| 1 | The characteristics of CO_2 assimilation of photosynthesis and chlorophyll fluorescence in transgenic PEPC rice显示文摘With PEPC, PPDK, NADP-ME and PEPC+ PPDK transgenic and untransformed rice (Orysa sativa L.), the activities of related C4 photosynthesis enzymes, the chlorophyll fluorescence parameters, CO2 exchange and other physiological indexes were compared, in which the physiological characteristics of PEPC transgenic rice were mainly studied. The results were as follows: (i) The activities of PEPC in PEPC transgenic rice were 20-fold higher than those in untransformed rice; the light-saturation photosynthetic rates and the carboxylation efficiency of PEPC transgenic rice were increased by 55% and 50% more than those of untransformed rice, respectively, while the CO2 compensation point decreased by 27%. | 2 | Protective mechanism of desiccation tolerance in Reaumuria soongorica: Leaf abscission and sucrose accumulation in the stem显示文摘Reaumuria soongorica (Pall.) Maxim., a perennial semi-shrub, is widely found in semi-arid areas in northwestern China and can survive severe desiccation of its vegetative organs. In order to study the protective mechanism of desiccation tolerance in R. soongorica, diurnal patterns of net photosynthetic rate (Pn), water use efficiency (WUE) and chlorophyll fluorescence parameters of Photosystem II (PSII), and sugar content in the source leaf and stem were investigated in 6-year-old plants during progressive soil drought imposed by the cessation of watering. The results showed that R. soongorica was char-acterized by very low leaf water potential, high WUE, photosynthesis and high accumulation of sucrose in the stem and leaf abscission under desiccation. The maximum Pn increased at first and then de-clined during drought, but intrinsic WUE increased remarkably in the morning with increasing drought stress. The maximal photochemical efficiency of PSII (Fv/Fm) and the quantum efficiency of noncyclic electric transport of PSII(ΦPSII) decreased significantly under water stress and exhibited an obvious phenomenon of photoinhibition at noon. Drought stressed plants maintained a higher capacity of dis-sipation of the excitation energy (measured as NPQ) with the increasing intensity of stress. Conditions of progressive drought promoted sucrose and starch accumulation in the stems but not in the leaves. However, when leaf water potential was less than –21.3 MPa, the plant leaves died and then abscised. But the stem photosynthesis remained and, afterward the plants entered the dormant state. Upon re-watering, the shoots reactivated and the plants developed new leaves. | 4 | Global patterns of the responses of leaf-level photosynthesis and respiration in terrestrial plants to experimental warming显示文摘Aims The balance between leaf photosynthesis and respiration of terrestrial plants determines the net carbon(C)gain by vegetation and consequently is important to climate–C cycle feedback.This study is to reveal the global patterns of the responses of leaf-level net photosynthesis and dark respiration to elevated temperature.Methods Data for leaf-level net photosynthesis rate(P_(n))and dark respiration rate(R_(d))in natural terrestrial plant species with standard deviation(or standard error or confidence interval)and sample size were collected from searched literatures on Web of Science.Then a metaanalysis was conducted to estimate the effects of experimental warming on leaf-level P_(n) and R_(d) of terrestrial plants.Important findings Across all the plants included in the analysis,warming enhanced P_(n) and R_(d) significantly by 6.13 and 33.14%,respectively.However,the responses were plant functional type(PFT)specific.Specifically,photosynthesis of C_(4) herbs responded to experimental warming positively but that of C_(3) herbs did not,whereas their respiratory responses were similar,suggesting C_(4) plants would benefit more from warming.The photosynthetic response declined linearly with increasing ambient temperature.The respiratory responses linearly enhanced with the increase in warming magnitude.In addition,a thermal acclimation of R_(d),instead of P_(n),was observed.Although greater proportion of fixed C was consumed(greater R_(d)/P_(n) ratio),warming significantly enhanced the daily net C balance at the leaf level.This provides an important mechanism for the positive responses of plant biomass and net primary productivity to warming.Overall,the findings,including the contrastive responses of different PFTs and the enhancement in daily leaf net C balance,are important for improving model projection of the climate–C cycle feedback. | Junyi Liang Jianyang Xia Lingli Liu Shiqiang Wan | 2013 | Journal of Plant Ecology2013,6,6: | 18 |
