| 1 | High expression of ACE2 receptor of 2019-nCoV on the epithelial cells of oral mucosa显示文摘It has been reported that ACE2 is the main host cell receptor of 2019-nCoV and plays a crucial role in the entry of virus into the cell to cause the final infection. To investigate the potential route of 2019-n Cov infection on the mucosa of oral cavity, bulk RNA-seq profiles from two public databases including The Cancer Genome Atlas(TCGA) and Functional Annotation of The Mammalian Genome Cap Analysis of Gene Expression(FANTOM5 CAGE) dataset were collected. RNA-seq profiling data of 13 organ types with para-carcinoma normal tissues from TCGA and 14 organ types with normal tissues from FANTOM5 CAGE were analyzed in order to explore and validate the expression of ACE2 on the mucosa of oral cavity. Further, single-cell transcriptomes from an independent data generated in-house were used to identify and confirm the ACE2-expressing cell composition and proportion in oral cavity. The results demonstrated that the ACE2 expressed on the mucosa of oral cavity. Interestingly, this receptor was highly enriched in epithelial cells of tongue. | 2 | Clinical significance of K-ras and BRAF mutations in Chinese colorectal cancer patients显示文摘AIM:To identify and assess mutations in the K-ras and BRAF genes in a cohort of Chinese patients with colorectal cancer (CRC) for their association with various clinicopathological parameters and prognosis.METHODS:Genomic DNA was isolated from frozen tissues.Pyrosequencing analysis was conducted to detect mutations in the K-ras (codons 12,13,and 61) and BRAF genes (codon 600).Statistical analysis was carried out using SPSS-15.0 software.RESULTS:Among the 118 colorectal cancer patients,we detected 41 (34.7%) mutations in the K-ras gene.Mutation frequencies at codon 12 and codon 13 were 23.7% (28/118) and 10.2% (12/118),respectively.Only one patient harbored a point mutation at codon 61 (0.8%,1/118).Gender was the only factor that showed an obvious relationship with K-ras gene mutation (female 44.7% vs male 28.2%,P=0.037).Other clinicopathological features,such as age,location of the tumor,tumor differentiation,Tumor,Node and Metastases classification,and the Union for International Cancer Control staging,showed no positive relationship with K-ras gene mutations.No significant correlation was observed between the presence of K-ras mutations (codons 12,13,and 61) and the survival of the patients.BRAF mutations were rare,and only two patients (1.7%) harbored a detectable mutation at codon 600.CONCLUSION:K-ras gene mutation is a common event in our 118 Chinese CRC patients,with an obvious relationship with gender.However,it seems not to be an independent prognostic factor in CRC patients.The BRAF gene is rarely mutated in Chinese CRC patients. | Hong Shen Ying Yuan Han-Guang Hu Xiao-Xian Ye Mo- Dan Li Xian Zhong Wei-Jia Fang Shu Zheng | 2011 | World Journal of Gastroenterology2011,17,6: | 29 |
| 3 | Biofeedback therapy for chronic pelvic pain syndrome显示文摘Aim: To evaluate the efficacy of biofeedback therapy in patients with chronic pelvic pain syndrome(CPPS). Methods: From November 2001 to April 2002, patients visiting the Urological Outpatient Clinic of this Hospital were evaluated by means of the National Institute of Health Chronic Prostatitis Symptom Index (NIH-CPSI)and classified by the NIH classification standard. Sixty-two patients of CPPS category Ⅲ were involved in this study. All patients had been treated by conventional approaches such as antibiotics and alpha-blockers for more than half a year without any improvement. The expressed prostatic secretion results were as follows: WBC 5 to 9/high power field, lipid +-+++ and bacterial culture negative. Their NIH-CPSI were 12-40. All the 62 cases complained of micturitional irritation (frequency, urgency, splitted stream and sense of residual urine), 32 cases, of pain or discomfort at the testicular, penile, scrotal, pelvic or rectal region and 13 cases, of white secretion-dripping. The patients were treated by the Urostym Biofeedback equipment (Laborie Co., Canada) 5 times a week for 2 weeks with a stimulus intensity of 15 mA-23 mA and duration of 