| 3 | Gaseous sulfur trioxide induced controllable sulfonation promoting biomineralization and osseointegration of polyetheretherketone implants显示文摘Fabricating a desired porous structure on the surface of biomedical polyetheretherketone(PEEK)implants for enhancing biological functions is crucial and difficult due to its inherent chemical inertness.In this study,a porous surface of PEEK implants was fabricated by controllable sulfonation using gaseous sulfur trioxide(SO3)for different time(5,15,30,60 and 90 min).Micro-topological structure was generated on the surface of sulfonated PEEK implants preserving original mechanical properties.The protein absorption capacity and apatite forming ability was thus improved by the morphological and elemental change with higher degree of sulfonation.In combination of the appropriate micromorphology and bioactive sulfonate components,the cell adhesion,migration,proliferation and extracellular matrix secretion were obviously enhanced by the SPEEK-15 samples which were sulfonated for 15 min.Finding from this study revealed that controllable sulfonation by gaseous SO3 would be an extraordinarily strategy for improving osseointegration of PEEK implants by adjusting the microstructure and chemical composition while maintaining excellent mechanical properties. | Teng Wan Zixue Jiao Min Guo Zongliang Wang Yizao Wan Kaili Lin Qinyi Liu Peibiao Zhang | 2020 | Bioactive Materials2020,5,4: | 5 |

CMA activity declines with age and is altered in neurodegenerative diseases. Its impairment leads to the accumulation of aggregated proteins, some of which may be directly tied to the pathogenic processes of neurodegenerative diseases. Its induction may accelerate the clearance of pathogenic proteins and promote cell survival, representing a potential therapeutic approach for the treatment of neurodegenerative diseases. | 7 | Long noncoding RNA UC.98 stabilizes atherosclerotic plaques by promoting the proliferation and adhesive capacity in murine aortic endothelial cells显示文摘Pathological studies have shown that the vulnerability of plaques affects outcomes in patients with atherosclerosis(AS),a chronic inflammatory disease and common cause of morbidity and mortality worldwide.Although emerging technologies have enabled early diagnosis of AS with high-risk vulnerable plaques,more accurate and noninvasive diagnostic methods are urgently required.To this end,molecules involved in genetic or epigenetic regulation of the vulnerability of atherosclerotic plaques have been extensively studied.Here,we evaluated long noncoding RNA(IncRNA)variability by microarray assay in murine aortic endothelial cells(MAECs)bearing wulnerable plaques and identified the novel functional IncRNA UC.98,whose expression pattern was associated with the vulnerability of atherosclerotic plaques.Consisient with this,clinical statistics comparing the peripheral blood specimens from sets of patients with AS with or without vulnerable plaques confirmed the linear relationship between the expression pattern of UC.98 and plaque instabilty.Moreover,MTT assays and western blot analysis showed that silencing of intrinsic UC 98 in MAECs not only suppressed cell proliferation but also decreased the expressions of vascular cell adhesion molecule-1 and intercellular adhesion molecule-1,thereby inactivating the nuclear factorkB pathway.In conclusion,our results highlighted the pivotal role of UC.98 in regulating the vulnerability of plaques during AS progression and suggested that UC.98 may be a biomarker of the early diagnosis and prognosis of AS with vulnerable plaques and a potential therapeutic target for slowing AS progression. | Zixu Fan Ying Zhang Danrui Xiao Jianwei Ma Hua Liu Linghong Shen Min Zhang Ben He | 2020 | Acta Biochimica et Biophysica Sinica2020,52,2: | 3 |

| 16 | Integrated microdevice with a windmill-like hole array for the clog-free,efficient,and self-mixing enrichment of circulating tumor cells显示文摘Circulating tumor cells(CTCs)have tremendous potential to indicate disease progression and monitor therapeutic response using minimally invasive approaches.Considering the limitations of affinity strategies based on their cost,effectiveness,and simplicity,size-based enrichment methods that involve low-cost,label-free,and relatively simple protocols have been further promoted.Nevertheless,the key challenges of these methods are clogging issues and cell aggregation,which reduce the recovery rates and purity.Inspired by the natural phenomenon that the airflow around a windmill is disturbed,in this study,a windmill-like hole array on the SU-8 membrane was designed to perturb the fluid such that cells in a fluid would be able to self-mix and that the pressure acting on cells or the membrane would be dispersed to allow a greater velocity.In addition,based on the advantages of fluid coatings,a lipid coating was used to modify the membrane surface to prevent cell aggregation and clogging of the holes.Under the optimal conditions,recovery rates of 93%and 90%were found for A549 and HeLa cells in a clinical simulation test of our platform with a CTC concentration of 20-100 cells per milliliter of blood.The white blood cell(WBC)depletion rate was 98.7%(n=15),and the CTC detection limit was less than 10 cells per milliliter of blood(n=6).Moreover,compared with conventional membrane filtration,the advantages of the proposed device for the rapid(2 mL/min)and efficient enrichment of CTCs without clogging were shown both experimentally and theoretically.Due to its advantages in the efficient,rapid,uniform,and clog-free enrichment of CTCs,our platform offers great potential for metastatic detection and therapy analyses. | Hao Li Jinze Li Zhiqi Zhang Zhen Guo Changsong Zhang Zixu Wang Qiuquan Guo Chao Li Chuanyu Li Jia Yao Anran Zheng Jingyi Xu Qingxue Gao Wei Zhang Lianqun Zhou | 2022 | Microsystems & Nanoengineering2022,8,1: | 1 |
