1Screening of stable internal reference genes by quantitative real-time PCR in humpback grouper Cromileptes altivelis显示文摘Humpback grouper Cromileptes altivelis is one commercial fish with considerable economic value.To determine the expression stabilities of six commonly used internal reference genes in C.altivelis challenged by Vibrio harveyi and viral nervous necrosis virus(VNNV)through quantitative real-time PCR(qRT-PCR),the expression levels of selected genes in five immune organs stimulated with pathogenic infection were carefully evaluated using algorithms of geNorm,NormFinder,and BestKeeper.The results show that the expre ssion stabilities of the six candidate inte rnal reference genes were diffe re nt.Under no rmal physiological conditions,RPL13 were identified as the most stably expressed genes among five different immune organs(liver,spleen,kidney,intestine,and gill).After V.harveyi stimulation,RPL13,RPL13,EF1 A,RPL13,and EF1 A were identified by geNorm,NormFinder,and BestKeeper as the most stable genes in liver,spleen,kidney,intestine,and gill,respectively.Combining these three algorithms suggested that under stimulation of VNNV,RPL13,EF1 A,Actin,RPL13,and Actin were as the most stable genes in liver,spleen,kidney,intestine,and gill,respectively.These results suggest that specific experiment conditions and tissue types shall be considered when selecting the reference genes in qRT-PCR analysis.This study provided a solid foundation for future studies on gene expression of C.altivelis under different conditions.Xiaojuan CHEN Yun SUN Panpan ZHANG Jianlong LI Haiping LI Caoying WEI Zhenjie CAO Yongcan ZHOU 2021Journal of Oceanology and Limnology2021,39,5:2
2Assessment of internal controls for data normalization of gene expression after different bacterial stimulation by quantitative real-time PCR in golden pompano Trachinotus blochii显示文摘Trachinotus blochii is one of the important commercial fish species.In this study,we aim to confirm the reliability reference genes in T.blochii during different bacterial challenge through quantitative real-time PCR(qRT-PCR).The expression of the seven selected genes in four immune organs(i.e.,spleen,kidney,intestine,and gill)stimulated with Vibrio harveyi,Edwardsiella tarda,and Streptococcus agalactiae were determined by qRT-PCR.The PCR data was analyzed using the geNorm and NormFinder algorithms.The results showed the selection of the internal controls should be tissue specific when studying gene expression in response to bacterial stimulation.After 48 h of stimulation with V.harveyi,geNorm ranked EF1 A/Actin,18 S rRNA/B2M,UBCE/B2M,and 18 S rRNA/B2M,as the most stably expressed genes in spleen,kidney,intestine,and gill,respectively.After 48 h of stimulation with E.tarda,geNorm ranked 18 S rRNA/EF1 A,18 S rRNA/B2M,B2M/RPL13,and 18 S rRNA/EF1 A,as the most stably expressed genes in spleen,kidney,intestine,and gill,respectively.After 48 h of stimulation with S.agalactiae,18 S rRNA/EF1 A,18 S rRNA/B2 M,B2 M/Actin,and 18 S rRNA/B2M were ranked as the most stably expressed genes in spleen,kidney,intestine,and gill,respectively.Compared to the results analyzed by geNorm,reference genes received similar rankings when using NormFinder software.The results showed that the reference genes appeared to be not only tissue specific,but also specific to the infecting species of bacteria.If one gene is preferred when T.blochii were infected by bacteria,18 S rRNA,B2M,B2M,18 S rRNA may be used in spleen,kidney,intestine,and gill,respectively.CHEN Xiaojuan ZHANG Xiaoqi SUN Yun TU Zhigang CAO Zhenjie WANG Shifeng ZHOU Yongcan 2020Journal of Oceanology and Limnology2020,38,2:1
