin-related protein 1(Drp1)is a protein mediating mitochondrial fission,with mitochondrial fusion that balances mitochondrial morphology and ensures mitochondria function,the present study was designed to investigate the possible role of Drp1 in endothelial cells involved in the neuroprotective effects of Dex in ischemic stroke.Our results showed that preconditioning with Dex reduced infarction volume,alleviated brain water content and BBB damage,and improved neurological scores in middle cerebral artery occlusion rats.Meanwhile,Dex enhanced cell activity and decreased cell apoptosis in oxygen-glucose deprivation human brain microvascular endothelial cells in vitro.These protective effects of Dex were correlated with the mitochondrial morphology integrality of endothelial cells,mediated by increased phosphorylation of serine 637 in Drp1,and could be reversed byα2-adrenergic receptor antagonist Yohimbine and AMP-activated protein kinase inhibitor Compound C.These findings suggest new molecular pathways involved in the neuroprotective effects of Dex in ischemic stroke.As Dex is routinely used as a sedative drug clinically,our findings provide molecular evidence that it has perioperative neuroprotection from ischemic stroke. | Wei Zhou Yunchun Zhang Yingfu Jiao Wen Yin Haiping Dong Saihong Xu Dan Tang Junli Jiang Jianlin Shao Zhenhong Wang Weifeng Yu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,9: | 2 |

| 19 | Species Diversity Based on Vertical Structure as Indicators of Artificial Restoration for Coniferous Forests in Southwest China显示文摘Five indices of species richness, species diversity and species evenness were used to assess α diversity of four types of coniferous forests undergone different artificial regeneration time. The study was based on the vertical structure of forests, involving growth-forms of total plants as well as vertical layers of woody plants. The results showed that, except for the fourth layer of woody plants, the vertical structure of the four forests exhibited significant difference in terms of their species richness, species diversity and species evenness, suggesting that α species diversity index provided more detailed information and was a better index reflecting the regeneration processes in this region.
