| 1 | Significance of dredging on sediment denitrification in Meiliang Bay,China:A year long simulation study显示文摘An experiment for studying the effects of sediment dredging on denitrification in sediments was carried out through a one-year incubation of undredged (control) and dredged cores in laboratory. Dredging the upper 30 cm of sediment can significantly affect physico-chemical characteristics of sediments. Less degradation of organic matter in the dredged sediments was found during the experiment. Denitrification rates in the sediments were estimated by the acetylene blockage technique, and ranged from 21.6 to 102.7 nmol N2/(g dry weight (dw)·hr) for the undredged sediment and from 6.9 to 26.9 nmol N2/(g dw·hr) for dredged sediments. The denitrification rates in the undredged sediments were markedly higher (p<0.05) than those in the dredged sediments throughout the incubation, with the exception of February 2006. The importance of various environmental factors on denitrification was assessed, which indicated that denitrification was regulated by temperature. Nitrate was probably the key factor limiting denitrification in both undredged and dredged sediments. Organic carbon played some role in determining the denitrification rates in the dredged sediments, but not in the undredged sediments. | 2 | Seasonal variation of potential denitrification rates of surface sediment from Meiliang Bay, Taihu Lake, China显示文摘The regulatory effects of environmental factors on denitrification were studied in the sediments of Meiliang Bay, Taihu Lake, in a monthly sampling campaign over a one-year period. Denitrification rates were measured in slurries of field samples and enrichment experiments using the acetylene inhibition technique. Sediment denitrification rates in inner bay and outer bay ranged from 2.8 to 51.5 nmol N2/(g dw (dry weight)·hr) and from 1.5 to 81.1 nmol N2/(g dw·hr), respectively. Sediment denitrification rates were greatest in the spring and lowest in the summer and early autumn, due primarily to seasonal differences in nitrate concentration and water temperature. For each site, positive and linear relationships were regularly observed between denitrification rate and water columnn itrate concentration. Of various environmental factors on denitrification that we assessed, nitrate was determined to be the key factor limiting denitrification rates in the sediments of Meiliang Bay. In addition, at the two sites denitrification rates were also regulated by temperature. The addition of organic substrates had no significant effect on denitrification rate, indicating that sediment denitrification was not limited by organic carbon availability in the sediments. | 3 | Time constraints for the closing of the Paleo-Asian Ocean in the Northern Alxa Region: Evidence from Wuliji granites显示文摘The Wuliji pluton in the Northern Alxa Region, Inner Mongolia, is the principal part of Shalazhashan Mountain. It belongs to the Zongnaishan-Shalazhashan Arc Zone, northwestern North China Plate, whose north is Engger Us Ophiolite Belt and south is Qagan Qulu Ophiolite Belt. The pluton was emplaced into Upper Carboniferous-Lower Permian Amushan Formation. According to the research about the original Carboniferous Amushan Formation, the lower and middle sections of the Carboniferous Amushan Formation consist of volcanic, clastic, and carbonate rocks, interpreted to represent the sedimentary association of a volcanic arc and back-arc basin; the upper section of the Amushan Formation is a molasse composed of silty shale, sandstone, gravel-bearing sandstone, and conglomerate. The Wuliji pluton consists mainly of biotite monzonitic granite, amphibole-bearing biotite monzonitic granite, and monzonitic granite. Geochemical analyses show that the pluton has both metaluminous and peraluminous characteristics, and on average has SiO2>70 wt%, Al2O3 >14 wt%, and high contents of Na2O+K2O (8.5 wt%), which define a calc-alkaline series. In addition, REE patterns show enrichment of LREE and weak negative Eu anomalies (δ Eu=0.3-1). Altogether, the samples are depleted in Nb, Ta, Ti, P, Sr, and Ba, and enriched in Rb, Th, and K. These geochemical traits are interpreted to reflect an arc component. A secondary ion mass spectrometry (SIMS) U-Pb zircon age of the biotite monzonitic Wuliji pluton in the Northern Alxa Region, Inner Mongolia, is 250.8±2.0 Ma (1σ). Samples have ε Nd (t) values between 0.1 and 1.3, which suggests that the granites were derived from mixing between the crust and mantle. Based on the SIMS age and geochemical characteristics, Wuliji granite is interpreted to be a post-collisional granite, the result of mantle-derived melt and assimilated juvenile arc crust. However, according to the newest international stratigraphic classification standard, the upper section of the Amushan Formation is Lower Permian in age, indicating that the back-arc basin had already closed in Early Permian. We conclude that the Paleo-Asian Ocean represented by the