3Protocol Format Extraction Based on an Improved CFSM Algorithm显示文摘As the information technology rapidly develops,many network applications appear and their communication protocols are unknown.Although many protocol keyword recognition based protocol reverse engineering methods have been proposed,most of the keyword recognition algorithms are time consuming.This paper firstly uses the traffic clustering method F-DBSCAN to cluster the unknown protocol traffic.Then an improved CFSM(Closed Frequent Sequence Mining)algorithm is used to mine closed frequent sequences from the messages and identify protocol keywords.Finally,CFGM(Closed Frequent Group Mining)algorithm is proposed to explore the parallel,sequential and hierarchical relations between the protocol keywords and obtain accurate protocol message formats.Experimental results show that the proposed protocol formats extraction method is better than Apriori algorithm and Sequence alignment algorithm in terms of time complexity and it can achieve high keyword recognition accuracy.Additionally,based on the relations between the keywords,the method can obtain accurate protocol formats.Compared with the protocol formats obtained from the existing methods,our protocol format can better grasp the overall structure of target protocols and the results perform better in the application of protocol reverse engineering such as fuzzing test.Peihong Lin Zheng Hong Lifa Wu Yihao Li Zhenji Zhou 2020China Communications2020,17,11:1

6Thrombin increases the expression of cholesterol 25-hydroxylase in rat astrocytes after spinal cord injury显示文摘Astrocytes are important cellular centers of cholesterol synthesis and metabolism that help maintain normal physiological function at the organism level.Spinal cord injury results in aberrant cholesterol metabolism by astrocytes and excessive production of oxysterols,which have profound effects on neuropathology.25-Hydroxycholesterol(25-HC),the main product of the membrane-associated enzyme cholesterol-25-hydroxylase(CH25H),plays important roles in mediating neuroinflammation.However,whether the abnormal astrocyte cholesterol metabolism induced by spinal cord injury contributes to the production of 25-HC,as well as the resulting pathological effects,remain unclear.In the present study,spinal cord injury-induced activation of thrombin was found to increase astrocyte CH25H expression.A protease-activated receptor 1 inhibitor was able to attenuate this effect in vitro and in vivo.In cultured primary astrocytes,thrombin interacted with protease-activated receptor 1,mainly through activation of the mitogen-activated protein kinase/nuclear factor-kappa B signaling pathway.Conditioned culture medium from astrocytes in which ch25h expression had been knocked down by siRNA reduced macrophage migration.Finally,injection of the protease activated receptor 1 inhibitor SCH79797 into rat neural sheaths following spinal cord injury reduced migration of microglia/macrophages to the injured site and largely restored motor function.Our results demonstrate a novel regulatory mechanism for thrombin-regulated cholesterol metabolism in astrocytes that could be used to develop anti-inflammatory drugs to treat patients with spinal cord injury.Chen Chen Huiyuan Ji Nan Jiang Yingjie Wang Yue Zhou Zhenjie Zhu Yuming Hu Yongjun Wang Aihong Li Aisong Guo 2023Neural Regeneration Research2023,18,6:1

8Initial experience of laparoendoscopic single-site radical prostatectomy with a novel purpose-built robotic system显示文摘Objective This prospective single-arm clinical trial aimed to evaluated the feasibility and safety of the application of the SHURUI system(Beijing Surgerii Technology Co.,Ltd.,Beijing,China),a novel purpose-built robotic system,in single-port robotic radical prostatectomy.Methods Sixteen patients diagnosed with prostate cancer were prospectively enrolled in and underwent robotic radical prostatectomy from October 2021 to August 2022 by the SHURUI single-port robotic surgical system.The demographic and baseline data,surgical,oncological,and functional outcomes as well as follow-up data were recorded.Results The mean operative time was 226.3(standard deviation[SD]52.0)min,and the mean console time was 183.4(SD 48.3)min,with the mean estimated blood loss of 116.3(SD 90.0)mL.The mean length of