20 minutes. Results: Sixty patients were significantly improved or cured, while no significant improvement in the remaining 2. No apparent side effect was observed. The NIH-CPSI dropped to 6 to 14 with an average reduction of 21 (P<0.01). In the 60 improved cases, pain was relieved after 2-3treatment courses and other symptoms disappeared after 4-5 courses. Conclusion: Biofeedback therapy is a safe and effective treatment for CPPS. Large randomized clinical trials are needed to confirm its efficacy and to explore the mechanism of action. | 4 | Indoor Allergen Levels and Household Distributions in Nine Cities Across China显示文摘Objective Chinese allergic subjects have high levels of sensitization to house dust mite(HDM) and other indoor allergens. This study quantifies common indoor allergen levels in Chinese households. Methods Dust samples were collected from nine cities. Major allergens Der p 1 and Der f 1 from Dermatophagoides pteronyssinus and D. farinae, and specific antigens of Blomia tropicalis, Tyrophagus putrescentiae, Acarus siro, and cockroach species Blattella germanica and Periplaneta americana were measured by ELISA. Results HDM allergens were found in dust samples from bedding in 95% of the Chinese households. The median levels varied from <0.006 to 9.2 μg/g of dust, depending on the city. The percentages of households having HDM allergen levels associated with the risk of developing allergy sensitization and asthma were 65% and 25%, respectively. Specific antigens of the storage mite and cockroach were only found in samples from the southern and tropical regions of China. Levels of mite allergens were generally higher in samples from bedding compared to samples from the living room, even for storage mites, whereas levels of cockroach antigens were higher in the living room samples. Conclusion HDM allergens are present in bedding dust samples from most Chinese households. Cities in southern and central China have relatively high levels of HDM major allergens compared to cities in northern and western China. | 6 | Application of gemstone spectral imaging for efficacy evaluation in hepatocellular carcinoma after transarterial chemoembolization显示文摘AIM: To assess the value of gemstone spectral imaging(GSI) in efficacy evaluation in hepatocellular cancer(HCC) after transcatheter arterial chemoembolization(TACE) treatment.METHODS: Thirty patients with HCC underwent GSI, including nonenhanced, arterial, portalvenous and delayed phase scans, after TACE treatment. Arterial phase images were acquired with GSI for reconstruction of virtual nonenhanced images and color overlay images. Digital subtraction angiography(DSA) was performed in all these patients. Two blinded and independent readers evaluated the data in two reading sessions; standard nonenhanced, arterial, portalvenous, and delayed phase images were read in session A, and the optimal monochromatic images, iodine/water based images and spectrum features were read in session B. Sensitivity and specificity were calculated with the DSA data as the reference standard. The sensitivity and specificity were compared using the χ2 test.RESULTS: DSA revealed 154 lesions in 30 patients, and 100 of them had blood supply. Overall sensitivity and specificity were 72%( 72/100) and 77.8 %(42/54) for session A, and 97%(97/100) and 94.4%(51/54) for session B, respectively. The sensitivity and specificity of the two reading sessions were significantly different(χ2 = 23.04, χ2 = 7.11, P < 0.05).CONCLUSION: Compared with conventional CT, GSI could significantly improve the detection of small and multiple lesions without increasing the radiation dose. | 9 | Head-related transfer function database and its analyses显示文摘Based on the measurements from 52 Chinese subjects (26 males and 26 females), a high-spatial-resolution head-related transfer function (HRTF) database with corre- sponding anthropometric parameters is established. By using the database, cues relating to sound source localization, including interaural time difference (ITD), interaural level difference (ILD), and spectral features introduced by pinna, are analyzed. Moreover, the statistical relationship between ITD