| 5 | Impacts of increased atmospheric CO_2 concentration on photosynthesis and growth of micro-and macro-algae显示文摘Marine photosynthesis drives the oceanic biological CO2 pump to absorb CO2 from the atmosphere, which sinks more than one third of the industry-originated CO2 into the ocean. The increasing atmos-pheric CO2 and subsequent rise of pCO2 in seawater, which alters the carbonate system and related chemical reactions and results in lower pH and higher HCO3- concentration, affect photosynthetic CO2 fixation processes of phytoplanktonic and macroalgal species in direct and/or indirect ways. Although many unicellular and multicellular species can operate CO2-concentrating mechanisms (CCMs) to util-ize the large HCO3- pool in seawater, enriched CO2 up to several times the present atmospheric level has been shown to enhance photosynthesis and growth of both phytoplanktonic and macro-species that have less capacity of CCMs. Even for species that operate active CCMs and those whose photo-synthesis is not limited by CO2 in seawater, increased CO2 levels can down-regulate their CCMs and therefore enhance their growth under light-limiting conditions (at higher CO2 levels, less light energy is required to drive CCM). Altered physiological performances under high-CO2 conditions may cause genetic alteration in view of adaptation over long time scale. Marine algae may adapt to a high CO2 oceanic environment so that the evolved communities in future are likely to be genetically different from the contemporary communities. | 6 | Improving water-use efficiency by decreasing stomatal conductance and transpiration rate to maintain higher ear photosynthetic rate in drought-resistant wheat显示文摘In wheat, the ear is one of the main photosynthetic contributors to grain filling under drought stress conditions. In order to determine the relationship between stomatal characteristics and plant drought resistance, photosynthetic and stomatal characteristics and water use efficiency(WUE) were studied in two wheat cultivars: the drought-resistant cultivar ‘Changhan 58' and the drought-sensitive cultivar ‘Xinong 9871'. Plants of both cultivars were grown in pot conditions under well-watered(WW) and water-stressed(WS) conditions. In both water regimes,‘Changhan 58' showed a significantly higher ear photosynthetic rate with a lower rate of variation and a significantly higher percentage variation of transpiration compared to control plants at the heading stage under WS conditions than did ‘Xinong 9871' plants. Moreover,‘Changhan 58' showed lower stomatal density(SD) and higher stomatal area per unit organ area(A) under both water conditions. Water stress decreased SD, A, and stomatal width(SW), and increased stomatal length in flag leaves(upper and lower surfaces) and ear organs(awn, glume,lemma, and palea), with the changes more pronounced in ear organs than in flag leaves.Instantaneous WUE increased slightly, while integral WUE improved significantly in both cultivars. Integral WUE was higher in ‘Changhan 58', and increased by a greater amount, than in‘Xinong 9871'. | 9 | Photosynthetic bicarbonate utilization in Porphyra haitanensis(Bangiales,Rhodophyta)显示文摘The activities of carbonic anhydrase (CA) and photosynthesis of Porphyra haitanensis were investigated in order to see its photosynthetic utilization of inorganic carbon source. Both intra- and extra-cellular CA activities existed in the thallus. CA inhibitors, acetazolamide (AZ) and ethoxy-zolamide (EZ), remarkably depressed the photosynthetic oxygen evolution in seawater of pH 8.2 and 10.0, and EZ showed stronger inhibition than AZ. The observed net photosynthetic rate in seawater of pH 8.2 was much higher than that of CO2 supply theoretically derived from spontaneous dehydration of HCO3. P. haitanensis also showed a rather high pH compensation point (9.9). The results demonstrated that P. haitanensis could utilize bicarbonate as the external inorganic carbon source for photosynthesis. | 10 | Cold acclimation improves photosynthesis by regulating the ascorbate–glutathione cycle in chloroplasts of Kandelia obovata显示文摘As the most northerly mangrove species in China, Kandelia obovata may undergo extreme cold event stress. Enhancing the cold tolerance of this species is crucial to its successful afforestation. This study aimed to determine the resistance of K. obovata seedlings to low temperature stress by cold acclimation and to explain the mechanisms for alleviating cold injury. To understand these mechanisms, seedlings that were acclimatized and not acclimatized were exposed to 5℃/- 2℃(day/night)for 48 h.Results showed that low temperature stress reduced leaf photosynthesis of non-acclimatized seedlings by inducing oxidative stress and structural damage to chloroplasts. These phenomena