| 1 | Chromosomal localization of the major ribosomal RNA genes in scallop Chlamys farreri显示文摘The chromosomes of Chlamys farreri were analyzed by means of silver staining and fluorescence in situ hybridization (FISH) with 18S—28S rDNA probe. Probe was made by PCR amplification of a DNA fragment containing internal transcribed spacers ITS1 between 18S and 5.8S ribosomal RNA gene, ITS2 between 5.8S and 28S ribosomal RNA gene and 5.8S rRNA gene, and labeled by PCR incorporation of bio-16-dUTP. FISH signals were located on the short arm of subtelocentric chromosome 10. After silver-staining, nucleolus organizer regions (NORs) could be observed on the telomere of the short arm of chromosome 10. | 2 | Long-Term Cognitive Improvement After Benfotiamine Administration in Patients with Alzheimer's Disease显示文摘To date,we still lack disease-modifying therapies for Alzheimer's disease(AD).Here,we report that long-term administration of benfotiamine improved the cognitive ability of patients with AD.Five patients with mild to moderate AD received oral benfotiamine(300 mg daily) over 18 months.All patients were examined by positron emission tomography with Pittsburgh compound B(PiB-PET) and exhibited positive imaging with pamyloid deposition,and three received PiB-PET imaging at follow-up.The five patients exhibited cognitive improvement as assayed by the Mini-Mental Status Examination(MMSE) with an average increase of 3.2 points at month18 of benfotiamine administration.The three patients who received follow-up PiB-PET had a 36.7%increase in the average standardized uptake value ratio in the brain compared with that in the first scan.Importantly,the MMSE scores of these three had an average increase of 3 points during the same period.Benfotiamine significantly improved the cognitive abilities of mild to moderate AD patients independently of brain amyloid accumulation.Our study provides new insight to the development of diseasemodifying therapy. | Xiaoli Pan Zhichun Chen Guoqiang Fei Shumei Pan Weiqi Bao Shuhua Ren Yihui Guan Chunjiu Zhong | 2016 | Neuroscience Bulletin2016,32,6: | 9 |

| 7 | Molecular identification of scallop planktonic larvae using species-specific microsatellites显示文摘The identification of scallop larvae is essential to understand the population structure and community dynamics and to assess the potential environmental impacts caused by scallop larvae released or escaped. However,the larvae identification by morphological characteristics is notoriously difficult,mainly due to the small size (usually being less than 150 μm) and vague morphological characteristics among different scallop species. A simple and accurate molecular method was developed to identify four economically farmed scallop species,the Zhikong scallop Chlamys farreri,the noble scallop C. nobilis,the bay scallop Argopecten irradians and the Yesso scallop Mizuhopecten yessoensis. The tests used the high degree of species-specific microsatellite markers,which was specified by transferability analyses,assessed by reference individuals and evaluated by BLAST searches. The sensitivity test indicated that the species-specific microsatellites were sensitive enough for the detection of 1%~2% larvae in mixed plankton samples. Larvae collected from scallop hatcheries and their effluents and from the artificially controlled crosses were well identified to the species/hybrid level. | 8 | Development of 101 Novel EST-Derived Single Nucleotide Polymorphism Markers for Zhikong Scallop (Chlamys farreri)显示文摘Zhikong scallop(Chlamys farreri) is an important maricultured species in China.Many researches on this species,such as population genetics and QTL fine-mapping,need a large number of molecular markers.In this study,based on the expressed sequence tags(EST),a total of 300 putative single nucleotide polymorphisms(SNPs) were selected and validated using high resolution melting(HRM) technology with unlabeled probe.Of them,101(33.7%) were found to be polymorphic in 48 individuals from 4 populations.Further evaluation with 48 individuals from Qingdao population showed that all the polymorphic loci had two alleles with the minor allele frequency ranged from 0.046 to 0.500.The observed and expected heterozygosities ranged from 0.000 to 0.925 and from 0.089 to 0.505,respectively.Fifteen loci deviated significantly from Hardy-Weinberg equilibrium and significant linkage disequilibrate was detected in one pair of markers.BLASTx gave significant hits for 72 of the 101 polymorphic SNP-containing ESTs.Thirty four polymorphic SNP loci were predicted to be non-synonymous substitutions as they caused either the change of codons(33 SNPs) or pretermination of translation(1 SNP).The markers developed can be used for the population studies and genetic improvement on Zhikong scallop. | LI Jiqin BAO Zhenmin LI Ling WANG Xiaojian WANG Shi HU Xiaoli | 2013 | Journal of Ocean University of China2013,12,3: | 2 |
