1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Nucleocytoplasmic Trafficking of the Arabidopsis WD40 Repeat Protein XIW1 Regulates ABI5 Stability and Abscisic Acid Responses显示文摘WD40 repeat-containing proteins(WD40 proteins)serve as versatile scaffolds for protein-protein interac-tions,modulating a variety of cellular processes such as plant stress and hormone responses.Here we report the identification of a WD40 protein,XIW1(for XPO1-interacting WD40 protein 1),which positively regulates the abscisic acid(ABA)response in Arabidopsis.XIW1 is located in the cytoplasm and nucleus.We found that it interacts with the nuclear transport receptor XPO1 and is exported by XPO1 from the nucleus.Mutation of XIW1 reduces the induction of ABA-responsive genes and the accumulation of ABA Insensitive 5(ABI5),causing mutant plants with ABA-insensitive phenotypes during seed germination and seedling growth,and decreased drought stress resistance.ABA treatment upregulates the expression of XIW1,and both ABA and abiotic stresses promote XIW1 accumulation in the nucleus,where it interacts with ABI5.Loss of XIW1 function results in rapid proteasomal degradation of ABI5.Taken together,these findings suggest that XIW1 is a nucleocytoplasmic shuttling protein and plays a positive role in ABA responses by interacting with and maintaining the stability of ABI5 in the nucleus.Xuezhong XU Wang Wan Guobin Jiang Yue Xi Haijian Huang Jiajia Cai Yanan Chang Cheng-Guo Duan Satendra KMangrauthia Xinxiang Peng Jian-Kang Zhu Guohui Zhu 2019Molecular Plant2019,12,12:8

6Hydrogen peroxide-generating nanomedicine for enhanced chemodynamic therapy显示文摘Chemodynamic therapy(CDT)is an emerging endogenous stimulation activated tumor treatment approach that exploiting iron-containing nanomedicine as catalyst to convert hydrogen peroxide(H_(2)O_(2))into toxic hydroxyl radical(·OH)through Fenton reaction.Due to the unique characteristics(weak acidity and the high H_(2)O_(2) level)of the tumor microenvironment,CDT has advantages of high selectivity and low side effect.However,as an important substrate of Fenton reaction,the endogenous H_(2)O_(2) in tumor is still insufficient,which may be an important factor limiting the efficacy of CDT.In order to optimize CDT,various H_(2)O_(2)-generating nanomedicines that can promote the production of H_(2)O_(2) in tumor have been designed and developed for enhanced CDT.In this review,we summarize recently developed nanomedicines based on catalytic enzymes,nanozymes,drugs,metal peroxides and bacteria.Finally,the challenges and possible development directions for further enhancing CDT are prospected.Peng Yu Xiaodong Li Guohui Cheng Xu Zhang Dan Wu Jin Chang Sheng Wang 2021Chinese Chemical Letters2021,32,7:1

9Inhibiting DNA methylation alleviates cisplatininduced hearing loss by decreasing oxidative stress-induced mitochondria-dependent apoptosis via the LRP1-PI3K/AKT pathway显示文摘Cisplatin-related ototoxicity is a critical side effect of chemotherapy and can lead to irreversible hearing loss.This study aimed to assess the potential effect of the DNA methyltransferase(DNMT)inhibitor RG108 on cisplatin-induced ototoxicity.Immunohistochemistry,apoptosis assay,and auditory brainstem response(ABR)were employed to determine the impacts of RG108 on cisplatin-induced injury in murine hair cells(HCs)and spiral ganglion neurons(SGNs).Rhodamine 123 and TMRM were utilized for mitochondrial membrane potential(MMP)assessment.Reactive oxygen species(ROS)amounts were evaluated by Cellrox green and Mitosox-red probes.Mitochondrial respiratory function evaluation was performed by determining oxygen consumption rates(OCRs).The results showed that RG108 can markedly reduce cisplatin induced damage in HCs and SGNs,and alleviate apoptotic rate by protecting mitochondrial function through preventing ROS accumulation.Furthermore,RG108 upregulated BCL-2 and downregulated APAF1,BAX,and BAD in HEI-OC1 cells,and triggered the PI3K/AKT pathway.Decreased expression of low-density lipoprotein receptor-related protein 1(LRP1)and high methylation of the LRP1 promoter were observed after cisplatin treatment.RG108 treatment can increase LRP1expression and decrease LRP1 promoter methylation.In conclusion,RG108 might represent a new potential agent for preventing hearing loss induced by cisplatin via activating the LRP1-PI3K/AKT pathway.Yingzi He Zhiwei Zheng Chang Liu Wen Li Liping Zhao Guohui Nie Huawei Li 2022Acta Pharmaceutica Sinica B2022,12,3:1

