1Toxic effects of chlortetracycline on maize growth,reactive oxygen species generation and the antioxidant response显示文摘The toxicity of chlortetracycline (CTC) on maize (Zea mays L.) growth and reactive oxygen species (ROS) generation was studied.The root and shoot lengths and fresh weights of maize seedlings were inhibited by CTC treatment (p < 0.05).Root length was more sensitive than other parameters with the EC 10 value of 0.064 mg/L.The spin trapping technique followed by electron paramagnetic resonance (EPR) analysis was used to quantify the ROS production.The ROS generated in maize roots after exposure to CTC was identified as hydroxyl radical (·OH).The EPR signal intensity correlated positively with the logarithm of CTC concentrations exposed (p < 0.05).The dynamic changes of malondialdehyde (MDA) contents and the antioxidative enzyme activities in maize roots were also determined.As compared to the control group,CTC was found to significantly increase MDA content.Treatment of maize roots with the ·OH scavenger sodium benzoate (SB) reduced the MDA content and enhanced the antioxidative enzyme activities.The results demonstrated the harmfulness of CTC at high dose to maize in the early developmental stage,and clarified that the inducement of ·OH is one of the mechanisms of CTC toxicity.Bei Wen Yu Liu Peng Wang Tong WU Shuzhen Zhang Xiaoquan Shan Jingfen Lu 2012Journal of Environmental Sciences2012,24,6:6
2Entinostat,a classⅠselective histone deacetylase inhibitor,plus exemestane for Chinese patients with hormone receptor-positive advanced breast cancer:A multicenter,randomized,double-blind,placebo-controlled,phase 3 trial显示文摘Entinostat plus exemestane in hormone receptor-positive(HR+)advanced breast cancer(ABC)previously showed encouraging outcomes.This multicenter phase 3 trial evaluated the efficacy and safety of entinostat plus exemestane in Chinese patients with HR+ABC that relapsed/progressed after≥1 endocrine therapy.Patients were randomized(2:1)to oral exemestane 25 mg/day plus entinostat(n=235)or placebo(n=119)5 mg/week in 28-day cycles.The primary endpoint was the independent radiographic committee(IRC)-assessed progression-free survival(PFS).The median age was 52(range,28—75)years and 222(62.7%)patients were postmenopausal.CDK4/6 inhibitors and fulvestrant were previously used in 23(6.5%)and 92(26.0%)patients,respectively.The baseline characteristics were comparable between the entinostat and placebo groups.The median PFS was 6.32(95%CI,5.30—9.11)and 3.72(95%CI,1.91—5.49)months in the entinostat and placebo groups(HR,0.76;95%CI,0.58—0.98;P=0.046),respectively.Grade≥3 adverse events(AEs)occurred in 154(65.5%)patients in the entinostat group versus 23(19.3%)in the placebo group,and the most common grade≥3 treatment-related AEs were neutropenia[103(43.8%)],thrombocytopenia[20(8.5%)],and leucopenia[15(6.4%)].Entinostat plus exemestane significantly improved PFS compared with exemestane,with generally manageable toxicities in HR+ABC(ClinicalTrials.gov#NCT03538171).Binghe Xu Qingyuan Zhang Xichun Hu Qing Li Tao Sun Wei Li Quchang Ouyang Jingfen Wang Zhongsheng Tong Min Yan Huiping Li Xiaohua Zeng Changping Shan Xian Wang Xi Yan Jian Zhang Yue Zhang Jiani Wang Liang Zhang Ying Lin Jifeng Feng Qianjun Chen Jian Huang Lu Zhang Lisong Yang Ying Tian Hongyan Shang 2023Acta Pharmaceutica Sinica B2023,13,5:3

43D-QSAR study on phenoxy-alkylamine compounds of α_1-adrenoceptor antagonist显示文摘The study of three-dimensional quantitative structure-activity relationship (3D-QSAR) of DDPH and its derivatives that have been known with their activity parameters has been developed using the comparative molecular field analysis (CoMFA) method. Here, (+)-DDPH crystal structure was selected as the active conformation model and comparisons between the influences of different charge calculation methods and grid setup were conducted. The coefficients of cross-validation (q2) and regression (r2) are 0.481 and 0.997, respectively. The standard error (SE) is 0.102. The research result suggests that the steric field makes more contributions to the activity than the electrostatic field. 8Blood oxygenation-level dependent functional MRI in evaluating the selective activation of motor cortexes associated with recovery of motor function in hemiplegic patients with ischemic stroke显示文摘BACKGROUND: Previous studies about blood oxygenation-level dependent (BOLD) functional MRI (fMRI) have indicated that the poststroke recovery of motor function is accompanied by the selective activation of motor cortexes with high correlation. OBJECTIVE: To evaluate the short-term outcomes after rehabilitative interventions with BOLD fMRI in hemiplegic patients with acute stroke, and analyze the correlation of the excitement of brain function in the passive and active movements of the affected limb with the recovery of motor function. DESIGN: A case observation. SETTING: Department of Neurology, Baotou Central Hospital. PARTICIPANTS: Thirty hemiplegic inpatients with ischemic stroke were selected from the Department of Neurology, Baotou Central Hospital from January to December in 2005, including 16 males and 14 females, aging 44-71 years with an average age of (56±5) years, and the disease course ranged from 12 to 72 hours. Inclusive criteria: In accordance with the diagnostic standard of ischemic stroke revised by the Fourth National Academic Meeting for Cerebrovascular Disease; Confirmed by cranial CT or MRI. They were all informed agreed with the detected items. METHODS: ① The Bobath technique was adopted in the rehabilitative interventions of the 30 patients, 30 minutes for each time, twice a day for three weeks continuously. ② The hand