1Morphological evolution of γ'precipitates in a nickel-based superalloy during various solution treatments显示文摘The morphological evolution of the γ' phase in nickel-based superalloy жc6y during various solution heat treatments was investigated. The significant changes of the γ' precipitates were observed in the solution-treated samples. The coarsening and dissolution of γ' phase simulta-neously occurred at intermediate temperatures. In some areas, the primary precipitates became blunt and the adjacent ones were intercon-nected with each other via a diffuse neck, indicating a coarsening process of the primary γ' population. The coarsening was dominated by the precipitate agglomeration mechanism (PAM) rather than by the well-known Ostwald ripening mechanism. In other areas, the partial dissolu-tion of the γ' precipitates began to occur, spreading gradually from dendrite cores to interdendritic regions. In addition, a flower-like γ' struc-ture was developed during the subsolvus solution treatments. 2Features of a rare advection-radiation fog event显示文摘To investigate effects of atmospheric pollutants on fog nature, a comprehensive in situ observation project was implemented in the northern suburb of Nanjing, in December of 2006. For December 24-27 there occurred a heavy fog lasting 4 d in succession. This event is of rare characteristics, namely long persistence, high concentration, tall fog top, acid fog water and explosive growth. Detailed analysis along with the causes of the fog was presented. The evidence suggests that the fog was generated by nighttime radiative cooling, maintained and developed under effects of warm, wet advection. 4An adaptive scaled boundary finite element method by subdividing subdomains for elastodynamic problems显示文摘The scaled boundary finite element method(SBFEM) is a semi-analytical numerical method,which models an analysis domain by a small number of large-sized subdomains and discretises subdomain boundaries only.In a subdomain,all fields of state variables including displacement,stress,velocity and acceleration are semi-analytical,and the kinetic energy,strain energy and energy error are all integrated semi-analytically.These advantages are taken in this study to develop a posteriori h-hierarchical adaptive SBFEM for transient elastodynamic problems using a mesh refinement procedure which subdivides subdomains.Because only a small number of subdomains are subdivided,mesh refinement is very simple and efficient,and mesh mapping to transfer state variables from an old mesh to a new one is also very simple but accurate.Two 2D examples with stress wave propagation were modelled.The results show that the developed method is capable of capturing propagation of steep stress regions and calculating accurate dynamic responses,using only a fraction of degrees of freedom required by adaptive finite element method.ZHANG ZiHua 1,2,YANG ZhenJun 2,LIU GuoHua 1 & HU YunJin 1 1 College of Civil Engineering and Architecture,Zhejiang University,Hangzhou 310058,China 2 School of Engineering,University of Liverpool,L69 3GQ,UK 2011Science China(Technological Sciences)2011,54,S1:4
5Experimental study of structural damage identification based on modal parameters and decay ratio of acceleration signals显示文摘A novel damage assessment method based on the decay ratio of acceleration signals(DRAS)was proposed.Two experimental tests were used to show the efficiency.Three beams were gradually damaged,and then the changes of dynamic parameters were monitored from initial to failure state.In addition,a new method was compared with the linear modal-based damage assessment using wavelet transform(WT).The results clearly show that DRAS increases in linear elasticity state and microcrack propagation state,while DRAS decreases in macrocrack propagation state.Preliminary analysis was developed considering the beat phenomenon in the nonlinear state to explain the turn point of DRAS.With better sensibility of damage than modal parameters,probably DRAS is a promising damage indicator in damage assessment.Zhigen WU Guohua LIU Zihua ZHANG 2011Frontiers of Structural and Civil Engineering2011,5,1:3
6Regulation of DNA double-strand break repair pathway choice:a new focus on 53BP1显示文摘Maintenance of cellular homeostasis and genome integrity is a critical responsibility of DNA double-strand break(DSB)signaling.P53-binding protein 1(53BP1)plays a critical role in coordinating the DSB repair pathway choice and promotes the non-homologous end-joining(NHEJ)-mediated DSB repair pathway that rejoins DSB ends.New insights have been gained into a basic molecular mechanism that is involved in 53BP1 recruitment to the DNA lesion and how 53BP1 then recruits the DNA break-responsive effectors that promote NHEJ-mediated DSB repair while inhibiting homologous recombination(HR)signaling.This review focuses on the up-and downstream pathways of 53BP1 and how 53BP1 promotes NHEJ-mediated DSB repair,which in turn promotes the sensitivity of poly(ADP-ribose)polymerase inhibitor(PARPi)in BRCA1-deficient cancers and consequently provides an avenue for improving cancer therapy strategies.Fan ZHANG Zihua GONG 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,1:2
