2Identification and expression profiling of Vigna mungo microRNAs from leaf small RNA transcriptome by deep sequencing显示文摘MicroRNAs(miRNAs) represent a class of small non-coding RNA molecules that play a crucial role in post-transcriptional gene regulation. Several conserved and species-specific miRNAs have been characterized to date, predominantly from the plant species whose genome is well characterized. However, information on the variability of these regulatory RNAs in economically important but genetically less characterized crop species are limited. Vigna mungo is an important grain legume, which is grown primarily for its protein-rich edible seeds. miRNAs from this species have not been identified to date due to lack of genome sequence information. To identify miRNAs from V. mungo, a small RNA library was constructed from young leaves. High-throughput Illumina sequencing technology and bioinformatic analysis of the small RNA reads led to the identification of 66 miRNA loci represented by 45 conserved miRNAs belonging to 19 families and eight non-conserved miRNAs belonging to seven families. Besides, 13 novel miRNAcandidates in V. mungo were also identified. Expression patterns of selected conserved, non-conserved, and novel miRNA candidates have been demonstrated in leaf, stem, and root tissues by quantitative polymerase chain reaction, and potential target genes were predicted for most of the conserved miRNAs. 4Adjustable Mechanism for Walking Robots with Minimum Number of Actuators显示文摘Recent literature on walking robots deals predominantly with multi-degrees-of-freedom leg mechanisms and machines capable of adopting several gaits.This paper explores the other end of the spectrum suggesting mechanisms derived from a four bar coupler curve for a one degree of freedom walking robot.Simulation of the walk indicates that body of the robot is able to move with low variation in velocity.The best strategy for changing the gait to enable the robot to walk over obstacles and the effect of change in length of different links are explored to open up the possibility of a two degree of freedom walking robot with the capability of changing its gait,suitable as a low cost unit for several applications.Such rugged units would permit the use of an IC engine as the primary source of power and could be of utility in installations where electronics may not be functional.In simple walking machines the foot of a leg is usually required to trace a D shaped curve with respect to the chassis.In this paper we begin with a Hoecken mechanism capable of tracing such a curve.The foot is required to move parallel to itself and the same could be achieved using a six or eight link mechanism.A few such devices have been synthesized in this paper and their motion properties compared.The study also covers the possibility of providing adjustments to vary the step length and height of the foot's movement.GUHA Anirban AMARNATH C 2011Chinese Journal of Mechanical Engineering2011,24,5:4
5Difficult-to-culture bacteria in the rhizosphere:The underexplored signature microbial groups显示文摘Microorganisms represent a substantial portion of the earth’s biodiversity and biomass, and the plant rhizosphere is an innate reservoir teeming with heterogeneous microbes predominated by bacterial communities. Rhizospheric microbial diversity (genetic, phenotypic, and metabolic) has been extensively studied to understand the key ecological roles played by the microbial members, including plant growth promotion. The application of 16S rRNA gene sequencing and next-generation sequencing (NGS) technologies has revolutionized the discovery of novel bacterial groups that have remained undetected by traditional cultivation-based approaches. Such technological advancements have opened new vistas in our current understanding of predominant but concealed and missed bacterial diversity referred to as difficult-to-culture bacterial lineages, especially the predominant phyla Acidobacteria, Verrucomicrobia,Planctomycetes, and Gemmatimonadetes. Regardless of their ubiquity and prevalence, little is known about their ecophysiology because of the non-availability of culturable members. More recently, there has been increased interest in understanding the cosmopolitan distribution and diversity of the difficult-to-culture bacteria, focusing on their role in driving complex plant-microbial interactions and mobilizing nutrients in soil and their potential as sources of novel bioactive metabolites. As an initial step, we review the distribution and significance of such bacterial phyla in soil, their ecophysiological roles, and their hidden plant growth promoting potential. 