1Intestinal endotoxemia as a pathogenetic mechanism in liver failure显示文摘Liver injury induced by various pathogenic factors (such as hepatitis virus, ethanol, drugs and hepatotoxicants, etc.)through their respective special pathogenesis is referred to as 'primary liver injury' (PLI). Liver injury resultedfrom endotoxin (lipopolysaccharide, LPS) and the activation of Kupffer cells by LPS while intestinal endotoxemia (IETM) occurred during the occurrence and development of hepatitis is named the 'secondary liver injury' (SLI).The latter which has lost their own specificities of primary pathogenic factors is ascribed to IETM. The 'secondary liver injury' is of important action and impact on development and prognosis of hepatitis. More severe IETM commonly results in excessive inflammatory responses, with serious hepatic necrosis,further severe hepatitis and even induces acute liver failure.The milder IETM successively precipitates a cascade,including repeated and persistent hepatocytic impairment accompanied by infiltration of inflammatory cells, hepatic fibrosis, cirrhosis and hepatocarcinoma. Generally, the milder IETM ends with chronic hepatic failure. If PLI caused by various pathogenic factors through their independent specific mechanismis regarded as 'the first hit' on liver, then SLI mediated by different chemical mediators from KCs activated by IETM in the course of hepatitis is 'the second hit' on liver. Thus, fusing and overlapping of the primary and scondary liver injuries determine and influeuce the complexity of the illness and outcome of the patient with hepatitis. For this reason, the viewpoint of 'SLI' induced by the 'second hit' on liver inflicted by IFTM suggests that medical professionals should attach great importance to both 'PLI'and 'SLI' caused by IETM. 2Screening and evaluation of human single-chain fragment variable antibody against hepatitis B virus surface antigen显示文摘BACKGROUND: Phage display technology has become a vital tool in studies aimed at identifying molecules binding to a specific target. It enables the rapid generation and selection of high affinity, fully human antibody product candidates to essentially any disease target appropriate for antibody therapy. In this study, we prepared the recombinant single-chain fragment variable ( ScFv) antibody to hepatitis B virus surface antigen (HBsAg) by the phage display technology for obtaining a virus-targeting mediator. METHODS: mRNA was isolated from B-lymphocytes from a healthy volunteer and converted into cDNA. The fragment variables of heavy and light chain were amplified separately and assembled into ScFv DNA with a specially constructed DNA linker by polymerase chain reaction. The ScFv DNA was ligated into the phagmid vector pCANT-AB5E and the ligated sample was transformed into competent E. coli TG1. The transformed cells were infected with M13K07 helper phage to form a human recombinant phage antibody library. The volume and recombinant rate of the library were evaluated by bacterial colony count and restriction analysis. After two rounds of panning with HBsAg. the phage clones displaying ScFv of the antibody were selected by enzyme-linked immunosorbant assay ( ELISA) from the enriched phage clones. The antigen binding affinity of the positive clone was detected by competition ELISA. HB2151 E. coli was transfected with the positive phage clone demonstrated by competition ELISA for production of a soluble form of the anti-HBsAg ScFv. ELISA assay was used to detect the antigen binding affinity of the soluble anti-HBsAg ScFv. Finally, the relative molecular mass of soluble anti-HBsAg ScFv was measured by SDS-PAGE. RESULTS: The variable heavy ( VH ) and variable light (VL) and ScFv DNAs were about 340bp, 320bp and 750bp, respectively. The volume of the library was up to 2 × 106 and 8 of 10 random clones were recombinants. Two phage clones could strongly compete with the original HBsAb for binding to HBsAg. Within 2 strong positive phage clones, the soluble anti-HBsAg ScFv from one clone was found to have the binding activity with HBsAg. SDS-PAGE showed that the relative molecular weight of soluble anti-HBsAg ScFv was 32 kDa. 3Hydrodynamics of a Newly China-Made Jiuling Bileaflet Heart Valve Prostheses显示文摘Jiuling bileaflet prosthetic heart valve is manufactured in compliance with the national standard of P.R.C. GB12279 