Identifying normal embryos from reciprocal translocation carriers by whole chromosome haplotyping显示文摘Reciprocal translocation is a chromosomal structural abnormality that arises when two non-homologous chromosomes rearrange and attach with each other, an incidence that occurs in about 1/500to 1/625 newborns (Mackie and Scriven, 2002). This event typically does not lead to any significant loss of genetic material, thus reciprocal translocation carriers do not exhibit any severe | Zhiqiang Yan Yuqian Wang Yanli Nie Xu Zhi Xiaohui Zhu Meng Qin Shuo Guan Yixin Ren Ying Kuo Di Chang Wei Chen Peng Yuan Liying Yan Jie Qiao | 2018 | Journal of Genetics and Genomics2018,45,9: | 4
Downregulation of MEIS1 mediated by ELFN1-AS1/EZH2/DNMT3a axis promotes tumorigenesis and oxaliplatin resistance in colorectal cancer显示文摘Oxaliplatin is widely used in the frontline treatment of colorectal cancer(CRC),but an estimated 50%of patients will eventually stop responding to treatment due to acquired resistance. This study revealed that diminished MEIS1 expression was detected in CRC and harmed the survival of CRC patients.MEIS1 impaired CRC cell viabilities and tumor growth in mice and enhanced CRC cell sensitivity to oxaliplatin by preventing DNA damage repair.Mechanistically, oxaliplatin resistance following MEIS1 suppression was critically dependent on enhanced FEN1 expression.Subsequently,we confirmed that EZH2-DNMT3a was assisted by lncRNA ELFN1-AS1 in locating the promoter of MEIS1 to suppress MEIS1 transcription epigenetically.Based on the above, therapeutics targeting the role of MEIS1 in oxaliplatin resistance were developed and our results suggested that the combination of oxaliplatin with either ELFN1-AS1 ASO or EZH2 inhibitor GSK126 could largely suppress tumor growth and reverse oxaliplatin resistance. This study highlights the potential of therapeutics targeting ELFN1-AS1 and EZH2 in cell survival and oxaliplatin resistance, based on their controlling of MEIS1 expression, which deserve further verification as a prospective therapeutic strategy. | Yimin Li Yaqi Gan Jiaxin Liu Juanni Li Zhengwei Zhou Ruotong Tian Ruizheng Sun Jiaqi Liu Qing Xiao Yuanyuan Li Pengyan Lu Yulong Peng Yuqian Peng Guang Shu Gang Yin | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 2
The effect of whey protein on viral infection and replication of SARS-CoV-2 and pangolin coronavirus in vitro显示文摘Dear Editor,Since severe acute respiratory syndrome coronavirus 2(SARSCoV-2)RNA has been detected in human breastmilk, infants’safety with breastmilk feeding is of great concern for women with coronavirus disease 2019(COVID-19).1 It is known that milk has antiviral properties.2 However, little is known about the antiviral property of human breastmilk to SARS-CoV-2 and its related pangolin coronavirus(GX_P2V).Here we present for the first time that whey protein from human breastmilk effectively inhibited both SARS-CoV-2 and GX_P2V by blocking viral attachment and viral replication at entry and even post entry. Moreover, human whey protein inhibited infectious virus production,as proved by the plaque assay. We found that whey protein from different species, such as cow and goat, also showed anti-coronavirus properties.Commercial bovine formula milk also showed similar anti-SARS-CoV-2 activity. | Huahao Fan Bixia Hong Yuqian Luo Qi Peng Liqin Wang Xiangshu Jin Yangzhen Chen Yunjia Hu Yi Shi Tong Li Hui Zhuang Yi-Hua Zhou Yigang Tong Kuanhui Xiang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 1
A single-cell transcriptome atlas of the aging human and macaque retina显示文摘The human retina is a complex neural tissue that detects light and sends visual information to the brain. However, the molecular and cellular processes that underlie aging primate retina remain unclear. Here,we provide a comprehensive transcriptomic atlas based on 119520 single cells of the foveal and peripheral retina of humans and macaques covering different ages. The molecular features of retinal cells differed between the two species, suggesting distinct regional and species specializations of the human and macaque retinae. In addition, human retinal aging occurred