1A study of recombinant protective H.Pylori antigens显示文摘AIM: To construct a recombinant vector which can express Mr 26 000 outer membrane protein (OMP) from Helicobacter pylori (Hp), and to obtain the vaccine protecting against Hp infection and a diagnostic reagent kit quickly detecting Hp infection.METHODS: The gene encoding the structural Mr 26000 outermembrane protein of Hp was amplified from Hpchromosomal DNA by PCR, and inserted in the prokaryoticexpression vector pET32a ( + ), which was transformed intothe Topl0 E. coli strain. Recombinant vector was selected,identified and transformed into BL-21(DE3) E. coli strain.The recombinant fusion proteins were expressed. Theantigenicity of recombinant protein was studied by ELISA orimmunoblotting and immunized Balb/c mice.RESULTS: The gene of Mr 26 000 OMP was amplified to be594 base pairs, 1.1% of the cloned genes was mutated and1.51% of amino acid residues was changed, but there washomogeneity between them. The recombinant fusion proteinencoded objective polypeptides of 198 amino acid residues,corresponding to calculated molecular masses of Mr 26000.The level of soluble expression products was about 38.96 %of the total cell protein. After purification by Ni-NTA agaroseresin columniation, the purity of objective protein becameabout 90 %. The EESA results showed that recombinantfusion protein could be recognized by patient serum infectedwith Hp and rabbit serum immunized with the recombinantprotein. 2Functional changes of dendritic cells derived from allogeneic partial liver graft undergoing acute rejection in rats显示文摘AIM: To investigate functional change of dendritic cells (DCs)derived from allogeneic partial liver graft undergoing acuterejection in rats.METHODS: Allogeneic (SD rat to LEW rat) whole and 50 %partial liver transplantation were performed. DCs from livergrafts 0 hr and 4 days after transplantation were isolated andpropagated in the presence of GM-CSFin vitro. Morphologicalcharacteristics of DCs propagated for 4 days and 10 dayswere observed by electron rmicroscopy. Phenotypical featuresof DCs propagated for 10 days were analyzed by flowcytometry. Expression of IL-12 protein and IL-12 receptormRNA in DCs propagated for 10 days was also measured byWestern blotting and semiquantitative RT-PCR, respectively.Histological grading of rejection were determined.RESULTS: Allogeneic whole liver grafts showed no featuresof rejection at day 4 after transplantation. In contrast,allogeneic partial liver grafts demonstrated moderate tosevere rejection at day 4 after transplantation. DCs derivedfrom allogeneic partial liver graft 4 days after transplantationexhibited typical morphological characteristics of DC after 4days' culture in the presence of GM-CSF. DCs from allogeneicwhole liver graft 0 hr and 4 days after transplantation didnot exhibit typical morphological characteristics of DC untilafter 10 days' culture in the presence of GM-CSF. 3Impact of simultaneous splenectomy and orthotopic liver transplantation in patients with end-stage liver diseases and splenic hyperfunction显示文摘BACKGROUND:Whether splenectomy can be performed simultaneously during liver transplantation in patients with end-stage liver diseases complicated by hypersplenism remains controversial. This study aimed to compare the impact of simultaneous splenectomy on high-and low-risk liver transplant patients with end-stage liver diseases and severe hypersplenism. METHODS:Forty-two patients with end-stage liver diseases complicated by severe hypersplenism who had undergone orthotopic liver transplantation were enrolled in this study. Splenectomy was performed in 19 of the patients. The 42 patients were grouped according to the risk of liver diseases and operations they received. Patients were considered to be at high-risk if they had at least one of the following conditions: preoperative prothrombin time >5 seconds, portal vein thrombosis, and severe perisplenitis. High-risk patients who had undergone splenectomy were classified into group A, whereas high-risk patients who had not undergone splenectomy were classified into group B. Low-risk patients who had undergone splenectomy were classified into group C, and low-risk patients who had spleen preservation were classified into group D. Operative time, intraoperative blood loss, postoperative bleeding, pulmonary infection, perioperative mortality, and postoperative platelet recovery were analyzed. RESULTS: Operative time and intraoperative blood loss were greater in group A than in groups B-D (P<0.01), but there was no significant difference between groups C and D (P>0.05). In group A, 3 patients had postoperative bleeding, 5 had pulmonary infection, and 2 had perioperative mortality,which was higher than any other group, but postoperative bleeding, pulmonary infection, and perioperative mortality were similar to those in groups C and D. In patients undergoing simultaneous splenectomy, platelet counts recovered within 6 months after surgery. Thrombocytopenia was sustained in 3 of the 23 patients who did not undergo simultaneous splenectomy. 