| 9 | Glutathione S-transferase genes in scallops and their diverse expression patterns after exposure to PST-producing dinoflagellates显示文摘The glutathione S-transferases(GSTs)are a superfamily of enzymes that function in cellular protection against toxic substances and oxidative stress.Bivalves could accumulate high concentration of paralytic shellfish toxins(PSTs)from harmful algae.To understand the possible involvement of GSTs in protecting bivalves during PST accumulation and metabolism,the GST genes were systemically analyzed in two cultured scallops,Azumapecten farreri and Mizuhopecten yessoensis,which were reported for PST deposition during harmful algae bloom.A total of 35 and 37 GSTs were identified in A.farreri(AfGSTs)and M.yessoensis(MyGSTs)genome,respectively,and the expansion of the sigma class from the cytosolic subfamily was observed.In both scallop species,sigma class GSTs showed higher expression than other members.The high GSTs expression was detected mainly during/after larvae stages and in the two most toxic organs,hepatopancreas and kidney.After ingesting PST-producing dinoflagellates,all the regulated AfGSTs in the hepatopancrcas were from the sigma class,but with opposite regulation pattern between Alexandrium catenella and A.minutum exposure.In scallop kidneys,where PSTs transformed into higher toxicity,more AfGSTs were regulated than in the hepatopancreas,and most of them were from the sigma class,with similar regulation pattern between A.catenella and A.minutum exposure.In M.yessoensis exposed to A.catenella,MyGST-σ2 was the only up-regulated MyGST in both hepatopancreas and kidney.Our results suggested the possible diverse function of scallop GSTs and the importance of sigma class in the defense against PSTs,which would contribute to the adaptive evolution of scallops in marine environments. | Jiarun Lou Jie Cheng Xiaogang Xun Xu Li Moli Li Xiangchao Zhang Tingting Li Zhenmin Bao Xiaoli Hu | 2020 | Marine Life Science & Technology2020,2,3: | 2 |
| 10 | An Efficient Procedure for Isolating Microsatellite DNAs from Sea Cucumber(Apostichopus japonicus)显示文摘The construction of enrichment library proves to be one of the efficient approaches for isolating microsatellites in this study. The genomic DNA of sea cucumber was digested with HaeIII and size-selected DNA fragments (250-700 bp) were ligated to an adaptor. Microsatellite-containing sequences were captured by using a combination of GA and CA probes, which were attached to a nylon membrane. The microsatellite enrichment library constructed in this study consisted of approximately 700 clones. Two hun-dred and thirty-two clones reacted positively after the library screening procedure. Of the 50 clones sequenced, all contained at least one microsatellite and one duplicate clone was found. | 11 | Development and Characterization of Microsatellite Markers for the Pacific Abalone (Haliotis discus) via EST Database Mining显示文摘The EST database of the Pacific abalone (Haliotis discus) was mined for developing microsatellite markers. A total of 1476 EST sequences were registered in GenBank when data mining was performed. Fifty sequences (approximately 3.4%) were found to contain one or more microsatellites. Based on the length and GC content of the flanking regions, cluster analysis and BLASTN, 13 microsatellite-containing ESTs were selected for PCR primer design. The results showed that 10 out of 13 primer pairs could amplify scorable PCR products and showed polymorphism. The number of alleles ranged from 2 to 13 and the values of Ho and He varied from 0.1222 to 0.8611 and 0.2449 to 0.9311, respectively. No significant linkage disequilibrium (LD) between any pairs of these loci was found, and 6 of 10 loci conformed to the Hardy-Weinberg equilibrium (HWE). | 15 | Rice FLOURY ENDOSPERM22, encoding a pentatricopeptide repeat protein, is involved in both mitochondrial RNA splicing and editing and is crucial for endosperm development显示文摘Most of the reported P-type pentatricopeptide repeat(PPR) proteins play roles in organelle RNA stabilization and splicing. However, P-type PPRs involved in both RNA splicing and editing have rarely been reported, and their underlying mechanism remains largely unknown. Here, we report a rice floury endosperm22(flo22) mutant with delayed amyloplast development in endosperm cells. Map-based cloning and complementation tests demonstrated that FLO22 encodes a mitochondrion-localized P-type PPR protein.Mutation of FLO22 resulting in defective transsplicing of mitochondrial nad1 intron 1 and perhaps causing instability of mature transcripts affected assembly and activity of complex Ⅰ, and mitochondrial morphology and function. RNA-seq analysis showed that expression levels of many genes involved in starch and sucrose metabolism were significantly down-regulated in the flo22mutant compared with the wild type, whereas genes related to oxidative phosphorylation and the tricarboxylic acid cycle were significantly upregulated. In addition to involvement in splicing as a P-type PPR protein, we found that FLO22 interacted with DYW3, a DYW-type PPR protein, and they may function synergistically in mitochondrial RNA editing. | 18 | Methods Comparison for Microsatellite Marker Development:Different Isolation Methods,Different Yield Efficiency显示文摘Microsatellite markers have become one kind of the most important molecular tools used in various researches. A large number of microsatellite markers are required for the whole genome survey in the fields of molecular ecology,quantitative genetics and genomics. Therefore,it is extremely necessary to select several versatile,low-cost,efficient and time-and labor-saving methods to develop a large panel of microsatellite markers. In this study,we used Zhikong scallop(Chlamys farreri) as the target species to compare the efficiency of the five methods derived from three strategies for microsatellite marker development. The results showed that the strategy of constructing small insert genomic DNA library resulted in poor efficiency,while the microsatellite-enriched strategy highly improved the isolation efficiency. Although the mining public database strategy is time-and cost-saving,it is difficult to obtain a large number of microsatellite markers,mainly due to the limited sequence data of non-model species deposited in public databases. Based on the results in this study,we recommend two methods,microsatellite-enriched library construction method and FIASCO-colony hybridization method,for large-scale microsatellite marker development. Both methods were derived from the microsatellite-enriched strategy.
