1One-year clinical study of NeuroR egen scaffold implantation following scar resection in complete chronic spinal cord injury patients显示文摘The objective of this clinical study was to assess the safety and feasibility of the collagen scaffold, Neuro Regen scaffold, one year after scar tissue resection and implantation. Scar tissue is a physical and chemical barrier that prevents neural regeneration. However, identification of scar tissue is still a major challenge. In this study, the nerve electrophysiology method was used to distinguish scar tissue from normal neural tissue, and then different lengths of scars ranging from 0.5–4.5 cm were surgically resected in five complete chronic spinal cord injury(SCI) patients. The NeuroR egen scaffold along with autologous bone marrow mononuclear cells(BMMCs), which have been proven to promote neural regeneration and SCI recovery in animal models, were transplanted into the gap in the spinal cord following scar tissue resection. No obvious adverse effects related to scar resection or Neuro Regen scaffold transplantation were observed immediately after surgery or at the 12-month follow-up. In addition, patients showed partially autonomic nervous function improvement, and the recovery of somatosensory evoked potentials(SSEP) from the lower limbs was also detected. 2Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. 3Xuebijing injection alleviates liver injury by inhibiting secretory function of Kupffer cells in heat stroke rats显示文摘OBJECTIVE:To evaluate the effects of Xuebijing(XBJ) injection in heat stroke(HS) rats and to investigate the mechanisms underlying these effects.METHODS:Sixty anesthetized rats were randomized into three groups and intravenously injected twice daily for 3 days with 4 mL XBJ(XBJ group) or phosphate buffered saline(HS and Sham groups) per kg body weight.HS was initiated in the HS and XBJ groups by placing rats in a simulated climate chamber(ambient temperature 40 ℃,humidity 60%).Rectal temperature,aterial pressure,and heart rate were monitored and recorded.Time to HS onset and survival were determined,and serum concentrations of tumor necrosis factor(TNF)-α,inRESULTS:Time to HS onset and survival were significantly longer in the XBJ than in the HS group.Moreover,the concentrations of TNF-α,IL-1β,IL-6,ALT and AST were lower and liver injury was milder in the XBJ than in the HS group.Heat-stress induced structural changes in KCs and hepatic cells were more severe in the HS than in the XBJ group and the concentrations of TNF-α,IL-β and IL-6 secreted by KCs were lower in the XBJ than in the HS group.CONCLUSION:XBJ can alleviate HS-induced systemic inflammatory response syndrome and liver injury in rats,and improve outcomes.These protective effects may be due to the ability of XBJ to inhibit cytokine secretion by KCs.Yi Chen Huasheng Tong Xingqin Zhang Liqun Tang Zhiguo Pan Zhifeng Liu Pengkai Duan Lei Su 2013Journal of Traditional Chinese Medicine2013,33,2:14

5Joint Inversion of the 3D P Wave Velocity Structure of the Crust and Upper Mantle under the Southeastern Margin of the Tibetan Plateau Using Regional Earthquake and Teleseismic Data显示文摘The special seismic tectonic environment and frequent seismicity in the southeastern margin of the Qinghai–Tibet Plateau show that this area is an ideal location to study the present tectonic movement and background of strong earthquakes in China's Mainland and to predict future strong earthquake risk zones. Studies of the structural environment and physical characteristics of the deep structure in this area are helpful to explore deep dynamic effects and deformation field characteristics, to strengthen our understanding of the roles of anisotropy and tectonic deformation and to study the deep tectonic background of the seismic origin of the block's interior. In this paper, the three-dimensional(3D) P-wave velocity structure of the crust and upper mantle under the southeastern margin of the Qinghai–Tibet Plateau is obtained via observational data from 224 permanent seismic stations in the regional digital seismic network of Yunnan and Sichuan Provinces and from 356 mobile China seismic arrays in the southern section of the north–south seismic belt using a joint inversion method of the regional earthquake and teleseismic data. The results indicate that the spatial distribution