1Identification of a denitrifying bacterium and verification of its anaerobic ammonium oxidation ability显示文摘A strain D3 of denitrifying bacterium was isolated from an anammox reactor,and identi-fied as Pseudomonas mendocina based on the morphological and physiological assay,Vitek test,Biolog test,(G+C) mol% content,and 16S rDNA phylogenetic analysis.As a typical denitrifying bac-terium,strain D3 achieved the maximal nitrate reduction rate of 26.2 mg/(L·d) at the nitrate concen-tration of 88.5 mg N/L.The optimal pH and growth temperature were 7.84 and 34.9℃,respectively.Strain D3 was able to oxidize ammonia under anaerobic condition.The maximum nitrate and ammo-nium utilization rates were 6.37 mg/(L·d) and 3.34 mg/(L·d) ,respectively,and the consumption ratio of ammonia to nitrate was 1:1.91.Electron microscopic observation revealed peculiar cell inclusions in strain D3.Because of its relation to anammox activity,strain D3 was presumed to be anammoxosome.The present investigation proved that denitrifying bacteria have the anammox ability,and the results have engorged the range of anammox populations.HU Baolan ZHENG Ping LI Jinye XU Xiangyang JIN Rencun 2006Science China(Life Sciences)2006,49,5:13
2Vertical quantitative and dominant population distribution of the bacteria isolated from the Muztagata ice core显示文摘Vertical distribution of the main bacteria isolated from the Muztagata ice core (about 22.4 m) was investigated by means of cultivation and 16S rRNA sequence analysis. The results showed that the amount of culturable bacteria fluctuated with ice core depth, and was more in dirty layer than in clean ice, which suggested the close corresponding relationship between high input of the bacteria deposited by wind and snowflow and dirty layer. Most of the bacteria were psychrophiles and psychrotolerants, including α- and γ-proteobacteria, Cryobacterium psychro-philum, CFB (Cytophaga-Flavobacterium-Bacteroides) group, high-G+C gram-positive bacteria (HGC). Acinetobacter sp. and HGC repeatly occurred in different ice depths, and their quantita-tive distribution was consistent with the change of the total amount of culturable bacteria with depth, which suggested the main bio-indicator; while Flavobacterium, Cryobacterium psychro-philum, and α-proteobacteria, also functioned as a secondary indicator of climatic and environ-mental changes. 3Identification and Practical Application of Silicate-dissolving Bacteria显示文摘Slime-forming bacteria were isolated from soils, rock surface and earthworm intestine, and their effects on dissolving silicate minerals and tomato growth were examined. One of the bacteria, Bacillus mucilaginosus RGBc13, had particularly strong ability to form slime and dissolve silicates. RGBc13 could also colonize and develop in both non-rhizosphere and rhizosphere soil. Total number of slime-forming bacteria increased from 2.9 × 103 cfu·g- 1and 8.4 × 103 cfu·g-1 to 9.6 × 106 cfu·g-1 and 6.0 × 107 cfu·g-1 in the non-rhizosphere and rhizosphere soils respectively. Potassium and phosphorus nutritional conditions in the rhizosphere were markedly improved through inoculation of this bacterium. Available K and P respectively increased from 25.86 and 3.63mg· kg-1 in the non-rhizosphere soil to 91.23 and 5.74mg· kg-1 in the rhizosphere soil. Tomato biomass increased by 125%, K and P uptakes were more than 150%, greater than the non- inoculation. 4Spectroscopic evidence for triplet excitation energy transfer among carotenoids in the LH2 complex from photosynthetic bacterium Rhodopseudomonas palustris显示文摘The LH2 complex from Rhodopsudomonas (Rps.) palustris is unique in the heterogeneous carotenoid compositions. The dynamics of triplet excited state Carotenoids (3Car*) has been investigated by means of sub-microsecond time-resolved absorption spectroscopy both at physiological temperature (295 K) and at cryogenic temperature (77 K). Broad and asymmetric Tn←T1 transient absorption was observed at room temperature following the photo-excitation of Car at 532 nm, which suggests the contribution from various carotenoid compositions having different numbers of conjugated C=C double bonds (NC=C). The triplet absorption bands of different