7Identifying EGFR-Expressed Cells and Detecting EGFR Multi-Mutations at Single-Cell Level by Microfluidic Chip显示文摘EGFR mutations companion diagnostics have been proved to be crucial for the efficacy of tyrosine kinase inhibitor targeted cancer therapies. To uncover multiple mutations occurred in minority of EGFR-mutated cells,which may be covered by the noises from majority of unmutated cells, is currently becoming an urgent clinical requirement. Here we present the validation of a microfluidic-chip-based method for detecting EGFR multimutations at single-cell level. By trapping and immunofluorescently imaging single cells in specifically designed silicon microwells, the EGFR-expressed cellswere easily identified. By in situ lysing single cells, the cell lysates of EGFR-expressed cells were retrieved without cross-contamination. Benefited from excluding the noise from cells without EGFR expression, the simple and cost-effective Sanger's sequencing, but not the expensive deep sequencing of the whole cell population, was used to discover multi-mutations. We verified the new method with precisely discovering three most important EGFR drugrelated mutations from a sample in which EGFR-mutated cells only account for a small percentage of whole cell population. The microfluidic chip is capable of discovering not only the existence of specific EGFR multi-mutations,but also other valuable single-cell-level information: on which specific cells the mutations occurred, or whether different mutations coexist on the same cells. 9The effect of slow shot speed and casting pressure on the 3D microstructure of high pressure die casting AE44 magnesium alloy显示文摘3D reconstruction and 2D observation were conducted to characterize the microstructure of the castings produced through high pressure die casting with different parameters.Our results indicate that shrinkage pores generally co-existed with externally solidified crystals(ESCs).In specimen produced without fast slow shot speed,big net-shrinkage pores accompanied with ESCs were found in the center of the specimen.When the casting pressurization was introduced,the shrinkage pores gathered to the specimen center and became much less due to the optimization of melt feeding.Much more porosity was found near the gate rather than in the middle of the rod bar,especially gas pores.Thefilling process simulation reveals that the middle position of the bars wasfirstlyfilled and followed by the near gate position accompanied with one intense turbulentflow.Chaosheng Ma Wenbo YU Tongtong Zhang Zihua Zhang Yihu Ma Shoumei Xiong 2023Journal of Magnesium and Alloys2023,11,2:2

15Characterization of Early Fatigue Microstructure in AISI 321 Steel Using Eddy Current Non-destructive Methodology显示文摘Accumulative damage during early stage of fatigue in AISI 321 steel was investigated by eddy current test, atomic force microscopy, X-ray diffraction and transmission electron microscopy. Surface slip, dislocation, and strain-induced martensite were determined as the main damage types. Moreover, damage during the fatigue was found to be increased with the increasing fatigue cycles and load amplitude. The contribution of strain-induced martensite to the total eddy current amplitude(V max) was enhanced with the increase in its volume fraction. 19ATF4/TXNIP/REDD1/mTOR signaling mediates the antitumor activities of liver X receptor in pancreatic cancers显示文摘Background:Limited by difficulties in early detection and availabilities of effective treatments,pancreatic cancer is a highly malignant disease with poor prognosis.Nuclear receptors are a family of ligand‐dependent transcription factors that are highly druggable therapeutic targets playing critical roles in human physiological and pathological development,including cancer.In this study,we explored the therapeutic potential as well as the molecular mechanisms of liver X receptor(LXR)agonist GW3965 in pancreatic cancer.Methods:Soft‐agar colony formation assay,xenograft tumors,Oligonucleotide microarray,Reverse transcription real‐time polymerase chain reaction,Western immunoblotting and Immunohistochemistry were used in this study.Results:We demonstrated pleotropic in vitro activities of GW3965 in pancreatic cell lines MIA PaCa‐2 and BXPC3 including reduction of cell viability,inhibition of cell proliferation,stimulation of cell death,and suppression of colony formation,which translated to significant inhibition of xenograft tumor growth in vitro.By mapping the gene expression profiles,we identified the up‐regulations of 188 and the down‐regulations of 92 genes common to both cell lines following GW3965 treatment.Genes responsive to GW3965 represent a variety of biological pathways vital for multiple cellular functions.Specifically,we identified that the activating transcription factor 4/thioredoxin‐interacting protein/regulated in development and DNA damage responses 1/mechanistic target of rapamycin(ATF4/TXNIP/REDD1/mTOR)signaling critically controls GW3965‐mediated regulation of cell proliferation/death.The significance of the ATF4/TXNIP/REDD1/mTOR pathway was further supported by associated expressions in xenograft tumors as well as human pancreatic cancer samples.Conclusions:This study provides the pre‐clinical evidence that LXR agonist is a promising therapy for pancreatic cancer.Zhikang Chen Xiaobo Lai Hui Ding Aijun Zhang Yufei Sun Jianhua Ling Paul J.Chiao Zihua Chen Xuefeng Xia 2022Cancer Innovation2022,1,1:1