4Construction of human eukaryotic expression plasmid vascular endothelial growth factor 165 and its expression in transfected vascular smooth muscles显示文摘BACKGROUND: The highly specific vascular endothelialgrowth factor (VEGF) induces the growth of vascular en-dothelial cell. This study was to construct the eukaryoticexpression plasmid of vascular endothelial growth factorl65(VEGF165) and observe its expression in vascular smoothmuscles (VSMCs).METHODS: The primers were designed and synthesizedaccording to the gene sequences of human VEGF165. TheVEGF165 gene was obtained from umbilic artery tissue bythe method of RT-PCR, then it was cloned to eukaryoticexpression plasmid pBudCE4.1 by recombination strategy.The eukaryotic expression plasmid named pBudCE4.1/VEGF165 was identified by restriction enzyme digestion,and was sequenced. The pBudCE4.1/VEGF165 was trans-fected into VSMCs by using lipofection. The VEGF165 ex-pression of mRNA and protein was detected by RT-PCRand Western blot respectively.RESULTS: VEGF165 was shown about 576bp by RT-PCR.Sequencing revealed the amplified VEGF165 gene was iden-tical with that in the GeneBank. Restrictive enzyme (HindBam HI) digestion analysis showed that recombinantexpression plasmid pBudCE4. l/tVEGF165 had been con-structed successfully. The expression of VEGF165 at mRNAand protein levels in the transformed VSMCs had beendemonstrated by RT-PCR and Western blot.CONCLUSIONS: The recombinant eukaryotic expressionplasmid pBudCE4.1/VEGF165 has been successfully con-structed and expressed in transformed VSMCs. 5Plasma hemoglobin concentration was related to estimated glomerular filtration rate in elderly patients with ischemic cardiomyopathy显示文摘Objectives To study the relationship between plasma hemoglobin concentration and estimated glomerular filtration rate (eGFR) in elderly patients with ischemic cardiomyopathy (ICM).Methods Clinical data of patients with coronary heart disease who were discharged from The First Affiliated Hospital,Chongqing Medical University between 2005 and 2007 were analyzed retrospectively. 6Construction of human VEGF165 gene eukaryotic expression plasmid and its effect on proliferation of vascular endothelial cells显示文摘After organ transplantation, rapid repair of injured vascular endothelial cell (VEC) is a key to prevent graft chronic dysfunction besides control of immunological rejection. Many studies have confirmed that vascular endothelial growth factor 165 (VEGF165) could accelerate the repair of VEC injury, decrease thrombosis and thrombotic occlusion, and inhibit hyperplasia of the intima. This study was designed to construct eukaryotic expression plasmid pBudCE4.1/VEGF165, and observe its effect on the prolife ration of VEC. METHODS:The VEGF165 gene cloned from human heart tissue by RT-PCR was cloned into eukaryotic expression plasmid pBudCE4.1. The recombinant expression plasmid pBudCE4.1/VEGF165 was identified by restriction enzyme (Hind III and BamH I) digestion analysis, and was sequenced. The pBudCE4.1/VEGF165 was introduced into VEC through lipofection transfection. The VEGF165 mRNA expression by Northern blot and VEGF165 protein expression was detected by immunocytochemical staining. The effect of expression protein on VEC proliferation was detected by flow cytometry. RESULTS:The RT-PCR product of the VEGF165 gene was about 576bp. Sequencing analysis revealed that the sequence of the amplified VEGF165 gene was identical with that in GenBank. Restrictive enzyme digestion analysis showed that recombinant expression plasmid pBudCE4.1/ tVEGF165 had been constructed successfully. The expression of VEGF165 at mRNA and protein levels in the transformed VSMCs had been demonstrated by Northern blot and immunocytochemical staining respectively. The expressed product of VEGF165 could notably accelerate the proliferation of VECs. CONCLUSIONS:pBudCE4.1/VEGF165 is successfully cons- tructed and is expressed in VECs. Expressed VEGF165 can accelerate the VEC proliferation. Through integrated simulation in computer, design efficiency and quality of oil tank is greatly improved. Adopting integrated CAD/CAE/CAM software to carry out integrated research to equ ipment and products, we will be able to take overall analysis in aspects of 3-D solid modeling, pre-assembly and strength, etc., to realize non-paper designi ng and parallel designing. 