| 2 | Impacts of nighttime post-anthesis warming on rice productivity and grain quality in East China显示文摘The impacts of nighttime post-anthesis warming on rice productivity and grain quality in East China were evaluated for two cultivars, II You 128, an indica rice, and Wuyunjing 7, a japonica rice. Warming by 3.0 °C stimulated the nighttime respiration rate and decreased the photosynthesis rate, resulting in significant decreases of 21.2% and 24.9% in aboveground biomass accumulation for II You 128 and Wuyunjing 7, respectively. Warming significantly reduced the rates of seed setting and grain filling, especially of inferior kernels(those lower in panicles), while the filling rate of superior kernels remained almost unchanged. As a result, 1000-grain weight and grain yield were respectively 3.7% and 30.0% lower for II You 128 and 12.8% and 34.3% for Wuyunjing 7 in warmed plots than in the unwarmed control. Nighttime warming also significantly reduced the grain milling and appearance quality of both varieties. More negative effects of warming on inferior than on superior kernels were found. | 4 | Rice FLOURY ENDOSPERM22, encoding a pentatricopeptide repeat protein, is involved in both mitochondrial RNA splicing and editing and is crucial for endosperm development显示文摘Most of the reported P-type pentatricopeptide repeat(PPR) proteins play roles in organelle RNA stabilization and splicing. However, P-type PPRs involved in both RNA splicing and editing have rarely been reported, and their underlying mechanism remains largely unknown. Here, we report a rice floury endosperm22(flo22) mutant with delayed amyloplast development in endosperm cells. Map-based cloning and complementation tests demonstrated that FLO22 encodes a mitochondrion-localized P-type PPR protein.Mutation of FLO22 resulting in defective transsplicing of mitochondrial nad1 intron 1 and perhaps causing instability of mature transcripts affected assembly and activity of complex Ⅰ, and mitochondrial morphology and function. RNA-seq analysis showed that expression levels of many genes involved in starch and sucrose metabolism were significantly down-regulated in the flo22mutant compared with the wild type, whereas genes related to oxidative phosphorylation and the tricarboxylic acid cycle were significantly upregulated. In addition to involvement in splicing as a P-type PPR protein, we found that FLO22 interacted with DYW3, a DYW-type PPR protein, and they may function synergistically in mitochondrial RNA editing. | 7 | Genetic dissection of top three leaf traits in rice using progenies from a japonica × indica cross显示文摘The size of the top three leaves of rice plants is strongly associated with yield; thus, it is important to consider quantitative traits representing leaf size(e.g.,length and width) when breeding novel rice varieties.It is challenging to measure such traits on a large scale in the field, and little is known about the genetic factors that determine the size of the top three leaves. In the present study, a population of recombinant inbred lines(RILs) and reciprocal single chromosomal segment substitution lines(SSSLs) derived from the progeny of a japonica Asominori ? indica IR24 cross were grown under four diverse environmental conditions. Six morphological traits associated with leaf size were measured,namely length and width of the flag, second and third leaves. In the RIL population, 49 QTLs were identified that clustered in 30 genomic region. Twenty-three of these QTLs were confirmed in the SSSL population. A comparison with previously reported genes/QTLs revealed eight novel genomic regions that contained uncharacterized ORFs associated with leaf size. | 9 | BRITTLE PLANT1 is required for normal cell wall composition and mechanical strength in rice显示文摘A series of nucleotide sugar interconversion enzymes(NSEs) generate the activated sugar donors required for biosynthesis of cell wall matrix polysaccharides and glycoproteins. UDPglucose 4-epimerases(UGEs) are NSEs that function in the interconversion of UDP-glucose(UDP-Glc) and UDP-galactose(UDP-Gal). The roles of UDP-glucose 4-epimerases in monocots remain unclear due to redundancy