and anthropometric parameters is estimated. It is proved that the mean values of maximum ITD for male and female are significantly different, so are those for Chinese and western sub- jects. The difference in ITD is due to the difference in individual anthropometric parameters. It is further proved that the spectral features introduced by pinna strongly depend on individual; while at high frequencies (f≥ 5.5 kHz), HRTFs are left-right asymmetric. | 10 | The prognostic value of serum C-reactive protein-bound serum amyloid A in early-stage lung cancer显示文摘Background:Elevated levels of serum C-reactive protein(CRP) have been reported to have prognostic significance in lung cancer patients.This study aimed to further identify CRP-bound components as prognostic markers for lung cancer and validate their prognostic value.Methods:CRP-bound components obtained from the serum samples from lung cancer patients or healthy controls were analyzed by differential proteomics analysis.CRP-bound serum amyloid A(CRP-SAA) was evaluated by coimmunoprecipitation(IP).Serum samples from two independent cohorts with lung cancer(retrospective cohort,242patients;prospective cohort,222 patients) and healthy controls(159 subjects) were used to evaluate the prognostic value of CRP-SAA by enzyme-linked immunosorbent assay.Results:CRP-SAA was identified specifically in serum samples from lung cancer patients by proteomic analysis.CRP binding to SAA was confirmed by co-IP in serum samples from lung cancer patients and cell culture media.The level of CRP-SAA was significantly higher in patients than in healthy controls(0.37 ± 0.58 vs.0.03 ± 0.04,P < 0.001).Elevated CRP-SAA levels were significantly associated with severe clinical features of lung cancer.The elevation of CRPSAA was associated with lower survival rates for both the retrospective(hazard ration[HR]= 2.181,95%confidence interval[CI]= 1.641-2.897,P < 0.001) and the prospective cohorts(HR = 2.744,95%CI = 1.810-4.161,P < 0.001).Multivariate Cox analysis showed that CRP-SAA was an independent prognostic marker for lung cancer.Remarkably,in stages l-ll patients,only CRP-SAA,not total SAA or CRP,showed significant association with overall survival in two cohorts.Moreover,univariate and multivariate Cox analyses also showed that only CRP-SAA could be used as an independent prognostic marker for early-stage lung cancer patients.Conclusion:CRP-SAA could be a better prognostic marker for lung cancer than total SAA or CRP,especially in earlystage patients. | Xue-Yan Zhang Ge Zhang Ying Jiang Dan Liu Man-Zhi Li Qian Zhong Shan-Qi Zeng Wan-Li Liu Mu-Sheng Zeng | 2015 | Chinese Journal of Cancer2015,34,8: | 10 |
| 11 | Construction of an integrated map and location of a bruchid resistance gene in mung bean显示文摘Bruchid beetle(Callosobruchus chinensis) poses a serious threat to the production and storage of mung bean(Vigna radiata). Mapping bruchid resistance(Br) will provide an important basis for cloning the responsible gene(s) and elucidating its functional mechanism, and will also facilitate marker-assisted selection in mung bean breeding. Here, we report the construction of the genetic linkage groups of mung bean and mapping of the Br1 locus using an RIL population derived from a cross between Berken, a bruchid-susceptible line, and ACC41, a bruchid-resistant line. A total of 560 markers were mapped onto 11 linkage groups,with 38.0% of the markers showing distorted segregation. The lengths of the linkage groups ranged from 45.2 to 117.0 c M with a total coverage of 732.9 c M and an average interval of1.3 c M between loci. Br1 was located on LG9 between BM202(0.7 c M) and Vr2-627(1.7 c M).Based on 270 shared SSR markers, most of the linkage groups were assigned to specific chromosomes. | 13 | Inhibition of phosphodiesterase 4 by FCPR16 protects SH-SY5Y cells against MPP^+ -induced decline of mitochondrial membrane potential and oxidative stress显示文摘Parkinson disease(PD) is a chronic neurodegenerative disorder caused by progressive dopaminergic neuronal death in the substantia nigra pars compacta within the midbrain.There still is no cure,effective treatments for PD,available therapies are only