of the P-wave velocity anomalies in the shallow upper crust is closely related to the surface geological structure, terrain and lithology. Baoxing and Kangding, with their basic volcanic rocks and volcanic clastic rocks, present obvious high-velocity anomalies. The Chengdu Basin shows low-velocity anomalies associated with the Quaternary sediments. The Xichang Mesozoic Basin and the Butuo Basin are characterised by lowvelocity anomalies related to very thick sedimentary layers. The upper and middle crust beneath the Chuan–Dian and Songpan–Ganzi Blocks has apparent lateral heterogeneities, including low-velocity zones of different sizes. There is a large range of low-velocity layers in the Songpan–Ganzi Block and the sub–block northwest of Sichuan Province, showing that the middle and lower crust is relatively weak. The Sichuan Basin, which is located in the western margin of the Yangtze platform, shows high-velocity characteristics. The results also reveal that there are continuous low-velocity layer distributions in the middle and lower crust of the Daliangshan Block and that the distribution direction of the low-velocity anomaly is nearly SN, which is consistent with the trend of the Daliangshan fault. The existence of the low-velocity layer in the crust also provides a deep source for the deep dynamic deformation and seismic activity of the Daliangshan Block and its boundary faults. The results of the 3D P-wave velocity structure show that an anomalous distribution of high-density, strong-magnetic and high-wave velocity exists inside the crust in the Panxi region. This is likely related to late Paleozoic mantle plume activity that led to a large number of mafic and ultra-mafic intrusions into the crust. In the crustal doming process, the massive intrusion of mantle-derived material enhanced the mechanical strength of the crustal medium. The P-wave velocity structure also revealed that the upper mantle contains a low-velocity layer at a depth of 80–120 km in the Panxi region. 8Transplantation of adult spinal cord grafts into spinal cord transected rats improves their locomotor function显示文摘Grafted embryonic central neural tissue pieces can recover function of hemisected spinal cord in neonatal rats and promote axonal growth in adults. However, spinal cord segments from adults have not been used as donor segments for allogeneic transplantation. Here, we utilized adult spinal cord tissue grafts(aSCGs) as donor constructs for repairing complete spinal cord injury(SCI). Moreover, to provide a favourable microenvironment for SCI treatment, a growth factor cocktail containing three growth factors(brain-derived neurotrophic factor, neurotrophin-3 and vascular endothelial growth factor), was applied to the aSCG transplants. We found that the locomotor function was significantly improved 12 weeks after transplantation of aSCGs into the spinal cord lesion site in adult rats. Transplantation of aSCGs combined with these growth factors enhanced neuron and oligodendrocyte survival and functional restoration. 9Recent Advances in Self-Supported Layered Double Hydroxides for Oxygen Evolution Reaction显示文摘Electrochemical water splitting driven by clean and sustainable energy sources to produce hydrogen is an efficient and environmentally friendly energy conversion technology.Water splitting involves hydrogen evolution reaction(HER)and oxygen evolution reaction(OER),in which OER is the limiting factor and has attracted extensive research interest in the past few years.Conventional noble-metal-based OER electrocatalysts like IrO_(2) and RuO_(2) suffer from the limitations of high cost and scarce availability.Developing innovative alternative nonnoble metal electrocatalysts with high catalytic activity and long-term durability to boost the OER process remains a significant challenge.Among all of the candidates for OER catalysis,selfsupported layered double hydroxides(LDHs)have emerged as one of the most promising types of electrocatalysts due to their unique layered structures and high electrocatalytic activity.In this review,we summarize the recent progress on self-supported LDHs and highlight their electrochemical catalytic performance.Specifically,synthesis methods,structural and compositional parameters,and influential factors for optimizing OER performance are discussed in detail.Finally,the remaining challenges facing the development of self-supported LDHs are discussed and perspectives on their potential for use in industrial hydrogen production through water splitting are provided to suggest future research directions.Libo Wu Luo Yu Xin Xiao Fanghao Zhang Shaowei Song Shuo Chen Zhifeng Ren 2020Research2020,,1:7