in a region-and cell-type-specific manner.Aging of human retina exhibited a foveal to peripheral gradient.MYO9A-rods and a horizontal cell subtype were greatly reduced in aging retina, indicating their vulnerability to aging. Moreover,we generated a dataset showing the cell-type-and region-specific gene expression associated with 55 types of human retinal disease, which provides a foundation to understanding of the molecular and cellular mechanisms underlying human retinal diseases.Such datasets are valuable to understanding of the molecular characteristics of primate retina,as well as molecular regulation of aging progression and related diseases. | Wenyang Yi Yufeng Lu Suijuan Zhong Mei Zhang Le Sun Hao Dong Mengdi Wang Min Wei Haohuan Xie Hongqiang Qu Rongmei Peng Jing Hong Ziqin Yao Yunyun Tong Wei Wang Qiang Ma Zeyuan Liu Yuqian Ma Shouzhen Li Chonghai Yin Jianwei Liu Chao Ma Xiaoqun Wang Qian Wu Tian Xue | 2021 | National Science Review2021,8,4: | 1
Isobaric vapor–liquid equilibrium for binary system of aniline+methyl-N-phenyl carbamate显示文摘In this study, the quasi-static ebulliometric method was used to measure both of the vapor pressures of methyl N-phenyl carbamate(MPC),and the isobaric vapor–liquid equilibrium(VLE) data of the aniline and MPC binary system. The measured vapor pressure data of MPC,at different temperature ranging from 369.60 to 389.54 K,fitted well with the Antoine equation. The VLE data for the aniline and MPC system at(2.00,4.00,6.00,7.00 and 8.00) k Pa were correlated by both of nonrandom two-liquid(NRTL) and Wilson models. The parameters of the two models were obtained by regressing the experimental data, with the absolute temperature deviations of 0.54 K and 0.53 K,respectively. The relative volatility of the binary system calculated was all far more than 1,which gives the conclusion that the high purity MPC can be separated from aniline and MPC binary system by rectification or distillation technology. | Yuqian Li Liguo Wang Peng He Kelin Huang Huiquan Li Yan Cao | 2016 | Chinese Journal of Chemical Engineering2016,24,10: | 1
Chromium trioxide modified spiro-OMeTAD for highly efficient and stable planar perovskite solar cells显示文摘Hole transporting materials(HTMs)play an unparalleled role in heightening the stability and photovoltaic performance of perovskite solar cells(PSCs).The organic small molecule spiro-OMeTAD is frequently utilized for HTM in PSCs.However, the raw spiro-OMeTAD without dopant would be harmful to the development of highly efficient PSCs,due to its unsatisfied hole mobility and conductivity. Therefore,we introduce an inorganic dopant(chromium trioxide,CrO_(3))into the lithium-salt doped spiro-OMeTAD.Because of the exclamatory oxidizability of CrO_(3),it can accelerate the oxidation of spiro-OMeTAD and thereby enhancing the hole mobility of HTM.The introduction of CrO_(3) not only substantially decreases the density of defects, but also adjusts spiro-OMeTAD energy band, and thus effectively suppresses the hysteresis and improving stability of PSCs.In the end,we obtained a power conversion efficiency(PCE)as high as 22.6%after doping CrO_(3) in spiro-OMeTAD.The facile, low cost and outstanding photovoltaic performance render CrO_(3) an excellent dopant for HTMs in PSCs. | Xiaobing Wang Jihuai Wu Yuqian Yang Guodong Li Zeyu Song Xuping Liu Weihai Sun Zhang Lan Peng Gao | 2021 | Journal of Energy Chemistry2021,30,10: | 1
Targeted gene panel provides advantages over whole-exome sequencing for diagnosing obesity and diabetes mellitus显示文摘A small fraction of patients diagnosed with obesity or diabetes mellitus has an underlying monogenic cause. Here,we constructed a targeted gene panel consisting of 83 genes reported to be causative for monogenic obesity or diabetes. We performed this panel in 481 patients to detect causative variants and compared these results with whole-exome sequencing(WES)data available for 146 of these patients. The coverage of targeted gene panel sequencing was significantly higher than that of WES.The diagnostic yield in patients sequenced by the panel was 32.9%with subsequent WES leading to three additional diagnoses with