| 3 | Dissection of cellular and molecular mechanisms of aristolochic acid-induced hepatotoxicity via single-cell transcriptomics显示文摘Background:Aristolochic acids(AAs),a class of carcinogenic and mutagenic natural products from Aristolochia and Asarum plants,are well-known to be responsible for inducing nephrotoxicity and urothelial carcinoma.Recently,accumulating evidence suggests that exposure to AAs could also induce hepatotoxicity and even hepatocellular carcinoma,though the mechanisms are poorly defined.Methods:Here,we aimed to dissect the underlying cellular and molecular mechanisms of aristolochic acid I(AAI)-induced hepatotoxicity by using advanced single-cell RNA sequencing(scRNA-seq)and proteomics techniques.We established the first single-cell atlas of mouse livers in response to AAI.Results:In hepatocytes,our results indicated that AAI activated NF-κB and STAT3 signaling pathways,which may contribute to the inflammatory response and apoptosis.In liver sinusoidal endothelial cells(LSECs),AAI activated multiple oxidative stress and inflammatory associated signaling pathways and induced apoptosis.Importantly,AAI induced infiltration of cytotoxic T cells and activation of proinflammatory macrophage and neutrophil cells in the liver to produce inflammatory cytokines to aggravate inflammation.Conclusions:Collectively,our study provides novel knowledge of AAs-induced molecular characteristics of hepatotoxicity at a singlecell level and suggests future treatment options for AAs associated hepatotoxicity. | Piao Luo Jiayun Chen Qian Zhang Fei Xia Chen Wang Yunmeng Bai Huan Tang Dandan Liu Liwei Gu Qingfeng Du Wei Xiao Chuanbin Yang Jigang Wang | 2022 | Precision Clinical Medicine2022,5,4: | 0 |
| 4 | Growth differentiation factor 11 promotes macrophage polarization towards M2 to attenuate myocardial infarction via inhibiting Notch1 signaling pathway显示文摘Background:Myocardial infarctions(MI)is a major threat to human health especially in people exposed to cold environment.The polarization of macrophages towards different functional phenotypes(M1 macrophages and M2 macrophages)is closely related to MI repairment.The growth differentiation factor 11(GDF11)has been reported to play a momentous role in inflammatory associated diseases.In this study,we examined the regulatory role of GDF11 in macrophage polarization and elucidated the underlying mechanisms in MI.Methods:In vivo,the mice model of MI was induced by permanent ligation of the left anterior descending coronary artery(LAD),and mice were randomly divided into the sham group,MI group,and MI+GDF11 group.The protective effect of GDF11 on myocardial infarction and its effect on macrophage polarization were verified by echocardiography,triphenyl tetrazolium chloride staining and immunofluorescence staining of heart tissue.In vitro,based on the RAW264.7 cell line,the effect of GDF11 in promoting macrophage polarization toward the M2 type by inhibiting the Notch1 Signaling pathway was validated by qRT-PCR,Western blot,and flow cytometry.Results:We found that GDF11 was significantly downregulated in the cardiac tissue of MI mice.And GDF11 supplementation can improve the cardiac function.Moreover,GDF11 could reduce the proportion of M1 macrophages and increase the accumulation of M2 macrophages in the heart tissue of MI mice.Furthermore,the cardioprotective effect of GDF11 on MI mice was weakened after macrophage clearance.At the cellular level,application of GDF11 could inhibit the expression of M1 macrophage(classically activated macrophage)markers iNOS,interleukin(IL)-1β,and IL-6 in a dose-dependent manner.In contrast,GDF11 significantly increased the level of M2 macrophage markers including IL-10,CD206,arginase 1(Arg1),and vascular endothelial growth factor(VEGF).Interestingly,GDF11 could promote M1 macrophages polarizing to M2 macrophages.At the molecular level,GDF11 significantly down-regulated the Notch1 signaling pathway,the activation of which has been demonstrated to promote M1 polarization in macrophages.Conclusions:GDF11 promoted macrophage polarization towards M2 to attenuate myocardial infarction via inhibiting Notch1 signaling pathway. | Manyu Gong Xuewen Yang Yaqi Wang Yanying Wang Dongping Liu Haodong Li Yunmeng Qu Xiyang Zhang Yanwei Zhang Han Sun Lei Jiao Ying Zhang | 2023 | Frigid Zone Medicine2023,3,1: | 0 |
| 5 | Study on the water-tree ageing characteristics of polyethylene/organic montmorillonite and crosslinked polyethylene/organic montmorillonite nanocomposites显示文摘The dispersion state of nanoparticles affects the properties of composites,and the crosslinking process of polymers affects the dispersion state of nanoparticles.In order to investigate the effects of crosslinking behaviour on the water-tree ageing characteristics of nanocomposite dielectrics at the molecular level,polyethylene/organic-montmorillonite(PE/organic montmorillonite(OMMT))and crosslinked polyethylene/organic-montmorillonite(XLPE/OMMT)nanocomposites were prepared by the melt blending method,after which the accelerated water-tree ageing experiments were conducted using a needle electrode.Fourier transform infrared spectroscopy,differential scanning calorimetry,successive self-nucleation annealing and scanning electron microscope(SEM)tests were conducted to characterise the properties of the samples before and after watertree ageing.The results indicate that the initiation probability and the length of water trees in XLPE/OMMT are less than those in PE/OMMT.The carbonyl index in the watertree area of the two nanocomposites increases,indicating that the water-tree ageing is the result of electrochemical degradation.The crystallisability of the water-tree area degrades,decreasing the crystallinity and the lamellar thickness.The growth of water trees destroys molecular chains and crystal structures,which results in local cracks.There are many holes for the water tree growth in the PE/OMMT specimen.However,the crosslinked meshwork not only restricts the movement of macromolecular chains but also increases the stability of the lamellar barrier effect of OMMT.The combination of the two effects significantly improves the water-tree resistance of the XLPE/OMMT nanocomposite. | Xiufeng Li Yunmeng Yang Qiang Wang Yuan Gao Wei Zhang Zhengdi Liu | 2023 | High Voltage2023,8,2: | 0 |