in the pathways. Here, we report a brittle plant(bp1) rice mutant that exhibits brittle leaves and culms at all growth stages. The mutant culms had reduced levels of rhamnogalacturonan I, homogalacturonan, and arabinogalactan proteins.Moreover, the mutant had altered contents of uronic acids, neutral noncellulosic monosaccharides, and cellulose. Map-based cloning demonstrated that OsBP1 encodes a UDPglucose 4-epimerase(OsUGE2), a cytosolic protein. We also show that BP1 can form homoand hetero-protein complexes with other UGE family members and with UDP-galactose transporters 2(OsUGT2) and 3(OsUGT3), which may facilitate the channeling of Gal to polysaccharides and proteoglycans. | 10 | Tetrapyrrole biosynthesis pathway regulates plastid-to-nucleus signaling by controlling plastid gene expression in plants显示文摘Plastid-to-nucleus retrograde signaling coordinates nuclear gene expression with chloroplast developmental status and is essential for the photoautotrophic lifestyle of plants.Previous studies have established that tetrapyrrole biosynthesis(TPB)and plastid gene expression(PGE)play essential roles in plastid retrograde signaling during early chloroplast biogenesis;however,their functional relationship remains unknown.In this study,we generated a series of rice TPB-related gun(genome uncoupled)mutants and systematically analyzed their effects on nuclear and plastid gene expression under normal conditions or when subjected to treatments with norflurazon(NF;a noncompetitive inhibitor of carotenoid biosynthesis)and/or lincomycin(Lin;a specific inhibitor of plastid translation).We show that under NF treatment,expression of plastid-encoded polymerase(PEP)-transcribed genes is significantly reduced in the wild type but is derepressed in the TPB-related gun mutants.We further demonstrate that the derepressed expression of PEPtranscribed genes may be caused by increased expression of the PEP core subunit and nuclear-encoded sigma factors and by elevated copy numbers of plastid genome per haploid genome.In addition,we show that expression of photosynthesis-associated nuclear genes(PhANGs)and PEP-transcribed genes is correlated in the rice TPB-related gun mutants,with or without NF or Lin treatment.A similar correlation between PhANGs and PGE is also observed in the Arabidopsis gun4 and gun5 mutants.Moreover,we show that increased expression of PEP-transcribed plastid genes is necessary for the gun phenotype in NF-treated TPB-related gun mutants.Further,we provide evidence that these TPB-related GUN genes act upstream of GUN1 in the regulation of retrograde signaling.Taken together,our results suggest that the TPB-related GUN genes control retrograde plastid signaling by regulating the PGE-dependent retrograde signaling pathway. | Yunlong Wang Yihua Wang Xiaopin Zhu Yulong Ren Hui Dong Erchao Duan Xuan Teng Huanhuan Zhao Rongbo Chen Xiaoli Chen Jie Lei Hang Yang Yunlu Tian Liangming Chen Xi Liu Shijia Liu Ling Jiang Haiyang Wang Jianmin Wan | 2023 | Plant Communications2023,4,1: | 0 |

| 14 | IR780/Gemcitabine-conjugated metal-phenolic network enhanced photodynamic cancer therapy显示文摘Photodynamic therapy (PDT) is a clinically approved cancer treatment that uses energy of light to generate active substances that cause damage to the cancer. Photosensitizers are employed to absorb light and generate toxic reactive oxygen species (ROS) to damage biomolecules like DNA. At the same time, some chemotherapy drugs like nucleotide analogues can provide mechanism-guided promotion in the treatment efficacy of PDT. However, the photosensitizer and chemotherapy drugs used in PDT is usually organic molecules, which suffers from bad solubility, fast clearance, and acute toxicity. To achieve targeted treatment, a reasonable delivery system is necessary. Therefore, we reported a metal-phenolic network where IR780 and gemcitabine were coupled chemically to overcome these shortcomings. The enhanced PDT effects can be realized by the promoted cell death both in vitro and in vivo. Moreover, the synergistic therapy also induced T-cell mediated anti-tumor immune response, which was significant for the inhibition of distant tumor growth.