were shown by increasing levels of malondialdehyde (MDA), O2-and H2O2, as well as decreasing enzyme activities in the ascorbate–glutathione (AsA-GSH) cycle. However, cold-acclimatized seedlings had improved photosynthetic rates and efficiency of photosystem II (PSII) under low temperature stress. Compared with non-acclimatized seedlings, leaves of coldacclimatized seedlings under low temperature stress for 48 h exhibited higher anti-oxidative enzyme activities, lower levels of O2^- and H2O2, less damage to chloroplast structure, and removed 33.7% of MDA at low temperature stress for 48 h. | 11 | A Synthetic Photorespiratory Shortcut Enhances Photosynthesis to Boost Biomass and Grain Yield in Rice显示文摘Several photorespiratory bypasses have been introduced into plants and shown to improve photosynthesis by increasing chloroplastic C02 concentrations or optimizing energy balance.We recently reported that an engineered GOC bypass could increase photosynthesis and productivity in rice.However,the grain yield of GOC plants was unstable,fluctuating in different cultivation seasons because of varying seed setting rates.In this study,we designed a synthetic photorespiratory shortcut(the GCGT bypass)consisting of genes en-coding Oryza sativa glycolate oxidase and Escherichia coli catalase,glyoxylate carboligase,and tartronic semialdehyde reductase.The GCGT bypass was guided by an optimized chloroplast transit peptide that targeted rice chloroplasts and redirected 75% of carbon from glycolate metabolism to the Calvin cycle,identical to the native photorespiration pathway.GCGT transgenic plants exhibited significantly increased biomass production and grain yield,which were mainly attributed to enhanced photosynthesis due to increased chloroplastic C02 concentrations.Despite the increases in biomass production and grain yield,GCGT transgenic plants showed a reduced seed setting rate,a phenotype previously reported for the GOC plants.Integrative transcriptomic,physiological,and biochemical assays revealed that photosynthetic car-bohydrates were not transported to grains in an efficient manner,thereby reducing the seed setting rate.Taken together,our results demonstrate that the GCGT photorespiratory shortcut confers higher yield by promoting photosynthesis in rice,mainly through increasing chloroplastic C02 concentrations. | Li-Min Wang Bo-Ran Shen Bo-Di Li Chuan-Ling Zhang Min Lin Pan-Pan Tong Li-Li Cui Zhi-Sheng Zhang Xin-Xiang Peng | 2020 | Molecular Plant2020,13,12: | 11 |

| 13 | Effect of Silicon on Growth and Development of Strawberry under Water Deficit Conditions显示文摘Water stress is a major factor that limits agricultural crop production. Silicon(Si) is generally considered as a beneficial element for the growth of higher plants, especially for those grown under stressful environment. This study was conducted to examine the effects of Si on growth and development of strawberry(Fragaria × ananassa ‘Camarosa') under water stress conditions. A factorial experiment, in a completely randomized design, was used to investigate the effects of three irrigation levels and four Si treatments consisting of 0, 5, 10, and 15 mmol·L^(-1) potassium silicate(K_2 SiO_3). The results showed that an increase in the levels of water stress caused a decrease in most of the quantitative characteristics such as specific leaf area, chlorophyll fluorescence, net photosynthesis rate, and stomata conductance; addition of Si significantly increased most of the mentioned factors; water stress increased electrolyte leakage, proline, and water use efficiency(WUE); Si treatment significantly decreased transpiration rate and improved chlorophyll content and WUE. Water stress stimulated mineral nutrient absorption whereas Si application decreased it under water stress. In conclusion, it was found that in most of the investigated factors, 10 mmol·L^(-1) potassium silicate had the best effect on growth and development of strawberry. | 17 | Suboptimal Temperature Acclimation Enhances Chilling Tolerance by Improving Photosynthetic Adaptability and Osmoregulation Ability in Watermelon显示文摘The temperature drop of plants from the optimal requirements can increase tolerance to severe chilling stress. Photosynthesis and osmoregulators were analyzed during chilling stress to explore the adaptation mechanisms that underlie the induction of chilling tolerance in response to suboptimal temperature. The relationships of these processes to suboptimal temperature acclimation in watermelon were then determined.Suboptimal temperature-acclimated watermelon plants demonstrated tolerance during chilling stress, as indicated by the decreased electrolyte leakage and malondialdehyde accumulation compared with those non-acclimated watermelon