carotenoids, which superimposed at room temperature, could be clearly distinguished upon decreasing the temperature down to 77 K. At room temperature the shorter-wavelength side of the main Tn←T1 absorption band decayed rapidly to reach a spectral equilibration with a characteristic time constant of~1 μs, the same spectral dynamics, however, was not observed at 77 K. The aforementioned spectral dynamics can be explained in terms of the triplet-excitation transfer among heterogeneous carotenoid compositions. Global spectral analysis was applied to the time-resolved spectra at room temperature, which revealed two spectral components peaked at 545 and 565 nm, and assignable to the Tn←T1 absorption of Cars with NC=C=11 and NC=C=13, respectively. Surprisingly, the decay time constant of a shorter-con- jugated Car, I.e. 0.72 μs (aerobic) and 1.36 μs (anaerobic), is smaller than that of a longer-con- jugated Car, I.e. 2.12 μs (aerobic) and 3.75 μs (anaerobic), which is contradictory to the general rule of carotenoids and relative polyenes. The results are explained in terms of triplet-excitation transfer among different types of Cars. 5Vertical distribution of phosphorus and P-dissolving/ decomposing bacteria in the sediment of Guanting reservoir显示文摘In this study, we determined pH, phosphorus content and the number of P-dis- solving/decomposing bacteria in relation to the depth in the sediment of Guanting reservoir in Beijing. The pH value was slightly increased from 8.0 in the surface to 8.5 in the bottle (69 cm in depth) of sediment. The highest P content, 1269 mg/kg, was detected in the 35 cm layer, followed by the 5 cm (993 mg/kg) and 69 cm (580 mg/kg) layers. The number of inorganic P-dissolving bacteria varied from 6(102 to 8(104 and the organic P-discomposing bacteria were from 1.9(103 to 6.3(104 per gram sediment in different depths, which were counted under 28℃ and 20℃ ( pH7.5 and 8.5. The number of P-dissolving/discomposing bacteria was directly correlated to the P content in each layer of sediments. The analysis of P-dissolving/decomposing ability of bacteria showed that some of these bacteria were also capable of accumulating P intracellularly. The intracellular P-accumulation was more efficient at lower temperature; in contrast, the activity of P-dissolving/decomposing was stronger at higher temperature. So the content of dissolved P in water body, or quality of water, could be affected by the change of temperature via the regulation of bacterial activity. 6The Conditions of Releasing Potassium by a Silicate- dissolving Bacterial Strain NBT显示文摘The potassium-releasing characteristics of a bacterium from different minerals were studied through pure culture and soil column experiments. The results showed that the strain NBT of tested strains had the highest potassium-releasing capacity. It released 35.2 mg/L after 7days of pure culture incubation at 28℃, 31.8% - 1203.7% more than other tested strains. Potassium released from the minerals was obviously affected by pH, aerobic condition, soil and mineral properties. The strain NBT had a much higher potential to release potassium in the pH 6.5 - 8.0 than other pHs. Living cell inoculation resulted in an increase of 84.8%- 127.9% compared with that of the dead cell inoculation. More aerobic condition produced more K than a less aerobic one. The potassium-releasing order was as follows: illite > feldspar> muscovite. Soil column experiment showed that the bacterial number increased from (2.6 - 3.0) × 106/g to (6.8 - 7.4) × 107/g. 7Effects of hydraulic retention time on nitrification activities and population dynamics of a conventional activated sludge system显示文摘The effects of hydraulic retention time (HRT) on the nitrification activities and population dynamics of a conventional activated sludge system fed with synthetic inorganic wastewater were investigated over a period of 260 days. When the HRT was gradually decreased from 30 to 5 h, the specific ammonium-oxidizing rates (SAOR) varied between 0.32 and 0.45kg NH4+-N (kg mixed liquor suspended solids (MLSS)·d)-1, and the specific nitrate-forming rates (SNFR) increased from 0.11 to 0.50kg NO3-N (kg MLSS·d)1l, showing that the decrease