11Suppression of murine breast cancer metastasis by selective inhibition of CXCR4 by synthetic polypeptide derived from viral macrophage inflammatory protein Ⅱ显示文摘Stromal cell-derived factor-1 and its receptor CXC chemokine receptor-4(CXCR4)have been implicated in breast cancer metastasis.A significant association between HER2 and CXCR4 expression has been observed in human breast tumor tissues,and overexpression of CXCR4 is essential for HER2-mediated tumor metastasis.Moreover,CXCR4 expression is low in normal breast tissues and high in malignant tumors,suggesting that a blockade of CXCR4 may limit tumor metastasis.The present study investigated the action of a synthetic antagonist 21-mer peptide derived from viral macrophage inflammatory protein Ⅱ against CXCR4(NT21MP)in inhibiting metastasis in vitro and in vivo.The results showed that chemotaxis of SKBR3 cells toward SDF-1αwas reduced by NT21MP in a dose-dependent manner(P<0.05).NT21MP inhibited tumor growth at 500μg/kg and in combination with Herceptin,the anti-HER2 antibody.The in vivo metastatic assay showed that NT21MP significantly inhibited pulmonary metastasis,and the number of metastatic tumor nodes on the surface of the lung was greatly decreased.Compared with the saline-treated control group,PCNA expression was dose-dependently decreased by NT21MP,the percentage of apoptotic cells was increased,and CXCR4 mRNA and protein expression were downregulated.In conclusion,NT21MP inhibits cellular proliferation,promotes apoptosis by downregulating CXCR4 expression,and suppresses the progression of primary and metastatic tumors.CXCR4 may be a useful therapeutic target for breast cancer,and NT21MP may serve as a potential target drug for the treatment of breast cancer metastasis.YANG QingLin DING YongXing CHEN ChangJie TANG Jie ZHANG Ju YANG ZhiFeng 2010Chinese Science Bulletin2010,55,20:6
12The novel OCT4 spliced variant OCT4B1 can generate three protein isoforms by alternative splicing into OCT4B显示文摘OCT4 is one of the key transcription factors in maintaining the pluripotency and self-renewal of embryonic stem (ES) cells.Human OCT4 can generate two isoforms OCT4A and OCT4B by alternative splicing.OCT4B1 is a recently discovered novel OCT4 spliced variant,which has been considered as a putative marker of stemness.Compared with the OCT4B mRNA,OCT4B1 mRNA is generated by retaining intron 2 as a cryptic exon which contains a TGA stop codon in it.As a result,the protein product of OCT4B1 mRNA could be truncated.Interestingly,we present here that OCT4B1 can indirectly produce the same protein products as OCT4B.We have demonstrated that OCT4B1 mRNA can be spliced into OCT4B mRNA,and encode three protein isoforms.The splicing of OCT4B1 mRNA into OCT4B mRNA can be remarkably inhibited by the mutation of the classical splicing site.Our result suggests that OCT4B mRNA may originate from OCT4B1 mRNA by alternative splicing.Yuan Gao Xia Wang Jin Han Zhifeng Xiao Bing Chen Guannan Su Jianwu Dai 2010Journal of Genetics and Genomics2010,37,7:6
13Virtual Machine-Based Task Scheduling Algorithm in a Cloud Computing Environment显示文摘Virtualization technology has been widely used to virtualize single server into multiple servers, which not only creates an operating environment for a virtual machine-based cloud computing platform but also potentially improves its efficiency. Currently, most task scheduling-based algorithms used in cloud computing environments are slow to convergence or easily fall into a local optimum. This paper introduces a Greedy Particle Swarm Optimization(G&PSO) based algorithm to solve the task scheduling problem. It uses a greedy algorithm to quickly solve the initial particle value of a particle swarm optimization algorithm derived from a virtual machine-based cloud platform. The archived experimental results show that the algorithm exhibits better performance such as a faster convergence rate, stronger local and global search capabilities, and a more balanced workload on each virtual machine. 