| 4 | MicroRNA-7-5p Inhibits Migration,Invasion and Metastasis of Intrahepatic Cholangiocarcinoma by Inhibiting MyD88显示文摘Background and Aims:Intrahepatic cholangiocarcinoma(ICC)is a malignant tumor derived from intrahepatic bile duct epithelial cells.Accumulating studies report that microRNAs are widely involved in tumor migration and metastasis by regulation of target genes.miR-7-5p has been confirmed to inhibit tumor metastasis and to be related to prognosis for several malignant tumors.Our study investigated the underlying functions of miR-7-5p in ICC.Methods:The expression of miR-7-5p in ICC tissues but also in ICC cell lines was analyzed by real-time PCR.By analyzing the relationship between the clinicopathological parameters of 60 ICC patients and the expression level of miR-7-5p,the effect of miR-7-5p on the prognosis was clarified.After transfected with miR-7-5p mimics or miR-7-5p inhibitor,cell counting kit-8 assay was applied to evaluate the cells proliferation,flow cytometry was applied to analyze the cells apoptosis,wound healing assay and transwell chamber assay were applied to analyze the cell invasion and migration.A luciferase reporter assay was identified the relationship of miR-7-5p and myeloid differentiation factor 88(MyD88).Western blotting was used to analyze the proteins expression.And immunochemistry was performed to determine the expression of MYD88 in ICC tissues.Results:Our data showed the expression of miR-7-5p was down-regulated not only in ICC tissues but also in ICC cell lines compared with normal controls.Low expression of miR-7-5p was notably associated with poor prognosis in ICC patients.miR-7-5p negatively regulated cell proliferation,migration,invasion and apoptosis in ICC cells.We further verified that MyD88 was a novel target of miR-7-5p and was significantly overexpressed in ICC tissues.Overexpression of MyD88 counteracted the effects of miR-7-5p in ICC cells.Conclusions:The present findings suggest that miR-7-5p plays a pivotal role in ICC invasion by regulating MyD88.Ampliative insight into the key factors of ICC invasion may result in the development of new treatment options for ICC. | Yi Tang Zhenyong Tang Jianrong Yang Tianqi Liu Yuntian Tang | 2021 | Journal of Clinical and Translational Hepatology2021,9,6: | 3 |

| 6 | Active dry yeast supplementation benefits ruminal fermentation,bacterial community,blood immunoglobulins,and growth performance in young dairy goats,but not for intermittent supplementation显示文摘This study evaluated the effects of active dry yeast(ADY)supplementation and supplementation stra-tegies on ruminal fermentation,bacterial community,blood metabolites,and growth performance in young dairy goats.Sixty young female Guanzhong dairy goats of similar age(4.00±0.50 months)and BW(19.65±0.41 kg)were randomly divided into 3 groups(n=20):(1)basal diet group(CON);(2)basal diet continuously supplemented with 3.0 g/goat per day commercial ADY(a proprietary strain of Saccharomyces cerevisiae with 5.0×10^(9)cfu/g)group(CSY);(3)basal diet with intermittently supple-mented ADY group(ISY;5 d supplementation with ADY at 4.5 g/goat per day following 5 d of no sup-plementation).The experiment lasted 67 d with the first 7 d as an adaptive period.Rumen fluid and blood samples were collected bi-weekly.Data were analyzed using the MIXED procedure combined with the SLICE option in SAS.Specific orthogonal contrasts of ADY vs.CON and CSY vs.ISY were also analyzed.During the experimental period,ADY supplementation resulted in greater DMI(P=0.03),ruminal ac-etate proportion(P<0.01)and acetylesterase activity(P=0.01),and blood contents of glucose(P=0.01)and IgM(P=0.02)and tended to have greater ADG(P=0.05)and paunch girth(P=0.06)than the CON,despite the propionate proportion(P=0.03)and contents of total protein(P=0.04)and IgA(P=0.03)being lower.The lower ruminal NH_(3)-N(P<0.01)and blood urea nitrogen(P=0.07)contents indicated greater nitrogen utilization with ADY supplementation.ADY supplementation showed persistent effects after it was stopped because the BW at 12 months of age(P=0.03)and birth weight of lambs(P=0.02)were greater than the CON.However,the