1Effects of AT1 receptor antagonist,Iosartan,on rat hepatic fibrosis induced by CCl_4显示文摘AIM To investigate effect of losartan,an AT1receptor antagonist,on hepatic fibrosis induced byCCl;and to determine whether or not AT1receptors are expressed on hepatic stellate cells,METHODS AND RESULTS Fifty male Sprague-Dawley rats,weighing(180±20)g,wererandomized into five groups(control group,modelgroup,and three losartan treated groups),inwhich all rats were given the subcutaneousinjection of 40% CCl4(every 3 days for 6 weeks)except for rats of control group.Rats of losartan-treated groups were treated with losartan(20 mg/kg,10 mg/kg,5 mg/kg,daily gavage),After 6weeks liver tissue and serum samples of all ratswere examined.Serum hyaluronic acid(HA),procollagen typeⅢ(PCⅢ)were detected byradioimmunoassays,van Giesion collagen stainingwas used to evaluate the extracellular matrix of ratswith liver fibrosis.The expression of AT1receptors,transforming growth factor-beta(TGF-β),and alpha-smooth muscle actin(a-SMA)inliver tissue were determined byimmunohistochemical techniques.Compared withmodel group,serum ALT and AST of losartan-treated groups were significantly reduced(t=4.20,P<0.01 and t=4.57,P<0.01).Serum HAand PCⅢalso had significant differences(t=3.53,P<0.01 and t=2.20,P<0.05).Thedegree of fibrosis was improved by losartan and correlated with the expressions of AT1 receptors,TGF-β,and α-SMA in liver tissue.CONCLUSION AT1 receptor antagonist,losartan,could limit the progression of the hepatic fibrosisinduced by CCl4.The mechanism may be related tothe decrease in the expression of AT1 receptorsand TGF-β,ameliorating the injury of hepatocytes;activation of local renin-angiotensin system mightrelate to hepatic fibrosis;and during progressionof fibrosis,activated hepatic stellate cells mightexpress AT1 receptors.Hong Shan Wei Ding Guo Li Han Ming Lu Yu Tao Zhan Zhi Rong Wang Xin Huang Jing Zhang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,4:42
2The regulatory role of AT 1 receptor on activated HSCs in hepat,c fibrogenesis,effects of RAS inhibitors on hepatic fibrosis induced by CCl_4显示文摘AIM To assess the effect of ACE inhibitor andAng Ⅱ type Ⅰ(AT1)receptor antagonist inpreventing hepatic fibrosis caused by CCl4administration in rats;to investigate whether ornot there are expression of AT 1 receptors onhepatic stellate cells;and to observe the effectof Ang Ⅱ on proliferation and ECM synthesis ofcultured HSCs.METHODS Studies were conducted in maleSprague-Dawley rats.Except for thehepatofibrotic model group and the controlgroup,in three treated groups,either enalapril(5 mg/kg),or Iosartan(10 mg/kg),or enalapril+Iosartan were given to the fibrotic rats bydaily gavage,and saline vehicle was given tomodel and normal control rats.After 6 weeks,liver fibrosis was assessed directly by hepaticmorphometric analysis,which has beenconsidered the gold standard for thequantification of fibrosis.The expressions of AT1 receptors and(α-mooth muscle actin,α-SMA)in liver tissue or isolated hepatic stellate cells(HSCs)were detected by immunohistochemicaltechniques.The effect of Ang Ⅱ on HSCproliferation was determined by MTT method.Effect of Ang Ⅱ on collagen synthesis of HSCswas determined by 3H-proline incorporation.RESULTS Contrasted to the fibrosis in rats ofthe model group,groups of rats treated with either enalapril or Iosartan,or a combination oftwo drugs showed a limited expansion of theinterstitium(4.23±3.70 vs 11.22±4.79,P<0.05),but no difference was observedamong three treated groups(5.38±3.43,4.96±2.96,4.23±2.70,P>0.05).Expression of AT 1receptors was found in fibrotic interstitium offibrotic rats,whereas in normal control rats theywere limited to vasculature only to a very slightdegree.AT 1 receptors were also expressed onactivated HSCs in the culture.At concentrationsfrom 10-9to 10-5mol/L,Ang Ⅱ stimulated HSCproliferation in culture in