1Comparative transcriptome analysis uncovers the regulatory functions of long noncoding RNAs in fruit development and color changes of Fragaria pentaphylla显示文摘To investigate the molecular mechanism underlying fruit development and color change,comparative transcriptome analysis was employed to generate transcriptome profiles of two typical wild varieties of Fragaria pentaphylla at three fruit developmental stages(green fruit stage,turning stage,and ripe fruit stage).We identified 25,699 long noncoding RNAs(lncRNAs)derived from 25,107 loci in the F.pentaphylla fruit transcriptome,which showed distinct stage-and genotype-specific expression patterns.Time course analysis detected a large number of differentially expressed protein-coding genes and lncRNAs associated with fruit development and ripening in both of the F.pentaphylla varieties.The target genes downregulated in the late stages were enriched in terms of photosynthesis and cell wall organization or biogenesis,suggesting that lncRNAs may act as negative regulators to suppress photosynthesis and cell wall organization or biogenesis during fruit development and ripening of F.pentaphylla.Pairwise comparisons of two varieties at three developmental stages identified 365 differentially expressed lncRNAs in total.Functional annotation of target genes suggested that lncRNAs in F.pentaphylla may play roles in fruit color formation by regulating the expression of structural genes or regulatory factors.Construction of the regulatory network further revealed that the low expression of Fra a and CHS may be the main cause of colorless fruit in F.pentaphylla.Lijun Bai Qing Chen Leiyu Jiang Yuanxiu Lin Yuntian Ye Peng Liu Xiaorong Wang Haoru Tang 2019Horticulture Research2019,6,1:5
2Effect of texture on deformation behavior of heterogeneous Mg-13Gd alloy with strength-ductility synergy显示文摘Heterostructure metals as a new class of materials can effectively break the traditional strength–ductility trade-off dilemma. In this study, the extruded sheet with the small extrusion ratio(ER) of 3.9(ER3.9)presented a heterogeneous lamella structure(HLS) and texture, where the fine dynamical recrystallized(DRXed) grains formed a random texture and coarse un-DRXed grains exhibited a strong basal texture.The ER3.9 sample presented an excellent combination of strength and ductility. The texture strengthening in coarse grains and hetero-deformation induced(HDI) strengthening contributed to the enhanced strength of the ER3.9 sample besides grain refinement. The improving ductility mainly stems from the weakened texture in fine grains. Interestingly, in coarse grains, the strong basal texture, the occurrence of cross slip, low stacking fault energy(SFE), and dislocation pinned by precipitates weaken the HDI hardening effect. While the traditional dislocation hardening mainly generated by fine grains dominates overall strain hardening. Meanwhile, the activation of non-basal slips, especially pyramidal slip,and the generation of cross slips in fine grains benefit for coordinating plastic deformation. The ability for coordinate plastic deformation in fine grains is higher than that of coarse grains, which was confirmed by the digital image correlation technology. 3MicroRNA-7-5p Inhibits Migration,Invasion and Metastasis of Intrahepatic Cholangiocarcinoma by Inhibiting MyD88显示文摘Background and Aims:Intrahepatic cholangiocarcinoma(ICC)is a malignant tumor derived from intrahepatic bile duct epithelial cells.Accumulating studies report that microRNAs are widely involved in tumor migration and metastasis by regulation of target genes.miR-7-5p has been confirmed to inhibit tumor metastasis and to be related to prognosis for several malignant tumors.Our study investigated the underlying functions of miR-7-5p in ICC.Methods:The expression of miR-7-5p in ICC tissues but also in ICC cell lines was analyzed by real-time PCR.By analyzing the relationship between the clinicopathological parameters of 60 ICC patients and the expression level of miR-7-5p,the effect of miR-7-5p on the prognosis was clarified.After transfected with miR-7-5p mimics or miR-7-5p inhibitor,cell counting kit-8 assay was applied to evaluate the cells proliferation,flow cytometry was applied to analyze the cells apoptosis,wound healing assay and transwell chamber assay were applied to analyze the cell invasion and migration.A luciferase reporter assay was identified the relationship of miR-7-5p and myeloid differentiation factor 88(MyD88).Western blotting was used to analyze the proteins expression.And immunochemistry was performed to determine the expression of MYD88 in ICC tissues.Results:Our data showed the expression of miR-7-5p was down-regulated not only in ICC tissues but also in ICC cell lines compared with normal controls.Low expression of miR-7-5p was notably associated with poor prognosis in ICC patients.miR-7-5p negatively regulated cell proliferation,migration,invasion and apoptosis in ICC cells.We further verified that MyD88 was a novel target of miR-7-5p and was significantly overexpressed in ICC tissues.Overexpression of MyD88 counteracted the effects of miR-7-5p in ICC cells.Conclusions:The present findings suggest that miR-7-5p plays a pivotal role in ICC invasion by regulating MyD88.Ampliative insight into the key factors of ICC invasion may result in the development of new treatment options for ICC.Yi Tang Zhenyong Tang Jianrong Yang Tianqi Liu Yuntian Tang 2021Journal of Clinical and Translational Hepatology2021,9,6:3
4Multi-objective vehicle rebalancing for ridehailing system using a reinforcement learning approach显示文摘The problem of designing a rebalancing algorithm for a large-scale ridehailing system with asymmetric(unbalanced)demand is considered here.We pose the rebalancing problem within a semi Markov decision problem(SMDP)framework with closed queues of vehicles serving stationary,but asymmetric demand,over a large city with multiple stations(representing neighborhoods).We assume that the passengers queue up at every station until they are matched with a vehicle.The goal of the SMDP is to minimize a convex combination of the waiting time of the passengers and the total empty vehicle miles traveled.The resulting SMDP appears to be difficult to solve yielding closed-form expression for the optimal rebalancing strategy.Consequently,we use a deep reinforcement learning algorithm to determine the approximately optimal solution to the SMDP.We show through extensive Monte Carlo simulations that the trained policy outperforms other well-known state-dependent rebalancing strategies.Yuntian Deng Hao Chen Shiping Shao Jiacheng Tang Jianzong Pi Abhishek Gupta 2022Journal of Management Science and Engineering2022,7,2:0