two novel genes. In total,178 variants in 83 genes were detected in 146 patients by targeted sequencing.Three of the 178 variants were missed by WES,although the WES-only approach had a similar diagnostic yield. For the 335 samples only receiving targeted sequencing, the diagnostic yield was 32.2%.In conclusion, taking into account the lower costs, shorter turnaround time, and higher quality of data, targeted sequencing is a more effective screening method for monogenic obesity and diabetes compared to WES.Therefore, this approach could be routinely established and used as a first-tier test in clinical practice for specific patients. | Hairong Yu Haoyong Yu Rong Zhang Danfeng Peng Dandan Yan Yunjuan Gu Yuqian Bao Weiping Jia Hong Zhang Cheng Hu | 2023 | Journal of Molecular Cell Biology2023,15,6: | 0
T cell cross-reactivity in autoimmune-like hepatitis triggered by COVID-19显示文摘Over 1000 cases of pediatric hepatitis of unknown etiology have been reported worldwide since the first case was reported in the UK.To date, the etiology of pediatric hepatitis remains unknown and controversial.Adenovirus was first suspected to be the cause as it was present in the blood samples of the majority of cases.Partial cases have also been tested positive for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)[1].However,it is still unclear how these viruses contribute to pediatric hepatitis. In the case of a pediatric patient with SARS-CoV-2 infection, the liver biopsy showed acute submassive hepatocyte necrosis, accompanied by a significant increase in T cell infiltration[2].Furthermore,CD8+T cell dominant hepatitis induced by coronavirus disease 2019(COVID-19)vaccination has also been recently reported[3].Although it is known that T cell receptors(TCRs)can discriminate between self-and non-self-antigens,it is now well-accepted that TCRs exhibit cross-reactivity toward similar and even distinct antigen peptides[4].Thus,we hypothesized that following SARS-CoV-2 infection or vaccination,T cells carrying TCRs that recognize self-antigens undergo clonal expansion, which could eventually result in the onset of autoimmune-like hepatitis(Figure 1A). | Yisu Liu Yuqian Wang Zhiqiang Peng Guideng Li Jianwei Wang | 2023 | hLife2023,1,1: | 0
Dose-related effects of norepinephrine on early-stage endotoxemic shock in a swine model显示文摘Background The benefits of early use of norepinephrine in endotoxemic shock remain unknown. We aimed to elucidate the effects of different doses of norepinephrine in early-stage endotoxemic shock using a clinically relevant large animal model. Methods Vasodilatory shock was induced by endotoxin bolus in 30 Bama suckling pigs.Treatment included fluid resuscitation and administration of different doses of norepinephrine,to induce return to baseline mean arterial pressure(MAP).Fluid management, hemodynamic, microcirculation, inflammation, and organ function variables were monitored. All animals were supported for 6 h after endotoxemic shock. Results Infused fluid volume decreased with increasing norepinephrine dose.Return to baseline MAP was achieved more frequently with doses of 0.8µg/kg/min and 1.6µg/kg/min(P<0.01).At the end of the shock resuscitation period, cardiac index was higher in pigs treated with 0.8µg/kg/min norepinephrine(P<0.01),while systemic vascular resistance was higher in those receiving 0.4µg/kg/min(P<0.01).Extravascular lung water level and degree of organ edema were higher in animals administered no or 0.2µg/kg/min norepinephrine(P<0.01),while the percentage of perfused small vessel density(PSVD)was higher in those receiving 0.8µg/kg/min(P<0.05)and serum lactate was higher in the groups administered no and 1.6µg/kg/min norepinephrine(P<0.01).Conclusions The impact of norepinephrine on the macro-and micro-circulation in early-stage endotoxemic shock is dose-dependent,with very low and very high doses resulting in detrimental effects.Only an appropriate norepinephrine dose was associated with improved tissue perfusion and organ function. | Hui Xiang Yuqian Zhao Siqing Ma Qi Li Kianoush B.Kashani Zhiyong Peng Jianguo Li Bo Hu | 2023 | Journal of Intensive Medicine2023,3,4: | 0
