| 1 | Community structure, dispersal ability and functional profiling of microbiome existing in fat body and ovary of the brown planthopper, Nilaparvata lugens显示文摘The endosymbionts play vital roles in growth, development and reproduction in insects. Yeast-like endosymbionts (YLSs) have been well studied in Nilaparvata lugens (N. lugens), but little is known about the tissue-specific bacterial microbiomes, especially on the microbial intersection among internal tissues. Here, the correlation of microbial composition, structure, dispersal ability and functional profiling were illuminated in two tissues, the fat body and ovary in N. lugens. A total of 11 phyla and 105 genera were captured from all samples;Firmicutes and Proteobacteria were the most predominant and accounted for more than 99% in all samples. However, the relative abundance ofFirmicutes and Proteobacteria was significantly different in ovary and fat body through Fisher's Least Significant Difference test. Microbial diversity but not the richness index in the two tissues exhibited significant difference. Furthermore, the microbial community structure of the ovary and fat body were primarily determined by tissue quality. Firmicutes showed strong dispersal ability between ovary and fat body based on the quantitative null model assessing, indicating the frequent interaction of these microbiomes in the two tissues. In addition, the Kyoto Encyclopedia of Genes and Genomes pathways of microbial participation were delineated. The ten most abundant pathways counted for over 46% of the annotation and were shared between the two tissues, mainly containing Energy Metabolism and Amino Acid Metabolism/Biosynthesis. | 2 | Exploration of the key microbes and composition stability of microbial consortium GF-20 with efficiently decomposes corn stover at low temperatures显示文摘The microbial consortium GF-20(GF-20) can efficiently decompose corn stover at low temperatures. The present study explored the key microbes of GF-20 and evaluated different culture conditions on its composition stability to promote the utilization of corn stover decomposing microbes in low temperature regions. GF-20 was subcultured to the 15 th generation under different temperatures, pHs, carbon, and nitrogen sources. Then, the dynamics of fermenting pH, cellulose enzyme activities, carbohydrate concentration, and oxidation reduction potential were determined to estimate the degradation efficiency of corn stover with GF-20. Furthermore, the structural stability and functional microbes of GF-20 were identified on the basis of PCR-denaturing gradient gel electrophoresis(DGGE) profiling and principal component analysis. The results showed that the offspring of GF-20 subcultured under different temperatures(4–30°C) and pH(6.0–9.0) conditions maintained stable growth, decomposition function, and composition structure. Furthermore, consortia GF-20 had a stable composition structure, which induced GF-20 to secrete cellulose and promote substrate decomposition as corn stover and ammonium were used as sources of carbon and nitrogen, respectively. | 5 | Effects of biochar and litter on carbon and nitrogen mineralization and soil microbial community structure in a China fir plantation显示文摘To investigate the effects of biochar addition(1 or 3%)to the soil of a China fir plantation with or without litter,we conducted a 90-day incubation experiment.We also studied the C and N dynamics and the microbial community structure of the soil.In soil without litter,the application of biochar at a rate of 3%significantly decreased CO2 emissions,while addition of 1%biochar had no effect.Biochar application did not affect the net N mineralization rate but significantly reduced the NH4?concentration after 90 days.In litter-enriched soil,biochar application had no significant effect on total CO2 emissions;however,application of 3%biochar significantly reduced the net N mineralization rate.Biochar application to soil with or without litter immediately reduced the dissolved organic carbon(DOC)concentration independent of the application rate,which was primarily due to sorption of DOC by the biochar.Phospholipid fatty acid analysis demonstrated that both concentrations of added biochar to soil(with or without litter)altered the soil microbial community structure at the end of incubation,although the effect of biochar was not as strong as the effect of time or litter application.The effect of biochar addition alone on microbial community structure was inconsistent over time.Litter added to soil significantly increased fungi and reduced Gram-positive bacteria.In the presence of litter,biochar applied at both 1%and 3%significantly increased(p<0.05)the proportion of actinomycete only at day 90.Our results indicate biochar as a potentially effective measure for C sequestration in the test soil of a China fir plantation,even in the presence of litter. | Ying Li Chuifan Zhou Yunxiao Qiu Mulualem Tigabu Xiangqing Ma | 2019 | Journal of Forestry Research2019,30,5: | 3 |
