1Biological drug and drug delivery-mediated immunotherapy显示文摘The initiation and development of major infammatory diseases,i.e.,cancer,vascular infammation,and some autoimmune diseases are closely linked to the immune system.Biologics-based immunotherapy is exerting a critical role against these diseases,whereas the usage of the immunomodulators is always limited by various factors such as susceptibility to digestion by enzymes in vivo,poor penetration across biological barriers,and rapid clearance by the reticuloendothelial system.Drug delivery strategies are potent to promote their delivery.Herein,we reviewed the potential targets for immunotherapy against the major infammatory diseases,discussed the biologics and drug delivery systems involved in the immunotherapy,particularly highlighted the approved therapy tactics,and finally offer perspectives in this feld.Qingqing Xiao Xiaotong Li Yi Li Zhenfeng Wu Chenjie Xu Zhongjian Chen Wei He 2021Acta Pharmaceutica Sinica B2021,11,4:15
2Astronomical tuning and magnetostratigraphy of Neogene biogenic reefs in Xisha Islands, South China Sea显示文摘Biogenic reefs are one of two major depositional types in the South China Sea, and are constructed by coral, algae and bryozoa. The West Pacific is a major area of biogenic reef development and plays a critical role in the global carbon cycle. However, the lack of geochronological studies in previous works inhibits our understanding of their contributions. Herein, we conduct a cyclostratigraphic and magnetostratigraphic study on Neogene biogenic reefs using the XK–1 core that was drilled at the Shidao Island,Xisha(Paracel) Islands. The main findings of this study are:(1) the establishment of reliable magentostratigraphy for Ledong, Huangliu, Meishan and Sanya Formations;(2) the magnetic susceptibility variation can be inferred as growth index and tuned to the 405–ka long eccentricity cycle;(3) the astronomical geochronology suggests that the bottom ages for Ledong, Yinggehai, Huangliu, Meishan,and Sanya Formations are 2.2 Ma, 5.7 Ma, 10.4 Ma, 16.6 Ma, and 24.3 Ma, respectively; and (4) Earth's eccentricity and obliquity played predominant roles in biogenic reef establishment on orbital to tectonic timescales. 3Hypertrophic adenoids in patients with nasopharyngeal carcinoma:appearance at magnetic resonance imaging before and after treatment显示文摘Introduction:Patients with nasopharyngeal carcinoma(NPC) sporadically develop abnormal adenoids.Nasopharyngeal adenoids are usually included in the gross tumor volume(GTV) but may have different therapeutic responses than tumor tissue.Therefore,distinguishing adenoids from tumor tissue may be required for precise and efficient chemoradiotherapy and radiotherapy.We characterized nasopharyngeal adenoids and investigated the therapeutic responses of NPC and nasopharyngeal adenoids using magnetic resonance imaging(MRI).Methods:MRI data from 40 NPC patients with a coexisting adenoid mass before and after treatment were analyzed.The features of the adenoid masses,including location,striped appearance,size,interface,symmetry/asymmetry,and cysts,were evaluated.Treatment response were scored according to the World Health Organization guidelines.Results:A striped appearance was observed in 36 cases before treatment and in all cases after treatment.In these 36 cases,the average GTVs including and excluding the uninvolved adenoids were 19.8 cm^3 and 14.8 cm^3,respectively.The average percentage change after excluding the uninvolved adenoids from the GTV was 31.0%.Stable disease in the adenoids was identified in 27(96.4%) of 28 patients after neoadjuvant chemotherapy,while NPC clearly regressed.Partial adenoid responses were identified in 33(82.5%) of 40 patients at 3 months after chemoradiotherapy or radiotherapy,whereas complete tumor responses were achieved in all patients.Six months after treatment,the adenoids continued to atrophy but did not disappear,and tumor recurrence was not found.Conclusions:Nasopharyngeal adenoids and carcinoma tissue in NPC patients can be distinguished by using MRI and have different responses to chemoradiotherapy and radiotherapy.These findings contribute to better delineating the GTV of NPC,based on which spatially optimized strategies can be developed to render precise and efficient chemoradiotherapy and radiotherapy.Additionally,we observed a clear difference in the responses of these two tissue types to current therapies.This finding may reduce or avoid unnecessary biopsies or overtreatment.Yao-Pan Wu Pei-Qiang Cai Li Tian Jie-Hua Xu Richard Alan Mitteer Jr Yi Fan Zhenfeng Zhang 2015Chinese Journal of Cancer2015,34,3:4
4Changes in progenitors and differentiated epithelial cells of neonatal piglets显示文摘This study aimed to assess the changes of small intestinal morphology,progenitors,differentiated epithelial cells,and potential mechanisms in neonatal piglets.Hematoxylin and eosin staining of samples from 36 piglets suggested that dramatic changes were observed in the jejunum crypts depth and crypt fission index of neonatal piglets(P<0.001).The number of intestinal stem cells(ISC)tended to increase(P<0.10),and a decreased number of enteroendocrine cells appeared in the jejunal crypt on d 7(P<0.05).Furthermore,the mRNA expression of jejunal chromogranin A(ChgA)was down-regulated in d 7 piglets(P<0.05).There was an up-regulation of the adult ISC marker gene of SPARC related modular calcium binding 2(Smoc2),and Wnt/b-catenin target genes on d 