in HRT led to a significant increase in the nitrite oxidation activity. According to fluorescence in situ hybridization (FISH) analysis results, the proportion of ammonia-oxidizing bacteria (AOBs) among the total bacteria decreased from 33% to 15% with the decrease in HRT, whereas the fraction of nitrite-oxidizing bacteria (NOBs), particularly the fast-growing Nitrobacter sp., increased significantly (from 4% to 15% for NOBs and from 1.5% to 10.6% for Nitrobacter sp.) with the decrease in HRT, which was in accordance with the changes in SNFR. 8Study on low-temperature lipase of psychrophilic bacterium 2-5-10-1 isolated from deep sea of Southern Ocean显示文摘A strain of psychrophilic bacterium, 2-5-10-1, which produces low-temperature lipase, is isolated from the deep sea of Prydz Bay in Southern Ocean. The highest lipase secretion of this strain is observed at 5 ℃ and this temperature is also for optimal growth. Tween 80 and olive oil enhance secretion of lipase. The optimal temperature and pH for lipase activity are 35 ℃ and 7.5 ℃ respectively. At 0℃, the lipase still has 37% relative enzyme activity. The lipase shows high thermolability, more than 50% activity lost after incubation at 60 ℃ for 15 min. EDTA has no effect on lipase activity, indicating the lipase activity is independent of divalent cation. 9Extracellular enzymatic activities of cold-adapted bacteria from polar oceans and effect of temperature and salinity on cell growth显示文摘The potential of 324 bacteria isolated from different habitats in polar oceans to produce a variety of extracellular enzymatic activities at low temperature was investigated. By plate assay, lipase, protease, amylase, gelatinase, agarase, chitinase or cellulase were detected. Lipases were generally present by bacteria living in polar oceans. Protease-producing bacteria held the second highest proportion in culturable isolates. Strains producing amylase kept a relative stable proportion of around 30% in different polar marine habitats. All 50 Arctic sea-ice bacteria producing proteases were cold-adapted strains, however, only 20% were psychrophilic. 98% of them could grow at 3% NaCl, and 56% could grow without NaCl. On the other hand, 98% of these sea-ice bacteria produced extracellular proteases with optimum temperature at or higher than 35℃, well above the upper temperature limit of cell growth. Extracellular enzymes including amylase, agarase, cellulase and lipase released by bacteria from seawater or sediment in polar oceans, most expressed maximum activities between 25 and 35℃. 10Fossil bacteria in Xuanlong iron ore deposits of Hebei Province显示文摘Discovered in Early Proterozoic Xuanlong iron ore deposits are six genera of fossil iron bacteria, i. e. sphere (coenobium of) rod-shaped (monomer) Naumanniella, ellipsoid elliptical Ochrobium,sphere spherical Siderocapsa and chain spherical Siderococcus, chain rod-shaped Leptothrix and Lieskeella, and six genera of fossil blue bacteria, namely sphere spherical Gloeocapsa, Synechocystis and Globobacter, chain spherical Anabaena and Nostoc, and con-strictive septate tubular Nodularia. The biomineralized monomers and coenobia of the two cate-gories of bacteria, together with hematite plates made up the bacteria pelletal, bacteria silky, bacteria fibrous and clasty bacteria pelletal textural lamina. The bacteria pelletal laminae com-bined with other bacteria laminae to make up oncolite, stromatolite and laminate. The precipita-tion of iron oxide was accelerated due to iron and blue bacteria cohabiting on microbial film or mat. 11Community diversity of bacteria in digestive tract of mud snail (Bullacta exarata Philippi) and its rearing shoal显示文摘The bacterial flora in the digestive tract of B. exarata Philippi and its rearing shoal were investigated, respectively. A total of 107 strains of heterotrophic bacteria, isolated from crop, stomach and intestine, mainly belong to genera Photobacterium, Bacillus, Pseudomonas and Vibrio. Varieties of bacteria in crop were significantly more than that in stomach and in intestine. A total of 173 strains of bacteria were isolated from the rearing shoal, belonging to 13 genera. The 5 predominant genera are Photobacterium, Bacillus, Pseudomonas, Vibrio and some genera of Enterobacteriaceae. 