1Construction of a Genetic Linkage Map of Mango Based on SRAP, AFLP and ISSR Markers显示文摘A hybrid F_1 population was obtained by crossing between mango cultivars‘Jinhuang'and‘Guifei'. Among the hybrid population,98 F_1 plants were selected as the mapping population. The molecular genetic map of mango was constructed by linkage analysis by SRAP,AFLP and ISSR markers using Joinmap 4.0 software. The genetic linkage map consisted of 33 linkage groups,with a total genetic distance of 1561. 1 cM. Moreover,the genetic linkage map involved 245 polymorphic markers,including 149 SRAP markers,90 AFLP markers and 6 ISSR markers,with an average genetic distance of 6. 37 cM. 2Molecular Cloning and Sequence Analysis of C4H Gene of Mangifera indica L.显示文摘Cinnamate-4-hydroxylase( C4H) is a key enzyme in phenylpropanoid pathway in plants. Its activity and abundance directly affect the biosynthesis of flavonoids and aromatic compounds. In this study,degenerate primers were designed according to previously reported C4 H gene sequences to clone C4H cDNA sequence with 3'and 5'RACE-PCR from mango( Mangifera indica L). The full-length cD NA of M. indica C4H is 1 680 bp long. Its open reading frame( ORF)is 1 518 bp,encoding a protein of 505 amino acids with a predicted molecular weight of 58. 08 kDa. The isoelectric point of the predicted protein is 9. 52. Functional prediction showed that this gene is mainly located in mitochondria. In addition,the tertiary structure of the protein was built using SWISS-MODEL,and the results showed that the protein has three possible conformations. Phylogenetic analysis based on C4H protein sequences revealed that M. indica has a close genetic relationship with olive( Canarium album) and cocoa( Theobroma cacao). By analyzing the expression level of C4H gene in three colored mango cultivars,we found that that the expression level of C4 H gene in Guifei( with red peel) was the highest,and that in Guiqi( with green peel) was the lowest. 3Effect of Internal Ethylene and 1-methylcyclopropene on Ripening of Papaya Fruit(Carica papaya L.)显示文摘With papaya fruit( Carica papaya) as an experimental material,variations of its physiological indices were investigated through hypobaric hypoxia( HH,10 kPa,1. 5 kPa O_2) treatment followed by fumigation with 1-methylcyclopropene( 1-MCP),so as to discuss the effect of interaction between 1-MCP and internal ethylene on ripening of papaya fruit during postharvest storage. The results indicated that HH treatment significantly reduced internal ethylene concentration( IEC) by 57. 6%. However,there were no remarkable differences in other physiological indices between various treatments and control( CK). The 1-MCP and HH+ 1-MCP treatments could significant postpone the softening process and color change of papaya fruit. The two treatments achieved the effects of significantly inhibiting the occurrence of fruit disease and the increase of pericarp cell membrane permeability,effectively delaying the variation of soluble solid and Vc contents,and better maintaining soluble solid and Vc contents in fruit,thereby better keeping nutritional substances and flavor quality in fruit in later storage period and prolonging the storage time of fruit.Huande FENG Zhiguo DANG Yeyuan CHEN 2018Agricultural Biotechnology2018,7,5:0