capable of offering temporary and symptomatic relief to the patients.There are certain patents that claim phosphodiesterase(PDE) inhibitors as possible anti-PD drugs,PDE4 is a promising target for the treatment of PD and the underlying mechanism has not yet been well elucidated.PDE4 is an enzyme that specifically hydrolyzes intracellular cyclic adenosine monophosphate(cAMP)throughout the body,including the brain.Most of the available PDE4 inhibitors exert unpleasant and serious side effects,such as emesis and nausea,which hinder its clinical application.Therefore,more efforts are needed before PDE4 inhibitors with high therapeutic indices are available for treatment of PD.FCPR16 is a novel PDE4 inhibitor with little emetic potential,which exhibits excellent enzyme inhibition activity(IC50=90 nmol·L^(-1)).METHODS SH-SY5 Y cell was induced with 1-methyl-4-phenylpyridinium(MPP+)to mimic PD cell injury in vitro,and CCK-8 assay was used to investigate the viability effects of different concentration of FCPR16(3.1-50 μmol·L^(-1)) on MPP+-injured SH-SY5 Y cells.Detection of apoptosis was performed by flow cytometry.The level of ntracellular reactive oxygen species was detected with the fluorescent probe DCFH-DA,and the mitochondrial membrane potential of cells in different experimental groups was detected with the JC-1 fluorescent probe.AO staining and Lysotracker Red staining were used to detect the intracellular antophagy changes.The expression of apoptosis related proteins,autophagy and other related signal molecules were demonstrated by Western blotting.Different cellular signaling pathway inhibitors were used to invesitigate the specific cellular mechanisms of FCPR16 protecting MPP+-induced cell injury.RESULTS FCPR16(12.5-50 μmol·L^(-1)) dose-dependently reduced MPP+-induced decline of cell viability,accompanied by reductions in nuclear condensation and lactate dehydrogenase release.The level of cleaved caspase 3 and the ratio of Bax/Bcl-2 were also decreased after treatment with FCPR16 in MPP+-treated cells.Furthermore,FCPR16(25 μmol·L^(-1)) significantly suppressed the accumulation of reactive oxygen species(ROS),prevented the decline of mitochondrial membrane potential(Δψm) and attenuated the expression of malonaldehyde level.Further studies disclosed that FCPR16 enhanced the levels of cA MP and the exchange protein directly activated by cA MP(Epac) in SHSY5 Y cel s.Western blotting analysis revealed that FCPR16 increased the phosphorylation of c AMP response element-binding protein(CREB) and protein kinase B(Akt)down-regulated by MPP+in SHSY5 Y cells.Moreover,the inhibitory effects of FCPR16 on the production of ROS and Δψm loss could be blocked by PKA inhibitor H-89 and Akt inhibitor KRX-0401.CONCLUSION The novel PDE4 inhibitor FCPR16 can protect against damaging pathways including oxidative stress,mitochondrial dysfunction and apoptosis in SH-SY5 Y cells.FCPR16 preventes MPP+-induced neurotoxicity through activation of cAMP/PKA/CREB and Epac/Akt signaling pathways.These may lead to develop mechanism based therapeutics and improved pharmacotherapy for PD.It is reasonable to assume that FCPR16 is a potential candidate for the prevention and treatment of PD. | ZHONG Jia-hong XIE Jin-feng XIAO Jiao LI Dan ZHOU Zhong-zhen WANG Hai-tao XU Jiang-ping | 2018 | 中国药理学与毒理学杂志2018,32,9: | 9 |
| 14 | Microbiologically influenced corrosion of titanium caused by aerobic marine bacterium Pseudomonas aeruginosa显示文摘Microbiologically influenced corrosion(MIC) is a big threat to the strength and safety of many metallic materials used in different environments throughout the world. The metabolites and bioactivity of the microorganisms cause severe deterioration on the metals. In this study, MIC of pure titanium(Ti) was studied in the presence of a highly corrosive aerobic marine bacterium Pseudomonas aeruginosa. The results obtained from electrochemical test showed that Ti was corrosion resistant in the abiotic culture medium after 14 d, while the increased corrosion current density(i_(corr)) obtained from polarization curves and the decreased charge transfer resistance(R_(ct)) from electrochemical impedance spectroscopy(EIS)indicated