ISY did not show those benefits and had significantly lower relative abundances of fiber-degrading related bacteria than the CSY.In conclusion,ADY supplementa-tion,especially continuously supplemented,may enhance ADG and ADG:DMI ratio by improving DMI,ruminal cellulolytic bacteria abundance and enzyme activity,nitrogen utilization,and immune status.These findings provide a theoretical basis for the rational application of ADY and have important practical implications for the design of nutritional strategies in growing dairy goats. | Jun Zhang Yuntian Yang Xinjian Lei Yannan Wang Yanhua Li Zhiqiang Yang Junhu Yao | 2023 | Animal Nutrition2023,,2: | 1 |

| 9 | Protonic recognition and assembly for the creation of porous Bronsted acid catalysts with enhanced catalytic efficiency显示文摘Due to the high local concentration of substrates in confined space, porous solid Bronsted acids have been extensively explored for efficient acid-catalyzed reaction. However, the porous structures with strong Bronsted acids lack long-term stability due to chemical hydrolysis. Moreover, the products inhibition effect in confined rigid cavities severely obstructs subsequent catalysis. Here, tubular Bronsted acid catalyst with unique recognition of protons was presented by self-assembly of p H-responsive aromatic amphiphiles. The responsive assembly could mechanically transfer hydrogen ions from low-concentration acidic solution into tubular defined pores, thereby producing effective catalytic activity for Mannich reactions in mildly acidic solution. Notably, the tubular catalyst unfolded into flat sheets upon addition of triethylamine for efficient release of products, which could be recovered by subsequent acidification and the catalytic activity still remained. RT-PCR, immunohistochemistry and Western blot were employed to detect the mRNA expression and protein expression of MMP20. Compared with the corresponding normal esophagus tissues, results showed the common trend of the dramatical overexpression of MMP20 in the esophageal cancer tissues. The analysis of MMP20 expression in different clinical early stages of esophageal cancer by RT-PCR indicated that MMP20 was much more overexpressed in the esophageal cancer with invasion than in the early stages. This suggested that MMP20 might play an important role in the progression of esophageal cancer. | 12 | Transient synthesis of carbon-supported high-entropy alloy sulfide nanoparticles via flash Joule heating for efficient electrocatalytic hydrogen evolution显示文摘High entropy alloys(HEA)are frequently employed as catalysts in electrocatalytic hydrogen evolution.However,the traditional high entropy alloy synthesis methods are time-consuming,energy-intensive,and environmentally polluting,which limits their application in the hydrogen evolution reaction(HER).This study leveraged the capabilities of flash Joule heating(FJH)to synthesize carbon-supported high-entropy alloy sulfide nanoparticles(CC-S-HEA)on carbon cloth(CC)with good self-standing properties within 300 ms.The carbon thermal shock generated by the Joule heating could pyrolyze the sulfur source into gas,resulting in numerous pore structures and defects on CC,forming an S-doped carbon substrate(CC-S).Then the S atoms were used to stably anchor the metal atoms on CC-S to form high-density uniformly dispersed HEA particles.The electrochemical test results demonstrated that CC-S-HEA prepared at 60 V flash voltage had HER performance comparable to Pt/C.The density functional theory(DFT)calculation indicated that the S atoms on CC-S accelerated the electron transfer between the carbon substrate and HEA particles.Moreover,the unique electronic structure of CC-S-HEA was beneficial to H*adsorption and promoted catalytic kinetics.The simplicity and versatility of FJH synthesis are of great significance for optimizing the synthesis of HEA and improving the quality of HEA products,which provides a broad application prospect for the synthesis of nanocatalysts with efficient HER performance. | Yuntian Liao Rongtao Zhu Wenjun Zhang Haiyang Zhu Yang Sun Jiale Chen Zhenhua Dong Ronghuan Lv | 2024 | Nano Research2024,17,4: | 0 |