11HOXC4 up-regulates NF-κB signaling and promotes the cell proliferation to drive development of human hematopoiesis, especially CD43+ cells显示文摘The hematopoietic function of HOXC4 has not been extensively investigated.Our research indicated that induction of HOXC4 in co-culture system from D10 significantly promoted productions of most hematopoietic progenitor cells.CD34−CD43+cells could be clearly classified into CD34−CD43^(low) and CD34−CD43^(high) sub-populations at D14.The former cells had greater myelogenic potential,and their production was not significantly influenced by induction of HOXC4.By contrast,the latter cells had greater potential to differentiate into megakaryocytes and erythroid cells,and thus had properties of erythroid–megakaryocyte common progenitors,which abundance was increased by∼2-fold when HOXC4 was induced from D10.For CD34−CD43^(low),CD34+CD43+,and CD34−CD43^(high) sub-populations,CD43 level served as a natural index for the tendency to undergo hematopoiesis.Induction of HOXC4 from D10 caused more CD43+cells sustain in S-phase with up-regulation of NF-κB signaling,which could be counteracted by inhibition of NF-κB signaling.These observations suggested that promotion of hematopoiesis by HOXC4 is closely related to NF-κB signaling and a change in cell-cycle status,which containing potential of clinical applications.Jiahui Zeng Wencui Sun Jing Chang Danying Yia Lijiao Zhu Yonggang Zhang Xu Pan Ya Zhou Mowen Lai Guohui Bian Qiongxiu Zhou Jiaxin Liu Bo Chen FengMa 2020Blood Science2020,2,4:1
12Overexpression of HOXA9 upregulates NF-κB signaling to promote human hematopoiesis and alter the hematopoietic differentiation potentials显示文摘Background: The HOX genes are master regulators of embryogenesis that are also involved inhematopoiesis. HOXA9 belongs to a cluster of HOX genes that play extensively studied roles inhematopoiesis and leukemogenesis.Methods: We established HOXA9-inducible human embryonic stem cells (HOXA9/hESCs) with normalpluripotency and potential for hematopoiesis, which could be used to analyze gene function with highaccuracy. HOXA9/hESCs co-cultured with aorta–gonad–mesonephros-derived stromal cells (AGM-S3) wereinduced to overexpress HOXA9 with doxycycline (DOX) at various times after hematopoiesis started andthen subjected to flow cytometry.Results: Induction of HOXA9 from Day 4 (D4) or later notably promoted hematopoiesis and also increasedthe production of CD34+ cells and derived populations. The potential for myelogenesis was significantlyelevated while the potential for erythrogenesis was significantly reduced. At D14, a significant promotion ofS phase was observed in green fluorescent protein positive (GFP+) cells overexpressing HOXA9. NF-κBsignaling was also up-regulated at D14 following induction of HOXA9 on D4. 13RUNX1-205, a novel splice variant of the human RUNX1 gene, has blockage effect on mesoderm-hemogenesis transition and promotion effect during the late stage of hematopoiesis显示文摘Runt-related transcription factor 1(RUNX1)is required for definitive hematopoiesis;however,the functions of most human RUNX1 isoforms are unclear.In particular,the effects of RUNX1-205(a novel splice variant that lacks exon 6 in comparison with RUNX1b)on human hematopoiesis are not clear.In this study,a human embryonic stem cell(hESC)line with inducible RUNX1-205 overexpression was established.Analyses of these cells revealed that induction of RUNX1-205 overexpression at early stage did not influence the induction of mesoderm but blocked the emergence of CD34+cells,and the production of hematopoietic stem/progenitor cells was significantly reduced.In addition,the expression of hematopoiesis-related factors was downregulated.However,these effects were abolished when RUNX1-205 overexpression was induced after Day 6 in co-cultures of hESCs and AGM-S3 cells,indicating that the inhibitory effect occurred prior to generation of hemogenic endothelial cells,while the promotive effect could be observed during the late stage of hematopoiesis.This is very similar to that of RUNX1b.Interestingly,the mRNA expression profile of RUNX1-205 during hematopoiesis was distinct from that of RUNX1b,and the protein stability of RUNX1-205 was much higher than that of RUNX1b.Thus,the function of RUNX1-205 in normal and diseased models should be further explored.Wencui Sun Jiahui Zeng Jing Chang Yuan Xue Yonggang Zhang Xu Pan Ya Zhou Mowen Lai Guohui Bian Qiongxiu Zhou Jiaxing Liu Bo Chen Feng Ma 2020Journal of Molecular Cell Biology2020,12,5:0
