1The Ap-2α/Elk-1 axis regulates Sirpα-dependent tumor phagocytosis by tumor-associated macrophages in colorectal cancer显示文摘The inhibitory receptor signal regulatory protein-α(Sirpα)is a myeloid-specific immune checkpoint that engages the“don’t eat me”signal CD47,which is expressed on tumor and normal tissue cells.However,the profile and regulatory mechanism of Sirpαexpression in tumor-associated macrophages(TAMs)are still not clear.Here,we found that the expression of Sirpαin TAMs increased dynamically with colorectal cancer(CRC)progression.Mechanistically,CRC cell-derived lactate induced the nuclear translocation of the transcription factor Ap-2αfrom the cytoplasm in TAMs.Ap-2αfunctioned as a transcription factor for Elk-1 by binding to the conserved element GCCTGC located at−1396/−1391 in the mouse Elk-1 promoter.Subsequently,the Elk-1 protein bound to two conserved sites,CTTCCTACA(located at−229/−221)and CTTCCTCTC(located at−190/−182),in the mouse Sirpαpromoter and promoted Sirpαexpression in TAMs.Functionally,the macrophage-specific knockout of Ap-2αnotably promoted the phagocytic activity of TAMs and suppressed CRC progression,whereas these effects were prevented by the transgenic macrophage-specific expression of Elk-1,which regulated TAM phagocytosis and CRC development in a Sirpα-dependent manner.Furthermore,we showed that Elk-1 expression was positively correlated with Sirpαexpression in TAMs and was associated with poor survival in CRC patients.Taken together,our findings revealed a novel mechanism through which CRC evades innate immune surveillance and provided potential targets for macrophage-based immunotherapy for CRC patients.Xiaojiao Wang Xi Luo Chuan Chen Ye Tang Lian Li Banghui Mo Houjie Liang Songtao Yu 2020Signal Transduction and Targeted Therapy2020,5,1:2
2Study of Helicity in Typhoon Rainfall Regions——A Case Study of Typhoon 'Muifa'显示文摘Using the center of figure method and center of mass method,precipitation and heavy precipitation processes caused by No.9 typhoon 'Muifa' in 2011 were analyzed based on helicity,and the mechanism of typhoon rainfall was interpreted from the relation between helicity and energy. The results showed that horizontal helicity checked the diffusion of nonlinear energy and maintained the development of the system. Precipitation mainly appeared on the southeast of vertical helicity,while heavy precipitation mainly occurred on its southwest. Compared with the center of figure method,the center of mass method had good results,especially for heavy precipitation. 3Phosphatidylserine externalized on the colonic capillaries as a novel pharmacological target for IBD therapy显示文摘Inflammatory bowel disease(IBD)is a chronic and relapsing disorder for many people associated with poor health.Although there are some clinical drugs for IBD treatment,the development of effective therapeutics on IBD patients has always been necessary.Here,we show that externalized phosphatidylserine(PS)is observed on the surface of colonic capillaries.Annexin A5(ANXA5)with high affinity for PS has a good targeting to the colon and effectively alleviates experimental colitis.In contrast,ANXA5 mutant(A5m)lacking the PS-binding ability,has no accumulation in the colon and no therapeutic effects on colitis.Mechanistic investigations indicate that ANXA5 reduces the inflammatory cell infiltration by inhibiting endothelial cell activation dependent on PS-binding ability.With the increasing of PS exposure on activated HUVECs(human umbilical vein endothelial cells),ANXA5 binding induces the internalization of TLR4 via PS-dependent endocytosis.We provide new insights on the molecular mechanism of ANXA5 for its anti-inflammatory effect.Our data suggest that PS-externalization is a potential target of ANXA5 aiming at targeted drug delivery(TDD)for IBD treatment.Xuerui Zhang Lulu Song Lin Li Banghui Zhu Lina Huo Zhaoqing Hu Xinran Wang Jie Wang Mengyue Gao Jing Zhang Zichun Hua 2021Signal Transduction and Targeted Therapy2021,6,7:1
4Analysis of Characteristic and Causes of a Strong Cooling Process in China in the Early Winter of 2020/2021显示文摘The characteristics and causes of a drop in temperature during a cold wave process in the early winter of 2020/2021 were analyzed.The results show that the air temperature at 700-600 hPa over China was firstly and mostly influenced by the cold wave process,and then the cold air gradually extended to the lower layer,causing the most severe cooling in North China and its nearby areas.During the cold wave,the longitude of the upper-level jet over the Chinese mainland was larger;the Ural blocking high and the East Asian trough were stronger,so that the geopotential height gradient between the two was also significantly larger;the meridional air flow was abnormally strong,which was conducive to the southward transport of cold air from the middle and high latitudes.Results of the diagnostic analysis further show that the outbreak of the cold wave and the negative temperature tendency anomaly in the key area were mainly caused by the meridional temperature horizontal advection anomaly,while the temperature rise accompanied by abnormal air subsidence compensated for the abnormal decrease in temperature,which was conducive to the gradual rise of temperature in the key area.Ju WANG Tianju WANG Hong HUANG Banghui HU Xuezhong WANG 2023Meteorological and Environmental Research2023,14,1:0