plants. Chilling-induced photoinhibition and reduction in CO2 assimilation rate were alleviated after suboptimal temperature acclimation. The xanthophyll cycle level was enhanced by improving thermal dissipation ability and avoiding light damage. Consequently, the chilling tolerance of suboptimal temperature-acclimated watermelon plants was enhanced. The osmoregulation ability induced by suboptimal temperature acclimation protected watermelon plants against chilling injury because of the accumulation of small molecular substances, such as soluble sugar and proline. The protein levels of Rubisco activase(ClRCA) and the gene expression of the Benson–Calvin cycle simultaneously increased in suboptimal temperature-acclimated watermelon plants during chilling stress. | 18 | Precipitation regulates plant gas exchange and its long-term response to climate change in a temperate grassland显示文摘Aims Climate change largely impacts ecosystem carbon and water cycles by changing plant gas exchange,which may further cause positive or negative feedback to global climate change.However,long-term global change manipulative experiments are seldom conducted to reveal plant ecophysiological responses to climatic warming and altered precipitation regimes.Methods An 8-year field experiment with both warming and increased precipitation was conducted in a temperate grassland in northern China.We measured leaf gas exchange rates(including plant photosynthesis,transpiration and instantaneous water use efficiency[WUE])of two dominant plant species(Stipa sareptana var.krylovii and Agropyron cristatum)from 2005 to 2012(except 2006 and 2010)and those of other six species from 2011 to 2012.Important Findings Increased precipitation significantly stimulated plant photosynthetic rates(A)by 29.5%and 19.9%and transpiration rates(E)by 42.2%and 51.2%for both dominant species S.sareptana var.krylovii and A.cristatum,respectively,across the 8 years.Similarly,A and E of the six plant functional types were all stimulated by increased precipitation in 2011 and 2012.As the balance of A and E,the instantaneous WUEs of different plant species had species-specific responses to increased precipitation.In contrast,neither warming nor its interaction with increased precipitation significantly affected plant leaf gas exchange rates.Furthermore,A and E of the two dominant species and their response magnitudes to water treatments positively correlated with rainfall amount in July across years.We did not find any significant difference between the short-term versus long-term responses of plant photosynthesis,suggesting the flexibility of leaf gas exchange under climate change.The results suggest that changing precipitation rather than global warming plays a prominent role in determining production of this grassland in the context of climate change. | Bing Song Shuli Niu Shiqiang Wan | 2016 | Journal of Plant Ecology2016,9,5: | 8 |
| 19 | Enhanced tolerance to drought in transgenic rice plants overexpressing C_4 photosynthesis enzymes显示文摘Maize-specific pyruvate orthophosphate dikinase(PPDK) was overexpressed in rice independently or in combination with the maize C4-specific phosphoenolpyruvate carboxylase(PCK). The wild-type(WT) cultivar Kitaake and transgenic plants were evaluated in independent field and tank experiments. Three soil moisture treatments,well-watered(WW), moderate drought(MD) and severe drought(SD), were imposed from 9d post-anthesis till maturity. Leaf physiological and biochemical traits, root activities,biomass, grain yield, and yield components in the untransformed WT and two transgenic rice lines(PPDK and PCK) were systematically studied. Compared with the WT, both transgenic rice lines showed increased leaf photosynthetic rate: by 20%–40% under WW, by45%–60% under MD, and by 80%–120% under SD. The transgenic plants produced 16.1%,20.2% and 20.0% higher grain yields than WT under the WW, MD and SD treatments,respectively. Under the same soil moisture treatments, activities of phosphoenolpyruvate carboxylase(PEPC) and carbonic anhydrase(CA) in transgenic plants were 3–5-fold higher than those in WT plants. Compared with ribulose-1,5-bisphosphate carboxylase, activities of PEPC and CA were less reduced under both MD and SD treatments. The transgenic plants also showed higher leaf water content, stomatal conductance, transpiration efficiency, and root oxidation activity and a stronger active oxygen scavenging system than the WT under all soil moisture treatments, especially MD and SD. The results suggest that drought tolerance is greatly enhanced in transgenic rice plants overexpressing C4photosynthesis enzymes. This study was performed under natural conditions and normal planting density to evaluate yield advantages on a field basis.
