| 1 | Improvement of wheat drought and salt tolerance by expression of a stress- inducible transcription factorGmDREB of soybean (Glycinemax)显示文摘Under stress conditions such as droughthigh-salinity and low-temperature, the transcription factorof DREB (dehydration responsive element binding proteins)improved efficiently stress resistance by regulating the ex-pression of its downstream genes with various environmentastress resistance in plants. GmDREB gene (GenBank Acces-sion No. AF514908) encoding a stress-inducible transcriptionfactor was cloned by screening a cDNA library of Glycinemax cv. Jinong 27 with yeast one-hybrid method. GmDREBgene was 910 bp in length and encoded 174 amino acids con-taining a conserved AP2/EREBP DNA-binding domain of 58amino acids. Two conserved functional amino acids, valineand glutamic acid, were located on the 14th and the 19thamino acid residues in the conserved structural domain. Analkaline amino acid region (KKR) related to a nuclear local-ization signal was at the N-terminal, while an acidic aminoacid region (DDD) related to trans-activation was at theC-terminal. Plant expression vectors were constructed andtransformed into wheat by bombardment. In total, 13 trans-genic plants with Ubi::GmDREB and 11 transgenic plantswith rd29A::GmDREB were identified from 103 regenerationplants by molecular analysis. The drought and salt tolerancesof T1 transgenic lines with Ubi::GmDREB orrd29A::GmDREB were demonstrated to be improved ascompared to wild type. | 2 | Expression of an(E)-β-farnesene synthase gene from Asian peppermint in tobacco affected aphid infestation显示文摘Aphids are major agricultural pests that cause significant yield losses in crop plants each year.(E)-β-farnesene(EβF) is the main or only component of an alarm pheromone involved in chemical communication within aphid species and particularly in the avoidance of predation. EβF also occurs in the essential oil of some plant species, and is catalyzed by EβF synthase. By using oligonucleotide primers designed from the known sequence of an EβF synthase gene from black peppermint(Mentha × piperita), two cDNA sequences, MaβFS1 and MaβFS2, were isolated from Asian peppermint(Mentha asiatica). Expression pattern analysis showed that the MaβFS1 gene exhibited higher expression in flowers than in roots, stems and leaves at the transcriptional level. Overexpression of MaβFS1 in tobacco plants resulted in emission of pure EβF ranging from 2.62 to 4.85 ng d-1g-1of fresh tissue. Tritrophic interactions involving peach aphids(Myzus persicae), and predatory lacewing(Chrysopa septempunctata) larvae demonstrated that transgenic tobacco expressing MaβFS1 had lower aphid infestation. | 11 | Case report of antiseizure medicine-induced long QT syndrome and a literature review显示文摘Background To realize the clinical characteristics of long QT syndrome(LQTS)caused by antiseizure medicines(ASMs),and to improve the prevention and management of ASM-acquired QT syndrome.Case presentation A case of ASM-acquired QT syndrome was diagnosed and relevant literature was reviewed.The case was a 7-year-old boy who presented with a sudden onset of panic followed by changes in consciousness,with or without convulsions,lasting from tens of seconds to 3 min.The patient then received antiepileptic treatment with valproic acid,levetiracetam and oxcarbazepine and was seizure free for about a year.However,on August 12,2021,his illness flared up again.Electroencephalogram(EEG)showed the background activity was slow,and no obvious epileptic discharge was detected.But electrocardiogram(ECG)showed a surprisingly prolonged QT interval(770 ms).Torsades de Pointes was found during Holter monitoring,while electrolyte levels were normal.The ECG recordings gradually returned to normal after stopping ASMs.For literature search,only 21 related papers were obtained after reading titles and full-texts of 105 English-language papers retrieved using keywords'acquired QT interstitial syndrome/acquired Long QT Syndrome(aLQTS)'and'anti-epileptic seizure drugs/ASMs',in the databases of Wanfang,CNKI,Pubmed,and other databases,from publication year 1965 to October 26,2021.There are 12 types of drug-acquired LQTS caused by ASMs,most of which are Na+blockers,but LQTS caused by oxcarbazepine had not been reported previously.Conclusions ASMs such as oxcarbazepine can cause acquired LQTS.When Na+or K+channel blockers are used clinically,ECG should be reviewed regularly and abnormal ECG should be intervened in time to reduce iatrogenic accidents in patients with epilepsy. | Xiongying Yu Jian Zha Zhaoshi Yi Xiaoyan Li Hui Chen Yuxin Xu Jianmin Zhong | 2022 | Acta Epileptologica2022,5,4: | 0 |
| 12 | TaNAC48 positively regulates drought tolerance and ABA responses in wheat(Triticum aestivum L.)显示文摘NAC family transcription factors(TFs) are important regulators in plant development and stress responses. However, the biological functions of NAC TFs in wheat are rarely studied. In this study, 43 putative drought-induced NAC genes were identified from de novo transcriptome sequencing data of wheat following drought treatment. Twelve wheat NACs along with ten known stress-related NACs from Arabidopsis and rice were clustered into Group II based on a phylogenetic analysis. Ta NAC48, which showed a higher and constitutive expression level in Group Ⅱ, was selected for further investigation.Ta NAC48 transcript was up-regulated by drought, PEG, H_(2)O_(2) and abscisic acid(ABA) treatment and encoded a nuclear localized protein. Overexpression of Ta NAC48 significantly promoted drought tolerance with increased proline content, and decreased rates of water loss, malondialdehyde(MDA), H_(2)O_(2) and O_(2)^(-) content. Root length and a stomatal aperture assay confirmed that Ta NAC48-overexpression plants increased sensitivity to ABA. Electrophoretic mobility shift assay(EMSA) and luciferase reporter analysis indicated that Ta AREB3 could bind to a cis-acting ABA-responsive element(ABRE) on Ta NAC48 promoter and activate the expression of Ta NAC48.