a dose-dependentmanner.Increasing Ang Ⅱ concentrationsproduced corresponding increases in 3H-prolineincorporation.Differences among groups were significant.CONCLUSION Angiotensin-converting enzyme inhibitors and AT I blocker may slow the progression of hepatic fibrosis; activated HSCs express AT 1 receptors, and Ang Ⅱ can stimulate the proliferation and collagen synthesis of HSCs in a dose-dependent manner; and activation of RAS may be related to hepatic fibrogenesis induced by CCI4.Hong Shan Wei Han Ming Lu Ding Guo Li Yu Tao Zhan Zhi Rong Wang Xin Huang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,6:27

4Identification of coastal wetlands of international importance for waterbirds:a review of China Coastal Waterbird Surveys 2005–2013显示文摘Background:China's coastal wetlands belong to some of the most threatened ecosystems worldwide.The loss and degradation of these wetlands seriously threaten waterbirds that depend on wetlands.Methods:The China Coastal Waterbird Census was organized by volunteer birdwatchers in China's coastal region.Waterbirds were surveyed synchronously once every month at 14 sites,as well as irregularly at a further 18 sites,between September 2005 and December 2013.Results:A total of 75 species of waterbirds met the 1 % population level Ramsar listing criterion at least once at one site.The number of birds of the following species accounted for over 20 % of the total flyway populations at a single site:Mute Swan(Cygnus olor),Siberia Crane(Grus leucogeranus),Far Eastern Oystercatcher(Haematopus osculans),Bar-tailed Godwit(Limosa lapponica),Spotted Greenshank(Tringa guttifer),Great Knot(Calidris tenuirostris),Spoon-billed Sandpiper(Calidris pygmeus),Saunders' s Gull(Larus saundersi),Relict Gull(Larus relictus),Great Cormorant(Phalacrocorax carbo),Eurasian Spoonbill(Platalea leucorodia),Black-faced Spoonbill(Platalea minor) and Dalmatian Pelican(Pelecanus crispus).A total of 26 sites supported at least one species of which their number met the1 % criterion.Forty-two species met the 1 % criterion in the Yellow River Delta,Shandong;29 at the Cangzhou coast,Hebei and 26 species at the Lianyungang coast,Jiangsu.Conclusions:The results highlight the international importance of China's coastal wetlands for waterbirds.This study also demonstrates that participation of local birdwatchers in waterbird surveys results in data that are invaluable not only for understanding the current status of waterbirds in China's coastal regions but also for waterbird conservation and management.Qingquan Bai Jianzhong Chen Zhihong Chen Guotai Dong Jiangtian Dong Wenxiao Dong Vivian Wing Kan Fu Yongxiang Han Gang Lu Jing Li Yang Liu Zhi Lin Derong Meng Jonathan Martinez Guanghui Ni Kai Shan Renjie Sun Suixing Tian Fengqin Wang Zhiwei Xu Yat-tung Yu Jin Yang Zhidong Yang Lin Zhang Ming Zhang Xiangwu Zeng 2015Chinese Birds2015,,3:20
5Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 9Impact of Traffic Emissions on Local Air Quality and the Potential Toxicity of Traffic-related Particulates in Beijing, China显示文摘Objective Air-borne particulates from different sources could have different physicochemical properties and inflammatory potentials. This study aims to characterize the chemical compositions and the toxicity of ambient particulate matter (PM) associated with traffic emissions. Methods The concentrations of trace elements, organic carbon (OC), elemental carbon (EC) and polycyclic aromatic hydrocarbons (PAHs) in PM 2.5 and PM 10 were measured in samples collected at sites in Beijing, China. Their toxic effects on the pulmonary system of rats were investigated. Biochemical parameters (LDH, T-AOC, TP) and inflammatory cytokine(IL-6, IL-1, TNF-α) levels were measured in the lungs of rats exposed to traffic-related PM. Oxidative damage was observed. PM samples were taken from a near road site and an off road site in summer time in 2006. Results The concentrations of the USEPA priority pollutant PAHs in both PM 10 and PM 2.5 were higher (299.658 and 348.412) at the near road site than those (237.728 and 268.472) at the off road site. The similar trend was observed for the concentrations of trace elements in PM. Compared to coarse particles (PM 10 ), fine particles (PM 2.5 ) have a greater adsorption capacity to enrich toxic elements than inhalable particles. Decrease in antioxidant capacity and an increase in the amount of lipid peroxidation products in rat lung tissues was observed. 