Lignin Interaction with Cellulase during Enzymatic Hydrolysis显示文摘The conversion of lignocellulosic biomass into biofuels or biochemicals typically involves a pretreatment process followed by the enzyme-catalyzed hydrolysis of cellulose and hemicellulose components to fermentable sugars. Many factors can contribute to the recalcitrance of biomass,e.g.,the lignin content and structure, crystallinity of cellulose, degree of fiber polymerization, and hemicellulose content, among others. However, nonproductive binding between cellulase and lignin is the factor with the greatest impact on enzymatic hydrolysis. To reduce the nonproductive adsorption of enzymes on lignin and improve the efficiency of enzymatic hydrolysis, this review comprehensively summarized the progress that has been made in understanding the interactions between lignin and enzymes.Firstly, the effects of pretreatment techniques on lignin content and enzymatic hydrolysis were reviewed. The effects of lignin content and functional groups on enzymatic hydrolysis were then summarized. Methods for the preparation and characterization of lignin films were assessed. Finally, the methods applied to characterize the interactions between lignin and cellulase were reviewed, and methods for decreasing the nonproductive binding of enzymes to lignin were discussed. This review provides an overview of the current understanding of how lignin hinders the enzymatic hydrolysis of lignocellulosic biomass, and provides a theoretical basis for the development of more economical and effective methods and additives to reduce the interaction of lignin and enzymes to improve the efficiency of enzymatic hydrolysis. | Mingfu Li Qingtong Zhang Changzhou Chen Shuangfei Wang Douyong Min | 2019 | Paper And Biomaterials2019,4,4: | 12
Experiment on Hydraulic Fracturing in Rock and Induced Earthquake显示文摘Experiment on rock hydraulic fracturing strength under different confining pressures was conducted on a series of test specimens with various pre-cracks prepared from 7 types of rock. Combining the data of an actual reservoir-induced earthquake with the experimental results of the contemporary tectonic stress field according to the theory of rock strength and the principle and method of rock fracture mechanics, the authors tentatively investigated the earthquakes induced by pore-water pressure in rock and obtained the initial results as follows: (1) One type of induced earthquake may occur in the case of larger tectonic stress on such weak planes that strike in similar orientation of principle tectonic compressional stress in the shallows of the rock mass; the pore-water pressure σp may generate tensile fracture on them and induce small earthquakes; (2) Two types of induced earthquake may occur in the case of larger tectonic stress, i.e.,① on such weakness planes that strike in similar orientation of principle tectonic compressional stress, σ1, in the shallows of the rockmass, the pore-water pressure, σp, may generate tensile fracture on them and induce small earthquakes; ② When the tectonic stress approximates the shear strength of the fracture, the pore-water pressure σp may reduce the normal stress, σn, on the fracture face causing failure of the originally stable fracture, producing gliding fracture and thus inducing an earthquake. σp may also increase the fracture depth, leading to an induced earthquake with the magnitude larger than the previous potential magnitude; (3) There is a depth limit for each type of rock mass, and no induced earthquake will occur beyond this limit. | Yan Yuding Li Yalin Zhang Zhuan Ouyang Lisheng Xie Mingfu | 2005 | Earthquake Research in China2005,19,4: | 4
Serum semaphorin 4C as a diagnostic biomarker in breast cancer:A multicenter retrospective study显示文摘Background:To date, there is no approved blood-based biomarker for breast cancer detection.Herein,we aimed to assess semaphorin 4C(SEMA4C),a pivotal protein involved in breast cancer progression,as a serum diagnostic biomarker. Methods:We included 6,213 consecutive inpatients from Tongji Hospital,Qilu Hospital, and Hubei Cancer Hospital.Training cohort and two validation cohorts were introduced for diagnostic exploration and validation.A pan-cancer cohort was used to independently explore the diagnostic potential of SEMA4C among solid tumors.Breast cancer patients who underwent mass excision prior to modified radical mastectomy were also analyzed. We hypothesized that increased pretreatment serum SEMA4C levels, measured using optimized in-house enzymelinked immunosorbent assay kits, could detect breast cancer. The endpoints were diagnostic performance, including area under the receiver operating characteristic curve(AUC),sensitivity, and specificity.Post-surgery pathological diagnosis was the reference standard and breast cancer staging followed the TNM classification. There was no restriction on disease stage for eligibilities. Results:We included 2667 