7(P<0.05).These results were further verified in vitro enteroid culture experiments.A mass of hollow spheroids was cultured from the fetal intestine of 0-d-old piglets(P<0.001),whereas substantial organoids with budding and branching structures were cultured from the intestine of 7-d-old piglets(P<0.001).The difference was reflected by the organoid budding efficiency,crypt domains per organoid,and the surface area of the organoid.Furthermore,spheroids on d 0 had more Ki67-positive cells and enteroendocrine cells(P<0.05)and showed a decreasing trend in the ISC and goblet cells(P<0.10).Moreover,the mRNA expression of spheroids differed markedly from that of organoids,with low expression of intestinal differentiation gene(Lysozyme;P<0.05),epithelial-specific markers(Villin,E-cadherin;P<0.05),and adult ISC markers(leucine-rich repeat-containing G protein-coupled receptor 5[Lgr5],Smoc2;P<0.001),and upregulation of fetal marker(connexin 43[Cnx43];P<0.05).The mRNA expression of relevant genes was up-regulated,and involved in Wnt/b-catenin,epidermal growth factor(EGF),Notch,and bone morphogenetic protein(BMP)signaling on d 7 organoids(P<0.05).Spheroids displayed low differentiated phenotype and high proliferation,while organoids exhibited strong differentiation potential.These results indicated that the conversion from the fetal progenitors(spheroids)to adult ISC(normal organoids)might largely be responsible for the fast development of intestinal epithelial cells in neonatal piglets.Lanmei Yin Jun Li Yitong Zhang Qing Yang Cuiyan Yang Zhenfeng Yi Yuebang Yin Qiye Wang Jianzhong Li Nengshui Ding Zhigang Zhang Huansheng Yang Yulong Yin 2022Animal Nutrition2022,,1:3
5Step-wise CAG@PLys@PDA-Cu^(2+)modification on micropatterned nanofibers for programmed endothelial healing显示文摘Native-like endothelium regeneration is a prerequisite for material-guided small-diameter vascular regeneration.In this study,a novel strategy is proposed to achieve phase-adjusted endothelial healing by step-wise modification of parallel-microgroove-patterned(i.e.,micropatterned)nanofibers with polydopamine-copper ion(PDA-Cu^(2+))complexes,polylysine(PLys)molecules,and Cys-Ala-Gly(CAG)peptides(CAG@PLys@PDA-Cu^(2+)).Using electrospun poly(L-lactide-co-caprolactone)random nanofibers as the demonstrating biomaterial,step-wise modification of CAG@PLys@PDA-Cu^(2+)significantly enhanced substrate wettability and protein adsorption,exhibited an excellent antithrombotic surface and outstanding phase-adjusted capacity of endothelium regeneration involving cell adhesion,endothelial monolayer formation,and the regenerated endothelium maturation.Upon in vivo implantation for segmental replacement of rabbit carotid arteries,CAG@PLys@PDA-Cu^(2+)modified grafts(2 mm inner diameter)with micropatterns on inner surface effectively accelerated native-like endothelium regeneration within 1 week,with less platelet aggregates and inflammatory response compared to those on non-modified grafts.Prolonged observations at 6-and 12-weeks post-implantation demonstrated a positive vascular remodeling with almost fully covered endothelium and mature smooth muscle layer in the modified vascular grafts,accompanied with well-organized extracellular matrix.By contrast,non-modified vascular grafts induced a disorganized tissue formation with a high risk of thrombogenesis.In summary,step-wise modification of CAG@PLys@PDA-Cu^(2+)on micropatterned nanofibers can significantly promote endothelial healing without inflicting thrombosis,thus confirming a novel strategy for developing functional vascular grafts or other blood-contacting materials/devices.Bingcheng Yi Boya Zhou Zhenfeng Song Lei Yu Wenbo Wang Wei Liu 2023Bioactive Materials2023,,7:1
6Pulmonary endothelium-targeted nanoassembly of indomethacin and superoxide dismutase relieves lung inflammation显示文摘Lung inflammation is an essential inducer of various diseases and is closely related to pulmonary-endothelium dysfunction.Herein,we propose a pulmonary endothelium-targeted codelivery system of anti-inflammatory indomethacin(IND)and antioxidant superoxide dismutase(SOD)by assembling the biopharmaceutical SOD onto the“vector”of rod-like pure IND crystals,followed by coating with anti-ICAM-1 antibody(Ab)for targeting endothelial cells.The codelivery system has a 237 nm diameter in length and extremely high drug loading of 39%IND and 2.3%SOD.Pharmacokinetics and biodistribution studies demonstrate the extended blood circulation and the strong pulmonary accumulation of the system after intravenous injection in the lipopolysaccharide(LPS)-induced inflammatory murine model.Particularly,the system allows a robust capacity to target pulmonary endothelium mostly due to the rod-shape and Ab coating effect.In vitro,the preparation shows the synergistic antiinflammatory and antioxidant effects in LPS-activated endothelial cells.In vivo,the preparation exhibits superior pharmacodynamic efficacy revealed by significantly downregulating the inflammatory/oxidative stress markers,such as TNF-a,IL-6,COX-2,and reactive oxygen species(ROS),in the lungs.In conclusion,the codelivery system based on rod-like pure crystals could well target the pulmonary endothelium and effectively alleviate lung inflammation.The study offers a promising approach to combat pulmonary endothelium-associated diseases.Yi Yang Makhloufi Zoulikha Qingqing Xiao Feifei Huang Qi Jiang Xiaotong Li Zhenfeng Wu Wei He 2023Acta Pharmaceutica Sinica B2023,13,11:0
