1Promotion of spinosad biosynthesis by chromosomal integration of the Vitreoscilla hemoglobin gene in Saccharopolyspora spinosa显示文摘To promote spinosad biosynthesis by improving the limited oxygen supply during high-density fermentation of Saccharopolyspora spinosa, the open reading frame of the Vitreoscilla hemoglobin gene was placed under the control of the promoter for the erythromycin resistance gene by splicing using overlapping extension PCR. This was cloned into the integrating vector pSET152, yielding the Vitreoscilla hemoglobin gene expression plasmid pSET152EVHB. This was then introduced into S. spinosa SP06081 by conjugal transfer, and integrated into the chromosome by site-specific recombination at the integration site ΦC31 on pSET152EVHB. The resultant conjugant, S. spinosa S078-1101, was genetically stable. The integration was further confirmed by PCR and Southern blotting analysis. A carbon monoxide differential spectrum assay showed that active Vitreoscilla hemoglobin was successfully expressed in S. spinosa S078-1101. Fermentation results revealed that expression of the Vitreoscilla hemoglobin gene significantly promoted spinosad biosynthesis under normal oxygen and moderately oxygen-limiting conditions (P<0.01). 2Phase demodulation method based on a dual-identical-chirped-pulse and weak fiber Bragg gratings for quasi-distributed acoustic sensing显示文摘A phase demodulation method for quasi-distributed acoustic sensing(DAS)systems based on a dual-identicalchirped-pulse and weak fiber Bragg gratings(WFBGs)is proposed.Compared to the use of Rayleigh backscattering light in optical fibers,the implementation of WFBGs can contribute to obtaining an optical signal with a higher signal-to-noise ratio(SNR).The dual-identical-chirped-pulse is generated by a time-delay fiber,and the sinusoidal carrier is generated by the interference between the two chirped pulses reflected by adjacent WFBGs.The phase of the sinusoidal carrier represents the dynamic strain change posed on the sensing fiber.Discrete Fourier transform is used to directly retrieve the phase information.The performance of the phase demodulation from interference signals under different sinusoidal carrier frequencies and SNRs is numerically investigated.The piezoelectric transducer is employed to emulate the sound in the experiment to verify the effectiveness of our method.It is shown that the dynamic strain can be well reconstructed at the end of a 101.64 km fiber when the signal SNR is down to 3.234 d B.Our proposed method enables the application of the long-distance sensing in quasi-DAS systems.Guanhua Liang Junfeng Jiang Kun Liu Shuang Wang Tianhua Xu Wenjie Chen Zhe Ma Zhenyang Ding Xuezhi Zhang Yongning Zhang Tiegen Liu 2020Photonics Research2020,8,7:6
3Effects of ionic strength and temperature on the aggregation and deposition of multi-walled carbon nanotubes显示文摘The aggregation and deposition of carbon nanotubes(CNTs) determines their transport and fate in natural waters.Therefore,the aggregation kinetics of humic-acid treated multi-walled carbon nanotubes(HA-MWCNTs) was investigated by time-resolved dynamic light scattering in NaCl and CaCl_2 electrolyte solutions.Increased ionic strength induced HA-MWCNT aggregation due to the less negative zeta potential and the reduced electrostatic repulsion.The critical coagulation concentration(CCC) values of HA-MWCNTs were 80 mmol/L in NaCl and 1.3 mmol/L in CaCl_2 electrolyte,showing that Ca^(2+) causes more serious aggregation than Na^+.The aggregation behavior of HA-MWCNTs was consistent with Derjaguin-Landau-Verwey-Overbeek theory.The deposition kinetics of HA-MWCNTs was measured by the optical absorbance at 800 ran.The critical deposition concentrations for HA-MWCNT in NaCl and CaCl_2 solutions were close to the CCC values,therefore the rate of deposition cannot be increased by changing the ionic strength in the diffusion-limited aggregation regime.The deposition process was correlated to the aggregation since larger aggregates increased gravitational deposition and decreased random Brownian diffusion.HA-MWCNTs hydrodynamic diameters were evaluated at 5,15 and 25℃.Higher temperature caused faster aggregation due to the reduced electrostatic repulsion and increased random Brownian motion and collision frequency.HA-MWCNTs aggregate faster at higher temperature in either NaCl or CaCl_2electrolyte due to the decreased electrostatic repulsion and increased random Brownian motion.Our results suggest that CNT aggregation and deposition are two correlated processes governed by the electrolyte,and CNT transport is favored at low ionic strength and low temperature.Lixin Wang Xuezhi Yang Qi Wang Yuxuan Zeng Lei Ding Wei Jiang 2017Journal of Environmental Sciences2017,29,1:5