10Seroepidemiological Investigation of Lyme Disease and Human Granulocytic Anaplasmosis among People Living in Forest Areas of Eight Provinces in China显示文摘Objective Lyme disease and Human granulocytic anaplasmosis are tick‐borne diseases caused by Borrelia burgdorferi and Anaplasma phagocytophilum respectively. We have investigated infection and co-infection of the two diseases in the population of forest areas of eight provinces in China by measuring seroprevalence of antibodies against B. burgdorferi and A. phagocytophilum. Methods Forest areas in 8 provinces were chosen for investigation using whole sampling and questionnaire survey methods. 3 669 serum samples from people in the forest areas were tested for the presence of antibodies by indirect immunofluorescent assay (IFA). Results Seroprevalence against B. burgdorferi was 3% to 15% and against A. phagocytophilum was 2% to 18% in the study sites in the 8 provinces in China. We also found co-infection of B. burgdorferi and A. phagocytophilum in 7 of the 8 provinces (the exception being the Miyun area in Beijing). The seroprevalence for both B. burgdorferi and A. phagocytophilum was significantly higher among people exposed to ticks than among people who were not exposed to ticks. 11Topographic differentiations of biological soil crusts and hydraulic properties in fixed sand dunes, Tengger Desert显示文摘Biological soil crusts(BSCs) play an important role in surface soil hydrology. Soils dominated with moss BSCs may have higher infiltration rates than those dominated with cyanobacteria or algal BSCs. However, it is unnown whether improved infiltration in moss BSCs is accompanied by an increase in soil hydraulic conductivity or water retention capacity. We investigated this question in the Tengger Desert, where a 43-year-old revegetation program has promoted the formation of two distinct types of BSCs along topographic positions, i.e. the moss-dominated BSCs on the interdune land and windward slopes of the fixed sand dunes, and the algal-dominated BSCs on the crest and leeward slopes. Soil water retention capacity and hydraulic conductivity were measured using an indoor evaporation method and a field infiltration method. And the results were fitted to the van Genuchten–Mualem model. Unsaturated hydraulic conductivities under greater water pressure(<–0.01 MPa) and water retention capacities in the entire pressure head range were higher for both crust types than for bare sand. However, saturated and unsaturated hydraulic conductivities in the near-saturation range(>–0.01 MPa) showed decreasing trends from bare sand to moss crusts and to algal crusts. 12Combined early fluid resuscitation and hydrogen inhalation attenuates lung and intestine injury显示文摘AIM:To study the effects of combined early fluid resuscitation and hydrogen inhalation on septic shockinduced lung and intestine injuries.METHODS:Wistar male rats were randomly divided into four groups:control group(Group A,n = 15);septic shock group(Group B,n = 15);early fluid resuscitation-treated septic shock group(Group C,n = 15);and early fluid resuscitation and inhalation of 2% hydrogentreated septic shock group(Group D,n = 15).The activity of hydroxyl radicals,myeloperoxidase(MPO),superoxide dismutase(SOD),diamine oxidase(DAO),and the concentration of malonaldehyde(MDA) in