| 6 | Long-term submergence of non-methanogenic oxic upland field soils helps to develop the methanogenic archaeal community as revealed by pot and field experiments显示文摘The community structure of methanogenic archaea is relatively stable,i.e.,it is sustained at a high abundance with minimal changes in composition,in paddy field soils irrespective of submergence and drainage.In contrast,the abundance in non-methanogenic oxic soils is much lower than that in paddy field soils.This study aimed to describe methanogenic archaeal community development following the long-term submergence of non-methanogenic oxic upland field soils in pot and field experiments.In the pot experiment,a soil sample obtained from an upland field was incubated under submerged conditions for 275 d.Soil samples periodically collected were subjected to culture-dependent most probable number(MPN)enumeration,polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE)analysis of archaeal 16 S r RNA gene,and quantitative PCR analysis of the methyl-coenzyme M reductase alpha subunit gene(mcr A)of methanogenic archaea.The abundance of methanogenic archaea increased from 102 to 103 cells g-1 dry soil and 104 to 107 copies of mcr A gene g-1 dry soil after submergence.Although no methanogenic archaeon was detected prior to incubation by the DGGE analysis,members from Methanocellales,Methanosarcinaceae,and Methanosaetaceae proliferated in the soils,and the community structure was relatively stable once established.In the field experiment,the number of viable methanogenic archaea in a rice paddy field converted from meadow(reclaimed paddy field)was monitored by MPN enumeration over five annual cycles of field operations.Viability was also determined simultaneously in a paddy field where the plow layer soil from a farmer’s paddy field was dressed onto the meadow(dressed paddy field)and an upland crop field converted from the meadow(reclaimed upland field).The number of viable methanogenic archaea in the reclaimed paddy field was below the detection limit before the first cultivation of rice and in the reclaimed upland field.Then,the number gradually increased over five years and finally reached 103–104 cells g-1 dry soil,which was comparable to that in the dressed paddy field.These findings showed that the low abundance of autochthonous methanogenic archaea in the non-methanogenic oxic upland field soils steadily proliferated,and the community structure was developed following repeated and long-term submergence.These results suggest that habitats suitable for methanogenic archaea were established in soil following repeated and long-term submergence. | Takeshi WATANABE Susumu ASAKAWA Koichi HAYANO | 2020 | Pedosphere2020,30,1: | 2 |

| 9 | Removal of tetrachlorobisphenol A and the effects on bacterial communities in a hybrid sequencing biofilm batch reactor-constructed wetland system显示文摘Tetrachlorobisphenol A (TCBPA) released into the sewage may cause environmental pollution and health risk to human beings. The objective of this study was to investigate the removal ofTCBPA and bacterial community structures in a laboratory-scale hybrid sequencing biofilm batch reactor (SBBR)- constructed wetland (CW) system. The results showed that the removal efficiency of chemical oxidation demand (COD), ammonia, total nitrogen and total phosphorus in the SBBR-CW system was 96.7%, 97.3%, 94.4%, and 8&6% respectively. At the stable operation stage, the system obtained a 71.7%±1.8% of TCBPA removal efficiency with the influent concentration at 200μg/L. Illumina MiSeq sequencing of 16S rRNA gene revealed that the presence of TCBPA not only reduced the bacterial diversity in the SBBR-CW system, but also altered the composition and structure of bacterial community. After the addition of TCBPA, Proteobacteria increased from 31.3% to 38.7%, while Acidobacteria and Parcubacteria decreased greatly in the SBBR. In contrast, Acidobacteria replaced Proteobacteria as the dominant phylum in the upper soils of CW. The results indicated that TCBPA stimulated the growth of GAOs in the SBBR without deteriorating the phosphorus removal due to the presence of sufficient carbon sources.