the accelerated corrosion of Ti caused by P. aeruginosa biofilm. For further confirmation of the above results, the surface of Ti was investigated using scanning electron microscopy(SEM), confocal laser scanning microscopy(CLSM) and X-ray photoelectron spectroscopy(XPS). According to the XPS results, TiO_2 was formed in both abiotic and biotic conditions, while unstable oxide Ti_2O_3 was detected in the presence of P. aeruginosa, leading to the defects in the passive film and localized corrosion. Pitting corrosion was investigated with the help of CLSM, and the largest pit depth found on Ti surface immersed in P. aeruginosa was 1.2 μm. | 15 | Development and Validation of a Model for Predicting Diabetic Nephropathy in Chinese People显示文摘Objective To develop a risk model for predicting later development of diabetic nephropathy(DN)in Chinese people with type 2 diabetes mellitus(T2DM)and evaluate its performance with independent validation.Methods We used data collected from the project‘Comprehensive Research on the Prevention and Control of Diabetes’,which was a community-based study conducted by the Jiangsu Center for Disease Control and Prevention in 2013.A total of 11,771 eligible participants were included in our study.The endpoint was a clear diagnosis of DN.Data was divided into two components:a training set for model development and a test set for validation.The Cox proportional hazard regression was used for survival analysis in men and women.The model’s performance was evaluated by discrimination and calibration.Results The incidence(cases per 10,000 person-years)of DN was 9.95(95%CI;8.66-11.43)in women and 11.28(95%CI;9.77-13.03)in men.Factors including diagnosis age,location,body mass index,high-density-lipoprotein cholesterol,creatinine,hypertension,dyslipidemia,retinopathy,diet control,and physical activity were significant in the final model.The model showed high discrimination and good calibration.Conclusion The risk model for predicting DN in people with T2DM can be used in clinical practice for improving the quality of risk management and intervention. | MIAO Dan Dan PAN En Chun ZHANG Qin SUN Zhong Ming QIN Yu WU Ming | 2017 | Biomedical and Environmental Sciences2017,30,2: | 8 |

| 18 | Sensory innervation around immediately vs. delayed loaded implants: a pilot study显示文摘Although neurophysiological and psychophysical proof of osseoperception is accumulating, histomorphometric evidence for the neural mechanisms of functional compensation following immediate and delayed implant loading is still lacking. For this randomized split-mouth study, six mongrel dogs randomly received one of four treatment protocols at 36 implant-recipient sites over 16 weeks(third maxillary incisor, third and fourth mandibular premolar): immediate implant placement and immediate loading(IIP1IL);delayed implant placement and delayed loading(DIP1DL); delayed implant placement and immediate loading(DIP1IL); and natural extraction socket healing(control). Histomorphometry was performed in the peri-implant bone and soft tissues within 300 mm around the implants. Immunocytochemistry and transmission electron microscopy were used to confirm the presence of neural structures and to reveal their ultrastructural characteristics, respectively. Myelinated nerve fibres densely populated the peri-implant crestal gingival and apical regions, although they were also identified in the woven bone and in the osteons near the implant threads. Compared with the control group in the mandible, the group that received IIP1 IL showed a higher innervation(in Nmm22, 5.9461.12 vs. 3.1560.63,P,0.001) and smaller fibre diameter(in mm, 1.3760.05 vs. 1.6460.13, P50.016), smaller axon diameter(in mm, 0.8960.05 vs.1.2460.10, P50.009) and g-ratio(0.6460.04 vs. 0.7660.05, P,0.001) in the middle region around the implants. Compared with DIP1 IL in the mandible, IIP1 IL had a higher nerve density(in Nmm22, 13.2362.54 vs. 9.6461.86, P50.027), greater fibre diameter(in mm, 1.3260.02 vs. 1.2060.04, P50.021), greater axon diameter(in mm, 0.9260.01 vs. 0.8960.03, P50.035) and lower g-ratio(0.6960.01 vs. 0.7460.01, P50.033) in the apical region around the implants.
