| 1 | Modern stalagmite oxygen isotopic composition and its implications of climatic change from a high-elevation cave in the eastern Qinghai-Tibet Plateau over the past 50 years显示文摘An oxygen isotope record of a stalagmite from Huanglong Cave in the eastern Qinghai-Tibet Plateau dated with 230Th and 210Pb methods provides variations of the Asian monsoon with an average resolu-tion of 1 year over the past 50 years. This study shows that the δ18O of dripwater in the cave represents the annual mean δ18O of local meteoric precipitation and the stalagmites were deposited in isotopic equilibrium. A comparison of the stalagmite δ18O record with instrumentally meteorological data indi-cates that shifts of the δ18O are largely controlled by the amount effect of meteoric precipitation con-veyed through the southwest monsoon(the Indian monsoon) and less affected by temperature. Therefore,the variations of δ18O record reflect the changes in monsoon precipitation on inter-annual time scales under the influence of the southwest monsoon. Like many other stalagmite δ18O records in the Asian monsoon regions,the δ18O record of the stalagmite from Huanglong Cave also reveals a gradually enriched trend during the past 50 years,i.e. relatively enriched in 18O. This trend may indicate the decline of the Asian monsoon intensity which is consistent with the decrease of monsoon indices. | 2 | Asian summer monsoon precipitation recorded by stalagmite oxygen isotopic composition in the western Loess Plateau during AD1875―2003 and its linkage with ocean-atmosphere system显示文摘Based on 5 high-precision 230Th dates and 103 stable oxygen isotope ratios (δ18O) obtained from the top 16 mm of a stalagmite collected from Wanxiang Cave,Wudu,Gansu,variation of monsoonal precipita-tion in the modern Asian Monsoon (AM) marginal zone over the past 100 years was reconstructed. Comparison of the speleothem δ18O record with instrumental precipitation data at Wudu in the past 50 years indicates a high parallelism between the two curves,suggesting that the speleothem δ18O is a good proxy for the AM strength and associated precipitation,controlled by 'amount effect' of the pre-cipitation. Variation of the monsoonal precipitation during the past 100 years can be divided into three stages,increasing from AD 1875 to 1900,then decreasing from AD 1901 to 1946,and increasing again thereafter. This variation is quite similar to that of the Drought/Flooding index archived from Chinese historical documents. This speleothem-derived AM record shows a close association with the Pacific Decadal Oscillation (PDO) between AD 1875 and 1977,with higher monsoonal precipitation corre-sponding to cold PDO phase and vice versa at decadal timescale. The monsoonal precipitation varia-tion is out of phase with the PDO after AD 1977,probably resulting from the decadal climate jump in the north Pacific occurring at around AD 1976/77. These results demonstrate a strong linkage between the AM and associated precipitation and the Pacific Ocean via ocean/atmosphere interaction. | 3 | Establishing a human pancreatic stem cell line and transplanting induced pancreatic islets to reverse experimental diabetes in rats显示文摘The major obstacle in using pancreatic islet transplantation to cure type I and some type II diabetes is the shortage of the donors. One of ways to overcome such obstacle is to isolate and clone pancreatic stem cells as 'seed cells' and induce their differentiation into functional islets as an abundant trans-plantation source. In this study, a monoclonal human pancreatic stem cell (mhPSC) line was obtained from abortive fetal pancreatic tissues. Pancreatic tissues were taken from abortive fetus by sterile procedures, and digested into single cells and cell clusters with 0.1% type IV collagenase. Cultured in modified glucose-low DMEM with 10% fetal bovine serum (FBS), these single cells and cell clusters adhered to culture dishes, and then primary epidermal-like pancreatic stem cells started to clone. After digesting with 0.25% trypsin and 0.04% EDTA, fibroblasts and other cells were gradually eliminated and epithelioid pancreatic stem cells were gradually purified during generations. Using clone-ring selection, the mhPSCs were obtained. After addition of 10 ng/mL epidermal growth factor (EGF) in cell culture medium, the mhPSCs quickly grew and formed a gravelstone-like monolayer. Continuously proliferated, a mhPSC line, which was derived from a male abortive fetus of 4 months old, has been passed through 50 generations. More than 1×109 mhPSCs were cryo-preserved in liquid nitrogen. Karyotype analysis showed that the chromosome set of the mhPSC line was normal diploid. Immunocytochemistry results demonstrated that the mhPSC line was positive for the pdx1, glucagon, nestin and CK19, and negative for the insulin, CD34, CD44 and CD45 protein expression. RT-PCR revealed further that the mhPSCs expressed transcription factors of the pdx1, glucagon, nestin and CK19. Also, in vitro induced with β-mercaptoethanol, the mhPSCs differentiated into nerve cells that expressed the NF protein. Induced with nicotinamide, the mhPSCs differentiated into functional islet-like clusters, as identified by di-thizone staining, which expressed the transcription factor of the insulin and secreted the insulin and C-peptide. | 9 | Reliability analysis of stochastic structural system considering static strength, stiffness and fatigue显示文摘Multi-failures are possible to appear in the process of using the structural system, such as dead load failure, fatigue failure and stiffness failure. The expression of residual resistance is given based on the impact of random crack propagation in- duced by