15Efficient inversions and duplications of mammalian regulatory DNA elements and gene clusters by CRISPR/Cas9显示文摘The human genome contains millions of DNA regulatory elements and a large number of gene clusters,most of which have not been tested experimentally.The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated nuclease 9(Cas9)programed with a synthetic single-guide RNA(sgRNA)emerges as a method for genome editing in virtually any organisms.Here we report that targeted DNA fragment inversions and duplications could easily be achieved in human and mouse genomes by CRISPR with two sgRNAs.Specifically,we found that,in cultured human cells and mice,efficient precise inversions of DNA fragments ranging in size froma few tens of bp to hundreds of kb could be generated.In addition,DNA fragment duplications and deletions could also be generated by CRISPR through trans-allelic recombination between the Cas9-induced double-strand breaks(DSBs)on two homologous chromosomes(chromatids).Moreover,junctions of combinatorial inversions and duplications of the protocadherin(Pcdh)gene clusters induced by Cas9 with four sgRNAs could be detected.In mice,we obtained founders with alleles of precise inversions,duplications,and deletions of DNA fragments of variable sizes by CRISPR.Interestingly,we found that very efficient inversions were mediated by microhomology-mediated end joining(MMEJ)through short inverted repeats.We showed for the first time that DNA fragment inversions could be transmitted through germlines in mice.Finally,we applied this CRISPR method to a regulatory element of the Pcdha cluster and found a new role in the regulation of members of the Pcdhg cluster.This simple and efficient method should be useful in manipulating mammalian genomes to study millions of regulatory DNA elements as well as vast numbers of gene clusters.Jinhuan Li Jia Shou YaGuo Yuanxiao Tang YonghuWu Zhilian Jia Yanan Zhai Zhifeng Chen Quan Xu Qiang Wu 2015Journal of Molecular Cell Biology2015,7,4:4

17Comparison of yield performance between direct-seeded and transplanted double-season rice using ultrashort-duration varieties in central China显示文摘Labor scarcity requires double-season rice to be planted by direct seeding as an alternative to transplanting. Only ultrashort-duration varieties can be used in direct-seeded, double-season rice(DSD) in central China where thermal time is limited. Whether ultrashort-duration varieties grown in DSD can be as productive and efficient in nitrogen(N) use as transplanted double-season rice(TPD) remains unclear. Field experiments were conducted in Hubei province, central China with two establishment methods(DSD,TPD) and three N rates in the early and late seasons of 2017 and 2018. Nitrogen treatments included zero-N control(N0), total N rate of 60 kg N ha;with equal splits at basal, midtillering, and panicle initiation(N1), and weekly N application at 15 kg ha;from seeding/transplanting to heading(N2). Both early-and late-season rice under DSD matured within 95 days, on average 9 days shorter than rice under TPD. The grain yield of DSD was comparable to or higher than that of TDP in both seasons, although the daily yield was significantly higher under DSD than under TDP. Before heading, DSD had higher leaf area,stem number, intercepted radiation, and radiation use efficiency than TPD, which compensated for the negative effect of short growth duration on biomass production. Total dry weight and harvest index under DSD were comparable to or higher than those under TDP. In general, the recovery efficiency of fertilizer-N under DSD was higher than that under TPD, but the reverse was true for physiological N use efficiency. 18Thermal Analysis of a Volumetric Solar Receiver显示文摘The volumetric receiver has received wide attention due to its high thermal efficiency. This paper studied a new type of a solid-liquid composite volumetric receiver. The heat transfer in a solid-liquid composite volumetric solar receiver was analyzed using a one-dimensional unsteady simulation model of the solid-liquid receiver. The model included absorption of the incident solar radiation by the glass window, the silicon carbide porous ceramic heat absorber panel and the water. The results were verified against experimental data for a volumetric receiver and the error did not exceed 10%.