12Biological characteristics of marine bacterium S-9801 strain and its culture conditions of pigment production显示文摘Strain of Flavobacterium sp.(S-9801),was screened from 207 strains of marine bacteria isolated from the Bohai Sea continental shelf and the Zhujiang Estuary,for its red pigment production.The biological characteristics of strain S-9801 and culture conditions of pigment production have been checked out in this study. The color of the bacterial colony on 2216E medium was from coccineus to rose bengal. Optimum culture conditions were sodium chloride concentration(g/dm3),10~30;pH,3~8;temperature,25~28℃;tryptone and yeast extract as nitrogen sources and glucose as carbon source. 13Applicability and Limits of Bioremediation of Contaminated Groundwater by Organic Compounds显示文摘This paper gives in some ways a broad look at the Biorcmediation Technology in the treatment of polluted groundwater. Environmentalists and Hydrologeologists around the world, especially in the developed countries welcomed this revolutionizing technique at a moment when other methods were beconling rather expensive and sources of secondary and more challenging pollution problems across sole fresh groundwater. Bioremediation of contaminated groundwater is based on the use of bacteria which breakdown organic matters to more stable forms, which will not create nuisance or give off foul odors. The Applicability of this technology at a specific site lies in the understanding of the site’s hydrogeologic, physio- chemical backgrounds and the knowledge of the properties of the designated bacteria colonies, which would likely stabilize the contaminants. These are key points, which determine the success of the all process. 14Study on the Development of Lactic Acid Bacteria in Bread Dough Enriched with Zinc and Selenium显示文摘The incorporation of lyophilized lactic acid bacteria in dough formulation is a current trend in bread making in order toachieve quality improvement and extension of shelf life of bread. Another major aspect for today's bread making industry is anincrease of the biological value of bread, by addition of mineral substances that are deficient in bread (such as zinc and selenium).This is an effective approach to combat the existing mineral deficiency in population nutrition. The aim of the present study is toinvestigate the development of lactic acid bacteria in a medium enriched with zinc and selenium. It was found that the addition ofZnSO4-7H20 in an amount of 0.174 g/kg flour and Na2SeO3.5H20 in an amount of 880.28 μg/kg flour (alone and in combination) didnot inhibit the development of lactic acid bacteria used as starter cultures in bread dough formulation. It was found that the growth ofthe lactococcal component of the applied starters is dominant. 15Experimental study on reduction of U (Ⅵ) by an anaerobic bacterium, Shewanella putrefaciens: Application to sandstone-hosted interlayer oxidation-zone type uranium deposits, China显示文摘An experimental study on reduction of U (VI) by anaerobic bacteria, Shewanella putrefaciens, is first reported here in China. The experimental conditions were: 35℃ and pH = 7.0—7.4, corresponding to a physicochemical environments in which the sandstone-hosted in- terlayer oxidation-zone type uranium deposit formed in Northwest China’s Xinjiang. Bacteria adopted in the present experiment, Shewanella putrefaciens, occur extensively in natural envi- ronment. Our study shows that nano-crystal precipitates of uraninite quickly occurred on the surface of the cells within one week. It was found that the pitchblende was characterized by a random arrangement of uraninite nanocrystals (2—4 nm) in it, significantly different from natural pitchblende in which uraninite nanocrystals are arranged in order. Finally, a possible mechanism of uranium biomineralization by microorganisms in the deposits is discussed. Our investigation may supply a technical train of thoughts for bioremediation of nuclear-contaminated water envi- ronments and for underground dissolving extraction of the sandstone-hosted uranium ores.L.L.Barton 2005Science China Earth Sciences2005,48,3:0