90. This mechanical heart valve substitute is constructed entirely of solid low temperature pyrolytic carbon. There is no graphite substrate within the leaflets or orifice. This prosthesis presents with satisfactory histocompatibility and induces little hemocoagulation around it. Objective: This study aimed to assess the hydrodynamic function of the newly China made Jiuling bileaflet heart valve prostheses. Methods: Jiuling valve has been tested in a pulsatile flow simulator in the aortic position. The testing condition was set at the pulsatile frequency of 55, 75, and 100 beats/min and a constant cardiac output of 4 L/min. The mean pressure differences, leakage volumes and closing volumes across each valve, and the effective orifice areas have been analyzed. Results: The Jiuling heart valves prosthesis had the lowest pressure difference at any given tissue annulus diameter. The mean pressure difference of all the Jiuling valves decreases with increasing the tissue annulus diameter or pulse frequency, and is in the range of below 10 mmHg. CarboMedics (CarboMedics, Inc., Austin, Tex.) is similar to that of the 21 mm and 23 mm Jiuling heart valves at the given pulse rate under 4 L/min. C L tilting disc valve (Lanzhou CarboMedics Inc., China) is similar to that of the 23 mm Jiuling heart valve. The effective orifice area of the Jiuling mechanical valve increases with increasing the tissue annulus diameter or pulse frequency. CarboMedics and C L tilting disc valve is similar to that of the 21 mm and 23 mm Jiuling heart valves. The Closing volume of the Jiuling prosthetic cardiac valves increases with increasing the tissue annulus diameter, but decreases with increasing the pulse frequency, and is in the range of below 11% (normalized by stroke volume). CarboMedics is below 16.5% of stroke volume; and C L tilting disc valve is below 7.5%. The leakage volume of the Jiuling prosthetic cardiac valves increases with increasing the tissue annulus diameter, but decreases with increasing the pulse frequency, and are in the range of below 28.9% (normalized by stroke volume). The leakage volume of CarboMedics is higher than that of the same sized 25 mm Jiuling valve, and the C L tilting disc valve is also higher than that of Jiuling valve at 4 L/min. Conclusion: For a given tissue diameter, the Jiuling valve showed significantly better hemodynamic characteristics. The material used in the construction is biocompatible, durable, and thromboresistant. 4Construction of recombinant plasmid and prokaryotic expression in E. Coli and biological activity analysis of human placenta arresten gene显示文摘BACKGROUND: The proliferation and metastasis of cancers depend on angiogenesis. This property provides the feasibility for the treatment of cancer by inhibition of angiogenesis, and many angiogenic inhibitors have been demonstrated to effectively inhibit angiogenesis and consequently the growth of solid cancer. As for the newly identified angiogenesis inhibitor, arresten, some studies have found its high activity on restrainting tumor vessel. This study was to assess the anti-angiogenic activity of arresten. METHODS: The arresten gene was obtained from a healthy puerpera’s placenta tissue by the reverse transcriptase-polymerase chain reaction (RT-PCR) method, and molecular cloning to prokaryotic expression plasmid pBV220 by recombination strategy. The prokaryotic expression plasmid pBV220/arr was identified by restriction enzyme digestion and sequenced. The pBV220/arr was transformed into E. coli JM109, DH5α, BL21 and BL21 (DE3) by the CaCl2 transformation method. The arresten expression level was detected by SDS-PAGE. The expressed product was purlfled, re-naturalized and detected for its biological activity of inhibiting the angiogenesis of chorioallantoic membrane (CAM). RESULTS: The arresten gene was cloned and pBV220/arr was constructed. The arresten expression level of protein was highly increased after pBV220/arr was transformed into E. coli BL21 (DE3). SDS-PAGE showed that the expressed arresten proteins were mainly inclusion bodies and had a molecular weight of 26 kDa. The expressed arresten protein showed evident biological activities. 