| 1 | Curative effect of assisted reproduction technology by Traditional Chinese Medicine multi-channel interventional therapy on 95 cases of in vitro fertilization and embryo transfer failure显示文摘OBJECTIVE: To evaluate the curative effect of Traditional Chinese Medicine(TCM) multi-channel interventional therapy on women with Assisted Reproductive Technology(ART) failure, to compare the curative effect of the dual therapy and triple therapy on women with ART Failure, and to choose the best TCM interventional therapeutic plan.METHODS: The 95 cases with ART Failure from West China second University Hospital of Sichuan University meeting the inclusion criteria were randomly divided into three groups:dual therapy group(31 cases), triple therapy group(33 cases)and control group(31 cases). According to the intervene treatment of in vitro Fertilization and Embryo Transfer(IVF-ET) long cycle scheme, the control group wait naturally for 3 months before IVFET. The dual therapy group take TCM prescriptionⅡ of cultivating emotion and assisting reproduction and auricular acupoint therapy for 3 months before IVF-ET, then Western Medicine treatment progestin supporting as well as auricular application and Antai Recipe after IVF-ET transplantation.The triple therapy group take TCM prescription Ⅱof cultivated emotion and assisted reproduction,auricular acupoint therapy and retention enema of TCM, and combination treatment the same as dual therapy group after transplantation. The natural pregnancy number, the period condition of secondary IVF-ET and the improvement of the kidney deficiency, liver depression and blood stasis syndrome among those three groups were compared.RESULTS: It was showed from analysis in 95 cases with ART failure that the number of natural preg-nancy was as followings: 3 patients from the dual therapy group, 10 patients from the triple therapy group, and no patient from the control group. The comparison among three groups have statistical significance(P < 0.05). The treatment group is superior to the control group, while the triple therapy is superior to the dual therapy. The comparison of the condition of the fertility rate, clinical pregnancy rate, biochemical pregnancy rate and early abortion rate during the period of secondary IVF-ET between pre-therapy and post-treatment of both the dual therapy group and the triple therapy group have statistical significance(P < 0.05). The comparison of the improvement of the kidney deficiency,liver depression and blood stasis syndrome between pre-therapy and post-treatment of both the dual therapy group and the triple therapy group have statistical significance(P < 0.05). | 6 | A PD-L1-targeting chimeric switch receptor enhances efficacy of CAR-T cell for pleural and peritoneal metastasis显示文摘Pleural and peritoneal metastasis accompanied by malignant pleural effusion(MPE)or malignant ascites(MA)is frequent in patients with advanced solid tumors that originate from the lung,breast,gastrointestinal tract and ovary.Regional delivery of CAR-T cells represents a new strategy to control tumor dissemination in serous cavities.However,malignant effusions constitute an immune-suppressive environment that potentially induces CAR-T cell dysfunction.Here,we demonstrated that the anti-tumor cytotoxicity of conventional 2nd-generation CAR-T cells was significantly inhibited by both the cellular and non-cellular components of MPE/MA,which was primarily attributed to impaired CAR-T cell proliferation and cytokine production in MPE/MA environment.Interestingly,we found that PD-L1 was widely expressed on freshly-isolated MPE/MA cells.Based on this feature,a novel PD-L1-targeting chimeric switch receptor(PD-L1.BB CSR)was designed,which can bind to PD-L1,switching the inhibitory signal into an additional 4-1BB signal.When co-expressed with a 2nd-generation CAR,PD-L1.BB CSR-modified CAR-T cells displayed superior fitness and enhanced functions in both culture medium and MPE/MA environment,causing rapid and durable eradication of pleural and peritoneal metastatic tumors in xenograft models.Further investigations revealed elevated expressions of T-cell activation,proliferation,and cytotoxicity-related genes,and we confirmed that PD-L1 scFv and 4-1BB intracellular domain,the two important components of PD-L1.BB CSR,were both necessary for the functional improvements of CAR-T cells.Overall,our study shed light on the clinical application of PD-L1.BB CSR-modified dual-targeting CAR-T cells.Based on this study,a phase I clinical trial was initiated in patients with pleural or peritoneal metastasis(NCT04684459). | Qizhi Ma Xia He Benxia Zhang Fuchun Guo Xuejin Ou Qiyu Yang Pei Shu Yue Chen Kai Li Ge Gao Yajuan Zhu Diyuan Qin Jie Tang Xiaoyu Li Meng Jing Jian Zhao Zeming Mo Ning Liu Yao Zeng Kexun Zhou Mingyang Feng Weiting Liao Wanting Lei Qiu Li Dan Li Yongsheng Wang | 2022 | Signal Transduction and Targeted Therapy2022,7,12: | 0 |
| 7 | Risk Assessment and Change Monitoring of Soil and Water Loss in Ruijin City Based on RS and GIS显示文摘The land use information extraction technology for the high-resolution remote sensing images of the Gaofen No. 1 satellite was constructed. According to the spectral,band,texture and shape attributes,land use types were divided,and the changing laws of land use types were analyzed. Afterwards,according to the Table of Grading Standard of Soil Erosion Intensity( SL190-96),as well as vegetation coverage index NDVI and slope,the risks of soil and water loss were assessed. Meanwhile,the level,scale,location and scope of changes in the risks of soil and water loss were monitored by using spatial visualization and spatial statistical techniques. The results showed that the area of areas without soil erosion and moderate soil erosion areas decreased obviously from 2015 to 2017,and the decreases were up to 22. 929 3 and 13. 626 3 km^2 respectively. The area of mild soil erosion areas increased fast,and the increase reached 31. 140 0 km^2. The area of extremely strong soil erosion areas increased by7. 267 4 km^2. In the city,moderate and strong soil erosion areas reduced,while extremely strong soil erosion patches increased fast,which was mainly related to road construction and construction and development of orchards. The extremely strong soil erosion areas were distributed in the shape of a banded loop,surrounded the suburbs of the city,and shrank towards the center of Ruijin City.