Engger Us Ophiolite Belt subducted southward in Late Carboniferous, at the same time that the trench-arc-basin system formed in the Northern Alxa Region. The Paleo-Asian Ocean was closed in Early Permian and the Northern Alxa Region entered a post-collisional period in the Late Permian, as indicated by the Wuliji granites. | 5 | Type 1 CD8^+ T Cells are Superior to Type 2 CD8^+ T Cells in Tumor Immunotherapy due to Their Efficient Cytotoxicity,Prolonged Survival and Type 1 Immune Modulation显示文摘CD8+ cytotoxic T(Tc) cells play a crucial role in host immune responses to cancer,and in this context,adoptive CD8+ Tc cell therapy has been studied in numerous animal tumor models. Its antitumor efficacy is,to a large extent,determined by the ability of Tc cells to survive and infiltrate tumors. In clinical trials,such in vitro-activated T cells often die within hours to days,and this greatly limits their therapeutic efficacy. CD8+ Tc cells fall into two subpopulations based upon their differential cytokine secretion. In this study,we in vitro generated that ovalbumin(OVA) -pulsed dendritic cell(DCOVA) -activated CD8+ type 1 Tc(Tc1) cells secreting IFN-γ,and CD8+ type 2 Tc(Tc2) cells secreting IL-4,IL-5 and IL-10,which were derived from OVA-specific T cell receptor(TCR) transgenic OT I mice. We then systemically investigated the in vitro and in vivo effector function and survival of Tc1 and Tc2 cells,and then assessed their survival kinetics after adoptively transferred into C57BL/6 mice,respectively. We demonstrated that,when compared to CD8+ Tc2,Tc1 cells were significantly more effective in perforin-mediated cytotoxicity to tumor cells,had a significantly higher capacity for in vivo survival after the adoptive T cell transfer,and had a significantly stronger therapeutic effect on eradication of well-established tumors expressing OVA in animal models. In addition,CD8+ Tc1 and Tc2 cells skewed the phenotype of CD4+ T cells toward Th1 and Th2 type,respectively. | 6 | Mycorrhizal symbiosis modulates the rhizosphere microbiota to promote rhizobia–legume symbiosis显示文摘Plants establish symbioses with mutualistic fungi,such as arbuscular mycorrhizal(AM)fungi,and bacteria,such as rhizobia,to exchange key nutrients and thrive.Plants and symbionts have coevolved and represent vital components of terrestrial ecosystems.Plants employ an ancestral AM signaling pathway to establish intracellular symbioses,including the legume–rhizobia symbiosis,in their roots.Nevertheless,the relationship between the AM and rhizobial symbioses in native soil is poorly understood.Here,we examined how these distinct symbioses affect root-associated bacterial communities in Medicago truncatula by performing quantitative microbiota profiling(QMP)of 16S rRNA genes.We found that M.truncatula mutants that cannot establish AM or rhizobia symbiosis have an altered microbial load(quantitative abundance)in the rhizosphere and roots,and in particular that AM symbiosis is required to assemble a normal quantitative root-associated microbiota in native soil.Moreover,quantitative microbial co-abundance network analyses revealed that AM symbiosis affects Rhizobiales hubs among plant microbiota and benefits the plant holobiont.Through QMP of rhizobial rpoB and AM fungal SSU rRNA genes,we revealed a new layer of interaction whereby AM symbiosis promotes rhizobia accumulation in the rhizosphere of M.truncatula.We further showed that AM symbiosis-conditioned microbial communities within the M.truncatula rhizosphere could promote nodulation in different legume plants in native soil.Given that the AM and rhizobial symbioses are critical for crop growth,our findings might inform strategies to improve agricultural management.Moreover,our work sheds light on the co-evolution of these intracellular symbioses during plant adaptation to native soil conditions. | Xiaolin Wang Huan Feng Yayu Wang Mingxing Wang Xingguang Xie Huizhong Chang Like Wang Jicheng Qu Kai Sun Wei He Chunyan Wang Chuanchao Dai Zhaohui Chu Changfu Tian Nan Yu Xuebin Zhang Huan Liu Ertao Wang | 2021 | Molecular Plant2021,14,3: | 6 |

The results showed that the optimal retention time of the heating up of capillary columns was 5 min. Of the seven fibers used in this work,DVB/CAR/PDMS was found to be the most suitable to extract the aroma volatile compounds in Fuji apple. A total number of 43 volatile compounds were identified according to their retention time on capillary columns and their mass spectra,including eleven esters,ten alcohols,ten aldehydes,seven alkenes,two anhydrides,one ketone,one phenol and one ether. In addition to lifestyle modifications, pharmacotherapy has become an important strategy to control weight gain. However, most of the anti-obesity drugs often show poor outcome for weight-loss and cause severe adverse effects. This review surveys recent advances in nanomedicine as an emerging strategy for obesity treatment with an emphasis on the enhanced therapeutic efficiency and minimized side effects.
