15High-throughput screening of SARS-CoV-2 main and papain-like protease inhibitors显示文摘The global COVID-19 coronavirus pandemic has infected over 109 million people,leading to over 2 million deaths up to date and still lacking of effective drugs for patient treatment.Here,we screened about 1.8 million small molecules against the main protease(M^(pro))and papain like protease(PL^(pro)),two major proteases in severe acute respiratory syndrome-coronavirus 2 genome,and identified 1851M^(pro)inhibitors and 205 PL^(pro)inhibitors with low nmol/l activity of the best hits.Among these inhibitors,eight small molecules showed dual inhibition effects on both M^(pro)and PL^(pro),exhibiting potential as better candidates for COVID-19 treatment.The best inhibitors of each protease were tested in antiviral assay,with over 40%of M^(pro)inhibitors and over 20%of PL^(pro)inhibitors showing high potency in viral inhibition with low cytotoxicity.The X-ray crystal structure of SARS-CoV-2 M^(pro)in complex with its potent inhibitor 4a was determined at 1.8Åresolution.Together with docking assays,our results provide a comprehensive resource for future research on anti-SARS-CoV-2 drug development.Yi Zang Mingbo Su Qingxing Wang Xi Cheng Wenru Zhang Yao Zhao Tong Chen Yingyan Jiang Qiang Shen Juan Du Qiuxiang Tan Peipei Wang Lixin Gao Zhenming Jin Mengmeng Zhang Cong Li Ya Zhu Bo Feng Bixi Tang Han Xie Ming-Wei Wang Mingyue Zheng Xiaoyan Pan Haitao Yang Yechun Xu Beili Wu Leike Zhang Zihe Rao Xiuna Yang Hualiang Jiang Gengfu Xiao Qiang Zhao Jia Li 2023Protein & Cell2023,14,1:0
16Map-based cloning and CRISPR/Cas9-based editing uncover BoNA1 as the causal gene for the no-anthocyanin-accumulation phenotype in curly kale (Brassica oleracea var. sabellica)显示文摘Brassica oleracea comprises several important vegetable and ornamental crops,including curly kale,ornamental kale,cabbage,broccoli,and others.The accumulation of anthocyanins,important secondary metabolites valuable to human health,in these plants varies widely and is responsible for their pink to dark purple colors.Some curly kale varieties lack anthocyanins,making these plants completely green.The genetic basis of this trait is still unknown.We crossed the curly kale inbred line BK2019(without anthocyanins)with the cabbage inbred line YL1(with anthocyanins)and the Chinese kale inbred line TO1000(with anthocyanins)to generate segregating populations.The no-anthocyanin trait was genetically controlled by a recessive gene,bona1.We generated a linkage map and mapped bona1 to a 256-kb interval on C09.We identified one candidate gene,Bo9g058630,in the target genomic region;this gene is homologous to AT5G42800,which encodes a dihydroflavonol-4-reductase-like(DFR-like)protein in Arabidopsis.In BK2019,a 1-bp insertion was observed in the second exon of Bo9g058630 and directly produced a stop codon.To verify the candidate gene function,CRISPR/Cas9 gene editing technology was applied to knock out Bo9g058630.We generated three bona1 mutants,two of which were completely green with no anthocyanins,confirming that Bo9g058630 corresponds to BoNA1.Different insertion/deletion mutations in BoNA1 exons were found in all six of the other no-anthocyanin kale varieties examined,supporting that independent disruption of BoNA1 resulted in no-anthocyanin varieties of B.oleracea.This study improves the understanding of the regulation mechanism of anthocyanin accumulation in B.oleracea subspecies.Kaiwen Yuan Xinyu Zhao Wenru Sun Limei Yang Yangyong Zhang Yong Wang Jialei Ji Fengqing Han Zhiyuan Fang Honghao Lv 2023Horticulture Research2023,10,8:0

18La ions-enhanced NH_(3)-SCR performance over Cu-SSZ-13 catalysts显示文摘Lanthanum(La)ions are generally recognized to cause a decline of the catalytic performance for Cu-SSZ-13 zeolite in the selective catalytic reduction of NO_(x)with NH_(3)(NH_(3)-SCR).Herein,we demonstrate that the NH_(3)-SCR performance and hydrothermal stability of Cu-La-SSZ-13 zeolites can be enhanced with the incorporation of a small amount of La ions.The incorporation of La ions into SSZ-13 favors more Z_(2)Cu^(2+)ions at six-membered rings(6MRs),which results in higher hydrothermal stability of Cu-La-SSZ-13 than that of Cu-SSZ-13.The NO conversion of Cu-La-SSZ-13 achieves 5%–10%higher than that of Cu-SSZ-13 at the temperature range of 400–550℃ after hydrothermal ageing.While introducing excess amount of La ions in SSZ-13 may cause the formation of inactive CuO_(x),leading to the decrease of catalytic activity and hydrothermal stability.Notably,the low-temperature activity of Cu-SSZ-13 with a low Cu content(≤2 wt.%)can be boosted by the introduction of La ions,which is largely due to the improved redox ability of Cu active sites modified by La ions.Density functional theory(DFT)calculations indicate that La ions prefer to locate at eight-membered rings(8MRs)and thus promoting the formation of more Z_(2)Cu^(2+)ions.Meanwhile,the existence of La ions in SSZ-13 inhibits the dealumination process and the transformation from Z_(2)Cu^(2+)to CuO_(x),resulting in its enhanced hydrothermal stability.The present work sheds a new insight into the regulation of secondary metal cations for promoting high NH_(3)-SCR performance over Cu-SSZ-13 zeolite catalysts.Mengyang Chen Wenru Zhao Yingzhen Wei Jinfeng Han Junyan Li Chang Sun Donghai Mei Jihong Yu 2023Nano Research2023,16,10:0