4The role of β18-β19 loop structure in insecticidal activity of Cry1Ac toxin from Bacillus thuringiensis显示文摘The β18-β19 loop in domain III of Cry1Ac toxin is unique among Bacillus thuringiensis Cry proteins. In this study, the role of the loop structure in insecticidal activity of Cry1Ac toxin was investigated. Alanine scanning mutations within the loop were initially generated and most mutants were over-expressed and reduced toxicity at different degrees, except mutant N546A that showed almost 2 times enhanced toxicity against Helicoverpa armigera larvae. Further mutagenic analysis of N546 revealed that a charged amino acid in this position would cause very unfavorable influence on insecticidal activity. In addition, the deletion of N546 led to protein instability because of destruction of the loop integrity. Besides, mutant W544F was much more toxic than W544Y, indicating that hydrophobic nature of the position was important for maintaining the stability and activity of Cry1Ac protein. 5Thermo-Economic Modeling and Evaluation of Physical Energy Storage in Power System显示文摘In order to assess the electrical energy storage technologies,the thermo-economy for both capacity-type and power-type energy storage are comprehensively investigated with consideration of political,environmental and social influence.And for the first time,the Exergy Economy Benefit Ratio(EEBR)is proposed with thermo-economic model and applied to three different storage systems in various scenarios,including pumped storage,compressed air energy storage and flywheel energy storage.The impact of the total system efficiency,annual utilization hour,life time,and other key factors are also analyzed.The results show that the EEBRs of pumped storage and compressed air energy storage under peak load shaving condition and flywheel energy storage under frequency modulation service condition are all larger than zero,which means they are all thermo-economically feasible.With extra consideration of political,environmental and social impact,the exergy cost could reduce by about 25%and the EEBR doubles.The sensitivity analysis indicates the similarity and diversity of influence to EEBR between capacity-type and power-type energy storage systems.The former is that energy efficiency is the dominated factor for all three storage systems.The latter is that the difference of exergy benefit mode causes variety in other major factors.For energy-type storage system,like pumped storage and compressed air storage,the peak-to-valley price ratio is very sensitive in energy arbitrage.For power-type storage system,like flywheel storage,the mileage ratio is in leading position in auxiliary service benefit by mileage.In the three cases studied,the pumped storage has the best thermo-economy;the compressed air energy storage is the second,and the flywheel energy storage is the third.The main reason is that the pumped storage has the least non-exergy cost,and flywheel has the most.HU Shan LIU Chang DING Jie XU Yujie CHEN Haisheng ZHOU Xuezhi 2021Journal of Thermal Science2021,30,6:4

16Analysis on Composition and Content of Glucosinolates in Broccoli Flowers and Leaves显示文摘Glucosinolate composition and content were evaluated in flowers and leaves of 12 different broccoli varieties. 18Study on the Complete Mitochondrial Genome of Qaidam Cattle(Bos taurus)显示文摘Qaidam cattle(Bos taurus) is an important endemic breed in Northwest China, which is mainly distributed in the northwest of Qinghai-Tibet Plateau. It has strong adaptive ability to plateau and swamp conditions, such as cold tolerance and insect resistance. In this study, the first complete mitochondrial genome of Qaidam cattle was reported. The circular double-stranded genome is 16 340 bp in size, and contains 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a D-loop region. The overall nucleotide composition is 33.4% A, 27.2% T, 26.0% C and 13.4% G, with a total A +T content of60.6%. The gene order and composition are similar to those of other B. taurus breeds.
