METHODS Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97. andbiological characteristics of the target clones selected by in vivo screening were studied.``RESULTS Two clones with high MHCC97-H and IowMHCC9--L1 metastatic potential were isolated from theparent cell line. Compared with MHCC97-L. MHCC97-H hadsmaller cell size average cell diameter 43 um vs 50 μmand faster in vitro and in vivo growth rate tumor celldoubling time was 34.2 h vs 60.0 h. | 4 | Source mechanism of strong aftershocks (M_s≥5.6) of the 2008/05/12 Wenchuan earthquake and the implication for seismotectonics显示文摘Dozens of >M5, hundreds of >M4, and much more >M3 aftershocks occurred after the 2008/05/12 Wenchuan earthquake, which were well recorded by permanent and portable seismic stations. After relocated with P arrival, the >M3 aftershocks show two trends of distribution, with most of the aftershocks located along the north-east strike consistent with Longmenshan fault system, yet there is a north-west trend around the epicenter. It seems that substantially more aftershocks occur in regions with crystalline bedrocks. Then we collected waveform data from National Digital Seismograph Network and regional seismograph network of China, and employed 'Cut and Paste' method to obtain focal mechanisms and depths of the big aftershocks (M≥5.6). While most of those aftershocks show thrust mechanism, there are some strike slip earthquakes in the northern-most end of the rupture. Focal mechanisms show that the events located on the southern part of central Beichuan-Yingxiu Fault (BY) are mainly thrust earthquakes, which is consistent with initial mechanism of the main shock rupture. In the north part the aftershocks along the BY are also dominated by thrust slip, which is quite different from the right slip rupture of the main shock. Around Qingchuan-Pingwu Fault, the focal mechanisms are dominated by right-slip rupture with large depths (~18 km). So we suspected that in the north part the main shock might rupture on two faults: Beichuan Fault and Qingchuan-Pingwu Fault. | 7 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 8 | Strongly peraluminous granites of Mesozoic in Eastern Nanling Range, southern China: Petrogenesis and implications for tectonics显示文摘The strongly peraluminous granites (SPGs) of Eastern Nanling Range (ENR) are a characteristic of all bearing highly aluminous minerals, such as muscovite±Al-rich biotite±tourmaline±garnet, and lack of cordierite. In respect of petrography, geochemistry, Nd isotope, and single grain zircon U-Pb dating, the representative granite bodies of them are studied. The research shows that these granites were emplaced in two stages, namely 228-225 Ma BP and J2-3 159-156 Ma BP, belonging to Indosinian and early Yanshanian periods, respectively, and they have low εNd(t) values (-10.6--11.1), high A/CNK, Rb/Sr ratios and tDM values (1887-1817 Ma), and REE's tetrad effect (TE1, 3=1.13-1.34). In comparison with related geology, petrology and chronology of granites in adjacent regions, it is suggested that Indosinian SPGs of ENR formed in the circumstance of post-collisional extension 20 Ma after the major collision of Indosinian Movement (258-243 Ma BP) in Indo-China Peninsula, and early Yanshanian SPGs formed in the background of back-arc extension setting controlled by paleo-Pacific tectonic domain, and J1, the interval of two stages, is the interim from Tethyan to Pacific tectonic domains in South China. | 10 | Genetic basis and breeding application of clonal diversity and dual reproduction modes in polyploid Carassius auratus gibelio显示文摘A unisexual species is generally associated with polyploidy, and reproduced by a unisexual reproduction mode, such as gyno- genesis, hybridogenesis or parthenogenesis. Compared with other unisexual and polyploid species, gibel carp (Carassius au- ratus gibelio) has a higher ploidy level of hexaploid. It has undergone several successive rounds of genome polyploidy, and experienced an additional, more recent genome duplication event. More significantly, the dual reproduction modes, including gynogenesis and sexual reproduction, have been demonstrated to coexist in the polyploid gibel carp. This article reviews the genetic basis concerning polyploidy origin, clonal diversity and dual reproduction modes, and outlines the progress in new va- riety breeding and gene identification involved in the reproduction and early development. The data suggests that gibel carp are under an evolutionary trajectory of diploidization. | 13 | Vegetation and soil carbon storage in China显示文摘This study estimated the current vegetation and soil carbon storage in China using a biogeochemical model driven with climate, soil and vegetation data at 0.5°latitude-longitude grid spatial resolution. The results indicate that the total carbon storage in China's vegetation and soils was 13.33 Gt C and 82.65 Gt C respectively, about 3% and 4% of the global total. The nationally mean vegetation and soil carbon densities were 1.47 kg C/m2 and 9.17 kg C/m2, respectively, differing greatly in various regions affected by climate, vegetation, and soil types. They were generally higher in the warm and wet Southeast China and Southwest China than in the arid Northwest China; whereas vegetation carbon density was the highest in the warm Southeast China and Southwest China, soil carbon density was the highest in the cold Northeast China and southeastern fringe of the Qinghai-Tibetan Plateau. | 15 | Geomorphologic evidence of phased uplift of the northeastern Qinghai-Tibet Plateau since 14 million years ago显示文摘A typical sequence of fluvial terraces and aeolian deposits overlying these terraces were multidisciplinary investigated. New evidences for uplift process of the northeastern Qinghai-Tibetan Plateau in the past 14 million years were obtained. At least 11 river terraces along Huangshui, the first-class tributary of Yellow River, at the Xining-Huzhu