5Growth and Morphological Differences between the Sexes and Female Fecundity of Ocadia sinensis显示文摘In this study,artificially hatched Ocadia sinensis individuals were cultured for 60 months,to record the growth and development process and investigate the morphological characteristics between the sexes and individual fecundity of O. sinensis. Results showed that the main morphological differences between male and female individuals are concentrated in secondary sexual characteristics: female turtle individuals have flat or slightly prominent sternites,short tails,short length from anal pore to sternite trailing edge; male turtle individuals have concave sternites,relatively stout tails,large length from anal pore to sternite trailing edge. There were significant differences in the growth of male and female individuals,in the first 12 months,female individuals grew faster than male individuals, with greater carapace length,carapace width and weight,but the differences between male and female individuals were not significant; in the late stage,the growth rate of female individuals was much higher than that of male individuals; by the end of the experiment,the average weight of female individuals was 3. 86 times of that of male individuals. 6Composition dependent intrinsic defect structures in ASnO3(A=Ca,Sr,Ba)显示文摘Perovskite stannates are promising semiconductors that are widely used in optoelectronic devices.Here,the composition dependent intrinsic point defects of stannate perovskites ASnO3(A=Ca,Sr,Ba)are studied by first-principles calculations.The preferences of defects under stoichiometric and nonstoichiometric conditions are unsealed,meanwhile the charge states of each intrinsic defect varying with the change of electron Fermi energy are clarified.For stoichiometric BaSnO3 and SrSnO3,the Schottky defect complexes are predicted as the most stable defect structure,while the antisite defect complexes are the most stable one in CaSnO3.In nonstoichiometric ASnO3,excessive AO is beneficial to the formation of oxygen vacancies and A-Sn antisite defects in all ASnO3;while the Ca interstitial is another major defect existing in CaSnO3.In the case of SnO2 excess,the predominant defects are the Sn-A antisite defects and A vacancies.Since the functionality of these perovskite oxides is closely related to the types and concentrations of their point defects,the present results are expected to guide the future experiments to optimize the function of the perovskite oxides by tailoring the intrinsic defects through controlling the composition of AO and SnO2.Yuchen Liu Yu Zhou Dechang Jia Juanli Zhao Banghui Wang Yuanyuan Cui Qian Li Bin Liu 2020Journal of Materials Science & Technology2020,42,7:0
7Prediction of Apple Fruit Quality by Soil Nutrient Content and Artificial Neural Network显示文摘The effect of soil nutrient content on fruit yield and fruit quality is very important.To explore the effect of soil nutrients on apple quality we investigated 200 fruit samples from 40 orchards in Feng County,Jiangsu Province.Soil mineral elements and fruit quality were measured.The effect of soil nutrient content on fruit quality was analyzed by artificial neural network(ANN)model.The results showed that the prediction accuracy was highest(R2=0.851,0.847,0.885,0.678 and 0.746)in mass per fruit(MPF),hardness(HB),soluble solids concentrations(SSC),titratable acid concentration(TA)and solid-acid ratio(SSC/TA),respectively.The sensitivity analysis of the prediction model showed that soil available P,K,Ca and Mg contents had the greatest impact on the quality of apple fruit.Response surface method(RSM)was performed to determine the optimum range of the available P,K,Ca,and Mg contents in orchards In Feng County,which were 10∼20 mg⋅kg^(−1),170∼200 mg⋅kg^(−1),1000∼1500 mg⋅kg^(−1),and 80∼200 mg⋅kg^(−1),respectively.The research also concluded that improving the content of available P and available Ca in orchard soil was crucial to improve apple fruit quality in Feng County,Jiangsu Province.Mengyao Yan Xianqi Zeng Banghui Zhang Hui Zhang Di Tan Binghua Cai Shenchun Qu Sanhong Wang 2023Phyton-International Journal of Experimental Botany2023,92,1:0
8Error Inhomogeneity in the Computation of Spherical Mean Displacement显示文摘The traditional method for computing the mean displacement in latitude–longitude coordinates is a spherical meridional–zonal resultant displacement method(MRDM), which regards the displacement as the resultant vector of the meridional and zonal displacement components. However, there are inhomogeneity and singularity in the computation error of the MRDM, especially at high latitudes. Using the NCEP/NCAR long-term monthly mean wind and idealized wind fields, the inhomogeneity in the MRDM was accessed by using a great circle displacement computing method(GCDM) for non-iterative cases. The MRDM and GCDM were also compared for iteration cases by taking the trajectories from a three-time level reference method as the real trajectories. In the horizontal direction, the GCDM assumes that an air particle moves along its locating great circle and that the magnitude of the displacement equals the arc length of the great circle. The inhomogeneity of the MRDM is evaluated in terms of the horizontal distance error from the products of wind speed, lapse time, and angle difference from the GCDM displacement orient.The non-iterative results show that the mean horizontal displacement computed through the MRDM has both computational and analytical errors. The displacement error of the MRDM depends on the wind speed, wind direction, and the departure latitude of the air particle. It increases with the wind speed and the departure latitude. The displacement magnitude error has a four-wave pattern and the displacement direction error has a two-wave feature in the definition range of the wind direction. The iterative result shows that the displacement magnitude error and angle error of the MRDM and GCDM with respect to the reference method increase with the lapse time and have similar distribution patterns.