the lung and intestinal tissue were assessed according to the corresponding kits.Hematoxylin and eosin staining was carried out to detect the pathology of the lung and intestine.The expression levels of interleukin(IL)-6,IL-8,and tumor necrosis factor(TNF)-α in lung and intestine tissue were detected by enzyme-linked immunosorbent assay method.The expression levels of Fas and Bcl2 in lung tissues were determined by immunohistochemistry and Western blotting.RESULTS:Septic shock elicited a significant increase in the levels of MDA(10.17 ± 1.12 nmol/mg protein vs 2.98 ± 0.64 nmol/mg protein) and MPO(6.79 ± 1.02 U/g wet tissue vs 1.69 ± 0.14 U/g wet tissue) in lung tissues.These effects were not significantly decreased by Group C pretreatment,but were significantly reduced by Group D pretreatment(MDA:4.45 ± 1.13 nmol/mg protein vs 9.56 ± 1.37 nmol/mg protein;MPO:2.58 ± 0.21 U/g wet tissue vs 6.02 ± 1.16 U/g wet tissue).The activity of SOD(250.32 ± 8.56 U/mg protein vs 365.78 ± 10.26 U/mg protein) in lung tissues was decreased after septic shock,and was not significantly increased by Group C pretreatment,but was significantly enhanced by Group D pretreatment(331.15 ± 9.64 U/mg protein vs 262.98 ± 5.47 U/mg protein).Histological evidence of lung hemorrhage,neutrophil infiltration and overexpression of IL-6,IL-8,and TNF-α was observed in lung tissues,all of which were attenuated by Group C and further alleviated by Group D pretreatment.Septic shock also elicited a significant increase in the levels of MDA,MPO and DAO(6.54 ± 0.68 kU/L vs 4.32 ± 0.33 kU/L) in intestinal tissues,all of which were further increased by Group C,but significantly reduced by Group D pretreatment.Increased Chiu scoring and overexpression of IL-6,IL-8 and TNF-α were observed in intestinal tissues,all of which were attenuated by Group C and further attenuated by Group D pretreatment.CONCLUSION:Combined early fluid resuscitation and hydrogen inhalation may protect the lung and intestine of the septic shock rats from the damage induced by oxidative stress and the inflammatory reaction.Wei Liu Li-Ping Shan Xue-Song Dong Xiao-Wei Liu Tao Ma Zhi Liu 2013World Journal of Gastroenterology2013,19,4:12
13Expression changes of activin A in the development of hepatic fibrosis显示文摘AIM To examine the expression of activin A, amember of the transforming growth factor (TGF-β) superfamily, recently has been reported to beoverexpressed in liver cirrhosis, in the course ofcarbon tetrachloride-induced rat hepaticfibrosis.METHODS Hepatic fibrosis was induced in ratsby subcutaneous injections of 40% carbontetrachloride oily solution for a period of 1 to 7weeks. At the end of 1, 2, 3, 4, 5, 6 and 7 weeksafter carbon tetrachloride injections, the ratswere killed in group (6- 10 rats each time) forstudy. The activin A messenger RNA expressionand its protein localization were assessed bysemi-quantitative reverse transcriptionpolymerase chain reaction ( RT-PCR ) andimmunohistochemistry.RESULTS The normal rat liver expressedactivin A mRNA and protein, and its expressionwas transiently decreased and becameundetectable after carbon tetrachlorideinjections for 2 or 3 weeks and then increasedgradually. After injection of carbon tetrachloridefor 6 and 7 weeks, activin A mRNA and proteinexpressions were significantly enchanced in ratliver. Compared with that of the normal ratliver. Activin A mRNA expression levels in ratsreceiving carbon tetrachloride injections for 6and 7 weeks were 1.6 and 2.2 times that of thosein normal rat liver respectively (0.456±0.094 vs0.286± 0.0670, P<0.01; 0.620± 0.134 vs 0.286±± 0.0670, P<0.01). Immunohistochemistryshowed that activin A expressed in hepatocytesof normal liver, and its expression wasdecreased in rats receiving carbon tetrachloridefor 2 or 3 weeks. 