postoperative hospital stay was 4.50(SD 0.97)days.Two patients had postoperative complications(Clavien-Dindo Grade II),and both patients improved after conservative treatment.All patients’postoperative prostate-specific antigen levels decreased to below 0.2 ng/mL 1 month after discharge.The mean prostate-specific antigen level further decreased to a mean of 0.0219(SD 0.0641)ng/mL 6 months after surgery.Thirty days postoperatively,12 out of 16 patients reported using no more than one urinary pad per day,and all patients reported satisfactory urinary control without the need for pads 6 months after surgery.Conclusion The SHURUI system is safe and feasible in performing radical prostatectomy via both transperitoneal and extraperitoneal approaches.Tumor control and urinary continence were satisfying for patients enrolled in.The next phase involves conducting a large-scale,multicenter randomized controlled trial to thoroughly assess the effectiveness and safety of the new technology in a broader population.Zheng Wang Chao Zhang Chengwu Xiao Yang Wang Yu Fang Baohua Zhu Shouyan Tang Xiaofeng Wu Hong Xu Yi Zhou Lingfen Wu Zhenjie Wu Bo Yang Yi He Yi Liu Linhui Wang 2023Asian Journal of Urology2023,10,4:0
9Confining Li_(2)O_(2) in tortuous pores of mesoporous cathodes to facilitate low charge overpotentials for Li-O_(2) batteries显示文摘Achieving low charge overpotentials represents one of the most critical challenges for pursuing highperformance lithium-oxygen(Li-O_(2))batteries.Herein,we propose a strategy to realize low charge overpotentials by confining the growth of lithium peroxide(Li_(2)O_(2))inside mesoporous channels of cathodes(CMK-8).The CMK-8 cathode with tortuous pore structures can extend the diffusion distance of lithium superoxide(LiO_(2))in the mesoporous channels,facilitating the further reduction of LiO_(2) to lithium peroxide(Li_(2)O_(2))inside the pores and preventing them to be diffused out of the pores.Therefore,Li_(2)O_(2) is trapped in the mesoporous channels of CMK-8 cathodes,ensuring a good Li_(2)O_(2)/CMK-8 contact interface.The CMK-8 electrode exhibits a low charge overpotential of 0.43 V and a good cycle life for 72 cycles with a fixed capacity of 500 m Ah g^(-1) at 0.1 A g^(-1).This study proposes a strategy to achieve a low charge overpotential by confining Li_(2)O_(2) growth in the mesoporous channels of cathodes.Yin Zhou Yong Zhao Zhenjie Liu Zhangquan Peng Li Wang Wei Chen 2021Journal of Energy Chemistry2021,30,4:0
10Comparative Study on Riparian Vegetation Buffers in Different Land Uses显示文摘This study focused on riparian vegetation buffer zones on the basis of low influence development(LID) and landscape restoration. Three types of land were selected to conduct comparative study. The types are buffers in urban districts,buffers in rural areas,and buffers in natural regions. These types basically reflect major ecological issues in different landuses. The follow conclusions were drawn:(1) there are different ecological issues in these 3 types,making the functions of buffers different.(2) The 3 types demand different ecological issues to be solved: Type A,functionally-diverse riparian landscape should be planned to enrich recreational activities and improve river's condition. Type B,agricultural areasource pollution needs to be cut down to sustain the stability of water conservation and ecological system. 11Enhancement of permittivity and energy storage efficiency of poly (vinylidene fluoride-chlorotrifluoroethylene) by uniaxial stretching显示文摘Dielectric polymer film capacitors with a high-power density as well as efficient charge and discharge rates have great potential for application to fulfill the miniaturized and lightweight requirements of the electronic and stationary power systems.It was reported that the elastic recovery rate and energy storage density of poly(vinylidene fluoride-chlorotrifluoroethylene)[P(VDF-CTFE)]polymer film can be enhanced through thermostatic uniaxial stretching.But it is unknown