6Protein transduction domain of membrane penetrating peptide can efficiently deliver DNA and protein into mouse liver for gene therapy显示文摘BACKGROUND: The development of a harmless and effi- cient nonviral gene delivery system that can facilitate the penetration of nucleic acids through the plasma membrane is a key to successful gene therapy. The aim of this study was to test a nonviral gene transferring vector’s function of delivering DNA into liver cells to provide an important clue for gene transfer in liver gene therapy. METHODS: The complex of DNA and DNA delivering protein was injected into mice through their tail veins. Then the mice were killed and their liver tissue was sec- tioned. The gene transferring results were detected using a confocal laser scanning microscope. RESULTS: Fluorescence analysis indicated that both DNA- membrane penetrating peptide (MPP) complex and DNA- hepatocyte specific receptor binding domain ( HSRBD) - MPP complex could go into liver cells. The fluorescence value of liver cells in the DNA-HSRBD-MPP group was higher than that in the DNA-MPP group. CONCLUSIONS; MPP can successfully deliver DNA and protein into cells, and MPP with a HSRBD can specifically deliver DNA into liver cells. 7吡格列酮对体外培养大鼠破骨细胞样细胞分化及活性的影响显示文摘目的观察吡格列酮对大鼠破骨细胞样细胞(OLC)分化及活性的影响,探讨PPARγ2活化与破骨细胞之间的关系。方法用NF—KB受体活化因子配体(RANKL)及巨噬细胞集落刺激因子(M-CSF)协同诱导大鼠骨髓单个核细胞(BMC)向OLC分化,同时加入1、5、10μmol/L盐酸吡格列酮干预。应用RT—PCR分析OLC分化过程中PPARγ2及NF-κB受体活化因子(RANK)mRNA的表达,结合细胞染色及抗酒石酸酸性磷酸酶(TRAP)活性观察吡格列酮对OLC分化的影响,通过骨吸收陷窝分析及整合素B3(CD61)平均荧光强度的检测比较吡格列酮对OLC活性的影响。结果(1)对OLC分化的影响:培养7d时10μmol/L吡格列酮组与其它组相比OLC数量及TRAP活性明显减少(P〈0.01或P〈0.05),表明吡格列酮部分抑制OLC分化且此作用与剂量相关;RT—PCR结果显示在诱导分化早期,不同浓度吡格列酮呈剂量依赖性上调PPARγ2的表达水平,但随OLC的成熟其表达渐减弱,而RANK表达在5及10μmol/L吡格列酮干预组培养的各时间点均明显下调,与对照及1μmol/L干预组相比有显著差异(P〈0.05或P〈0.01);(2)对OLC活性的影响:骨吸收陷窝数量及吸收面积在吡格列酮5、10μmol/L组明显减少,与另两组相比差异显著(P〈0.05或P〈0.01),培养早期这两组细胞CD61平均荧光强度均明显下调(P〈0.05或P〈0.01)。结论吡格列酮激活PPARγ2转录活性可部分抑制大鼠骨髓OLC的分化及活性。朱亦堃 乔振华 山西医科大学第二医院内分泌科,太原030001周永安 朱镭 郗光霞 史书红 赵宝珍 郭志新 李兴 刘素筠 2008中华内分泌代谢杂志2008,24,4:3
8Adenoma of the posterior urethra:131 case report显示文摘A case-report on adenoma of the posterior urethra. Methods: In 131 cases of adenoma of the posterior urethra, aged 17- 79 (mean: 36.4) years, a detailed medical history was taken and urinalysis, urethroscopy, and prostatic specific antigen (PSA) immunohistochemical staining were performed. They were then treated with transurethral resection (TUR) or transurethral electric coagulation (TUEC). Results: Hemospermia occurred in 51% of the cases, hematuria in 38 %, bood overflow from the urethral orifice in 6 %, and dysuria in 5 %. The position of the tumor was at or around the verurumontanum. The appearance of the rumor was similar to those of a papilla, a villus, a dactyl or polyp, or simply an engorgement. The tumor contained glandular alveoli and adeno-epithelial cells. PSA immunohistochemistry was positive in the cytoplasm and nucleus of the adeno-epithelial cell. One hundred and tweenty-nine cases were cured after TUR or TUEC, while 2 patients recurred and were operated again. Conclusion: Adenoma of the posterior urethra is a common cause of hemospermia and hematuria in young men. Urethroscopic examination and biopsy are the principal diagnostic measures. 9STUDY ON THE PROPERTIES OF DIFFERENT ACTIVATED CARBON FIBERS AND THEIR ADSORPTION CHARACTERISTICS FOR FORMALDEHYDE显示文摘Porous structure and surface chemistry of activated carbon fibers obtained by different precursors and activation methods were investigated. Adsorption isotherms were characterized by nitrogen adsorption at 77K over a relative pressure range from 10-6 to 1. The regularization method according to Density Functional Theory (DFT) was employed to calculate the pore size distribution in the samples. Their specific surface areas were calculated by BET method, micro pore volume and micro porous specific surface area calculated by t-plot method and MPD by Horvath-Kawazoe equation. Micropore volume of rayon-based ACF was higher than that of other samples. The static and dynamic adsorption capacity for formaldehyde on different ACFs was determined. The results show that steam activated Rayon-based ACFs had higher adsorption capacity than that of steam and KOH activated PAN-ACFs. Breakthrough curves illustrated that Rayon-ACFs had longer breakthrough time, thus they possessed higher adsorption capacity for formaldehyde than that of PAN-A CFs. The entire sample had small adsorption capacity and short breakthrough time for water. Rayon-ACFs had excellent adsorption selectivity