1Tailoring the Meso-Structure of Gold Nanoparticles in Keratin-Based Activated Carbon Toward High-Performance Flexible Sensor显示文摘Flexible biosensors with high accuracy and reliable operation in detecting pH and uric acid levels in body fluids are fabricated using well-engineered metaldoped porous carbon as electrode material.The gold nanoparticles@N-doped carbon in situ are prepared using wool keratin as both a novel carbon precursor and a stabilizer.The conducting electrode material is fabricated at 500℃ under customized parameters,which mimics A-B type(two different repeating units) polymeric material and displays excellent deprotonation performance(pH sensitivity).The obtained pH sensor exhibits high pH sensitivity of 57 mV/pH unit and insignificant relative standard deviation of 0.088%.Conversely,the composite carbon material with sp^2 structure prepared at 700℃ is doped with nitrogen and gold nanoparticles,which exhibits good conductivity and electrocatalytic activity for uric acid oxidation.The uric acid sensor has linear response over a range of 1-150 μM and a limit of detection 0.1 μM.These results will provide new avenues where biological material will be the best start,which can be useful to target contradictory applications through molecular engineering at mesoscale.Aniruddha BPatil Zhaohui Meng Ronghui Wu Liyun Ma Zijie Xu Chenyang Shi Wu Qiu Qiang Liu Yifan Zhang Youhui Lin Naibo Lin Xiang Yang Liu 2020Nano-Micro Letters2020,12,9:3

8Emergent superconducting fluctuations in compressed kagome superconductor CsV_(3)Sb_(5)显示文摘The recent discovery of superconductivity(SC)and charge density wave(CDW)in kagome metals AV3Sb5(A=K,Rb,Cs)provides an ideal playground for the study of emergent electronic orders.Application of moderate pressure leads to a two-dome-shaped SC phase regime in CsV_(3)Sb_(5) accompanied by the destabilizing of CDW phase.Nonetheless,the nature of this pressure-tuned SC state and its interplay with the CDW are yet to be explored.Here,we perform soft point-contact spectroscopy(SPCS)measurements in CsV_(3)Sb_(5) to investigate the evolution of superconducting order parameter with pressure.Surprisingly,we find that the superconducting gap is significantly enhanced between the two SC domes,at which the zero-resistance temperature is suppressed and the transition is remarkably broadened.Moreover,the temperature-dependence of the SC gap in this pressure range severely deviates from the conventional Bardeen-Cooper-Schrieffer(BCS)behavior,evidencing for strong Cooper pair phase fluctuations.These findings reveal the complex intertwining of the CDW with SC in the compressed CsV_(3)Sb_(5),suggesting striking parallel to the cuprate superconductor La2xBaxCuO4.Our results point to the essential role of charge degree of freedom in the development of intertwining electronic orders,and thus provide new constraints for theories.Xikai Wen Fanghang Yu Zhigang Gui Yuqing Zhang Xingyuan Hou Lei Shan Tao Wu Ziji Xiang Zhenyu Wang Jianjun Ying Xianhui Chen 2023Science Bulletin2023,,3:1

11Characteristics of homologous recombination repair pathway genes mutation in ovarian cancers显示文摘Background:Few studies have investigated the characteristics of non‐BRCA homologous recombination repair(HRR)pathway somatic mutations,and the impact of these mutations on efficacy of treatment in ovarian cancer patients is not clear.Therefore,we conducted this study to analyze the frequency and spectrum of somatic mutations in HRR pathway genes in patients with ovarian cancer and to examine the relationships between somatic mutations in HRR pathway genes and their effects on the efficacy of platinum‐based chemotherapy.Methods:We performed targeted sequencing of 688 genes related to the occurrence,development,treatment,and prognosis of solid tumors.Somatic mutations were identified by paired analysis of tumor tissue and germline DNA in blood cells.Results:A total of 38 patients with ovarian cancer were included in the study,and 35(92.1%)patients were diagnosed with high‐grade serous carcinoma.All patients exhibited somatic mutations in the tumor tissue samples.The commonly mutated genes were TP53(73.7%),BRCA2(55.3%),NF1(52.6%),BRCA1(47.4%),and CDH1(47.4%).Overall,71.1%of the patients exhibited mutation in at least one HRR pathway gene.The most frequently altered HRR genes were BRCA2(55.3%),followed by BRCA1(47.4%),ATM(44.7%),BARD1(42.1%),and CHEK1(36.8%).The median progression‐free survival(PFS)in patients with HRR pathway mutation was 36.0 months compared with 13.6 months in patients with no HRR pathway mutation(hazard ratio[HR],0.25;95%confidence interval[CI],0.08–0.77;p=0.016).Patients harboring BRCA1/2 and/or CDK12 mutations displayed a longer PFS(median,36.0 months)compared with patients with no BRCA1/2 or CDK12 mutation(median,13.6 months;HR,0.21;95%CI,0.07–0.61;p=0.004).In multivariate analysis Cox proportional hazards models,after adjustment for tumor stage at diagnosis and histology of initial diagnosis,patients with HRR pathway mutation had a longer PFS than patients with HRR wild‐type genes(p=0.006).Conclusions:HRR pathway somatic mutations are common in Chinese patients with ovarian cancer.HRR pathway somatic mutations were associated with improved sensitivity to platinum-based chemotherapy.Large-scale prospective studies are needed to verify our findings.Zongbi Yi Min Chen Shaoxing Sun Chunxu Yang Zijie Mei Hui Yang Qingming Xiang Hui Qiu 2022Cancer Innovation2022,1,3:0
