1Oleic Acid and Eicosapentaenoic Acid Reverse Palmitic Acid-induced Insulin Resistance in Human HepG2 Cells via the Reactive Oxygen Species/JUN Pathway显示文摘Oleic acid(OA),a monounsaturated fatty acid(MUFA),has previously been shown to reverse saturated fatty acid palmitic acid(PA)-induced hepatic insulin resistance(IR).However,its underlying molecular mechanism is unclear.In addition,previous studies have shown that eicosapentaenoic acid(EPA),aω-3 polyunsaturated fatty acid(PUFA),reverses PA-induced muscle IR,but whether EPA plays the same role in hepatic IR and its possible mechanism involved need to be further clarified.Here,we confirmed that EPA reversed PA-induced IR in HepG2 cells and compared the proteomic changes in HepG2 cells after treatment with different free fatty acids(FFAs).A total of 234 proteins were determined to be differentially expressed after PA+OA treatment.Their functions were mainly related to responses to stress and endogenous stimuli,lipid metabolic process,and protein binding.For PA+EPA treatment,the PA-induced expression changes of 1326 proteins could be reversed by EPA,415 of which were mitochondrial proteins,with most of the functional proteins involved in oxidative phosphorylation(OXPHOS)and tricarboxylic acid(TCA)cycle.Mechanistic studies revealed that the protein encoded by JUN and reactive oxygen species(ROS)play a role in OA-and EPA-reversed PA-induced IR,respectively.EPA and OA alleviated PA-induced abnormal adenosine triphosphate(ATP)production,ROS generation,and calcium(Ca^(2+))content.Importantly,H_(2)O_(2)-activated production of ROS increased the protein expression of JUN,further resulting in IR in HepG2 cells.Taken together,we demonstrate that ROS/JUN is a common response pathway employed by HepG2 cells toward FFA-regulated IR.Yaping Sun Jifeng Wang Xiaojing Guo Nali Zhu Lili Niu Xiang Ding Zhensheng Xie Xiulan Chen Fuquan Yang 2021Genomics, Proteomics & Bioinformatics2021,19,5:6
2Long-range atmospheric transport and alpine condensation of short-chain chlorinated paraffins on the southeastern Tibetan Plateau显示文摘Pristine alpine regions are ideal regions for investigating the long-range atmospheric transport and cold trapping effects of short chain chlorinated paraffins(SCCPs).The concentrations and alpine condensation of SCCPs were investigated in lichen samples collected from the southeastern Tibetan Plateau.The concentrations of SCCPs ranged from 3098 to 6999 ng/g lipid weight(lw)and appeared to have an increasing trend with altitude.For congeners,C10 dominated among all the congener groups.The different environmental behavior for different congener groups was closely related to their octanol-air partition coefficient(Koa).C10 congeners showed an increasing trend with altitude,whereas C13 congeners were negatively correlated with altitude.Volumetric bioconcentration factors(BCF)of SCCPs reached 8.71 in lichens,which were higher than other semivolatile organic compounds(SVOCs)such as organochlorine pesticides(OCPs),polybrominated diphenyl ethers(PBDEs),polychlorinated biphenyls(PCBs),and hexabromocyclododecane(HBCD).These results suggested that SCCPs were prone to accumulate in the lichen from the air and provided evidence for the role of lichens as a suitable atmospheric indicator in the Tibetan Plateau.Huijuan Li Duo Bu Yan Gao Nali Zhu Jing Wu Xiangfeng Chen Jianjie Fu Yawei Wang Aiqian Zhang Guibin Jiang 2021Journal of Environmental Sciences2021,33,1:1