about the relationship between the stretching rate and above properties.In this study,we investigated the effect of different stretching rates on the conformation,elastic recovery,dielectric constant,and energy storage density of stretched P(VDF-CTFE)polymer films.It was found that the stretching rate significantly affected the formation of polarβ-crystal phase,causing different dielectric properties.The degrees of elastic recovery of P(VDF-CTFE)film vary with stretching rates.Among them,the elastic recovery rate of the P(VDF-CTFE)94/6 film is 46.5%at a stretching rate of 15 mm/min,the dielectric constant is 12.25 at 100 Hz,and the energy density reaches 3.95 J/cm^(3) with the energy loss of 39%at 200 MV/m field.Zhenji Zhou Weimin Xia Jing Liu Na Tian Caiyin You 2023Journal of Advanced Dielectrics2023,13,1:0
12Yiqi Qingre Xiaozheng formula protects against diabetic nephropathy by restoring autophagy in mice显示文摘Objective:To evaluate the protective effect of Yiqi Qingre Xiaozheng formula(YQQRXZF)via the regulation of autophagy in diabetic nephropathy(DN)mice induced by injection of streptozotocin(STZ)after high fat diet(HFD).Methods:The composition of YQQRXZF was analyzed by high performance liquid chromatographyelectrospray ionization/mass spectrometry(HPLC-ESI/MSn).The DN model was induced by intraperitoneally injection of 50 mg/kg STZ within 5 days,followed by HFD feeding for 8 weeks in C57BL/6J mice.Mice were randomly separated into DN group,YQQRXZF group,irbesartan group,and control group.Blood glucose was calculated,and body weight was measured every 2 weeks.An enzyme-linked immunosorbent assay was used to measure the urinary albumin-to-creatinine ratio(uACR)before and after treatment,and the serum concentrations of total cholesterol(TC),low-density lipoprotein(LDL),triglycerides(TG),high-density lipoprotein(HDL),blood urea nitrogen(BUN),and serum creatinine(Scr)were measured.In addition,hematoxylin-eosin(H&E)staining,periodic acid-Schiff(PAS)staining,Masson's trichrome staining,and transmission electron microscopy(TEM)were used to observe pathological changes in renal tissue.Autophagy levels were determined by immunofluorescence staining.Results:In this study,21 dominant chemical constituents were identified in YQQRXZF.The treatment group reduced u ACR in a more significant way than the DN group(P=.018).The treatment group reduced TC,TG,and LDL concentrations after YQQRXFF treatment(P=.021,P=.014 and P=.026,respectively).H&E,PAS,and Masson staining showed that pathological damage to mice kidneys improved,the volume of renal glomeruli was reduced,and glomerular sclerosis and fibrosis were reduced.Immunofluorescence analysis revealed that expressions of LC3-Ⅱprotein increased in the treatment group(P=.013).In contrast,the protein expressions of P62 were reduced after treatment(P=.025).Na Sun Nan Yang Jingwei Zhou Ying He Jian Wang Yingnan Liang Run Dai Jiajia Bai Zhenjie Chen 2023Journal of Traditional Chinese Medical Sciences2023,10,3:0
13Selection of the appropriate reference genes by quantitative real-time PCR in leopard coral groupers Plectropomus leopardus显示文摘Leopard coral groupers(Plectropomus leopardus),commercially bred in South China,are a significant economical fish species.In this study,by means of quantitative real-time PCR(qRT-PCR)technology,we assessed the stability of six common reference genes expression and selected the appropriate reference genes in leopard coral groupers with or without Vibrio harveyi stimulation at different time points.These data produced by qRT-PCR was handled via the geNorm,NormFinder,and BestKeeper software.The results revealed all the examined reference genes had manifest tissue-specific expression in different tissues.Prior to V.harveyi stimulation,RPL13 gene was the appropriate reference gene among eleven tissue types(blood,spleen,hepatopancreas,kidney,stomach,gill,heart,skin,muscle,intestine,brain)in leopard coral groupers.Under V.harveyi stimulation,the most reliable