9Friction and Wear Behaviors of Nano-Silicates in Water显示文摘Nano-metric magnesium silicate and zinc silicate with particle size of about 50-70nm were prepared in water by the method of chemical deposition. The antiwear and friction reducing abilities of the nano-silicates,as well as their composites with oleic acid tri-ethanolamine(OATEA) ,were evaluated on a four-ball friction tester. The topographies and tribochemical features of the worn surfaces were analyzed by scanning electron microscope(SEM) and X-ray photoelectron spectroscope(XPS) . Results show that nano-silicates alone provide poor antiwear and friction reducing abilities in water,but exhibits excellent synergism with OATEA in reducing friction and wear. The synergism in reducing friction and wear between nanosilicates and OATEA does exist almost regardless of particle sizes and species,and may be attributed,on one hand,to the formation of an adsorption film of OATEA,and,on the other hand,to the formation of tribochemical species of silicon dioxide and iron oxides on the friction surfaces. 10Design and Research of the EQ6105DTAA Diesel Engine显示文摘The EQ6105DTAA diesel engine which first pattern en gi ne is EQD6105T is developed through the original EQ6102 diesel engine and other advanced engine structures. This paper analyses performance parameters, general layout and parts design process of the diesel engine. The development cycle is s horten by CAD/CAE/CAM technology. Through experiment, the general performance of the engine is in keeping ahead in our country. With boosting mid-cooling technology and related designing correction in EQ6105 DTAA diesel engine, it had obtained better motivity and economy. The full load s teady smog emission and smog emission during simulated free accelerating are all meeting with GB14761.6-93, GB3847-1999 limit requirement. The prototype had p assed reliability test and has reliable parts. It performance indexes are in the leading position in same diesel engine in China. The 13 working conditions gas pollute and particle discharging in this pro totype can meet the limit requirement of GB17691-2001, phase I. 11Analysis of generation of cumulative second harmonics of Lamb modes in a layered planar structure显示文摘A technique for analyzing the nonlinear generation of the cumulative second har-monics of generalized Lamb modes in a layered planar structure is developed. A theoretical model for nonlinear generalized Lamb mode propagation in a layered planar structure has been established, based on a partial plane wave approach. The nonlinearity is treated as a second-order perturbation of the linear elastic response. This model reveals some interesting features of the physics of the cumulative second harmonic generation. Although Lamb mode propagation is dispersive in a layered structure, the results of this analysis show that the amplitudes of the second harmonics do accumulate with propagation distance under certain special conditions. On the basis of the boundary and initial conditions of excitation, the formal solution of the cumulative second harmonic has been derived. 13Protective effect of glucocorticoid-free immunosuppressive regimen in allogenic islet transplantation显示文摘BACKGROUND: The most common complication after allogenic islet transplantation is rejection. This study was to evaluate the effect of anti-rejection of glucocorticoid-free immunosuppressive regimen on allogenic islet transplantation. METHODS: Tacrolimus(FK506)+mycophenolate mofetil (MMF) and FK506+MMF+prednisone (Pred) were administered respectively for 2 weeks to inhibit rejection after allogenic islet transplantation in rats, which were compared with the control group. The concentrations of blood glucose, insulin and C-peptide were determined dynamically in recipients and the sites of transplantation were observed morphologically. RESULTS: As compared with the control group without immunosuppressive agents, FK506+MMF and FK506+MMF+Pred could prolong the survival time of grafts significantly. There were many morphologically intact islets in the liver of recipients 2 months after transplantation. Group FK506+MMF kept normal levels of blood glucose, insulin and C-peptide beyond 60 days after transplantation. In contrast, group FK506+MMF+Pred secreted less C-peptide(P<0.05) and maintained a higher level of blood glucose concentration (P<0.01) after the operation. There was no significant difference in insulin concentrations between the two groups. The level of blood glucose beyond the first 2 weeks after drug withdrawal in group FK506+MMF+Pred decreased obviously (P<0.05), and the secretion of insulin and C-peptide increased. These results were compared with those the first 2 weeks after transplantation and the first 2 weeks after drug withdrawal. CONCLUSIONS: Both regimens of FK506+MMF and FK506+MMF+Pred could provide effective immunosup-pression. 