region are identified. While the first one (T1) is classified as an accumulation terrace, the others are all basement river terraces, which consist of the Tertiary sandstone and siltstone bedrock, fluvial gravel and pebbles and the overlying aeolian loess-Red Clay deposit. Samples from the aeolian deposits were examined for paleomagnetic stratigraphic reconstruction (1030 samples), luminescence dating (16 samples), magnetic susceptibility and grain-size distribution (more than 4000 samples). The luminescence dating and stratigraphic correlation suggest that terraces of T11, T10,T8, T7, T3, T2, T1 were formed at 14, 11.3, 1.55, 1.2, 0.15, 0.07 and 0.01 million years ago, respectively. Sedimentological analysis and geomorphological observation indicate that formation and evolution of these terraces were mainly driven by tectonic uplift. Therefore, the terrace sequence provides an ideal geological record of the uplift process of the northeastern Qinghai-Tibet during the past 14 million years, and the timings of the terraces formation are regarded as the timings of tectonic uplift. The significant uplifting events took place at 14, 11.3, 1.2 and 0.15 million years ago, respectively. The fluvial incision at the Xining-Huzhu region is less than 100 m during a period of ~12 million years in the Miocene era (between the T11 and T9), while the Huangshui River had incised 432 m during the past 1.2 million years (from T7 to the present floodplain). The river incision process clearly demonstrates that accelerated rising of the northeastern Qinghai-Tibet Plateau during the late Cenozoic, and provides new evidence of previous thoughts. There was a significant readjustment of the fluvial catchment during 1.55-1.2 million years ago: before this time, the paleoriver flowed to southwest. After this time the Huangshui River flows to southeast. | 16 | Analysis on the ecological benefits of the stream water conveyance to the dried-up river of the lower reaches of Tarim River,China显示文摘This paper analyzes the monitored data of the 4 times of stream water conveyances to the river section where the stream flow was cut-off, of 9 groundwater-monitoring sections and 18 vegetation plots in the lower reaches of Tarim River. The results show that the groundwater depth in the lower reaches of Tarim River rose from 9.87 m before the conveyances to 7.74 m and 3.79 m after the first and second conveyances, 3.61 and 3.16 m after the 2 phases of the third conveyance, and 2.66 m after the fourth conveyance. The transverse response scope of groundwater level was gradually enlarged along both sides of the channel of conveyances, i.e., from 450 m in width after the first conveyance to 1050 m after the fourth conveyance, but the response degree of groundwater level was reduced with the increase of the distance away from the channel of conveyances. The composition, distribution and growth status of the natural vegetation are directly related to the groundwater depth. The indexes of Simpson’s biodiversity, McIntosh’s evenness and Margalef’s richness, which reflect the change of the quantity of species and the degree of biodiversity, are reduced from 0.70, 0.48 and 0.90 to 0.26, 0.17 and 0.37 re- spectively along with the drawdown of groundwater level from the upper reaches to the lower reaches. After the stream water conveyances, the natural vegetation in the lower reaches is saved and restored along with the rise of groundwater level, the response scope of vegetation is gradually enlarged, i.e., from 200— 250 m in width after the first conveyance to 800 m after the fourth conveyance. However, there is still a great disparity to the objective of protecting the “Green Corridor”in the lower reaches of Tarim River. | 19 | Effects of N Forms and Rates on Vegetable Growth and Nitrate Accumulation显示文摘Experiments were carried out on a vegetable field with Peking cabbage (Brassica pekinensis (Lour.)Rupr.), cabbage (Brassica chinensis var. Oleifera Makino and nemoto), green cabbage (Brassica chinensis L.), spinach (Spinacia oleracea L.) and rape (Brassica campestris L.) to study the effects of N forms and N rates on their growth and nitrate accumulation. The results indicated that application of ammonium chloride,ammonium nitrate, sodium nitrate and urea significantly increased the yields and nitrate concentrations of Peking cabbage and spinach. Although no significant difference was found in the yields after application of the 4 N forms, nitrate N increased nitrate accumulation in vegetables much more than ammonium N. The vegetable yields were not increased continuously with N rate increase, and oversupply of N reduced the plant growth, leading to a yield decline. This trend was also true for nitrate concentrations in some vegetables and at some sampling times. However, as a whole, the nitrate concentrations in vegetables were positively correlated with N rates. Thus, addition of N fertilizer to soil was the major cause for increases in nitrate concentrations in vegetables. | 20 | INTERFACE REACTION IN MAGNETIC MULTILAYERS显示文摘Ta/NiO/NiFe/Ta multilayers were prepared by rf reactive and dc magnetron sputtering. The exchange coupling field (Hex) between NiO and NiFe reached 120Oe. The composition and chemical states at the interface region of NiO/NiFe were studied using the x-ray photoelectron spectroscopy (XPS) and peak decomposition technique. The results show that there are two thermodynamically favorable reactions at NiO/NiFe interface: NiO+Fe=Ni+FeO and 3NiO+2Fe = 3Ni+Fe2O3. The thickness of the chemical reaction as estimated by angle-resolved XPS was about 1-1.5nm. These interface reaction products are magnetic defects, and we believe that the Hex and the coercivity (Hc) of NiO/NiFe are affected by these defects. Moreover, the results also show that there is an 'intermixing layer' at the Ta/NiO (and NiO/Ta) interface due to a thermodynamically favorable reaction: 2Ta+5NiO=5Ni+ Ta2O5.