reference genes varied from tissue to tissue and were closely hinged upon different time points.At 6-h post-bacterial injection,the appropriate reference genes in hepatopancreas,spleen,kidney,and gill were Actin,B2M,UBCE,and Actin,respectively.At 9-and 12-h post-bacterial injection,the appropriate reference genes in hepatopancreas,spleen,kidney,and gill were RPL13,Actin,Actin,and Actin,respectively.If one reference gene was preferable,RPL13,Actin,Actin,and Actin could be selected as the reference gene in hepatopancreas,spleen,kidney,and gill of leopard coral groupers after V.harveyi infection,respectively.Expression profiles of two target genes(IL-6 and NK-lysin)were used to further validate reliability of these selected appropriate candidates.This study will lay a solid foundation for the future research on qRTPCR analysis of gene expression in leopard coral groupers.Han ZHANG Haoran YANG Pengsuo LI Na ZHAO Xiaojuan CHEN Yongcan ZHOU Zhenjie CAO Yun SUN 2023Journal of Oceanology and Limnology2023,41,3:0
14Facile construction of a multilayered interface for a durable lithium‐rich cathode显示文摘Layered lithium-rich manganese-based oxide(LRMO)has the limitation of inevitable evolution of lattice oxygen release and layered structure transformation.Herein,a multilayer reconstruction strategy is applied to LRMO via facile pyrolysis of potassium Prussian blue.The multilayer interface is visually observed using an atomic-resolution scanning transmission electron microscope and a high-resolution transmission electron microscope.Combined with the electrochemical characterization,the redox of lattice oxygen is suppressed during the initial charging.In situ X-ray diffraction and the high-resolution transmission electron microscope demonstrate that the suppressed evolution of lattice oxygen eliminates the variation in the unit cell parameters during initial(de)lithiation,which further prevents lattice distortion during long cycling.As a result,the initial Coulombic efficiency of the modified LRMO is up to 87.31%,and the rate capacity and long-term cycle stability also improved considerably.In this work,a facile surface reconstruction strategy is used to suppress vigorous anionic redox,which is expected to stimulate material design in high-performance lithium ion batteries.Zhou Xu Yifei Yuan Qing Tang Xiangkun Nie Jianwei Li Qing Sun Naixuan Ci Zhenjie Xi Guifang Han Lijie Ci Guanghui Min 2023Carbon Energy2023,5,9:0
15Metallocorrole-based porous organic polymers as a heterogeneous catalytic nanoplatform for efficient carbon dioxide conversion显示文摘Metallocorrole macrocycles that represent a burgeoning class of attractive metal-complexes from the porphyrinoid family,have attracted great interest in recent years owing to their unique structure and excellent performance revealed in many fields,yet further functionalization through incorporating these motifs into porous nanomaterials employing the bottom-up approach is still scarce and remains synthetically challenging.Here,we report the targeted synthesis of porous organic polymers(POPs)constructed from custom-designed Mn and Fe-corrole complex building units,respectively denoted as CorPOP-1(Mn)and CorPOP-1(FeCl).Specifically,the robust CorPOP-1(Mn)bearing Mn-corrole active centers displays superior heterogeneous catalytic activity toward solvent-free cycloaddition of carbon dioxide(CO_(2))with epoxides to form cyclic carbonates under mild reaction conditions as compared with the homogeneous counterpart.CorPOP-1(Mn)can be easily recycled and does not show significant loss of reactivity after seven successive cycles.This work highlights the potential of metallocorrole-based porous solid catalysts for targeting CO_(2) transformations,and would provide a guide for the task-specific development of more corrole-based multifunctional materials for extended applications.Yanming Zhao Yunlei Peng Chuan Shan Zhou Lu Lukasz Wojtas Zhenjie Zhang Bao Zhang Yaqing Feng Shengqian Ma 2022Nano Research2022,15,2:0