6Experimental and analytical investigation on metal damage suffered from simulated lightning currents显示文摘The damage of two typical metal materials, Al alloy 3003 and steel alloy Q235 B, subjected to four representative lightning current components are investigated by laboratory and analytical studies to provide fundamental data for lightning protection. The four lightning components simulating the natural lightning consist of the first return stroke, the continuing current of interval stroke, the long continuing current, and the subsequent stroke, with amplitudes 200 k A, 8 k A,400 A, and 100 k A, respectively. The damage depth and area suffered from different lightning components are measured by the ultrasonic scanning system. And the temperature rise is measured by the thermal imaging camera. The results show that, for both Al 3003 and steel Q235 B, the first return stroke component results in the largest damage area with damage depth0.02 mm uttermost. The long continuing current component leads to the deepest damage depth of 3.3 mm for Al 3003 and much higher temperature rise than other components. The correlation analysis between damage results and lightning parameters indicates that the damage depth has a positive correlation with charge transfer. 8Hemodynamic effects of propranolol with spironolactone in patients with variceal bleeds: A randomized controlled trial显示文摘AIM: To study the hemodynamic effects of spironolactone with propranolol vs propranolol alone in the secondary prophylaxis of variceal bleeding. METHODS: Thirty-five cirrhotics with variceal bleeding randomly received propranolol (n = 17: Group A) or spironolactone plus propranolol (n = 18: Group B). Hemodynamic assessment was performed at baseline and on the eighth day. RESULTS: Spironolactone with propranolol caused a greater reduction in the hepatic venous pressure gradient than propranolol alone (26.94% vs 10.2%; P < 0.01). Fourteen out of eighteen patients on the combination treatment had a reduction in hepatic venous pressure gradient to ≤ 12 mmHg or a 20% reduction from baseline in contrast to only six out of seventeen (6/17) on propranolol alone (P < 0.05). CONCLUSION: Spironolactone with propranolol results in a better response with a greater reduction in hepatic venous pressure gradient in the secondary prophylaxis of variceal bleeding. A greater number of patients may be protected by this combination therapy than by propranolol alone. 12Liver abscess caused by Burkholderia pseudomallei in a young man: A case report and review of literature显示文摘Pyogenic liver abscess is a common entity in Indian subcontinent and is mostly caused by gram negative bacteria. Melioidosis is not commonly seen in India and only a few cases are reported. It can give rise to multiple abscesses at different sites including liver. We report a case of isolated liver abscess caused by Burkholderia pseudomallei(B. pseudomallei) in a 29-yearold recently diagnosed diabetic, immunocompetent male. Diagnosis was made by imaging and culture of pus aspirated from the abscess and he was treated with percutaneous pigtail catheter drainage followed by antibiotics(meropenem and trimethoprim-sulphmethoxazole). Melioidosis is an emerging infection in India and has high mortality rate, so early diagnosis and promptmanagement is warranted which requires clinical vigilance and an intensive microbiological workup. 13Pulsed laser deposited Al-doped ZnO thin films for optical applications显示文摘Highly transparent and conducting Al-doped Zn O(Al:Zn O) thin films were grown on glass substrates using pulsed laser deposition technique.The profound effect of film thickness on the structural, optical and electrical properties of Al:Zn O thin films was observed. The X-ray diffraction depicts c-axis, plane(002) oriented thin films with hexagonal wurtzite crystal structure. Al-doping in Zn O introduces a compressive stress in the films which increase with the film thickness. AFM images reveal the columnar grain formation with low surface roughness. The versatile optical properties of Al:Zn O thin films are important for applications such as transparent electromagnetic interference(EMI) shielding materials and solar cells. The obtained optical band gap(3.2–3.08 e V) was found to be less than pure Zn O(3.37 e V) films. The lowering in the band gap in Al:Zn O thin films could be attributed to band edge bending phenomena. The photoluminescence spectra gives sharp visible emission peaks, enables Al:Zn O thin films for light emitting devices(LEDs) applications.
