2Exploiting sound for emerging applications of extracellular vesicles显示文摘Extracellular vesicles are nano-to micro-scale,membrane-bound particles released by cells into extracellular space,and act as carriers of biomarkers and therapeutics,holding promising potential in translational medicine.However,the challenges remain in handling and detecting extracellular vesicles for disease diagnosis as well as exploring their therapeutic capability for disease treatment.Here,we review the recent engineering and technology advances by leveraging the power of sound waves to address the challenges in diagnostic and therapeutic applications of extracellular vesicles and biomimetic nanovesicles.We first introduce the fundamental principles of sound waves for understanding different acoustic-assisted extracellular vesicle technologies.We discuss the acoustic-assisted diagnostic methods including the purification,manipulation,biosensing,and bioimaging of extracellular vesicles.Then,we summarize the recent advances in acoustically enhanced therapeutics using extracellular vesicles and biomimetic nanovesicles.Finally,we provide perspectives into current challenges and future clinical applications of the promising extracellular vesicles and biomimetic nanovesicles powered by sound.Zhuhao Wu Hongwei Cai Chunhui Tian Zheng Ao Lei Jiang Feng Guo 2024Nano Research2024,17,2:1

7An Alternative to the Adversarial:Studies on Challenges of Court-appointed Experts显示文摘At present,experts have become a mainstay of modern litigation,although criticisms suggest that the problems of how to fit expert knowledge comfortably into the method of adversarial fact-finding are numerous,significant,and without simple solutions.Concerns about partisanship and lack of scientific competence by adjudicators to evaluate contradictory expert testimony have been widely recognized in the traditional use of party-called expert witnesses.While such concerns cannot be wholly ameliorated,there may be alternative mechanisms that can help.One solution would be to call for the use of neutral court-appointed experts,to create a nonpartisan source of expert knowledge.A system of neutral court-appointed experts is an advisory tribunal to the court that could deliver“those general truths,applicable to the issue,which they may treat as final and decisive.”However,no matter in which country,the choice of appointing neutral experts still seems to be a rare option for trial judges to consider and exercise.An obvious question would be:Why are neutral experts not used more frequently at trial?This paper did a study on court-appointed experts,with a focus on challenges that such mechanism faces.Part Ⅰ examines problems in the traditional use of expert witnesses in an adversarial system.Part Ⅱ discusses the incentives to make greater use of court-appointed experts in a typical adversarial system and to what extent such mechanism would solve difficulties within the traditional use of party-called expert witnesses.Part Ⅲ further explores and analyzes obstacles that a typical neutral expert system nowadays encounters when it operates in practice.Taking all analysis together,Part IV makes an overall evaluation of the mechanism of court-appointed experts.Zhuhao Wang 2016Journal of Forensic Science and Medicine2016,2,1:0
8Replacing alkyl side chain of non-fullerene acceptor with siloxane-terminated side chain enables lower surface energy towards optimizing bulk-heterojunction morphology and high photovoltaic performance显示文摘Towards a good control of the morphology of bulk-heterojunction(BHJ)active layers for polymer solar cells(PSCs),selecting an appropriate side chain for a polymer donor and a nonfullerene acceptor(NFA)is very crucial.In this work,two novel NFAs i-IE-4F and i-IESi-4 F comprising alkyl and siloxane-terminated side chains on the central indacenodithiophene(IDT)core,respectively,were synthesized.Attaching the siloxane-terminated side chain to i-IESi-4 F affords surface energy(γ)of33.32 mN/m,much lower than that of 39.83 mN/m for i-IE-4F,supplying a big chance to tune miscibility with a polymer donor.Two fluorobenzotriazole-based polymer donors J52 and PBZ-2Si bearing alkyl and siloxane-terminated side chains,respectively,showγvalues of 36.08 and 33.10 mN/m,respectively.The estimated Flory-Huggins interaction parameters(χD,A)indicate that the i-IESi-4 F is more miscible than i-IE-4F in pairing with J52 or PBZ-2Si.The resulting i-IESi-4 F-based blend film exhibits low film roughness and accompanies obviously improved BHJ uniformity.In PSCs,the J52:i-IESi-4F and PBZ-2Si:i-IESi-4 F active layers display power conversion efficiencies(PCEs)of 12.67%and 14.54%,respectively,all remarkably higher than PCEs≤7.34%of the i-IE-4F-based active layers.Interestingly,the PBZ-2Si:i-IESi-4 F active layer,a donor:acceptor blend system comprising siloxane-terminated side chains(DSi:ASimatching)with the highest BHJ miscibility due to the combinatory effect of the side chains,shows the highest efficiency,as supported by efficient exciton dissociation,the lowest bimolecular recombination,and the optimal charge transport.Our results demonstrate that attaching siloxane-terminated side chains to NFAs,as a side chain engineering,has big potential in lowering their surface energy towards fine control of BHJ morphology and leading to a better donor:acceptor blend system.Rihang Qiu Zhuhao Wu Suhan Li Haiying Jiang Qian Wang Yinchu Chen Xuanchen Liu Lianjie Zhang Junwu Chen 2021Science China Chemistry2021,64,7:0

10Engineering human spinal microphysiological systems to model opioid-induced tolerance显示文摘pioids are commonly used for treating chronic pain.However,with continued use,they may induce tolerance and/or hyperalgesia,which limits therapeutic efficacy.The human mechanisms of opioid-induced tolerance and hyperalgesia are significantly understudied,in part,because current models cannot fully recapitulate human pathology.Here,we engineered novel human spinal microphysiological systems(MPSs)integrated with plug-and-play neural activity sensing for modeling human nociception and opioid-induced tolerance.Each spinal MPS consists of a flattened human spinal cord organoid derived from human stem cells and a 3D printed organoid holder device for plug-and-play neural activity measurement.We found that the flattened organoid design of MPSs not only reduces hypoxia and necrosis in the organoids,but also promotes their neuron maturation,neural activity,and functional development.We further demonstrated that prolonged opioid exposure resulted in neurochemical correlates of opioid tolerance and hyperalgesia,as measured by altered neural activity,and downregulation ofμ-opioid receptor expression of human spinal MPSs.The MPSs are scalable,cost-effective,easy-to-use,and compatible with commonly-used well-plates,thus allowing plug-and-play measurements of neural activity.We believe the MPSs hold a promising translational potential for studying human pain etiology,screening new treatments,and validating novel therapeutics for human pain medicine.Hongwei Cai Zheng Ao Chunhui Tian Zhuhao Wu Connor Kaurich Zi Chen Mingxia Gu Andrea G.Hohmann Ken Mackie Feng Guo 2023Bioactive Materials2023,,4:0