motor recovery of the stroke patients was graded by the Brunnstrom stages (Ⅰ-Ⅵ), and be able to grasp various objects and extend for the whole range was taken as grade Ⅵ. ③ The patients were examined with fMRI BOLD before rehabilitation and 3 weeks after rehabilitation. All the patients were trained with finger movements, the distracting thoughts should be eliminated as much as possible especially during the movement phase, the patients should highly concentrate on the hand movements. The range for the finger movements should be as large as possible with moderate frequency. The hand movements should be 10 s with an interval of 30 s. Te AVANTO 1.5T MRI machine (Siemens Company, Germany) was used for scanning, the 30 s before fMRI BOLD scanning was the adaptation phase for the patients, and the finger movements were alternated for 6-10 times till the end of the scanning. MAIN OUTCOME MEASURES: Differences after rehabilitation evaluated by the activations of different sensorimotor cortex in the hemiplegic patients with acute stroke. RESULTS: ① In passive and active movements of the affected hand, 5 cases of Brunnstrom grade Ⅰ orⅡ were manifested by the activation of unilateral sensorimotor cortex (SMC) of the hemiplegic side, and they were Brunnstrom grade Ⅱ after 3-week rehabilitation, and 1 case with activations of bilateral SMC recovered to Brunnstrom grade Ⅲ. ② Among the patients of Brunnstrom gradeⅢ or Ⅳ, 5 cases were unilaterally activated, and had no changes after 3-week rehabilitation; Whereas of the 9 cases of bilateral activations, 3 recovered to grade Ⅵ and 6 recovered to grade Ⅴ after 3-week rehabilitation. ③ In the passive and active movements of the affected hand, all the cases of Brunnstrom grades Ⅴ and Ⅵ were manifested by the activations of bilateral SMC. After 3-week rehabilitation, 2 of the 4 cases of grade Ⅴ recovered to grade Ⅵ, and the other 2 recovered to normal; Whereas 5 of the 6 cases of grade Ⅵ recovered to normal, and the other 1 was still grade Ⅵ. CONCLUSION: BOLD fMRI showed that the outcomes of short-term rehabilitation in the patients with bilateral activations were obvious, whereas the outcomes were bad in the patients whose ipsilateral cortexes were activated. 9A novel dephosphorylation targeting chimera selectively promoting tau removal in tauopathies显示文摘Intraneuronal accumulation of hyperphosphorylated tau is a hallmark pathology shown in over twenty neurodegenerative disorders,collectively termed as tauopathies,including the most common Alzheimer's disease(AD).Therefore,selectively removing or reducing hyperphosphorylated tau is promising for therapies of AD and other tauopathies.Here,we designed and synthesized a novel DEPhosphorylation TArgeting Chimera(DEPTAC)to specifically facilitate the binding of tau to Ba-subunit-contalning protein phosphatase 2A(PP2A-Ba),the most active tau phosphatase in the brain.The DEPTAC exhibited high efficiency in dephosphorylating tau at multiple AD-associated sites and preventing tau accumulation both in vitro and in vivo.Further studies revealed that DEPTAC significantly improved microtubule assembly,neurite plasticity,and hippocampus-dependent learning and memory in transgenic mice with inducible overexpression of truncated and neurotoxic human tau N368.Our data provide a strategy for selective removal of the hyperphosphorylated tau,which sheds new light for the targeted therapy of AD and related-tauopathies.Jie Zheng Na Tian Fei Liu Yidian Zhang Jingfen Su Yang Gao Mingmin Deng Linyu Wei Jingwang Ye Honglian Li Jian-Zhi Wang 2021Signal Transduction and Targeted Therapy2021,6,8:0
10A New Hidden Markov Model for Protein Quality Assessment Using Compatibility Between Protein Sequence and Structure显示文摘Protein structure Quality Assessment(QA) is an essential component in protein structure prediction and analysis. The relationship between protein sequence and structure often serves as a basis for protein structure QA.In this work, we developed a new Hidden Markov Model(HMM) to assess the compatibility of protein sequence and structure for capturing their complex relationship. More specifically, the emission of the HMM consists of protein local structures in angular space, secondary structures, and sequence profiles. This model has two capabilities:(1) encoding local structure of each position by jointly considering sequence and structure information, and(2)assigning a global score to estimate the overall quality of a predicted structure, as well as local scores to assess the quality of specific regions of a structure, which provides useful guidance for targeted structure refinement. We compared the HMM model to state-of-art single structure quality assessment methods OPUSCA, DFIRE, GOAP,and RW in protein structure selection. 11Phylogeny of SARS-CoV as inferred from complete genome comparison显示文摘SARS-CoV, as the pathogeny of severe acute respiratory syndrome (SARS), is a mystery that the origin of the virus is still unknown even a few isolates of the virus were completely sequenced. To explore the genesis of SARS-CoV, the FDOD method previously developed by us was applied to comparing complete genomes from 12 SARS-CoV isolates to those from 12 previously identified coronaviruses and an unrooted phylogenetic tree was constructed. Our results show that all SARS-CoV isolates were clustered into a clique and previously identified coronaviruses formed the other clique. Meanwhile, the three groups of coronaviruses depart from each other clearly in our tree that is consistent with the results of prevenient papers. Differently, from the topology of the phylogenetic tree we found that SARS-CoV is more close to group 1 within genus coronavirus.