inpatients with breast lesions,2378 patients with other solid tumors, and 1168 healthy participants.Specifically,118 patients with breast cancer were diagnosed with stage 0(5.71%),620 with stage I(30.00%),966 with stage II(46.73%),217 with stage III(10.50%),and 8 with stage IV(0.39%).Patients with breast cancer had significantly higher serum SEMA4C levels than benign breast tumor patients and normal controls(P<0.001).Elevated serum SEMA4C levels had AUC of 0.920(95%confidence interval[CI]:0.900–0.941)and 0.932(95%CI:0.911–0.953)for breast cancer detection in the two validation cohorts. The AUCs for detecting early-stage breast cancer(n=366)and ductal carcinoma in situ(n=85)were 0.931(95%CI:0.916–0.946)and 0.879(95%CI:0.832–0.925),respectively.Serum SEMA4C levels significantly decreased after surgery, and the reduction was more striking after modified radical mastectomy, compared with mass excision(P<0.001).The positive rate of enhanced serum SEMA4C levels was 84.77%for breast cancer and below 20.75%for the other 14 solid tumors. Conclusions:Serum SEMA4C demonstrated promising potential as a candidate biomarker for breast cancer diagnosis. However, validation in prospective settings and by other study groups is warranted. | Ya Wang Long Qiao Jie Yang Xiong Li Yaqi Duan Jiahao Liu Shaoqi Chen Huayi Li Dan Liu Tian Fang Jingjing Ma Xiaoting Li Fei Ye Junxiang Wan Juncheng Wei Qin Xu Ensong Guo Ping Jin Mingfu Wu Lin Zhang Yun Xia Yaqun Wu Jun Shao Yaojun Feng Qing Zhang Zongyuan Yang Gang Chen Qinghua Zhang Xingrui Li Shixuan Wang Junbo Hu Xiaoyun Wang Mona P.Tan Kazuaki Takabe Beihua Kong Qifeng Yang Ding Ma Qinglei Gao | 2021 | Cancer Communications2021,41,12: | 3

Spontaneous movement of a retrotransposon generated genic dominant male sterility providing a useful tool for rice breeding显示文摘Male sterility in plants provides valuable breeding tools in germplasm innovation and hybrid crop production. However, genetic resources for dominant genic male sterility, which hold great promise to facilitate breeding processes, are extremely rare in natural germplasm. Here we characterized the Sanming Dominant Genic Male Sterility in rice and identified the gene SDGMS using a map-based cloning approach. We found that spontaneous movement of a 1978-bp long terminal repeat(LTR)retrotransposon into the promoter region of the SDGMS gene activates its expression in anther tapetum, which causes abnormal programmed cell death of tapetal cells resulting in dominant male sterility.SDGMS encodes a ribosome inactivating protein showing N-glycosidase activity. The activation of SDGMS triggers transcription reprogramming of genes responsive to biotic stress leading to a hypersensitive response which causes sterility. The results demonstrate that an ectopic gene activation by transposon movement can give birth to a novel trait which enriches phenotypic diversity with practical utility. | Conghao Xu Yifeng Xu Zhengji Wang Xiaoyu Zhang Yuying Wu Xinyan Lu Hongwei Sun Lei Wang Qinglu Zhang Qinghua Zhang Xianghua Li Jinghua Xiao Xu Li Mingfu Zhao Yidan Ouyang Xianbo Huang Qifa Zhang | 2023 | National Science Review2023,10,9: | 1

HIF-1α promotes SARS-CoV-2 infection and aggravates inflammatory responses to COVID-19显示文摘Cytokine storm induced by Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)is a major pathological feature of Coronavirus Disease 2019(COVID-19)and a crucial determinant in COVID-19 prognosis.Understanding the mechanism underlying the SARS-CoV-2-induced cytokine storm is critical for COVID-19 control. Here,we identify that SARS-CoV-2 ORF3a and host hypoxia-inducible factor-1α(HIF-1α)play key roles in the virus infection and pro-inflammatory responses. | Mingfu Tian Weiyong Liu Xiang Li Peiyi Zhao Muhammad Adnan Shereen Chengliang Zhu Shanyu Huang Siyu Liu Xiao Yu Miaomiao Yue Pan Pan Wenbiao Wang Yongkui Li Xulin Chen Kailang Wu Zhen Luo Qiwei Zhang Jianguo Wu | 2021 | Signal Transduction and Targeted Therapy2021,6,9: | 1
Effect of Shouldering Angle on Distribution of Thermal Stress in Sapphire Single Crystal Growth Using Improved Kyropoulos显示文摘A two-dimensional model was established in the rectangular co-ordinate to study the thermal stress in the sapphire single crystal grown by the improved Kyropoulos. In the simulation, the distribution, the maximum and minimum values of the thermal stress were calculated. In addition, the relationship between the thermal stress and the shouldering angles was obtained that for lower shouldering angles, the maximum of the thermal stress value is lower and the minimum value is higher. It indicates that the distribution of the thermal stress can be improved by decreasing the shouldering angles of the crystal during the growth process. To evaluate the model, the experiment was carried out and the results are in good agreement with the calculation. | Yao Tai Han Jiecai Zuo Hongbo Meng Songhe Zhang Mingfu | 2006 | Journal of Rare Earths2006,24,z1: | 1