7Facet-dependent transformation and toxicity of nanoscale zinc oxide in the synthetic saliva显示文摘The nanoscale zinc oxide(n-ZnO)was used in food packages due to its superior antibac terial activity,resulting in potential intake of n-ZnO through the digestive system,wherein n-ZnO interacted with saliva.In recent,facet engineering,a technique for controlling the exposed facets,was applied to n-ZnO,whereas risk of n-ZnO with specific exposed facets in saliva was ignored.ZnO nanoflakes(ZnO-0001)and nanoneedles(ZnO-1010)with the pri mary exposed facets of{0001}and{1010}respectively were prepared in this study,investigat ing stability and toxicity of ZnO-0001 and ZnO-1010 in synthetic saliva.Both ZnO-0001 and ZnO-1010 partially transformed into amorphous Zn_(3)(PO_(4))_(2)within 1 hr in the saliva even containing orgnaic components,forming a ZnO-Zn_(3)(PO_(4))_(2)core-shell structure.Neverthe less,ZnO-1010 relative to ZnO-0001 would likely transform into Zn_(3)(PO_(4))_(2),being attributed to superior dissolution of{1010}facet due to its lower vacancy formation energy(1.15 eV than{0001}facet(3.90 eV)).The toxicity of n-ZnO to Caco-2 cells was also dependent on the primary exposed facet;ZnO-0001 caused cell toxicity through oxidative stress,whereas ZnO-1010 resulted in lower cells viability than ZnO-0001 through oxidative stress and mem brane damage.Density functional theory calculations illustrated that·O_(2)^(-)was formed and released on{1010}facet,yet O_(2)^(2-)instead of·O_(2)^(-)was generated on{0001}facet,leading to low oxidative stress from ZnO-0001.All findings demonstrated that stability and toxicity of n-ZnO were dependent on the primary exposed facet,improving our understanding o health risk of nanomaterials.Xiang Zhang Yunpeng Huang Jikun Wang Jie Tang YangMei Nali Zhu Zhigang Li Lingxiangyu Li YaweiWang 2024Journal of Environmental Sciences2024,,5:0
8Mitochondrion targeting peptide-modified magnetic graphene oxide delivering mitoxantrone for impairment of tumor mitochondrial functions显示文摘In this study,we prepared mitochondrion targeting peptide-grafted magnetic graphene oxide(GO)nanocarriers for efficient impairment of the tumor mitochondria.The two-dimensional GOMNP-MitP nanosheets were synthesized by grafting magnetic y-Fe_(2)O_(3)to the surface of GO,followed by covalent modification of mitochondrion targeting peptide(MitP).GOMNP-MitP exhibited the high capacity of loading the anticancer drug mitoxantrone(MTX),and preferentially targeted the tumor mitochondria.With the aid of alternating magnetic field(AMF),the MTX-loading GOMNP-MitP released MTX to the mitochondria,severely impairing mitochondrial functions,including attenuation of ATP production,decrease in mitochondrial membrane potential(MMP),and further leading to activation of apoptosis.This study realized high-efficient mitochondrion-ta rgeting drug delivery for anticancer therapy by twodimensional nanoplatforms.Hangqi Zhu Bing Zhang Nali Zhu Mingchun Li Qilin Yu 2021Chinese Chemical Letters2021,32,3:0
9Effects of Forest Filtering and Cold Trapping on Polycyclic Aromatic Hydrocarbons Distribution in Southeast Tibet显示文摘New pollutants in remote regions have become a global issue of concern.This study collected soil,lichen,moss,and air samples from the high mountain area of southeastern Tibet to analyze the altitude pollution distribution of 16 polycyclic aromatic hydrocarbons(PAHs)and the related influencing factors.The results revealed that the average levels ofΣ6PAH in soil,moss,lichen,and air samples were 46.8 ng/g dry weight(dw),140.6 ng/g dw,712.0 ng/g dw,and 60.1 ng/m^(3),respectively.Naphthalene and phenanthrene were the most abundant individual isomers,and 2–4 rings PAHs accounted for approximately 90%of the totalΣPAH concentration in the samples.Source apportionment analysis highlighted the impact of atmospheric PAH inputs on this region.Octanol/air distribution factor(K_(oa)),altitude,plant coverage,and soil organic matter were the key determinants of PAH concentration along the altitudinal gradient.Across all the soil sampling sites,the average contribution of the forest filter effect(FFE)was greater than that of the mountain cold trapping effect based on the principal component analysis and multiple linear regression.The findings of this study strengthened the reasons for the preferential accumulation of organic pollutants in the high-altitude mountainous area,and provided important insights for effective environmental strategies of new pollutants.ZHU Nali LI Yanping JIANG Lu LI Lingxiangyu LI Zhigang WANG Yawei JIANG Guibin 2023Chemical Research in Chinese Universities2023,39,3:0