12An anti-inflammatory and neuroprotective biomimetic nanoplatform for repairing spinal cord injury显示文摘Spinal cord regeneration after a spinal cord injury (SCI) remains a difficult challenge due to the complicated inflammatory microenvironment and neuronal damage at the injury sites. In this study, retinoic acid (RA) and curcumin (Cur) were co-loaded with bovine serum albumin (BSA) self-assembly to obtain RA@BSA@Cur nanoparticles (NPs) for SCI treatment. Cur, as an antioxidant drug, facilitated reactive oxygen species (ROS) scavenging, and decreased the amount of inflammatory factors secreted by macrophages, while RA could enhance neurite extensions and neural differentiation. The constructed RA@BSA@Cur NPs not only induced polarization of macrophages toward pro-regenerative phenotypes and markedly reduced the inflammatory response of macrophages or microglia, but also increased neurite length in PC12 cells and neuronal differentiation of bone marrow mesenchymal stem cells, improved the differentiation of neural stem cells (NSCs) into β3- tubulin+ neurons, and reversed the pro-astrocyte differentiation effect of inflammatory cytokines on NSCs. In vivo experiments revealed that RA@BSA@Cur NPs regulated the phenotypic polarization of macrophages, inhibited the release of inflammatory mediators, promoted functional neuron regeneration and motor function, and further inhibited scar tissue formation. 13Magnetic field-induced electronic phase transition in the Dirac semimetal state of black phosphorus under pressure显示文摘Different instabilities have been confirmed to exist in the three-dimensional(3D) electron gas when it is confined to the lowest Landau level in the extreme quantum limit. The recently discovered 3D topological semimetals offer a good platform to explore these phenomena due to the small sizes of their Fermi pockets, which means the quantum limit can be achieved at relatively low magnetic fields. In this work, we report the high-magnetic-field transport properties of the Dirac semimetal state in pressurized black phosphorus. Under applied hydrostatic pressure, the band structure of black phosphorus goes through an insulator-semimetal transition. In the high pressure topological semimetal phase, anomalous behaviors are observed on both magnetoresistance and Hall resistivity beyond the relatively low quantum limit field, which is demonstrated to indicate the emergence of an exotic electronic state hosting a density wave ordering. 14Macrophage-derived exosomes modulate wear particle-induced osteolysis via miR-3470b targeting TAB3/NF-κB signaling显示文摘Aseptic prosthesis loosening(APL)is one of the most prevalent complications associated with arthroplasty.The main cause is the periprosthetic osteolysis induced by wear particles.However,the specific mechanisms of crosstalk between immune cells and osteoclasts/osteoblasts during osteolysis are unclear.In this study,we report the role and mechanism of macrophage-derived exosomes in wear particle-induced osteolysis.The results of exosomes up-taken experiments revealed that osteoblast and mature osteoclasts capture macrophage-derived exosomes(M-Exo).Next-generation sequencing and RT-qPCR on M-Exo revealed that exosomal microRNA miR-3470b was downregulated in wear particle-induced osteolysis.The results of analysis on Luciferase reporter assays/fluorescence in situ hybridization(FISH)/immunofluorescence(IF)/immunohistochemistry(IHC)and co-culture experiments demonstrated that wear particles induced osteoclast differentiation by increasing the expression of NFatc1 via M-Exo miR-3470b targeting TAB3/NF-κB signaling.We also illustrate that engineered exosomes enriching miR-3470b facilitated to suppressed the osteolysis;the microenvironment enriching with miR-3470b could suppress wear particle-induced osteolysis via inhibition of TAB3/NF-κB in vivo.In summary,our findings indicate that macrophage-derived exosomes transfer to osteoclasts to induce osteolysis in wear particle-induced APL.Engineering exosomes enriching with miR-3470b might be a novel strategy for the targeting treatment of bone resorption-related diseases.Baiqi Pan Ziji Zhang Xiaoyu Wu Guoyan Xian Xuantao Hu Minghui Gu Linli Zheng Xiang Li Lingli Long Weishen Chen Puyi Sheng 2023Bioactive Materials2023,,8:0