15Adoptive therapy with CAR redirected T cells for hematological malignancies显示文摘The survival of patients with hematological malignancies has been significantly improved due to the development of new therapeutic agents. However, relapse remains a major matter for concern. Recently, T cells engineered with chimeric antigen receptor(CAR) were reported to show unprecedented responses in a range of hematological malignancies. The persistence of the CAR-T cell can last for years and tends toward long-term antitumor memory by which relapses can be effectively prevented. The primary side effects that appear in most clinical trials are cytokine release syndrome and neurotoxicity. However, these symptoms can be treated and reversed. 16Prevalence and Genotypes of Rotavirus A and Human Adenovirus among Hospitalized Children with Acute Gastroenteritis in Fujian, China, 2009-2017显示文摘Gastroenteritis is an infectious diarrhea that has been considered as an important cause of hospitalizations and death in children aged < 5 years, particularly in developing countries. Unsanitary water, contaminated food, poor hygiene, and inadequate disposal of waste and feces are all risk factors for gastroenteritis, resulting in the higher incidence in developing countries. Gastroenteritis is generally caused by viral infections, among which rotavirus (RV) infections have been reported to be the most common, especially among young children aged < 5 years with acute gastroenteritis in Asia and Africa[1]. Other viruses associated with acute gastroenteritis include human Adenovirus (HAdV), Norovirus, Sapovirus (SaV), human Astrovirus (HAstV), and Aichi virus. Recent research has reported that adenovirus types 40 and 41, belonging to species F, cause gastroenteritis and were therefore termed as enteric adenoviruses. 17The coseismic displacement field of the Zhangbei-Shangyi earthquake mapped by differ-ential radar interferometry显示文摘The coscismic deformation produced by 1998 earthquake (M8 = 6.2) in Zhangbei-Shangyi of northern China is measured by the differential synthetic aperture radar interferometry (D-InSAR) technique using the European Remote Sensing satellite (ERS) SAR data. Interferograms are constructed from the ERS-1/2 SAR data by the three-pass method. The line-of-sight displacement map indicates that the deformation center of the earthquake is located at E114°20’, N40°57’, with the maximum uplift of 25 cm. The extent of the displacement is around 300 km2. The focal mechanism and earthquake-induced structures are analyzed based on the spatial distribution of the deformation. 20Preparation of Fe_3O_4@Au nano-composites by self-assembly technique for immobilization of glucose oxidase显示文摘Superparamagnetism amination nanocrystals Fe3O4 with 3-aminopropyltriethyloxy silane (APTES) were prepared by modified co-precipitation method. Next, 4―5 nm gold nanoparticles, prepared by classical Frens procedure, were coated on the surface of the amination Fe3O4 by self-assembly technology. The prepared Fe3O4@Au nanocomposite particles were investigated by transmission electron microscopy (TEM), UV-vis, infrared spectrum (FT-IR), and vibrating sample magnetometer (VSM) in order to eluci-date the morphology, optics and magnetic properties of the nanocomposites. Their uniform distribution of particle size,which is about 15 nm , and good magnetic responsiveness were observed. In view of the fact that Fe3O4 owns superparamagnetism and that nano-gold can readily combine with biological molecules, glucose oxidase (GOx) was chosen as a model to penetrate the condition of immobilizing enzyme, and enzymatic properties of resultant immobilized enzyme were studied as well. By systematic optimization, we established that at 28℃, and pH (5.5) and when mole ratio of Fe3O4:HAuCl4 was 0.5:1, the immobilization provided the best results. Finally, we are glad to find that the immobilized enzyme exhibits excellent thermostability in addition to its better stability than free enzyme.