the fatigue load on the critical limit stress and section modulus in this paper. The failure modes of every element of the structural system are analyzed under dead and fatigue loads, and the influence of the correlation of failure modes on reliability of the element is considered. Failure mechanism and the correlation of failure modes under dead and fatigue loads are discussed, and the method of reli- ability analysis considering static strength, fatigue and stiffness is given. A nu- merical example is analyzed, which indicates that the failure probability is different for different use life and the influence of dead and fatigue loads on reliability of the structural system is different as well. | 13 | Evaluation of Shale Reservoir Quality by Geophysical Logging for Shuijingtuo Formation of Lower Cambrian in Yichang Area, Central Yangtze显示文摘Taking the shale of Shuijingtuo Formation of Lower Cambrian in Yichang area as the research object,the shale reservoir characteristics are comprehensively evaluated and classified by fitting regression and formula calculation method in this study,using laboratory testing and geophysical logging data.The results show that the interpretation data of ECS(elemental capture spectroscopy)logging has a high correlation with the measured minerals data,which can be a good method to evaluate the minerals component of the shale.The calculated content of brittle minerals at the lower part of Shuijingtuo Formation is the relatively highest,generally more than 40%,which is the most favorable segment for fracturing.The correlation coefficient between the interpretation data of CMR(combinable nuclear magnetic resonance)logging and the result of laboratory porosity test is 0.97,which can effectively and accurately evaluate the reservoir porosity.The evaluation results show that the porosity of the lower member of Shuijingtuo Formation is generally greater than 3%,while that of the upper member is generally less than 3%.The lower segment is with the relative optimal physical conditions.There is a good correlation between the acoustic logging data and the gas bearing content testing results.A gas bearing content evaluation model is established.The results show that the gas bearing content of the lower 20 m shale is generally more than 2%,indicating that the lower part is a shale gas enrichment segment.Mechanical parameters such as Young modulus,Poisson ratio and brittleness index of shale reservoir are evaluated by using the logging data of P-wave time difference and S-wave time difference.The continuous 15 m shale at the lower part is with the relatively optimal low Poisson ratio,high Young modulus and high brittleness index,developing the optimum brittle condition.Based on the evaluation and classification of above parameters,the shale is divided into three types.The TypeⅠis the optimal,mainly located at the bottom.Its thickness is 8.5 m in total.The TypeⅡmainly develops at the middle part.The TypeⅢis the worst,mainly at the upper part. | Xiaohong Chen Lin Chen Shu Jiang An Liu Shengyuan Luo Hai Li Peijun Li Ping Chen | 2021 | Journal of Earth Science2021,32,4: | 3 |

| 15 | Hepatitis C virus may infect extrahepatic tissues in patients with hepatitis C显示文摘AIM To explore the status of extrahepatichepatitis C virus(HCV)infection and replicationin hepatitis C patients,and its potentialimplication in HCV infection and pathogenicity.METHODS By reverse-transcriptase poly-merase chain reaction(RT-PCR),in situhybridization(ISH)and immunohistochemistry,HCV RNA,HCV replicative intermediate(minus-strand of HCV RNA),and HCV antigens weredetected in 38 autopsy extrahepatic tissuespecimens(including 9 kidneys,9 hearts,9pancreas,5 intestines,2 adrenal glands,2spleens,1 lymph node,and 1 gallbladder)from 9hepatitis C patients,respectively;and thestatus of HCV replication in extrahepatic tissueswas studied.RESULTS By RT-PCR,all 9 patients werepositive for HCV RNA in kidney,heart,pancreas,and intestine,but only 6(66.7%)patients were positive for HCV replicativeintermediate.HCV RNA and HCV antigens weredetected in kidney,heart,pancreas,intestine,adrenal gland,lymph node,and gallbladder in 5(55.6%)and 6(66.7%)patients by ISH andimmunohistochemistry,respectively.HCV RNA and HCV antigens were not detected in theseextrahepatic organs in 3(33.3%)patients,although their livers were positive for HCV.HCVreplicative intermediate detected by RT-PCR wasconsistent with HCV RNA and HCV antigensdetected by ISH and immunohistochemistry(Kappa=0.42-0.75).HCV RNA and HCVantigens were detected in myocardial cells,epithelial cells of intestinal gladular,interstitialcells of kidney,epithelial cells of tubules andglomerulus,pancreas acinar cells and epithelialcells of pancreatic duct,epithelial cells ofmucous membrane sinus of gallbladder,cortexand medulla cells in adrenal gland,andmononuclear cells in lymph node.HCV RNA wasalso detected in bile duct epithelial cells,sinusoidal cells,and mononuclear cells in livertissues by ISH.CONCLUSION HCV can infect extrahepatictissues,and many various tissue cells maysupport HCV replication;extrahepatic HCVinfection and replication may be of'concomitantstate'in most of patients with hepatitis C.Theinfected extrahepatic tissues might act as areservoir for HCV,and play a role in both HCVpersistence and reactivation of infection.HCVas an etiologic agent replicating and expressingviral proteins in extrahepatic tissues itselfcontributes to extrahepatic syndromeassociated.HCV infection in a few patients withchronic HCV infection. | Fu Ming Yan An Shen Chen Fei Hao Xi Ping Zhao Chang Hai Gu Li Bin Zhao Dong Liang Yang Lian Jie Hao | 2000 | World Journal of Gastroenterology2000,6,6: | 2 |