14Analyzing ECS for Perinatal Pregnant and Fetus显示文摘Objective:Analyzing digital characteristics (chaotic and spectral features) of fetal and pregnant womans Heart Period Signal (HPS) can assess the autonomic function. Methods:Extracting and analyzing HPS of fetal and pregnant woman in perinatal period were realized by the method called visual programming. The hardware consists of electrode, amplifier, analog to digital converter (ADC) with 12bit, printer and microcomputer. Low level I/O operation for the ADC was finished by means of assembler in-line in Delphi (language) under Windows operating system. The software package was designed by Object Oriented Programming technique. So, all the system is a multi-media one and established under Windows. The subjects are in supine,electrocardiosignal (ECS) from the leads placed at symphysis pubica to inferior border of abdominal wall was acquired. ECS was preprocessed by wavelet filter. The sophisticated technique developed by our laboratory was used to accomplish analyzing digital characteristic of HPS. Results: The system could be used to assess fetal and pregnant womans autonomic function, furthermore, sympathetic and parasympathetic nervous function can be evaluated respectively and quantificationally.Discussions:(1) The system could be used to prognosis fetal distress. (2) Digital characteristic of fetal and other age segments HPS that vary with age could approve the viewpoint that there is a characteristic curve to suggest the physiological process of development, mature and senility, based on it, we could find the way of anti-senility. 16Labeling polypeptide with ^(99m)Tc and bioactivity get back显示文摘A method for labeling polypeptide (insulin) with technetium- 99(99mTc) was established without marked loss of biological activity. Following reduction of intrinsic disulfide bonds by mercaptoethanol and purification on a Sephadex G50 column, the polypeptide was labeled with 99mTc by transchelation from methylene diphosphonate (MDP). 99mTc labeled insulin was identified by thin layer chromatograph (TLC) and the change of blood sugar of mice injected, their hypoglycemic shock symptom was also observed. Six hours after labeling, the dissociation of labeled insulin was only 3%, Mom then on to 24 h, there was no more dissociation. The blood sugar concentration of mice injected with the mercaptoethanol-reduced insulin was (5.0±3.2)μmol.L-1, while those injected with the original insulin was (1.4±1.2)μmol.L-1, the difference was significant (Q test, p <0.01). Blood sugar concentration of the mice was 0.3±0.2μmol.L-1 for the labeled insulin, and was about the same with that for the original insulin. The labeling efficiency was 74.31% for the labeled insulin, whereas the original insulin cannot be labeled with 99mTc. The result suggests that while disulfide bonds of polypeptide were reduced by mercaptoethanol, it became free sulfhydryl group, and its bioactivity descended. Then free sulfhydryl group was chelated with 99mTc under mild condition, restablishing the disulfide bond, therefore, the bioactivity came back. 17Tribological performances of 3-(N, N- dialkyl dithiocarbamate-y1) propionic acids显示文摘Three kind of water-soluble additives, 3-(N, N- dialkyl dithiocarbamate-y1) propionicacid, were synthesized. Their tribological performances when added to water were evaluated on afour-ball tester and a ring-on-block machine. Results indicate that the hydrocarbon chain length ofthese compounds has significant influence on their tribological properties. When the number ofcarbon atom in alkyl group is equal to (or more than) 4, the compounds exhibit excellentload-carrying capacity, antiwear property and weld load. The antiwear action mechanisms of thepropionic acid compounds can be ascribed to the formation of adsorption film and chemical reac-tion film. It is in many ways similar to chemical heattreatment. A new concept, technology of on-line strengthening, was logically put forward. Based oncurrent gear surface treatment technology, the on-line strengthen of gear surface is proposed. Itsdesign method is established. 