for formaldehyde than PAN-ACFs. And the samples with high surface areas had relatively high adsorption capacity for formaldehyde. Elemental content of different ACFs were performed. Rayon-based ACFs contained mare oxygen than PAN-ACFs, which may be attributed to their excellent adsorption capacity for formaldehyde.H.Q. Rong, Z.Y. Ryu and J.T. 10CFD Model of Dense Gas-solid Systems in Jetting Fluidized Beds显示文摘A CFD code has been developed based on the conservation principles describing gas and solid flow in fluidized beds. This code is employed to simulate not only the spatiotemporal gas and solid phase velocities and voidage profiles in a two dimensional bed but also fluid dynamics in the jet region. The computational results show that gas flow direction is upward in the entire bed accompanied with random local circulations, whilst solid flow direction is upward at the center and downward near the wall. The radical reason of strong back mixing of solid particles and good transfer behavior between two phases is that the jet entrains solid particles. Numerical calculation indicates that gas velocity, solid velocity and pressure profile have a significant change when the voidage is 0 8. The simulated time averaged voidage profiles agree with the experimental results and simulated data reported by Gidaspow and Ettehadieh(1983). 14STUDY OF MICROSTRUCTURE ON PAN-ACFs PREPARED BY DIFFERENT ACTIVATION METHODS显示文摘Polyacrylonitrile (PAN) -based activated carbon fibers (ACFs) are amorphous graphitic carbon, consisting of sp2 hexagonal carbon layers with different sizes of pores ranging from micropores to macropores. The microstructure, including graphite-like micro-crystal, surface chemistry, pore size distributions and so on, of PAN activated carbon fibers prepared by different activation methods were investigated by element analysis (EA), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and infrared spectra. The XRD patterns show that PAN-ACFs activated by different methods are of amorphous diffraction characteristic but composed of very small graphitic crystallites. The surface functional groups were analyzed by XPS and FT-JR spectra. The samples activated by air possess relatively abundant oxygen functional groups, and the samples activated by potassium hydroxide possess less oxygen functional groups. 16Karyotypic analysis of intersexuality in Chinese from Taiyuan显示文摘Aim: To analyze the kayrotypic patterns of 33 cases of intersexuality in Chinese from Talyuan, China in order to further clarify its mechanism of development and the interrelationship between karyotype and phenotypic sex. Methods: High-resolution GTG-banding chromosome technique was used to analyze the karyotype patterns. Results: In these patients, 57.58% were male pseudohermaphrodites (46,XY), 18.18%, female pseudohermaphrodites (46, XX), 12.12%, true hemaphrodites, and 12.12%, other karyotypes. Although testes can be seen in 88.8% of karyotypes with Y chromosome, 73.68% of the patients were of female social sex. In 42.82% of patients the social sex is in conformity with their karyotypes. There were 2 cases of male pseudohermaphrodites, where the sex chromosome was normal, but abnormalities were found in chromosomes 9, 13, or 14. 17Propane Aromatization over Mo/HZSM-5 Catalysts显示文摘Impregnation, mechanical mixing and hydrothermal treatment methods were used to introducemolybdenum species into the HZSM-5 zeolite. The structure and surface acidity of the catalysts werestudied by means of XRD, FT-IR, NH3-TPD, TPR and XPS. The effects of Mo content and reaction timeon stream on the aromatization of propane were investigated. It was found that the performance of theMo/HZSM-5 catalyst prepared by the hydrothermal treatment method was much better than that of theother two catalysts. For example, under the reaction conditions of 823 K and 600 h-1, propane conversionand aromatics selectivity over the catalyst prepared by hydrothermal pretreatment could reach 89.17%and 78.56%, respectively. XRD and XPS results showed that the Mo species in the catalysts preparedby hydrothermal treatment were highly dispersed on the surface of the HZSM-5, and larger amounts ofthem could penetrate into the HZSM-5 channel, as compared with the other two kinds of catalysts. Thesefactors may be responsible for their high activities for propane aromatization.