14EXPORTIN 1A prevents transgene silencing in Arabidopsis by modulating nucleo-cytoplasmic partitioning of HDA6显示文摘In eukaryotic cells,transport of macromolecules across the nuclear envelope is an essential process that ensures rapid exchange of cellular components,including protein and RNA molecules.Chromatin regulators involved in epigenetic control are among the molecules exported across the nuclear envelope,but the significance of this nucleo-cytoplasmic trafficking is not well understood.Here,we use a forward screen to isolate XPO1 A(a nuclear export receptor in Arabidopsis)as an anti-silencing factor that protects transgenes from transcriptional silencing.Loss-of-function of XPO1 A leads to locus-specific DNA hypermethylation at transgene promoters and some endogenous loci.Wefound that XPO1 A directly interacts with histone deacetylase HDA6 in vivo and that the xpo1 a mutation causes increased nuclear retention of HDA6 protein and results in reduced histone acetylation and enhanced transgene silencing.Our results reveal a new mechanism of epigenetic regulation through the modulation of XPO1 A-dependent nucleo-cytoplasm partitioning of a chromatin regulator.Guohui Zhu Yanan Chang Xuezhong Xu Kai Tang Chunxiang Chen Mingguang Lei Jian-Kang Zhu Cheng-Guo Duan 2019Journal of Integrative Plant Biology2019,61,12:0
15Comprehensive evaluation and analysis of a nearly zero-energy building heating system using a multi-source heat pump in severe cold region显示文摘The integrated application of multi-energy coupled technology in nearly zero-energy building(NZEB)is promising from the perspective of low-carbon development to achieve the goal of net zero energy.PVT(photovoltaic/thermal),air,and ground sources were combined organically to establish an experimental platform of a multi-source heat pump(MSHP)system,which can realize flexible switching of multi-energy sources.The paper presents the analytical hierarchy process and fuzzy comprehensive evaluation method to comprehensively evaluate the five modes of the MSHP system with regard to energy,economic and environmental benefits.The results indicate that the waste heat of the PVT cavity can improve the coefficient of performance of the heat pump unit(COP)by approximately 8.0%.The initial investment in air source heat pump(ASHP)modes is lower than that of a coal-powered system.The ground source heat pump(GSHP)modes have high stability and their payback period is 8.81–10.66 years.The photovoltaic/thermal-dual source heat pump(PVT-DSHP)mode presents the most appropriate system applied in the NZEB in severe cold region,followed by the DSHP,GSHP,ASHP,and PVT-ASHP mode.When compared with other modes,COP,annual saving cost,carbon dioxide emission reduction,and comprehensive value of the PVT-DSHP mode have improved by 7.07%–29.57%,2.21%–23.88%,3.38%–14.83%,and 27.91%–52.62%,respectively.The study provides important insights into the practical application and sustainable development of multi-energy coupled systems in the NZEB in severe cold region.Lei Zhang Guohui Feng Ainong Li Kailiang Huang Shasha Chang 2023Building Simulation2023,16,10:0
16Inhibition of histone methyltransferase PRMT5 attenuates cisplatininduced hearing loss through the PI3K/Akt-mediated mitochondrial apoptotic pathway显示文摘This study aimed to evaluate the therapeutic potential of inhibiting protein arginine methyltransferase 5(PRMT5)in cisplatin-induced hearing loss.The effects of PRMT5 inhibition on cisplatin-induced auditory injury were determined using immunohistochemistry,apoptosis assays,and auditory brainstem response.The mechanism of PRMT5 inhibition on hair cell survival was assessed using RNA-seq and Cleavage Under Targets and Tagment-quantitative polymerase chain reaction(CUT&Tag-qPCR)analyses in the HEI-OC1 cell line.Pharmacological inhibition of PRMT5 significantly alleviated cisplatin-induced damage to hair cells and spiral ganglion neurons in the cochlea and decreased apoptosis by protecting mitochondrial function and preventing the accumulation of reactive oxygen species.CUT&Tag-qPCR analysis demonstrated that inhibition of PRMT5 in HEI-OC1 cells reduced the accumulation of H4R3me2s/H3R8me2s marks at the promoter region of the Pik3ca gene,thus activating the expression of Pik3ca.These findings suggest that PRMT5 inhibitors have strong potential as agents against cisplatininduced ototoxicity and can lay the foundation for further research on treatment strategies of hearing loss.Zhiwei Zheng Benyu Nan Chang Liu Dongmei Tang Wen Li Liping Zhao Guohui Nie Yingzi He 2023Journal of Pharmaceutical Analysis2023,13,6:0