19Low-intensity ultrasound for regeneration of injured peripheral nerve显示文摘BACKGROUND: Ultrasound is a kind of mechanical wave and characterized by mechanical effect,heat affect and physical and chemical effect.Ultrasound can promote regeneration of peripheral nerves after a slight injury based on its mechanical effect.However,whether it can promote regeneration of peripheral nerves after a severe injury or not is still unclear.OBJECTIVE: To study the effect of low-intensity ultrasound(LIU)on regeneration of injured peripheral nerve,throgh examining sciatic nerve function index,the sensory nerve conduction velocity and the thickness of myelin sheath.DESIGN: Single factor design of contrast observation.SETTING: Institute of Ultrasound Engineering,Chongqing Medical University.MATERIALS:A total of 64 female Wistar rats,of clean grade,age 3 moths,weighing 200-250g ,were provided by Experimental Animal Center of Chongqing Medical University. All rats were randomly divided into treatment group and control group with 32 in each group. In addition rats were observed at 4 time points, including 2,4,6 and 8 weeks,with 8 at each time point.The main equipments were detailed as follows:forceps (Medical Treatment Apparatus Company,Chongqing),low-intensity ultrasound treatment instrument(Institute of Ulrasound Engineering in Medicine),the analysis instrument of diagram resembles and arithmetic figure(the United States Bio-RAD Company),ultrasound coupling agent(Xunde Image material factory,Hangzhou),Osmium Tetraoxide(Next Chimicam,South Africa).METHODS:The experiment was carried out in Institute of Ultrasound Engineering of Chongqing Medical University from December 2003 to May 2004.The right sciatic nerves of 64 rats were crushed with forceps for 30 s to form the experimental animal models.Then they were treated at 3 days after operation.Rats in the treatment group received the LIU exposure.LIU was applied every other day to the crush site of rats,which had a spatial peak,time-averaged intensity of 0.25 W/cm2 operated at 1 MHz for 1 minute per application for total 8 weeks.At various stages after operation,the sciatic nerve function index(SFI).the sensory nerve conduction velocity and its histology were detected.Rats in the control received a sham exposure.MAIN OUTCOME MEASURES:SFI;sensory nerve conduction velocity;density of myelinated nerve fiber;velocity of nerve regeneration;histological examination.RESULTS: Among 64 Wistar rats, 2 were lost during the experiment and another 2 were supplied. ①Histological examination:Two weeks after treatment,degeneration of axis-cylinder and myelin sheath was obvious in treatment group as compared with that in control group. Within 4-8 weeks after treatment.regeneration of axis-cylinder and myelin sheath of nerve fiber was superior in treatment group to that in control group.At 8 weeks after treatment,axis-cylinder and myelin sheath in treatment group were closed to normal value.Quantity of nerve fiber was less in control group than that in treatment group and the arrangement was disorder.At 2,4 and 6 weeks after treatment,proliferation of Schwann cells was superior in treatment group to that in control group.At 6 and 8 weeks after treatment,proliferation of fiber tissue in nerve was severer in control group than that in treatment group.②SFI:At 4.6 and 8 weeks after treatment.SFI was higher in treatment group than that in control group(t=8.00,12.41m15.13,P<0.01).③Sensory nerve conduction velocity:At 2,4,6 and 8 weeks after treatment,sensory nerve conduction velocity was faster in treatment group than that in control group(t=11.74,10.81,3.51,P<0.01).④Density of myelinated nerve fiber:At 2,4,6 and 8 weeks after treatment,density of myelinated nerve fiber was higher in treatment group than that in control group(t=2.16,P<0.05;t=3.29,3.52,3.23,P<0.01).⑤Velocity of nerve regeneration:Velocity of nerve regeneration was(1.50+0.08)mm/d and(1.22±0.10)mm/d of treatment group and control group.respectively. 20Serum macrophage inflammatory protein-lαand high-sensitivity C-reactive protein levels in the paitents with obstructive sleep apnea-hypopnea syndrome and hypertension显示文摘Objectives To identify the effects of obstructive sleep apnea-hypopnea syndromemacrophage inflammatory protein -1α(MIP-1α) and high-sensitivity c-reactive protein(hs-CRP) levels,and its impact on cardiac structure and function in patients with hypertension.(OSAHS) on serum. Methods We studied 86 middle-aged subjects classified into four groups according to the absence or presence of OSAHS with and without hypertension.(1)OSAHS patients without hypertension(OSAHS group,n=29);(2)OSAHS patients with hypertension(OSAHS +HT group,n=27);(3) non-OSAHS patients with hypertension(HT group,n =27);(4)volunteers without OSAHS and hypertension(Control subjects, n=27).OSAHS patients were divided into mild,moderate and severe degree based on apnea hypopnea index(AHI).All participants underwent polysomnography and echocardiography. Serum MIP-1αand hs-CRP levels were tested by enzyme linked immunosorbent assay(ELISA).Results Body mass index(BMI),neck collar(NC),waist-to-hip ratio(WHR) in OSAHS group and OSAHS +HT group were significantly higher than those in Control group(PP<0.05).Serum MIP- 1αlevels in OSAHS+HT group was significant higher than HT groups(P<0.05).Serum MlP-1αlevels in those three groups were negative correlationwith AV(r=-0.238,P=0.08) and positively correlated with E/A ratio(r=0.307,P=0.02).
